RESUMO

Eukaryotic cells regulate various cellular processes through membrane-bound and membrane-less organelles, enabling active signal communication and material exchange. Lysosomes and lipid droplets are representative organelles, contributing to cell lipophagy when their interaction and metabolism are disrupted. Our limited understanding of the interacting behaviours and physicochemical properties of different organelles during lipophagy hinders accurate diagnosis and treatment of related diseases. In this contribution, we report a fluorescent probe, PTZ, engineered for dual-targeting of lipid droplets and lysosomes. PTZ can track liquid-liquid phase separation and respond to polarity shifts through ratiometric fluorescence emission, elucidating the lipophagy process from the perspective of organelle behavior and physicochemical properties. Leveraging on the multifunctionality of PTZ, we have successfully tracked the polarity and dynamic changes of lysosomes and lipid droplets during lipophagy. Furthermore, an unknown homogeneous transition of lipid droplets and lysosomes was discovered, which provided a new perspective for understanding lipophagy processes. And this work is expected to serve as a reference for diagnosis and treatment of lipophagy-related diseases.

RESUMO

Visualization of the mitochondrial state is crucial for tracking cell life processes and diagnosing disease, while fluorescent probes that can accurately assess mitochondrial status are currently scarce. Herein, a fluorescent probe named "SYN" was designed and prepared, which can target mitochondria via the mitochondrial membrane potential. Upon pathology or external stimulation, SYN can be released from the mitochondria and accumulate in the nucleolus to monitor the status of mitochondria. During this process, the brightness of the nucleolus can then serve as an indicator of mitochondrial damage. SYN has demonstrated excellent photostability in live cells as well as an extremely inert fluorescence response to bioactive molecules and the physiological pH environment of live cells. Spectroscopic titration and molecular docking studies have revealed that SYN can be lit up in nucleoli due to the high viscosity of the nucleus and the strong electrostatic interaction with the phosphate backbone of RNA. This probe is expected to be an exceptional tool based on its excellent imaging properties for tracking mitochondrial state in live cells.

RESUMO

The digital economy is an important engine for China's economic and social development, accelerating the development of agricultural digitization and promoting the integration of agricultural digitization. Low-carbon production is an inevitable trend in China's current economic development. The article takes 30 provinces (cities and districts) in China as research objects, constructing and measuring indicators of digitization level and carbon emission intensity of agricultural production from 2006 to 2018. It classifies agricultural production into planting and animal husbandry and uses a basic regression model to study their dynamic relationships. A mediating effect model is used to explore the specific mechanism path of the digital economy affecting carbon emission intensity, and a regional heterogeneity analysis is conducted. The study found that: (1) The level of digitalization can significantly reduce the carbon intensity of agricultural production; (2) Digitalization can reduce China's carbon intensity by promoting the level of agricultural technological inputs, the level of human capital and the urbanization rate. (3) There are regional and sectoral differences in the impact of digitization on the carbon intensity of agricultural production. The impact on the plantation sector is greater than that on the livestock sector, and the carbon reduction effect is slightly greater in the central and western regions than in the eastern regions.

RESUMO

The social integration in host cities of China's migrant population and its determinants has received much attention from researchers. However, few have directly addressed the family migration in differentiating migrants' social integration. This study employs descriptive statistics and multivariate regression to explore the social integration across different family migration types, based on the data of China Migrants Dynamic Survey 2013 and 2017 in Wuhan metropolitan area. The findings show that the family migration in Wuhan metropolitan area is prevalent, and the central city Wuhan has advantages both in the proportion of whole-family migration and the scores of migrants' social integration. In addition, the migrants' family migration is significantly positively associated with their social integration, but the effect is variance in the regression models of social integration's four dimensions. The findings reveal the Wuhan City's leading position in promoting migrants' social integration within the Wuhan metropolitan area. Furthermore, migration with more family members has higher levels of integration in economy and public service. This implies that the convenience provided by inflow cities' government for family migration is crucial. To provide high-quality public services is of great significance to attract and retain migrants and then promote their overall social integration.

RESUMO

Exploration of urban spatial connections and network structures of urban agglomeration in the Yangtze River Delta, as well as its influencing factors, is of great significance regarding optimization of the development pattern of the Yangtze River Delta urban agglomeration and promotion of regional high-quality development. Therefore, based on Baidu index data in 2015 and 2019, this paper first analyzes the spatiotemporal variation characteristics of information-flow connections in the Yangtze River Delta urban agglomeration. Then it uses social network analysis to explore the information-flow network structure in the Yangtze River Delta urban agglomeration, and finally explores the influencing factors of information-flow intensity in the Yangtze River Delta urban agglomeration. The main conclusions are as follows: (1) The total amount of information flow in the Yangtze River Delta urban agglomeration has had no obvious change, and the coverage of information flow in the central urban circle has expanded. (2) The network hierarchy presents a relatively stable "pyramid" distribution pattern, which tends to develop into a "spindle" pattern. (3) The overall network density of the Yangtze River Delta urban agglomeration is high and is increasing. The backbone network is a "triangle" structure. The central cities in the region are stable, and the subgroups are adjacent to each other geographically. (4) Gross Domestic Product, resident population of the region and the number of Internet broadband subscribers all have important effects on the total information flow, among which the number of Internet broadband subscribers has the greatest effect on the total information flow. In addition, urban functions and their positioning, urban events, history and culture, and other factors that are difficult to quantify also have a certain impact on the information-flow network among cities.

RESUMO

The expensive production of bioethanol is because it has not yet reached the 'THREE-HIGH' (High-titer, high-conversion and high-productivity) technical levels of starchy ethanol production. To cope with it, it is necessary to implement a high-gravity mash bioethanol production (HMBP), in which sugar hydrolysates are thick and fermentation-inhibitive compounds are negligible. In this work, HMBP from an atmospheric glycerol autocatalytic organosolv pretreated wheat straw was carried out with different fermentation strategies. Under an optimized condition (15% substrate concentration, 10 g/L (NH4)2SO4, 30 FPU/g dry matter, 10% (V/V) inoculum ratio), HMBP was at 31.2 g/L with a shaking simultaneous saccharification and fermentation (SSF) at 37 degrees C for 72 h, and achieved with a conversion of 73% and a productivity of 0.43 g/(L x h). Further by a semi-SFF with pre-hydrolysis time of 24 h, HMBP reached 33.7 g/L, the conversion and productivity of which was 79% and 0.47 g/(L x h), respectively. During the SSF and semi-SSF, more than 90% of the cellulose in both substrates were hydrolyzed into fermentable sugars. Finally, a fed-batch semi-SFF was developed with an initial substrate concentration of 15%, in which dried substrate (= the weight of the initial substrate) was divided into three portions and added into the conical flask once each 8 h during the first 24 h. HMBP achieved at 51.2 g/L for 72 h with a high productivity of 0.71 g/(L x h) while a low cellulose conversion of 62%. Interestingly, the fermentation inhibitive compound was mainly acetic acid, less than 3.0 g/L, and there were no other inhibitors detected, commonly furfural and hydroxymethyl furfural existing in the slurry. The data indicate that the lignocellulosic substrate subjected to the atmospheric glycerol autocatalytic organosolv pretreatment is very applicable for HMBP. The fed-batch semi-SFF is effective and desirable to realize an HMBP.
