RESUMO

BACKGROUND:Salvianolic acid B can inhibit cell damage induced by H2O2,effectively remove excess reactive oxygen species, and exert antioxidant properties. It has been used in the treatment of many diseases. However, there are relatively few studies on the role and mechanism of salvianolic acid B in intervertebral disc degeneration. OBJECTIVE:To observe the effect and mechanism of salvianolic acid B on oxidative stress-induced intervertebral disc degeneration by using gelatin methacryloyl hydrogel as a carrier through the in vitro cell experiment and the in vivo animal experiment. METHODS:The gelatin methacryloyl hydrogel(drug-loaded hydrogel)loaded with salvianolic acid B was prepared.(1)In vitro cell experiment:The lumbar nucleus pulposus cells of adult SD rats were isolated and extracted, and passage 3 nucleus pulposus cells were selected and divided into groups:Group A was added complete medium. In group B,a complete medium containing H2O2 was added.Group C was inoculated on methylacrylylated gelatin hydrogel and added with a complete medium containing H2O2.Group D was inoculated on methyl acrylyl gelatin hydrogel loaded with salvianolic acid B and added into a complete medium containing H2O2.The E group was inoculated on the methylacrylyl gelatin hydrogel loaded with salvianolic acid B,and the complete medium containing H2O2 and the complete medium containing TLR4 signaling pathway inhibitor were added.Cell proliferation, oxidative stress, inflammatory response, gene expression of cell matrix-associated proteins and the protein expression of TLR4/nuclear factor-kB signaling pathway were detected.(2)Animal in vivo experiment:Sixty adult SD rats were randomly divided into normal group, acupuncture group, acupuncture + salvianolic acid group, acupuncture + hydrogel group and acupuncture + loading potion gel group, with 12 rats in each group. The last four groups were treated with acupuncture to establish models of intervertebral disc degeneration and then injected with normal saline, salvianolic acid B solution,non-drug loaded gel and drug-loaded gel in turn.Imaging examination and pathological observation were performed 4 weeks after surgery. RESULTS AND CONCLUSION:(1)In vitro cell experiment:Compared with group A,the cell proliferation was decreased;the oxidative stress reaction and inflammation reaction were enhanced;the expression of extracellular matrix degrading enzymes(matrix metalloproteinase 3,matrix metalloproteinase 13,ADAMTS4,ADAMTS5)was increased in group B(P<0.05),and the synthesis of extracellular matrix(type Ⅱ collagen, proteoglycan)was decreased(P<0.05).The protein expression of the TLR4/nuclear factor-kB signaling pathway was increased(P<0.05).Compared with group B,the cell proliferation of groups D and E was increased, the oxidative stress response and inflammatory response were weakened, and the expression of extracellular matrix degrading enzymes(matrix metalloproteinase 3,matrix metalloproteinase 13,ADAMTS4,ADAMTS5)was decreased(P<0.05),and the synthesis of extracellular matrix was increased(P<0.05).The protein expression of TLR4/nuclear factor-kB signaling pathway was decreased(P<0.05),and the effect was more significant in group E.(2)Animal in vivo experiment:4 weeks after surgery, intervertebral disc height index, index of MRI and pathological and histological grading of the intervertebral disc had improved significantly in the acupuncture+drug-loaded hydrogel group, and simply injecting hydrogel or salvianolic acid B solution can to a certain extent improve the intervertebral disc degeneration, but they are not as good as the injection of the drug-loaded hydrogel.(3)It is concluded that gelatin methacryloyl hydrogel loaded with salvianolic acid B can inhibit oxidative stress and inflammation in the degenerated intervertebral disc tissue, inhibit the degradation of extracellular matrix, and alleviate the process of intervertebral disc degeneration, which may be accomplished by inhibiting the TLR4/nuclear factor-kB signaling pathway.

RESUMO

The objective of this study is to investigate the effect of chemokine CXCR4 on the apoptosis of gastric cancer cells through IL-6/STAT3 signaling pathway and to explore the related mechanisms.Fluorescence quantitative PCR and Western blot were performed to detect the mRNA and protein expression levels of the chemokine CXCR4 in human gastric cancer tissues and adjacent non-cancerous tissues(PCT).Next,CXCR4 knockdown and overexpression were achieved by transfecting SGC7901 gastric cancer cell line with lentiviral vectors.TUNEL staining was used to evaluate the apoptosis of SGC7901 cells, while MTT assay was employed to measure cell proliferation.Western blot was conducted to determine the expression of apoptosis-related proteins Bax and Bcl2.Further,enzyme-linked immunosorbent assay(ELISA)was employed to measure the secretion levels of inflammatory cytokines.Real-time fluorescent quantitative PCR(RT-PCR)was utilized to quantify the expression of IL-6 mRNA in the IL-6/STAT3 signaling pathway, and Western blot was performed to analyze the expression of STAT3-Ser727 protein. In addition, after knocking down CXCR4 in SGC7901 cells,IL-6/STAT3 signaling pathway agonist lipopolysaccharide(LPS)was transfected, while in CXCR4-overexpressing SGC7901 cells,IL-6/STAT3 signaling pathway inhibitors angoline or bruceantinol were transfected. Then TUNEL staining was used to assess cell apoptosis, and Western blotting was performed to examine the expression levels of apoptosis-related proteins Bax and Bcl2 in these cells.Data showed that the expression of immune chemokine CXCR4 was increased in gastric cancer tissues,as compared with adjacent non-cancerous tissues.Single-cell gel electrophoresis analysis indicated that knockdown or overexpression of CXCR4 do not induce DNA damage in SGC7901 cells.TUNEL staining,MTT cell proliferation assay and Western blotting demonstrated that knockdown of CXCR4 in SGC7901 cells promoted the apoptosis in SGC7901 cells, while overexpression of CXCR4 inhibited the apoptosis.ELISA showed that knockdown of CXCR4 in SGC7901 cells promotes the expression of pro-inflammatory factors IL-1β and TNF-α,while inhibited the expression of anti-inflammatory factors IL-10 and TGF-β.Conversely,overexpression of CXCR4 demonstrated opposite effects. Finally, the activation of the IL-6/STAT3 signaling pathway significantly reduced the apoptosis induced by knocking down CXCR4 in iSGC7901 cells, whereas the inhibition of IL-6/STAT3 signaling pathway can significantly suppressed the induction of SGC7901 cells proliferation induced by CXCR4 overexpress. In conclusion, immunochemokine CXCR4 regulates gastric cancer cell apoptosis and inflammatory cytokines secretion through IL-6/STAT3 signaling pathway.

RESUMO

Objective:To investigate the status quo of subclinical thyroid diseases in the faculty and staff of a university and to explore their affecting factors. Methods:A total of 4 219 faculty and staff members who met the exclusion criteria and underwent the health examination in the Community Health Service Center of Beijing Jiaotong University in 2021 were enrolled in the study. General clinical data and laboratory findings of the enrolled subjects were collected. According to the upper and low reference range of thyroid stimulating hormone (TSH) in our laboratory (0.35-5.5 μIU/ml), subjects were classified into subclinical hyperthyroidism group, subclinical hypothyroidism group and normal thyroidism group. The association of gender, age and body mass index (BMI), as well as the metabolic indices with the prevalence of subclinical thyroid disease was analyzed. Results:The prevalence rates of subclinical hypothyroidism and subclinical hyperthyroidism were 4.10% (173/4 219) and 0.69% (29/4 219), respectively. The prevalence of subclinical thyroid diseases in females was higher than that in males(5.90% (77/2 101) vs. 3.66%(125/2 018),χ 2=11.58, P<0.05); there was a significant difference in prevalence among different age groups(χ 2=39.49, P<0.05)and the prevalence increased with the age. There were significant differences in levels of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C), diastolic blood pressure(DBP), fasting blood glucose (FBG), free triiodothyronine (FT 3), free thyroxine (FT 4), thyroglobulin antibody (TGAb) and thyroid peroxidase antibody (TPOAb) among three groups ( P<0.05). TSH levels were positively correlated with the age ( r=0.58, P<0.001), and levels of TG ( r=0.66, P<0.001), TC ( r=0.67, P<0.001), LDL-C ( r=0.62, P<0.001), TPOAb ( r=0.78, P<0.001), TGAb ( r=0.77, P<0.001); was negatively correlated with FBG ( r=-0.50, P<0.001). Conclusion:The prevalence of subclinical thyroid diseases among faculty and staff of the studied university is relatively high, and it is related to gender, age, thyroid antibodies, blood glucose and lipid levels.

RESUMO

Fish trophic niches reflect important ecological interactions and provide insight into the structure of mangrove food webs. Few studies have been conducted in mangrove fish predators to investigate interpopulation trophic niches and ontogenetic shifts. Using stable isotope analysis and two complementary approaches, the authors investigated trophic niche patterns within and between two ontogenetic groups (juveniles and sub-adults) of a generalist predator (Acentrogobius viridipunctatus) in four mangroves with heterogeneous environmental conditions (e.g., tidal regimes, salinity fluctuations and mangrove tree community). The authors hypothesized that the trophic niche between populations would vary regionally and trophic position would increase consistently from juvenile to sub-adult stages. The results revealed that both δ13 C and δ15 N values varied greatly across populations and between ontogenetic groups, and complex spatio-ontogenetic variations were expressed by Layman's metrics. They also found some niche separation in space, which is most likely related to resource availability in spatially diverse ecosystems. In addition, trophic niche position increased consistently from juveniles to sub-adults, indicating ontogenetic feeding shifts. The isotopic plasticity index and Fulton's condition index also showed significant spatial-ontogenetic variation, which is consistent with optimal foraging theory. The findings highlight that trophic plasticity has a high adaptive value for mangrove fish predators in dynamic ecosystems.

RESUMO

Objective:To compare the efficacy and safety of minimally invasive puncture and drainage for hypertensive cerebral hemorrhage at different time windows. Methods:A total of 98 patients with spontaneous cerebral hemorrhage underwent minimally invasive puncture and drainage in our hospital from January 2016 to December 2019 were chosen. These patients were divided into 3 groups in accordance with operation time: an ultra-early-stage group (accepted surgery within3 h of onset), an early-stage group (accepted surgery within 3-24 h of onset), an acute-stage group (accepted surgery within 24-72 h of onset); the clinical data of these patients were retrospectively analyzed. The hematoma clearance, muscle strength improvement 14 d after surgery, consciousness improvement 14 d after surgery, National Institutes of Health Neurological Deficit Scale (NIHSS) scores 14 d after surgery, and re-hemorrhage were compared among the 3 groups. Results:In the ultra-early-stage group, there were 11 patients (84.6%), 2 patients (15.4%) and 0 patients (0%) with low, medium and high hematoma clearance rates. In the early-stage group, there were 20 patients (33.9%), 32 patients (54.2%) and 7 patients (11.9%) with low, medium and high hematoma clearance rates. In the acute-stage group, 8 patients (30.8%), 14 patients (53.8%) and 4 patients (15.4%) had low, medium and high hematoma clearance rates. The hematoma clearance rate in the early group and the acute group was higher than that in the ultra-early group. Fourteen d after surgery, the proportion of patients with muscle strength improvement in the early-stage group and acute-stage group was significantly higher than that in the ultra-early-stage group ( P<0.05); the proportion of patients with consciousness improvement in the early-stage group was significantly higher than that in the ultra-early-stage group and acute-stage group ( P<0.05); NIHSS scores of patients in the early-stage group were significantly lower than those in the ultra-early stage group and acute-stage group ( P<0.05). There were 4 patients with postoperative re-hemorrhage, including one from the ultra-early-stage group and 3 from the early group. Conclusion:In patients with cerebral hemorrhage, hematoma clearance is relatively good and postoperative recovery is good when minimally invasive puncture and drainage is performed within 3-24 h of onset.

RESUMO

Go is not only regarded as the most complex board game, but also viewed as a huge challenge for artificial intelligence (AI). A computer Go program, AlphaGo, using artificial neural networks and tree search defeated one of the best professional human players in March 2016, achieving an unprecedented performance in the game of Go. This victory represented a major milestone in AI research. AI would profoundly affect all fields in the days to come. On the other hand, traditional Chinese medicine (TCM) is thought to be of similar cultural background, traditional concept and way of thinking to Go. Consequently AI techniques may be employed in TCM research. This article presented a brief overview of recent progress in AI, and explored the feasibility of applying the techniques concerning AlphaGo to the diagnosis and treatment of TCM.

RESUMO

7 cm in diameter)of the breast were treated in our hospital by lumpectomy via a periareolar incision. The length of the incision was less than half of the perimeter of the areola. For multiple fibroadenomas, circumareolar incision(not longer than half of the nipple-areolar perimeter)was made in different quadrants according to the position of the main tumor,so that the multiple benign tumors could be integrally removed through the approach between the hypodermic fat and glands via radial incisions.Similarly, giant benign tumors were treated through the same approach, cut into pieces, and then removed. Results Totally 165 tumors were removed from the 46 patients with multiple fibroadenomas. In the resected masses,19 were from one patient, who had bilateral multiple breast tumors. The 2 patients with giant breast fibroadenoma were also cured by lumpectomy, the diameter of the largest tumor in the two was 8 cm,while the circumareolar incisions were shorter than 3.5 cm.No nipple necrosis occurred after the operation in our series.Follow-up was achieved in all of the patients for 1 month to 2 years(mean,14.4 months),during which,11 cases showed no incision scar,26 had non-notable scars,9 cases developed observable incision scars, and 2 cases had apparent scars owing to their susceptibility to scar. Conclusions Lumpectomy via minor circumareolar incision is convenient and effective for multiple or giant benign fibroadenoma of the breast with good cosmetic outcomes.
