RESUMO

In many jurisdictions foodservice workers are required to obtain food handler certification via written examination before being able to work. This study investigated the effect of the readability, or the ease in which one can read and comprehend written text, of food handler exam questions on exam performance. It was hypothesized that the reduction in cognitive load by improving the readability of exam questions would lead to improved scores. Participants received training in personal hygiene and basic food safety and were tested on their knowledge using questions that were worded using the traditional phrasing and updated phrasing that has improved readability. The results indicate that improved readability had a significant difference in the personal hygiene section but not on the basic food safety section. These results are due, in part, to the types of cognitive load (intrinsic vs. extraneous) that are required to solve different types of problems.

RESUMO

Visible light communications (VLC) (including LiFi) represent a subset of the broader field of optical wireless communications. Where narrow beams, typical of free space optical communications are largely free from interference. VLC encompasses use cases involving combined illumination and data access and supporting a wireless access point (AP) model. The use of many units provides scaling of spatial coverage for both lighting and data access. However, AP replication in close proximity creates many interference challenges that motivate the investigation embodied in this paper. In particular, we frame the interference challenge in the context of existing strategies for driving improvements in link performance and consider the impacts of multiple users, multiple sources and multiple cells. Lastly, we review the state of existing research in this area and recommend areas for further study. This article is part of the theme issue 'Optical wireless communication'.

Cell populations that are exclusively composed by descendants from a defined single parental progenitor are referred to as "monoclonal", "clonal" or more correctly as "clonally derived cell population". Clonal derivation of cell lines used in the manufacturing of recombinant biologics is a regulatory requirement that aims to ensure a robust process and consistent quality throughout the life cycle of a product. Clonal derivation of cell lines is usually ensured by the process (e.g. two subsequent rounds of limiting dilution). Here we present an approach to analytically assess the probability of clonal derivation of existing cell populations. Using target locus amplification (TLA) followed by next generation sequencing (NGS), unique genetic features can be identified, for example the integration site of the plasmid in the host cell genome or plasmid-plasmid junctions of the plasmids used for expression of recombinant biologics. Whereas a direct assessment of clonal derivation using TLA/NGS data is challenging due to limitations in specificity, confirmed clonally derived populations generated from the cell line population can be analyzed by qPCR for the presence of the unique genetic features identified by TLA/NGS. In the present study, a statistical analysis allowed the demonstration that two independently generated CHO cell lines were clonally derived with an upper 95% confidence interval limit of a potentially present contaminating population of 1.3%.

Recently with the advancement of solid state lighting and the application thereof to Visible Light Communications (VLC), the concept of Visible Light Positioning (VLP) has been targeted as a very attractive indoor positioning system (IPS) due to its ubiquity, directionality, spatial reuse, and relatively high modulation bandwidth. IPSs, in general, have four major components: (1) a modulation, (2) a multiple access scheme, (3) a channel measurement, and (4) a positioning algorithm. A number of VLP approaches have been proposed in the literature and primarily focus on a fixed combination of these elements and moreover evaluate the quality of the contribution often by accuracy or precision alone. In this article, we provide a novel two-phase indoor positioning algorithmic framework that is able to increase robustness when subject to insufficient anchor luminaries and also incorporate any combination of the four major IPS components.

RESUMO

MEMS mirrors are currently used in many applications to steer beams of light. An area of continued research is developing mirrors with varifocal capability that allows the beam to be shaped and focused. In this work, we study the varifocal capability of a 380 µm diameter, thermally actuated MEMS mirror with a ± 40° tip-tilt angle and a radius of curvature between -0.48 mm to 20.5 mm. Light is coupled to the mirror via a single mode optical fiber, similar to an indoor optical wireless communication architecture. The performance of the mirror is characterized with respect to (1) the profile of the reflected beam as the mirror deforms and (2) the mirror's impact when integrated into an optical communication system. We found that the mirror can focus light to a beam with a 0.18° half-angle divergence. Additionally, the ability to change the shape of fiberized light from a wide to narrow beam provides an unmatched level of dynamic control and significantly improves the bit error rate in an optical communication system.

A statistical modeling tool is presented that enables real-time viewing of how changes in method, process, and stability variability/bias impact product acceptance rate. The tool can be used to set and justify specifications. As needed, additional sources of variability/bias can be added to further optimize the tool's prediction power. The tool can be used to assess each manufacturing run to ensure the process is in control. Aberrant results can then be investigated to see what source of variability/bias may have changed. To enable continuous improvement, the impact of new processes, methods, or technologies can also be addressed and such changes justified.

RESUMO

MEMS micromirrors have proven to be very important optical devices with applications ranging from steerable mirrors for switches and cross-connects to spatial light modulators for correcting optical distortions. Usually beam steering and focusing are done with different MEMS devices and tilt angles in excess of 10 degrees are seldom obtained. Here we describe a single MEMS device that combines tip/tilt, piston mode and varifocal capability into a single, low cost device with very large tilt angles. Our device consists of a 400 micron diameter mirror driven with thermal bimorphs. We have demonstrated deflection angles of ± 40 degrees along both axes, a tunable focal length which varies between -0.48 mm to + 20.5 mm and a piston mode range of 300 microns - four separately controllable degrees of freedom in a single device. Potential applications range from smart lighting to optical switches and devices for telecom systems.

Visible light communications (VLC) technology permits the exploitation of light-emitting diode (LED) luminaries for simultaneous illumination and broadband wireless communication. Optical orthogonal frequency-division multiplexing (O-OFDM) is a promising modulation technique for VLC systems, in which the real-valued O-OFDM baseband signal is used to modulate the instantaneous power of the optical carrier to achieve gigabit data rates. However, a major design challenge that limits the commercialization of VLC is how to incorporate the industry-preferred pulse-width modulation (PWM) light dimming technique while maintaining a broadband and reliable communication link. In this work, a novel signal format, reverse polarity O-OFDM (RPO-OFDM), is proposed to combine the fast O-OFDM communication signal with the relatively slow PWM dimming signal, where both signals contribute to the effective LED brightness. The advantages of using RPO-OFDM include, (1) the data rate is not limited by the frequency of the PWM signal, (2) the LED dynamic range is fully utilized to minimize the nonlinear distortion of the O-OFDM communication signal, and (3) the bit-error performance is sustained over a large fraction of the luminaire dimming range. In addition, RPO-OFDM offers a practical approach to utilize off-the-shelf LED drivers. We show results of numerical simulations to study the trade-offs between the PWM duty cycle, average electrical O-OFDM signal power, radiated optical flux as well as human perceived light.

Changes in gait parameters have been shown to be an important indicator of several age-related cognitive and physical declines of older adults. In this paper we propose a method to monitor and analyze walking and cycling activities based on a triaxial accelerometer worn on one ankle. We use an algorithm that can (1) distinguish between static and dynamic functional activities, (2) detect walking and cycling events, (3) identify gait parameters, including step frequency, number of steps, number of walking periods, and total walking duration per day, and (4) evaluate cycling parameters, including cycling frequency, number of cycling periods, and total cycling duration. Our algorithm is evaluated against the triaxial accelerometer data obtained from a group of 297 middle-aged to older adults wearing an activity monitor on the right ankle for approximately one week while performing unconstrained daily activities in the home and community setting. The correlation coefficients between each of detected gait and cycling parameters on two weekdays are all statistically significant, ranging from 0.668 to 0.873. These results demonstrate good test-retest reliability of our method in monitoring walking and cycling activities and analyzing gait and cycling parameters. This algorithm is efficient and causal in time and thus implementable for real-time monitoring and feedback.

The development of functional activity monitors (FAMs) will allow rehabilitation researchers and clinicians to evaluate treatment efficacy, to monitor compliance to exercise instructions, and to provide real time feedback in the treatment of movement disorders during the performance of daily activities. The purpose of the present study was to develop and test a small sized wearable FAM system comprised of three sensors positioned on the sternum and both thighs, wireless Bluetooth transmission capability to a smartphone, and computationally efficient activity detection algorithms for the accurate detection of functional activities. Each sensor was composed of a tri-axial accelerometer and a tri-axial gyroscope. Computationally efficient activity recognition algorithms were developed, using a sliding window of 1 second, the variability of the tilt angle time series and power spectral analysis. In addition, it includes a decision tree that identifies postures such as sitting, standing and lying, walking at comfortable, slow and fast speeds, transitions between these functional activities (e.g, sit-to-stand and stand-to-sit), activity duration and step frequency. In a research lab setting the output of the FAM system, video recordings and a 3D motion analysis system were compared in 10 healthy young adults. The results show that the agreement between the FAM system and the video recordings ranged from 98.10% to 100% for all postures, transfers and walking periods. There were no significant differences in activity durations and step frequency between measurement instruments.

Recent evidence supports the use of certain soccer drills for combined technical and physical training. Therefore, it is important to be able to accurately monitor training intensity during soccer drills intended for physical development to allow the optimization of training parameters. Twenty-eight professional soccer players were assessed for heart rate (HR) and rating of perceived exertion (RPE) responses to 5 commonly used soccer training drills (2v2 to 8v8 drills). The responses of both HR and RPE differed significantly (p < 0.05) between the drills, generally showing an elevated response to drills involving lower player numbers. However, the 2v2 drill showed a significantly (p < 0.05) lower HR response (mean +/- SD: 88.7 +/- 1.2% HRmax) than 3v3 (91.2 +/- 1.3% HRmax) and 4v4 drills (90.2 +/- 1.6% HRmax). There was no significant correlation between the HR and RPE responses to the various drills (r = 0.60, p = 0.200). This poor relationship is probably because during the 2v2 drill, RPE was higher than during any of the other 6 drills, whereas HR was only fourth highest of the 6 drills. This demonstrates that HR and RPE are only poorly related during the intense drills used in this study, and that HR underestimates the intensity of the 2v2 drill. Heart rate demonstrated lower intersubject variability (1.3-2.2%) than RPE (5.1-9.9%). However, unlike HR, Borg 15-point RPE appears to be a valid marker of exercise intensity over a wide range of soccer training drills by maintaining validity in all drills and demonstrating acceptable intersubject variability. A combination of both HR- and RPE-based training load calculations appears optimal for use in soccer training.

The purpose of this study was to examine the physiological effects of different sprint repetition protocols on professional footballers. Of particular interest were the abilities of repeated sprint protocols to induce fatigue to an extent observed during competitive soccer. Six professional soccer players were assessed for fatigue rate and physiological responses of heart rate (HR), blood lactate (BLa), and rating of perceived exertion (RPE) during the performance of 4 repeated sprint drills, each totaling a sprint distance of 600 m. The 4 drills used 15- or 40-m sprints with 1:4 or 1:6 exercise: rest ratios. The 15-m sprint drill with 1:4 exercise:rest ratio induced the greatest fatigue (final sprint time 15% greater than initial sprint time) and greatest physiological responses. The 40-m sprint drill using a 1:4 exercise:rest ratio produced similar BLa and HR responses to the 15-m drill (13-14 mmol.L(-1) and 89% HRmax, respectively) but significantly lower RPE (mean +/- SD: 17.1 +/- 0.4 vs. 18.8 +/- 0.4, p < 0.05) and fatigue rates (11.1 vs. 15.0%, p < 0.01). Both sprint distance and exercise:rest ratio independently influenced fatigue rate, with the 15-m sprint distance and the 1:4 exercise:rest ratio inducing significantly (p < 0.01) greater fatigue than the 40-m sprint distance and the 1:6 exercise:rest ratio. The magnitude of fatigue during the 40- x 15-m sprint drill using a 1:6 exercise:rest ratio was 7.5%, which is close to the fatigue rate previously reported during actual soccer play. The present study is the first to examine both variations in sprint distances and rest ratios simultaneously, and the findings may aid the design of repeated sprint training for soccer.

Recent evidence suggests that certain soccer drills produce exercise intensities suitable for physical conditioning. However, it remains debatable whether soccer drills can provide a sufficiently unified exercise intensity among different players and on repetition of a drill, because movement patterns cannot be externally controlled during soccer drills. Good reliability and low variability of exercise intensity would enable all players to receive an appropriate training stimulus. The purpose of this study was to investigate intersubject variability and intrasubject reliability in exercise intensity during soccer drills. It was hypothesized that soccer drills that involve the highest exercise intensities would demonstrate the lowest intersubject variability and the highest intrasubject reliability. Heart rates of 23 professional soccer players were recorded during a range of soccer training drills. The drills consisted of 2 vs. 2 to 8 vs. 8 normal scoring games and 2 further possession games. Heart rate responses were examined for variability, reliability, and suitability for soccer endurance training. Coefficients of variation across players were less than 3% for all drills. Paired t-tests showed no significant differences in heart rate on repetition of the drills and 95% ratio limits of agreement were 1.8-3.8%. There were no significant correlations between exercise intensity and the statistical measures of variability and reliability. Several drills produced exercise intensities suitable for soccer endurance training with mean heart rate responses ranging from 87-91% HRmax. Soccer drills such as those used in the present study appear to be an adequate substitute for physical training without the ball and thus provide simultaneous skill and fitness training. The increase in training time spent developing technical ability and/or a reduction in total training time required may be useful for soccer teams.

The purpose of this study was to examine the effects of different modes of stretching within a pre-exercise warm-up on high-speed motor capacities important to soccer performance. Eighteen professional soccer players were tested for countermovement vertical jump, stationary 10-m sprint, flying 20-m sprint, and agility performance after different warm-ups consisting of static stretching, dynamic stretching, or no stretching. There was no significant difference among warm-ups for the vertical jump: mean +/- SD data were 40.4 +/- 4.9 cm (no stretch), 39.4 +/- 4.5 cm (static), and 40.2 +/- 4.5 cm (dynamic). The dynamic-stretch protocol produced significantly faster 10-m sprint times than did the no-stretch protocol: 1.83 +/- 0.08 seconds (no stretch), 1.85 +/- 0.08 seconds (static), and 1.87 +/- 0.09 seconds (dynamic). The dynamic- and static-stretch protocols produced significantly faster flying 20-m sprint times than did the no-stretch protocol: 2.41 +/- 0.13 seconds (no stretch), 2.37 +/- 0.12 seconds (static), and 2.37 +/- 0.13 seconds (dynamic). The dynamic-stretch protocol produced significantly faster agility performance than did both the no-stretch protocol and the static-stretch protocol: 5.20 +/- 0.16 seconds (no stretch), 5.22 +/- 0.18 seconds (static), and 5.14 +/- 0.17 seconds (dynamic). Static stretching does not appear to be detrimental to high-speed performance when included in a warm-up for professional soccer players. However, dynamic stretching during the warm-up was most effective as preparation for subsequent high-speed performance.

High-speed actions are known to impact soccer performance and can be categorized into actions requiring maximal speed, acceleration, or agility. Contradictory findings have been reported as to the extent of the relationship between the different speed components. This study comprised 106 professional soccer players who were assessed for 10-m sprint (acceleration), flying 20-m sprint (maximum speed), and zigzag agility performance. Although performances in the three tests were all significantly correlated (p < 0.0005), coefficients of determination (r(2)) between the tests were just 39, 12, and 21% for acceleration and maximum speed, acceleration and agility, and maximum speed and agility, respectively. Based on the low coefficients of determination, it was concluded that acceleration, maximum speed, and agility are specific qualities and relatively unrelated to one another. The findings suggest that specific testing and training procedures for each speed component should be utilized when working with elite players.

Because of its potential for antigenicity, theoretical concerns related to readministration of abciximab have been raised. We conducted the ReoPro Readministration Registry to assess the efficacy and safety of abciximab readministration. A total of 1,342 patients who underwent percutaneous coronary intervention and who received abciximab for at least a second time were recruited. Safety end points were hypersensitivity reactions, major bleeding, and thrombocytopenia (TCP). Human antichimeric antibody (HACA) titers were determined before and after readministration. Procedural success was 98% and was not influenced by the number of courses of abciximab or the presence of HACA. There were no cases of anaphylaxis. There were 5 minor allergic reactions, none of which required termination of the infusion. Clinically significant bleeding occurred in 31 patients (2.3%), including 1 (0.07%) with intracranial hemorrhage. TCP (<100 x 10(9)/L) developed in 5% of patients; profound TCP (<20 x 10(9)/L) occurred in 2%. In patients who received abciximab within 1 month of a previous treatment (n = 115), the risk of developing TCP and profound TCP was 16.5% and 12.2%, respectively. Having a positive HACA before readministration was not correlated with adverse clinical outcomes or bleeding, but was associated with TCP (14.1% vs 4.4%, p = 0.002) and profound TCP (5.6% vs 1.6%, p = 0.036). Readministration of abciximab can be accomplished without severe allergic responses and with a bleeding and efficacy profile similar to first-time administration. However, the rate of severe and profound TCP is increased relative to first-time administration, particularly when the time between treatments is <30 days or when HACA is present.
