The effect of lithium hydroxybutyrate on the development of the fetus and offsprings was studied on a model of alcohol intoxication of male rats. Under such conditions lithium hydroxybutyrate relieved completely the negative action of alcohol on the reproductive function, according to all parameters. The learning ability of the offsprings and their behavioral disorders in a stress situation caused by alcohol were normalized. Two-week administration of 100 mg/kg lithium hydroxybutyrate had no negative effect on the embryonal and postnatal development of the offsprings.

Phenazepam, 10 mg/kg/day, or phenobarbital, 30 mg/kg/day, administered to female rats during the entire pregnancy and 5 days after delivery during lactation induced behavioral changes in their offsprings at early (1.5-2 months) and later (4 months) ages. Phenobarbital produced a more marked effect on the central nervous system than phenazepam by causing impairments of extrapolatory reflexes and behavioral stereotype in a conflict situation or open field.

Examining the effects of phenazepam and phenobarbital on the course of pregnancy and the development of offsprings has revealed that phenobarbital that has an embryotoxic effect reduced the number of rat offsprings and diminished their weight, but failed to affect the behavioral responses of newborn rats. On the contrary, phenazepam had no embryotoxic effect, but on entering the newborn rats' organism with milk it inhibited some behavioral reactions in the early postnatal period.

The rat experiments studied the impact of inductors and inhibitors of enzymatic activity of xenobiotic metabolism on the pharmacological activity of sodium thiopental and ketamine and on the pharmacokinetics of sodium thiopental. Cocarboxylase, cobalt chloride and cimetidine enhanced the pharmacological action of sodium thiopental, but failed to exert any action on the effect of ketamine. The preparations of vitamin K, particularly vikasol and menadione sodium bisulfite, and phenobarbital attenuated the effects of these anesthetics, but tocopherol unchanged it. Phenobarbital and vikasol accelerated blood elimination of sodium thiopental, whereas cimetidine and cocarboxylase slowed down this process.

Experiments on rats with myocardial infarction induced by occlusion of the left coronary artery were made to explore the influence of calcium antagonists injected intraperitoneally for a long time (daily, for 21 days) on hemodynamics, contractile function and morphology of the heart muscle. It has been shown that verapamil (0.5 mg/kg), diltiazem (2 mg/kg) and nimodipine (20 micrograms/kg) do not exert under those conditions any noticeable effect on pump and contractile heart functions. Administration of verapamil in a doze of 2 mg/kg enhanced heart failure. The drugs under study normalized the response of contractile heart function to volumetric load, which is related to a considerable measure to the acceleration of reparative processes in the myocardium.

Administration of ethanol (5 g/kg/day, per os) to the pregnant rats evoked delayed impairments of the learning and memory in the offspring. Prenatal alcoholization of the animals attenuated the habituation of the exploration behavior in open field, impaired acquisition and retention of active avoidance in a shuttle box, increased slow activity of the EEG spectrum power, disturbed the function of the serotoninergic system in the brain cortex and of the dopaminergic system in the hippocamp. The new nootropic drug nooglutyl (N-5/hydroxynicotinoyl/-L-glutamic acid) administered in a dose of 25 mg/kg/day from the 8th to the 20th day of life prevented the above-mentioned delayed disturbances of higher integrative functions and biochemical processes in rat brain.

A study was made of the effect of zixorin, an inductor of liver cytochrome P-450, on the intensity of the immediate type allergic reaction--active food anaphylaxis in response to ovalbumin in guinea-pigs. Intraperitoneal injection of zixorin in a dose of 50 mg/kh for 3 days before administration of the antigen resolution dose was found to lower 2,3-fold the level of lethal outcomes due to the anaphylactic shock. It is suggested that zixorin may be used in multimodality treatment of food allergy.

In experiments on rats it was shown that alcohol administered intragastrically in a dose of 8 g/kg for 4 weeks produced long-term disturbances of CNS function in the offspring similar to those observed under clinical conditions. Early postnatal administration of lithium oxybutyrate (from the 8th through the 14th day of life) was found to prevent the development of the disturbances.

The preclinical study of safety of an antiarrhythmic drug bonnecor (GS-015) was carried out in the experiments on dogs. Administration of the drug in a dose of 5 mg/kg was found to exert no significant effect on functions of the vital organs. The dosage of 15 mg/kg can produce in dogs changes of the hematological and biochemical parameters and cause functional disorders of the cardiac muscle and death of the animals. The data obtained indicate that in dogs the drug possesses a little therapeutic range.

A preclinical study on safety of a new antialcoholic drug inmecarb was carried out on three species of experimental animals (rats, guinea pigs, dogs) receiving the drug at different doses including a subtoxic one (1/5 of LD50) for 6 months. The authors concluded that on the basis of a relatively low toxicity and the absence of specific toxicity and long-term side effects the drug may be recommended for clinical use as an antialcoholic agent.

During experiments on rats it was found that in alcoholized animals teturam on the whole did not potentiate and in some cases even attenuated the toxic effect of alcohol on the offspring development. The data confirm the idea about necessity of studying toxicity of drugs under the conditions corresponding to their clinical use.

Study of toxicological characteristic of teturam (200 mg/kg, administered into the stomach through a tube daily for 2 weeks) on the model of alcoholic intoxication in rats (ethanol, 8 g/kg, administered into the stomach through a tube daily for 1 month) confirmed the literature data on the drug's ability to enhance or maintain ethanol-induced pathological changes in the blood and internal organs.

It was found that lithium oxybutyrate suppresses voluntary consumption of 5% ethanol solution in rats with preliminarily formed alcohol motivation, slows down the formation of alcohol addiction under conditions of forced consumption of 5% ethanol solution and exerts a positive effect on general state of the animals subjected to a long-term action of ethanol by improving the conditioned reflex activity of the central nervous system and stimulating defensive-adaptational mechanisms of the organism.

The experiments on 10 dogs showed that replacement of sodium oxybutyrate for lithium oxybutyrate in the solution for preservation increases the period of preserving the structural and functional integrity of the donor kidney, contributes to its survival and delays the development of the transplant rejection.

The experiments on male rats showed that oral ethanol administered in a dose of 8 g/kg (2/3 of LD50) daily for a month produces toxic heart damage whose electrophysiological and morphological characteristics are similar to clinico-anatomical manifestations of human alcohol myocardiodystrophy. It was concluded that the model may be used for the search of new pharmacological agents for the treatment and prevention of alcohol myocardiodystrophy.
