RESUMO

Aggregation-induced emission enhancement (AIEE) phenomena received great popularity during the last decade but in most cases insights into the packing structure - fluorescence properties remained scarce. Here, an almost non-fluorescent merocyanine dye was equipped with large solubilizing substituents, which allowed the investigation of it's aggregation behaviour in unpolar solvents over a large concentration range (10-2 to 10-7 M). In depth analysis of the self-assembly process by concentration-dependent UV/Vis spectroscopy at different temperatures revealed a two-step anti-cooperative aggregation mechanism. In the first step a co-facially stacked dimer is formed driven by dipole-dipole interactions. In a second step these dimers self-assemble to give an oligomer stack consisting of about ten dyes. Concentration- and temperature-dependent UV/Vis spectroscopy provided insight into the thermodynamic parameters and allowed to identify conditions where either the monomer, the dimer or the decamer prevails. The centrosymmetric dimer structure could be proven by 2D NMR spectroscopy. For the larger decamer atomic force microscopy (AFM), diffusion ordered spectroscopy (DOSY) and vapour pressure osmometric (VPO) measurements consistently indicated that it is of small and defined size. Fluorescence, circular dichroism (CD) and circularly polarized luminescence (CPL) spectroscopy provided insights into the photofunctional properties of the dye aggregates. Starting from an essentially non-fluorescent monomer (Φ Fl = 0.23%) a strong AIEE effect with excimer-type fluorescence (large Stokes shift, increased fluorescence lifetime) is observed upon formation of the dimer (Φ Fl = 2.3%) and decamer (Φ Fl = 4.5%) stack. This increase in fluorescence is accompanied for both aggregates by an aggregation-induced CPL enhancement with a strong increase of the g lum from ∼0.001 for the dimer up to ∼0.011 for the higher aggregate. Analysis of the radiative and non-radiative decay rates corroborates the interpretation that the AIEE effect originates from a pronounced decrease of the non-radiative rate due to π-π-stacking induced rigidification that outmatches the effect of the reduced radiative rate that originates from the H-type exciton coupling in the co-facially stacked dyes.

RESUMO

A broadband, femtosecond transient circular dichroism (TRCD) spectrometer has been developed and tested in the wavelength range from 350 to 700 nm. The spectrometer uses a femtosecond probe white light with well-defined circular polarization. The latter is modulated by the polarization of a narrowband seed pulse. We have implemented a dual-beam probe geometry with phase-locked detection technique to increase the signal-to-noise ratio and to reduce optical artifacts. The spectrometer allows the acquisition of TRCD spectra with subpicosecond time resolution and typical noise levels of 10(-4) absorbance units. The performance of this instrument has been demonstrated on bis(merocyanine) nanorod aggregates in tetrahydrofurane/methylcyclohexane solution. The case study confirmed that this spectrometer is effective for the investigation of chiral properties in various molecular and nanostructural systems that have transient spectra in the UV-visible spectral range.

RESUMO

Examples of a novel class of tweezer molecules have been constructed through the tethering of two dipolar merocyanine chromophores by a naphthalenedimethylene or dimethylenediphenylmethane spacer. The electrostatic-interaction-directed self-assembly of these bis(merocyanine) tweezers affords centrosymmetric bimolecular complexes with very high dimerization constants of up to >10(9) M(-1), even in the good solvating solvent chloroform. This pronounced self-association of the bis(merocyanine) tweezers is attributed to the strongly dipolar nature of the merocyanine chromophores and a unique aggregate geometry of four pi-stacked chromophores with an alternating arrangement of their dipole moments. The structural assignment of the tetrachromophoric pi-stack has been accomplished by MALDI-TOF mass spectrometry and ROESY NMR spectroscopy. Furthermore, molecular modeling studies have accounted for the relationships between the dimerization constants and optical properties of the bimolecular complexes of the present bis(merocyanine) dyes and the structure of the spacer and the positions at which the merocyanine chromophores are attached.

RESUMO

This critical review provides an overview on the formation of pi-stacks of functional dyes in solution, aiming to acquaint young researchers with this topical research field and to stimulate further advance in supramolecular dye chemistry. Different mathematical models that have been proposed and applied for the description of aggregation equilibria of pi-systems in solution are discussed. The factors that have significant impact on the structural features of aggregates and the thermodynamics of pi-pi stacking such as electrostatic interactions, size and geometry of the dye molecules are covered in this review. A comparison of the binding strength is made for different classes of functional pi-conjugated systems, from simple benzene to more extended polycyclic hydrocarbon molecules, including triphenylenes and hexabenzocoronenes, heteroaromatic porphyrins and phthalocyanines, quadrupolar naphthalene and perylene bisimides, dipolar or even zwitterionic merocyanines and squaraines, and some macrocyclic dyes. Solvent effects on binding constants are analysed by linear free energy relationships with various solvent polarity scales (98 references with multiple entries).

RESUMO

We analyzed morphological and immunohistochemical features in 174 aggressive B-cell lymphomas of nodal and extranodal origin. Morphological features included presence or absence of a follicular component and cytologic criteria according to the Kiel classification, whereas immunohistochemical studies included expression of CD10, BCL-2, BCL-6, IRF4/MUM1, HLA-DR, p53, Ki-67 and the assessment of plasmacytoid differentiation. Patients were treated with a CHOP-like regimen. While the presence or absence of either CD10, BCL-6 and IRF4/MUM1 reactivity or plasmacytoid differentiation did not identify particular cytomorphologic or site-specific subtypes, we found that expression of CD10 and BCL-6, and a low reactivity for IRF4/MUM1 were favourable prognostic indicators. In contrast, BCL-2 expression and presence of a monotypic cytoplasmic immunoglobulin expression was associated with an unfavourable prognosis in univariate analyses. Meta-analysis of these data resulted in the development of a cumulative immunohistochemical outcome predictor score (CIOPS) enabling the recognition of four distinct prognostic groups. Multivariate analysis proved this score to be independent of the international prognostic index. Such a cumulative immunohistochemical scoring approach might provide a valuable alternative in the recognition of defined risk types of aggressive B-cell lymphomas.

RESUMO

A wave packet approach to the calculation and interpretation of circular dichroism (CD) spectra is applied to the spectroscopy of aggregates of a merocyanine dye. A combined analysis of absorption and CD spectra allows for the extraction of geometric information and excited state electronic coupling. It is shown that in the case of dimer aggregates of a chiral merocyanine dye, it is possible to infer the dynamics of an exciton transfer directly from the CD spectrum. This relation is established via the Fourier relation to a time-dependent correlation function reflecting the quantum dynamics in the dye aggregate.

RESUMO

CD5(+) diffuse large B-cell lymphomas (DLBLs) have recently been described as a particular subgroup of DLBLs. Classical banding and interphase cytogenetic analyses targeting ATM, TP53, and P16(INK4a) genes and the D13S25 locus from 13 CD5(+) DLBLs were compared with 55 CD5(-) DLBLs. Additionally, analysis of somatic mutations of the immunoglobulin heavy chain variable region (IgVH) genes were performed in CD5(+) DLBLs. CD5(+) DLBLs were somatically mutated (7 of 8 cases) and were negative for t(11;14)(q13;q32) and t(14;18)(q32;q21), whereas t(3;14)(q27;q32) was found in only one tumor. Trisomy 3 and gains on chromosomes 16/16p and 18/18q were significantly overrepresented in CD5(+) DLBLs. No ATM deletions were detected. The prevalence of deletions at the D13S25 locus was significantly higher in CD5(+) DLBLs (4 of 12 [33%]) compared with CD5(-) DLBLs (4 of 42 [10%]), as were p16(INK4a) deletions (33% versus 8%). On the basis of these findings, CD5(+) DLBLs are likely to arise from the same progenitor cell as the mutated variant of CD5(+) lymphocytic lymphoma/B-cell chronic lymphocytic leukemia (B-CLL).

RESUMO

Follicular lymphoma (FL) grades 1 and 2 are regarded as a distinct disease entity, whereas data suggest that FL grade 3 might be an inhomogeneous tumor category. To define the biologic spectrum of FL, 89 follicular lymphomas were studied for their cytologic composition, antigen expression, mitotic and proliferation indices, cytogenetics, and clinical data. In contrast to the homogeneous appearance of FL grades 1 and 2 (29 and 33 cases, respectively), 2 types of FL grade 3 were recognized. Eleven cases of FL 3a displayed structural features similar to those of FL 1 and 2 and were composed of centroblasts and centrocytes, whereas 16 cases of FL 3b, with (n = 4) or without (n = 12) a diffuse large B-cell lymphoma component (DLBL) (FL 3b +/- DLBL), consisted exclusively of blasts. In contrast to FL 3a, FL 3b +/- DLBL were CD10(+) in only 50% of cases and displayed plasmacytoid differentiation in 44% of cases. Although FL3a was t(14;18)+ in 8 of 11 (73%) cases, only 2 of 16 (13%) FL3b +/- DLBLs harbored this translocation. In contrast, chromosomal breaks at 3q27 were encountered in 7 of 16 (44%) FL 3b +/- DLBL in contrast to only 2 of 11 (18%) FL 3a, and the spectrum of secondary aberrations in FL 3b +/- DLBL was similar to that of diffuse large B-cell lymphoma. We conclude, therefore, that FL grade 3 is a heterogeneous disease group and that the distinction proposed in the new World Health Organization classification between FL 3a (with centrocytes) and FL3b (without centrocytes) is of biologic, and possibly clinical, importance.
