RESUMO

A hypothesis was tested that eggshell temperature manipulations during incubation and in ovo injection of thyroxine (T4) would help their progeny chicks to better survive the ascites-inducing condition during the growing period. In experiment 1, a total of 4,800 hatching eggs was randomly arranged in a 2 × 4 factorial design (8 replicates of 75 eggs per treatment), in which the eggs were incubated at a constant eggshell temperature (EST) of 37.8°C throughout the incubation period (CON) or were exposed to 15°C for one h on d 11, 13, 15, and 17 of incubation (EST manipulations; ESTM), and 4 treatment groups of 3 control groups (no injection; INJN, needle pricked; INJP, and sterilized distilled water injection; INJW) and one T4 treatment group (injected with sterilized distilled water containing 65 ng of T4; INJT4). In experiment 2, 240 one-day-old male broiler chicks from 2 temperature conditions and injection (INJN and INJT4) treatment groups were reared for 42 d in a completely randomized design with a 2 × 2 factorial arrangement. To induce ascites, all chicks were exposed to a 15°C room temperature from 14 d onwards. Results from experiment 1 showed that second-grade chicks and yolk sac weight were decreased, and body weight at hatch was increased in the ESTM and INJT4 groups. Also, final body weight was increased in the ESTM group. Ascites mortality rate was decreased in the ESTM and INJT4 groups. In the ESTM and INJT4 groups, the red blood cell (RBC) and the packed cell volume (PCV) count were decreased. In conclusion, the results showed that the ESTM and INJT4 treatments during incubation were associated with improved chick quality, productive performance of broilers, and a decreased incidence of cold-induced ascites in broiler chickens.

An experiment was conducted to evaluate the effects of a periodically low eggshell temperature exposure during incubation and dietary supplementation of arginine on performance, ascites incidence, and cold tolerance acquisition in broilers. A total of 2,400 hatching eggs were randomly assigned to 2 treatment groups (16 replicates of 75 eggs per treatment). The eggs were incubated at a constant eggshell temperature (EST) of 37.8ºC throughout the incubation period (CON) or were periodically exposed to 15°C for one hour on days 11, 13, 15, and 17 of incubation and the EST was measured (periodical low EST; PLE). After hatching, 240 one-day-old male broiler chicks from both treatment groups were reared for 42 d with or without dietary arginine supplementation in a completely randomized design with a 2 × 2 factorial arrangement. In order to induce ascites, all chicks were exposed to a 15°C room temperature from 14 d onwards. Results showed that second grade chicks and yolk sac weight were decreased, and final body weight was increased in the PLE group. Ascites mortality rate was decreased only in the PLE group and dietary arginine supplementation had no apparent effect. In the PLE group, the packed cell volume (PCV) percentage and red blood cell (RBC) count were decreased. In conclusion, the results showed that the PLE treatment during incubation was associated with improved hatchability, chick quality, and productive performance of broilers and decreased ascites incidence during post hatch cold exposure. Dietary arginine supplementation had no beneficial effects in cold exposed broilers.

Two experiments were conducted in which effects of floor eggs, washed floor eggs, and clean nest eggs were investigated on incubation characteristics and performance in later life of broiler chickens. In both experiments, a young and an older breeder flock were used in a 3×2 factorial design during incubation. In the second experiment, male and female chickens were reared separately until d 35 of age in floor pens. During this grow out trial, an extra group was created in which chickens obtained from clean nest eggs were mixed with chickens obtained from floor eggs, meaning that grow out period was set up as a 4×2×2 factorial design with 4 egg types, 2 breeder ages, and 2 sexes. In both experiments, fertility and hatchability of fertile eggs were lower in floor and washed eggs than in clean nest eggs (hatchability: experiment 1: 74.4 vs. 70.6 vs. 92.6% for floor eggs, washed floor eggs and clean nest eggs, respectively, P<0.001; experiment 2: 78.3 vs. 81.7 vs. 90.2%, respectively, P<0.001). In experiment 2, BW at d 0 of chickens obtained from clean nest eggs was higher than that of chickens from floor eggs and washed floor eggs (41.5 vs. 40.4 and 40.3 g, respectively; P<0.001). This difference disappeared during the grow out period and was absent at slaughter age at d 35 of age. Feed intake (FI), feed conversion ratio (FCR), and mortality during the grow out period were not affected by egg type. Incidence and severity of hock burns and footpad dermatitis were not affected by egg type or breeder age. Litter friability at d 35 of age tended to be lower in pens with chickens obtained from washed floor eggs compared to clean nest eggs. We conclude that incubation of floor eggs or washed floor eggs resulted in lower fertility and hatchability compared to clean nest eggs, but that performance during the grow out period was not affected.

In some European countries, footpad dermatitis (FPD) is measured as an indicator of broiler welfare. Prevalence and seasonal variation of FPD was determined within broiler flocks (fast-growing breeds) in the Netherlands. Samples were taken from 386 Dutch flocks at 8 slaughterhouses during a period of one year. Prevalence of footpad dermatitis was related to background information gathered using a food chain certification scheme to identify possible factors of influence. On average, 35.5% of the broilers had no lesions, whereas 26.1% and 38.4% had mild or severe lesions, respectively. Season, age, thinning of flocks, slaughter age, breed, slaughterhouse, and the interaction between thinning and slaughter age significantly affected severity of FPD. Peak flock FPD scores occurred in flocks where 1-d-old chicks were placed in March and December, whereas flocks placed in warm months, between June and August, displayed lower flock FPD scores. Generally, birds sent to slaughter when thinning a flock displayed less severe FPD than birds from completely depopulated flocks. Severity of FPD decreased with age. Because poultry farmer, hatchery, veterinary practice, and feed manufacturer were included in the model as random factors, it was only possible to assess their contribution relative to each other. The broiler farmer had the largest contribution. Also, a large contribution was found for hatchery, perhaps indicating that broiler quality is important. No relationship was observed between FPD and mortality. Across farms, less severe FPD was observed on farms using antibiotics. However, within farms, FPD was more common in flocks where antibiotics had been used compared with flocks that did not require antibiotic treatment. In conclusion, footpad dermatitis was frequently observed in Dutch fast-growing broiler flocks, and many factors had significant effects on severity of FPD, such as breed, thinning of flocks, age at slaughter, slaughter plant, and hatchery.

BACKGROUND: Recent publications report that up to 70% of total laboratory errors occur in the pre-analytical phase. Identification of specific problems highlights pre-analytic processes susceptible to errors. The rejection of unsuitable samples can lead to delayed turnaround time and affect patient care. METHODS: A retrospective audit was conducted investigating the rejection rate of routine blood specimens received at chemistry and haematology laboratories over a 2-week period. The reasons for rejection and potential clinical impact of these rejections were investigated. Thirty patient files were randomly selected and examined to assess the impact of these rejections on clinical care. RESULTS: A total of 32,910 specimens were received during the study period, of which 481 were rejected, giving a rejection rate of 1.46%. The main reasons for rejection were inappropriate clotting (30%) and inadequate sample volume (22%). Only 51.7% of rejected samples were repeated and the average time for a repeat sample to reach the laboratory was about 5 days (121 h). Of the repeated samples, 5.1% had results within critical values. Examination of patient folders showed that in 40% of cases the rejection of samples had an impact on patient care. CONCLUSIONS: The evaluation of pre-analytical processes in the laboratory, with regard to sample rejection, allowed one to identify problem areas where improvement is necessary. Rejected samples due to factors out of the laboratory's control had a definite impact on patient care and can thus affect customer satisfaction. Clinicians should be aware of these factors to prevent such rejections.

In the Dutch broiler chain, data are collected as a routine practice. However, there is wide variation in the content of data collected and in data collection systems. This variability hampers the use of field data in management information systems to support decisions. The objective of this study was to analyze the quality of data and to standardize the content of data sets in the broiler production chain. To evaluate the quality of data, data sets from 3 Dutch hatcheries, from 23,637 batches of eggs, were assessed. The quality of data was assessed intuitively based on 7 quality attributes. To standardize the content of the data set, a protocol was proposed and validated. The protocol was validated at 30 breeder farms, 3 hatcheries, and 104 broiler farms by using 3 quality attributes: consistency, uniformity, and completeness. Results of the data quality analysis of the 3 Dutch hatcheries showed that the data sets had some fields with inaccurate, incorrect, inconsistent, nonuniform, incomprehensible, missing relevant, or incomplete data. Results of the validation protocol were as follows: feedback was obtained from 23 (77%) breeder farms, 3 (100%) hatcheries, and 7 (7%) broiler farms. Of all the questions, on average 88% were answered on breeder farms; 57, 65, and 82% were answered at each of the 3 hatcheries, respectively; and 79% were answered on the broiler farms. Data collected at 2 hatcheries were more consistent than those collected at the third hatchery. Hatchery data were less consistent than breeder farm data, but the number of data entries at hatcheries far exceeded the number at the farm level. Data from the hatcheries, breeder farms, and broiler farms were not always uniform, possibly because of differences in management strategies. This protocol enables the listing of relevant and standard contents of a data set whereby information exchange along the chain can be simplified. However, it is recommended that the protocol be supplemented with some rules for data collection and management, for example, that variables must be recorded in the provided fields, and that a variable must have one and only one name or code, the same unit of measurement, and the same definition.

In practice, many hatchability and chick quality problems have been related to the control of embryo temperature (ET) during incubation. Within an incubator, set at a constant machine temperature (MT), ET can vary substantially. Embryo temperature is the result of the balance between heat transfer to and from the embryo and heat production (HP) of the embryo. We investigated which factors theoretically could account for the variation in ET within an incubator. First, the effects egg weight, MT, and oxygen availability on HP of embryos were quantified. Differences in HP could be due only to differences in the amount of energy utilized from the egg or to differences in the efficiency of the conversion of energy in the egg to energy in the chicken, indicated as E(YFB). Results of these analyses showed that differences in HP attributable to egg weight or oxygen availability were mainly a result of the amount of energy used from the egg constituents and not of a change in E(YFB). However, at a given MT, this variation in HP could account for a maximum increase in ET of only 1.21°C, suggesting that other factors played a role because in practice within an incubator, larger differences in ET have been found. The most important factor was probably the difference in air velocity within an incubator, resulting in differences in heat transfer. Because of this variation, ET varied within an incubator and with increasing ET, E(YFB) decreased, resulting in an even higher HP and consequently ET. We concluded that this theoretical approach could explain the wide variation in ET, and consequently could explain the negative effects of high ET on hatchability and chick quality found in the literature. This indicates that, in both practice and in incubation experiments, it is of great importance to realize that any factor affecting HP or heat transfer influences ET. We strongly suggest that ET (or eggshell temperature) be controlled in any incubation experiment involving hatchability or energy utilization.

AIMS OF THE STUDY: In South Africa, the genus Helichrysum is widely used in traditional medicine. The uses are well documented although renaming of species and the resulting confusing taxonomic nomenclature may cause uncertainty as to which specific species was referred to in some reports. The aim of this paper is to present a collated and coherent overview of the documented traditional uses of Helichrysum species and to update the botanical identity of previously studied species. MATERIALS AND METHODS: Databases (Scifinder, ISI Web of Knowledge) and several books were used to collect in information on South African Helichrysum species. RESULTS: The traditional uses, chemistry and biological activity of Helichrysum species have been summarized. It was attempted to give clarity as to exactly which species is refer to in the ethnobotanical literature. CONCLUSIONS: Although a large number of ethnopharmacological uses have been documented and the chemistry of the genus has been studied extensively, only a few South African species have been investigated for their biological activity.

Embryo development and heat production (HP) were studied in eggs of similar size (60 to 65 g) that were incubated at normal (37.8 degrees C) or high (38.9 degrees C) eggshell temperature (EST) and exposed to low (17%), normal (21%), or high (25%) O(2) concentration from d 9 through 19. High EST initially increased HP, but gradually O(2) became more important for HP than EST. Finally,HP was highest for the combination of high EST with high O(2) and lowest for the combination of high EST with low O(2). High EST decreased hatch time, BW, yolk free BW, and relative heart weight. The EST had no effect on residual yolk weight, chick length, or relative liver weight. Increased O(2) increased yolk free BW and chick length and decreased residual yolk weight at hatch. No interactions between EST and O(2) were observed with regard to embryo development and hatchling characteristics. If embryo development is reflected by HP, it can be concluded that high EST primarily increased embryonic development until the second week of incubation. During the third week of incubation, O(2) had a greater effect in determining embryo development than EST.

Two experiments were carried out to study embryonic metabolic responses to short-term temperature fluctuations in order to explore the possibilities of using embryonic metabolic responses as a tool to control the incubation process. In the first experiment, eggshell temperature (ET) in the control group was kept constant at 37.8 degrees C, and embryos in the experimental group were exposed to varying ET within the range of 36.8 to 38.8 degrees C using ET steps of 0.2 degrees C and time steps of 3 h. This was repeated in 3 periods between 6.5 and 9.5 d, 10.5 and 13.5 d, and 14.5 and 17.5 d. In the studied ET range, heat production (HP) increased linearly at 4.9% per 1 degrees C ET. In the second experiment, a standard machine temperature (MT) was used for the control group, and eggs in the experimental group were exposed to low (MT - 0.3 degrees C) or high (MT + 0.3 degrees C) temperatures for 1 h of time at d 8, 9, and 11 to 16. When MT was decreased, CO2 production initially increased at 0.5% and decreased thereafter. When MT was increased, CO2 production initially decreased at 0.4% and increased thereafter. It was concluded that embryonic HP responded linearly with short-term ET changes in the studied ET range of 36.8 to 38.8 degrees C. Changes in CO2 concentration due to short-term MT changes could not be explained by embryonic HP only. It can be speculated that blood flow through the chorio-allantoic membrane changes with MT, affecting heat transfer and diffusion of CO2. A second, delayed response to MT changes was in accordance with the findings in Experiment 1. Within the studied temperature range it will be difficult to use embryonic metabolic responses as a tool to control the incubation process. Because HP is linearly related to ET as in the studied temperature range, other factors such as O2 availability or CO2 release may limit embryo development at higher ET. At this moment, research on the effects of gas exchange at different temperatures on embryo development and survival is lacking.

Different incubation conditions can cause eggshell temperature (EST) to deviate from optimum. Two experiments were performed to determine the effect of low EST at the start of incubation and high EST at the end of incubation on hatchability, chick quality, 6-wk live performance, and breast meat yield of broiler chickens. In each experiment, 1,800 eggs from a single flock were divided and set into 2 setters. From 0 to 10 d of incubation, one setter was set to attain an EST of 36.6 degrees C (considered low), whereas the other was set to 37.8 degrees C (the control temperature). Using an infrared thermometer, EST was measured daily on a sample of eggs to ensure treatment intentions. On d 11 of incubation, the temperature of the low EST setter was increased to 37.8 degrees C in synchrony with the other setter until transfer. On d 18 of incubation, eggs from both setters were combined into 2 equal groups and transferred to hatchers. The EST in one hatcher was set to 37.8 degrees C (control) and in the other to 39.5 degrees C (considered high) until 21 d of incubation. Hatched males were placed in battery cages (Experiment 1) or floor pens (Experiment 2) and reared on common feeds to 1 or 6 wk of age, respectively. Low EST in the first 10 d of incubation reduced hatchability, increased BW and chick yield, and reduced 1-wk gain compared with the control EST. Throughout rearing, BW was reduced for low EST chicks compared with control EST chicks; consequently, carcass, fillet, and tender weights were also reduced. High EST in the hatcher increased hatchability, and reduced BW, chick yield, and 1-wk gain compared with control EST in the hatcher. By 3 wk of age, there was no difference in BW between chicks in high EST and control EST treatments. Subsequent carcass and processing yields were also similar. Incubation at the control EST of 37.8 degrees C, particularly from 0 to 10 d, resulted in the best performance overall.

An experiment was conducted to study the effect of egg size on embryo development, heat production, and energy partitioning between egg and hatchling. Small (56.1 +/- 0.12 g SEM) and large (70.0 +/- 0.11 g SEM) hatching eggs were incubated in climate respiration chambers, and eggshell temperature was maintained constant at 37.8 degrees C in both egg weight classes by adjusting machine temperature. Dry matter, ash, protein, and fat contents were determined in albumen, yolk, yolk-free body (YFB), and residual yolk (RY), and carbohydrate contents and caloric values were calculated. To achieve a constant eggshell temperature, machine temperature needed to be set lower from d 15 onward, coinciding with increased heat production in large eggs compared with small eggs. Selective nutrient uptake resulted in higher fat content and lower protein content in RY in chicks that hatched from small eggs compared with large eggs. The respiration quotient in small and large eggs was the same, and embryos in small and large eggs were equally efficient in the transfer of energy from egg to YFB. The surplus availability of nutrients in large eggs was therefore held responsible for the absolute and relative higher weight of RY in chicks that hatched from large eggs compared with small eggs.

Salvia species (sage) are well known in folk medicine throughout the world. In South Africa sage is used against fever and digestive disorders. Three closely related South African species (Salvia stenophylla, Salvia repens and Salvia runcinata) were investigated for their anti-oxidant (DPPH assay); anti-inflammatory (5-lipoxygenase and cyclo-oxygenase assays); antimalarial (tritiated hypoxanthine incorporation assay); antimicrobial (disc diffusion and micro-dilution assays) properties and toxicity profile (tetrazolium-based assay). The solvent extracts exhibited anti-oxidant, antimalarial and antibacterial and poor anti-inflammatory properties. The essential oils exhibited anti-inflammatory and antimalarial properties, but displayed poor anti-oxidant and antimicrobial activity. The extract of Salviastenophylla and the essential oil of Salvia runcinata displayed the highest toxicity profile. Overall, Salvia runcinata displayed the most favorable activity of all three taxa tested with an IC(50) value of 6.09 (anti-oxidant); 29.05 (antimalarial) and 22.82 microg/ml (anti-inflammatory). Analytical procedures (GC-MS and HPLC-UV) were employed to generate chromatographic profiles for the essential oils and solvent extracts respectively. The HPLC analysis revealed the presence of rosmarinic acid in all three taxa while carnosic acid was only present in Salvia repens and Salvia stenophylla. The GC-MS analysis showed that oils were qualitatively and quantitatively variable. beta-Caryophyllene was present in large amounts in all three taxa. Other components present include camphor, alpha-pinene and alpha-bisabolol. The results of the in vitro pharmacological activities provide a scientific basis to validate the use of these Salvia species in traditional medicine in South Africa.

An experiment was conducted to study the effects of different eggshell temperature (EST) profiles during incubation on embryo mortality, hatchability, and embryo development. Furthermore, chicks from different EST profiles were reared under low and high housing temperatures to investigate subsequent posthatch growth and rectal temperature. Two batches of eggs were used in this experiment. Hatching eggs were subjected to 36.7 or 37.8 degrees C EST during the first week, to 37.8 degrees C EST during the second week, and to 37.8 or 38.9 degrees C EST during the third week of incubation. Posthatch housing temperature decreased from 35 degrees C at d 1 to 30 degrees C at d 7 (high) or decreased from 30 degrees C at d 1 to 25 degrees C at d 7 (low). The difference between machine temperature and EST (DT) was used to illustrate the effect of EST on heat production during incubation. DT differed per batch, and was smallest when eggs were incubated at 36.7 degrees C instead of 37.8 degrees C during wk 1. High EST during wk 3 of incubation (38.9 degrees C instead of 37.8 degrees C) reduced DT only in batch 2. Embryo development was most retarded in eggs incubated at 36.7 degrees C EST compared with at 37.8 degrees C during the first week of incubation. However, highest hatchability and embryo development were always found when EST was maintained at 37.8 degrees C constantly throughout incubation. Chicks that hatched from eggs incubated at low EST during wk 1 of incubation had lower rectal temperature after hatching, especially under low housing temperatures, and this effect lasted until 7 d posthatch in batch 1. The highest rectal temperatures were always found in chicks incubated at 37.8 degrees C EST constantly throughout incubation. Eggs and chicks from different batches require different environmental conditions for optimal embryo development, hatchability, and posthatch growth. Rearing temperature and incubation conditions affect the ability of young chicks to maintain their rectal temperature during the first week posthatch.

Salts can damage building materials by chemical reactions or crystallization, which is a serious threat to cultural heritage. In order to develop better conservation techniques, more knowledge of the crystallization processes is needed. In a porous material, the size of a salt crystal is limited by the sizes of the pores. It has been predicted that as a consequence, the solubility of a salt increases with decreasing pore size. This increase seems to be related to an increase of the stress generated by a crystal on the pore wall. It has been suggested that the resulting stress could become high enough to induce failure. We have studied the crystallization of salts in porous materials with well-defined pore sizes. Samples were saturated at 40 degrees C with saturated Na2SO4 and Na2CO3 solutions. Next we have cooled the samples to 0 degrees C and waited for nucleation. After nucleation occurred, the solubility in the porous material was measured with nuclear magnetic resonance (NMR) as a function of the temperature. The measurements on Na2CO3 indeed show an increase in solubility with a decrease in pore size. For Na2SO4, we did not observe a pore size-dependent solubility. However, we have to remark that these results show a metastable crystal phase. The results can be used to calculate the actual pressure exerted by the crystals onto the pore wall.

Helichrysum species are used widely to treat various medical conditions. In this study, the anti-microbial, anti-oxidant (DPPH assay) and anti-inflammatory activity (5-lipoxygenase assay) of Helichrysum dasyanthum, Helichrysum felinum, Helichrysum excisum and Helichrysum petiolare were investigated. The essential oil compositions of these species were determined. The acetone and methanol extracts as well as the essential oils exhibited activity against Gram-positive bacteria, while both the methanol and acetone extracts of all four species were active in the anti-oxidant assay. The essential oils, on the other hand, displayed activity in the 5-lipoxygenase assay, which was used as an indication of anti-inflammatory activity. Two extracts exhibited promising activity in the anti-microbial assay, the acetone extract of Helichrysum dasyanthum with a MIC value of 15.63 microg/ml and the methanol extract of Helichrysum excisum with a MIC value of 62.5 microg/ml. The acetone extract of Helichrysum dasyanthum was the most active free radical scavenger in the DPPH assay (IC(50) of 9.53 microg/ml) while values for the anti-inflammatory activity of the essential oils ranged between 25 and 32 microg/ml. The essential oil compositions of three species (Helichrysum dasyanthum, Helichrysum excisum and Helichrysum petiolare) were dominated by the presence of monoterpenes such as alpha-pinene, 1,8-cineole and p-cymene. In the oil of Helichrysum felinum, monoterpenes were largely absent. Its profile consisted of a variety of sesquiterpenes in low concentrations with beta-caryophyllene dominating.
