RESUMO

MicroRNAs (miRNAs) are known to play a regulatory role in various cancers including laryngocarcinoma. MiR-29a-3p is a potential tumor-suppressive miRNA, but its function in laryngocarcinoma is unknown. The purpose of this study was to investigate the roles of miR-29a-3p in laryngocarcinoma. Prominin1 (PROM1) was predicted as a target gene of miR-29a-3p and this was verified using a luciferase reporter assay. Transfection of miR-29a-3p into two laryngocarcinoma cell lines indicated that miR-29a-3p could decrease cell proliferation and enhance the chemotherapy response by targeting PROM1. PROM1 expression was up-regulated in the laryngocarcinoma cells when miR-29a-3p was down-regulated. We found miR-29a-3p expression levels were lower in laryngocarcinoma tissues than in control tissues. We also found that miR-29a-3p expression was negatively correlated with PROM1 expression in laryngocarcinoma tissues. The study demonstrates that miR-29a-3p suppresses cell proliferation in laryngocarcinoma by targeting PROM1.

Chemokine and its receptors play important roles in laryngeal cancer development and progression. CCR6 is the receptor of CCL20 chemokine, but its function in laryngeal cancer is not known. The aim of this study is to explore the roles of CCR6 and its regulation mechanism in laryngeal cancer. We found CCR6 expression was higher in laryngeal cancer tissues compared with their normal controls, so did in laryngeal cancer cells. Cellular function indicated that down-regulation of CCR6 in laryngeal cancer cells could suppress cell proliferation and metastasis. Further research showed that CCR6 could activate p38, which was related with the changes of microRNA (miRNA) profile in laryngeal cancer cells. We also found that CCR6 was positively associated with miR-20a-5p, miR-489 and negatively related to miR-29-3p, miR-632 and miR-1276 in laryngeal cancer tissues.

MicroRNAs (miRNAs) play important roles in gene regulation during laryngocarcinoma. MiR-9 is a potential oncomiR, but its function in laryngocarcinoma is not known. The aim of this study is to investigate the roles of miR-9 in laryngocarcinoma. We found miR-9 expression was higher in laryngocarcinoma tissues compared with their normal controls, so did the laryngocarcinoma cells. Cellular function of miR-9 indicated that miR-9 restoration in laryngocarcinoma cells could promote cell proliferation and metastasis. Phosphatase and tensin homolog (PTEN) was predicted as a target gene of miR-9 and verified using luciferase reporter assay. PTEN expression was down-regulated in the laryngocarcinoma cells with miR-9 overexpression. We also found that miR-9 expression was negatively associated with PTEN expression in laryngocarcinoma tissues.

Many studies have investigated the association between CYP1A1 rs1048943 and rs4646903 polymorphisms and laryngeal cancer risk, but their results have been inconsistent. The PubMed and CNKI were searched for case-control studies published up to 01 July 2015. Data were extracted and pooled odds ratios (OR) with 95% confidence intervals (CI) were calculated. In this meta-analysis, we assessed 10 published studies involving comprising 748 laryngeal cancer cases and 1558 controls of the association between CYP1A1 rs1048943 and rs4646903 polymorphisms and laryngeal cancer risk. For CYP1A1 rs1048943 of the homozygote G/G and G allele carriers (A/G + G/G) versus A/A, the pooled ORs were 1.77 (95% CI = 1.28-2.81, P = 0.007 for heterogeneity) and 1.86 (95% CI = 1.45-2.40, P = 0.000 for heterogeneity). For CYP1A1 rs4646903 of the homozygote G/G and G allele carriers (A/G + G/G) versus A/A, the pooled ORs were 1.53 (95% CI = 1.31-2.21, P = 0.012 for heterogeneity) and 1.33(95% CI = 1.04-1.71, P = 0.029 for heterogeneity). In the stratified analysis by ethnicity, the significantly risks were found among Asians for both the G allele carriers and homozygote G/G. However, no significant associations were found in Caucasian population all genetic models. These results from the meta-analysis suggest that CYP1A1 rs1048943 and rs4646903 polymorphisms contribute to risk of laryngeal cancer among Asian populations.

OBJECTIVE: To study the expression of ICAM-1 in laryngeal carcinoma and it's significance. METHOD: ICAM-1 were detected in 78 cases laryngeal carcinoma, 30 cases dysplasia and 5 cases normal by SP immunohistochemistry. RESULT: ICAM-1 can be expressed on the lymphocytes of patients with laryngeal carcinoma, but not cells of normal, dysplasia and cancer. The first and second stages laryngeal carcinoma had stronger ICAM-1 expression than those of the third and fourth stages (P < 0.05). ICAM-1 expression in laryngeal carcinoma with cervical metastasis was higher than that without cervical metastasis (P < 0.05). Three and five years survival rate in patients with higher ICAM-1 expression were higher than those of patients with lower ICAM-1 expression (P < 0.05). CONCLUSION: These results implicate that the development, metastasis and prognosis of laryngeal carcinoma may be correlated to ICAM-1 expression of lymphocyte.

OBJECTIVE: To observe the effect of laser in treatment of obstructive sleep apnea-hypopnea syndrome (OSAHS). METHOD: Eighty-nine cases of OSAHS were treated by Laser assisted uvulopalptoplasty (LAUP). Among the cases, the soft palate and uvula were partly resected. The cureiform incise was made into the soft palate along the each side of uvula. RESULT: The follow-up period was six months, 76 cases received the detection of polysomnography. Eight cases were cured; 36 cases had notable effects; 25 cases had effects; 7 cases had no effects. The total ratio of validity was 90.8%. CONCLUSION: It has clinical value to use LAUP in treatment of OSAHS, and its advantage include hemostasia, solidfication, penetrability, clear visual threshold, lower complication and shortening the surgery time.
