RESUMO

Ionotropic receptors (IRs) play an important role in olfaction, but little is known in nondrosophila insects. Here, we report in vitro and in vivo functional characterization of IR75q.2 in the invasive moth pest Spodoptera frugiperda. First, 13 IRs (including four coreceptor IRs) were found specifically or highly expressed in adult antennae. Second, these IRs were tested for responding profiles to 59 odorants using the Xenopus oocyte expression system, showing that only SfruIR75q.2 responded to 8-10C fatty acids and their corresponding aldehydes, with SfruIR8a as the only coreceptor. Third, the three acids (especially nonanoic acid) showed repellent effects on moth's behavior and oviposition, but the repellence significantly reduced to the insects with IR75q.2 knockout by CRISPR/Cas9. Taken together, our study reveals the function of SfruIR75q.2 in perception of acid and aldehyde odorants and provides the first in vivo evidence for olfactory function of an odor-specific IR in Lepidoptera.

The primary advantage of a hollow structure is the likelihood of introducing diverse components in a single particle to achieve multiple missions. Herein, hollow microspheres with multicomponent nanocores (HMMNs) have been prepared based on a template-free strategy via a microwave-assisted hydrothermal treatment of Chlorella. The resulting HMMNs retain the near-spherical hollow morphology and functional groups of the cell wall of Chlorella, obviating the need for templates and chemical modification. The elements (iron, cobalt, calcium, magnesium, chlorine, and phosphorus) naturally present within the Chlorella cells react to form hydroxyapatite/chlorapatite and magnetic nanocores without the need for exogenous chemical reagents. The performances of HMMNs for cadmium ion (Cd2+) removal and antibiotic detection are explored. HMMNs exhibit relatively high adsorbance of Cd2+ (1035.8 mmol/kg) and can be easily recovered by application of an external magnetic field. Ion exchange with Ca2+ and Mg2+ is shown to be the main mechanism of Cd2+ elimination. In addition, HMMNs are a suitable carrier for the construction of a magnetic immunosensor, as demonstrated by the successful development of such an immunosensor with acceptable analytical performance for the detection of neomycin in milk samples. The versatile applications of HMMNs result from their multicomponent nanocores, hollow structure, and the functional groups on their shell. This work not only offers a simple and eco-friendly strategy for the fabrication of novel HMMNs but also provides a valuable advanced material for contaminant detection and heavy-metal removal.

The distribution in water and sediment, the sources/sinks and the risk of perfluorinated compounds (PFCs) in Lake Taihu, China were investigated. The total PFCs concentration was 164 to 299 ng L(-1) in water and 5.8 to 35 ng g(-1) (dw) in sediment. The highest concentrations of perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA) in water were 29.2 ng L(-1) and 136ngL(-1). PFOS was largely associated with sediment, whereas short chain PFCs predominated in water. The partition coefficient (Kd) was positively correlated with the organic carbon fraction (ƒoc) for PFOS but not for the other PFCs. The organic carbon normalized partition coefficient (Koc) increased by 0.51 log units for each additional CF2 moiety from perfluoro-butanesulfonate (PFBS) to PFOS. For the same chain length but different functional groups, the log Koc of PFOS was 1.35 units higher than PFOA. PFOS exhibited the highest affinity for sediment through the partition mechanism, and ƒoc affected the sediment as a sink of PFOS. Although there was no immediate health impact by the intake of the water alone, the consumption of aquatic products may cause potential health risks for animals/humans on the time scale of months to years. The relationship between the concentration, water-sediment distribution, bioaccumulation and toxicity should be considered in determining the water standards of PFCs.
