Objective: Cancer and its treatments affect patients' fertility potential. This study examined the prevalence of reproductive concerns and their relationship with metacognitions among Chinese young adult female cancer survivors (YAFCS). Methods: A total of 318 YAFCS (aged 18-39) completed an online survey from March to December 2021. Participants reported sociodemographic characteristics, reproductive concerns and metacognitions. Reproductive concerns were measured using the Reproductive Concerns after Cancer scale (RCAC), and metacognitions were measured by the Short Form of Metacognitions Questionnaire (MCQ-30). We used Pearson correlation analysis to examine associations between metacognitions and reproductive concerns across multiple domains and multivariable linear regression to determine the influencing factors of reproductive concerns. Results: The mean score of reproductive concern among YAFCS was 49.97 ± 12.52. A total of 57.9% of participants reported a high level of concern regarding at least one dimension of reproductive concerns, and they were most concerned about their child's health and least concerned about partner disclosure of fertility status. We also found a moderate association between RCAC and MCQ-30 scores (r = 0.408, p < 0.001). In multivariate analyses, metacognitions, especially negative metacognitive beliefs had an impact on reproductive concerns among YAFCS (p < 0.001). Conclusion: Higher reproductive concerns were associated with higher metacognitions among YAFCS, especially with negative metacognitive beliefs. Oncology professionals should pay attention to assessing reproductive concerns in patients who want to have children or who have no children. Moreover, metacognitive beliefs may be an intervention target for alleviating reproductive concerns among YAFCS.

OBJECTIVE: To investigate the effects of Eucommia ulmoides Oliv polysaccharide on the injury of interleukin-1ß(IL-1ß)-induced chondrocyte and its possible mechanism. METHODS: ATDC5 was treated with 10 µg/ml IL-1ß to establish osteoarthritis chondrocyte inflammation model, mouse chondrocyte ATDC5 were cultured in vitro and randomly divided into blank group, model group, model+Eucommia ulmoides Oliv polysaccharide low concentration group, model+Eucommia ulmoides Oliv polysaccharide medium concentration group and model+Eucommia ulmoides Oliv polysaccharide high concentration group. The cells in the blank group were cultured with conventional medium;the cells in the model group cells were cultured with a medium containing 10 ?g/ml IL-1ß, and the cells in the model+Eucommia ulmoides Oliv polysaccharide low concentration group, model+Eucommia ulmoides Oliv polysaccharide medium concentration group and model+Eucommia ulmoides Oliv polysaccharide high concentration group were co-cultured with medium containing 100, 200, 400 µg/ml Eucommia ulmoides Oliv polysaccharide and 10 µg/ml IL-1ß. After the cells of each group were cultured for 24 h, 48 h and 72 h, CCK-8 method was used to detect cell viability. After the cells of each group were cultured for 48 h, flow cytometry and DAPI staining were used to detect cell apoptosis;ELISA method was used to detect the expression of TNF-α, NO, IFN-γ and IL-6 in cells; DCFH-DA method was used to detect the content of ROS in cells;Western blot was used to detect the protein expression of TIMP-1, MMP-13 and NF-κB signaling pathway-related P65 and p-P65;Immunofluorescence staining was used to observe the localization of NF-κB P65 cells. RESULTS: Compared with the blank group, the ATDC5 cell viability and the protein expression of TIMP-1 in the model group reduced (P<0.05), while apoptosis rate, the levels of TNF-α, NO, IFN-γ and IL-6, the content of ROS, the protein expression of MMP-13 and p-P65, and the number of P65+ in the nucleus increased(P<0.05). Compared with the model group, the ATDC5 cell viability and the protein expression of TIMP-1 in the model+Eucommia ulmoides Oliv polysaccharide low concentration group, model+Eucommia ulmoides Oliv polysaccharide medium concentration group and model+Eucommia ulmoides Oliv polysaccharide high concentration group increased (P<0.05), while apoptosis rate, the levels of TNF-α, NO, IFN-γ and IL-6, the content of ROS, the protein expression of MMP-13 and p-P65, and the number of P65+ in the nucleus reduced (P<0.05). CONCLUSION: The results showed that Eucommia ulmoides Oliv polysaccharide could promote proliferation of IL-1ß-induced chondrocyte ATDC5 and inhibit its apoptosis, inflammatory response and matrix degradation. Its mechanism may be related to the inhibition of the activation of NF-κB pathway.

The aim of the present study was to clarify the roles of the mammalian target of rapamycin (mTOR) signalling pathway in follicular growth and development of thecal cells. Using in vivo-grown and in vitro-cultured ovaries, histological changes were evaluated using haematoxylin and eosin (HE) staining. Differentially expressed genes (DEGs) from 0 day post partum (d.p.p.) to 8 d.p.p. ovaries were screened by microarray and verified by quantitative real-time polymerase chain reaction. Forty-two DEGs related to cell proliferation and differentiation were screened out, with most DEGs being related to the to mTOR signalling pathway. Then, 3 d.p.p. ovaries were retrieved and used to verify the role of mTOR signalling in follicle and thecal cell development using its activators (Ras homologue enriched in brain (Rheb) and GTP) and inhibitor (rapamycin). The development of follicles and thecal cells was significantly impaired in ovaries cultured in vitro Day 3 to Day 8. In in vitro-cultured ovaries, Rheb and GTP (is 100ngmL-1 Rheb and 500ngmL-1 GTP for 48h) significantly increased follicle diameter, the percentage of primary and secondary follicles and the umber of thecal cells, and upregulated expression of mTOR, phosphorylated eukaryotic translation initiation factor 4E-binding protein 1 (4EBP1), eukaryotic initiation factor (eIF) 4F and cytochrome P450, family 17, subfamily A, polypeptide 1 (CYP17A1). Rapamycin (10nM rapamycin for 24h) had opposite effects to those of Rheb and GTP, and partly abrogated (significant) the effects of Rheb and GTP when added to the culture in combination with these drugs. Thus, mTOR signalling plays an important role in follicle growth and thecal cell development.

RESUMO

A new quinoline-based fluorescent probe was synthesized and characterized. Probe showed opposite fluorescence responses toward Cd(2+) (turn-on) and Hg(2+) (turn-off) in a 100% aqueous environment. X-ray crystallography analysis and TD-DFT calculations showed the different mechanisms of CHEF-based for -Cd(2+) and PET-based for -Hg(2+) caused by different binding modes and different electronic transfer patterns. The detection limits for the analysis of Cd(2+) and Hg(2+) ions were found to be 3.9 × 10(-8) M and 9.8 × 10(-7) M, respectively. Its in vivo sensitivities to Cd(2+) and Hg(2+) were demonstrated in the EC9706 cell line with the use of a laser scanning fluorescence microscope.

OBJECTIVE: To develop a method for the simultaneous determination of 44 organophosphorus pesticides in food by SPE disk extraction-capillary gas chromatography with pulsed flame photometric detection. METHODS: Organophosphorus pesticides in food were extracted ultrasonically with water. Then the extract was cleaned-up with SPE disk and eluted with ethyl acetate. Finally the eluent was condensed to 1mL under N2 at 55 degrees C. Gas chromatography was applied for quantitative detection of the organophosphorus pesticides in the sample. RESULTS: The linear range of the method for all the pesticides were in the range of 0.01-0.5 mg/kg with correlation coefficients of 0.992-1.000. The detection limits of the method were in the range of 0.0005-0.01 mg/kg. The recoveries for most pesticides were 60%-120% with relative standard deviations of less than 15%. CONCLUSION: The method is simple, sensitive, environmentally friendly and suitable for the determination of organophosphorous pesticides in food.

OBJECTIVE: To improve the understanding in pathology and provide steady proof for diagnosis and treatment of tethered cord syndrome (TCS), the ultrastructure of filum terminale (FT) with TCS will be researched, respectively. METHODS: Thirty-one fila were sectioned and divided into two groups according to diameter. Twenty-two fila thicker than 2 mm were placed into group 1; nine no thicker than 2 mm were placed into group 2. Their transverse and longitudinal sections were respectively submitted for scanning electron microscopy analysis. RESULTS: For the FTs with TCS, ultrastructural analysis confirmed the presence of abundant collagen bundles and adipocytes, sparse or invisible elastic and reticular fibers inside the FT, but the latter two fibers were still visible in the relative normal areas of FT; some abnormalities, such as thickening, distortion, adhesion or fusion to piece, relatively dense and disorderly arrangement at a different degree, were quite common for collagen fiber. Some areas looked blurry, and some crystallization appeared occasionally on their surface. For the FTs with TFTS, those changes as mentioned above were still exhibited, but slightly relatively. All the changes mentioned inside all the FTs may exist alone or concomitantly, rarely simultaneously. Moreover, they can be often seen focally or diffusely occasionally. CONCLUSIONS: Low elasticity resulting from the destruction of the three-dimensional structure of FT is involved in TCS. Early FT severing is a treatment of choice no matter whether its appearance looks normal.
