RESUMO

The development of electrochemical sensors for flutamide detection is a crucial step in biomedical research and environmental monitoring. In this study, a composite of Actinidia-derived carbon particles (CPs) and tungsten disulfide (WS2) was formed and used as an electrocatalyst for the electrochemical detection of flutamide. The CPs had an average diameter of 500 nm and contained surface hydroxyl and carbonyl groups. These groups may help anchor the CPs onto the WS2 platelets, resulting in the formation of a CPs-WS2 nanocomposite with a high surface area and a conducting network, enabling electron transfer. Using the CPs-WS2 composite supported at a glassy carbon electrode, a linear concentration range extending from 1 nM to 104 µM, a limit of detection of 0.74 nM, and a sensitivity of 26.9 ± 0.7 µA µM-1 cm-2 were obtained in the detection of flutamide in a phosphate buffer. The sensor showed good recovery, ranging from 88.47 to 95.02%, in river water samples, and exhibited very good selectivity in the presence of inorganic ions, including Al3+, Co2+, Cu2+, Fe3+, Zn2+, NO3 -, SO4 2-, CO3 2-, and Cl-.

Sulfonamides are a family of synthetic drugs with a broad-spectrum of antimicrobial activity. Like other antimicrobials, they have been found in aquatic environments, making their detection important. Herein, an electrochemical sensor was designed using tannic acid exfoliated few-layered MoS2 sheets, which were combined with a mixture of reduced graphene oxide (rGO) and graphite flakes (G). The rGO/G was formed using electrodeposition, by cycling from -0.5 to -1.5 V in an acidified sulfate solution with well dispersed GO and G. The exfoliated MoS2 sheets were drop cast over the wrinkled rGO/G surface to form the final sensor, GCE/rGO/G/ta-MoS2. The mixture of rGO/G was superior to pure rGO in formulating the sensor. The fabricated sensor exhibited an extended linear range from 0.1 to 566 µM, with a LOD of 86 nM, with good selectivity in the presence of various salts found in water and structurally related drugs from the sulfonamide family. The sensor showed very good reproducibility with the RSD at 0.48 %, repeatability and acceptable long term stability over a 10-day period. Good recovery from both tap and river water was achieved, with recovery ranging from 90.4 to 98.9 % for tap water and from 83.5 to 94.4 % for real river water samples.

Granulosa cells are essential for follicle initiation and development, and their abnormal function or apoptosis is a crucial factor leading to follicular atresia. A state of oxidative stress occurs when the balance between the production of reactive oxygen species and the regulation of the antioxidant system is disturbed. Oxidative stress is one of the most important causes of the abnormal function and apoptosis of granulosa cells. Oxidative stress in granulosa cells causes female reproductive system diseases, such as polycystic ovary syndrome and premature ovarian failure. In recent years, studies have confirmed that the mechanism of oxidative stress in granulosa cells is closely linked to the PI3K-AKT signaling pathway, MAPK signaling pathway, FOXO axis, Nrf2 pathway, NF-κB signaling pathway, and mitophagy. It has been found that drugs such as sulforaphane, Periplaneta americana peptide, and resveratrol can mitigate the functional damage caused by oxidative stress on granulosa cells. This paper reviews some of the mechanisms involved in oxidative stress in granulosa cells and describes the mechanisms underlying the pharmacological treatment of oxidative stress in granulosa cells.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) mainly invades the respiratory system, but may also cause various cardiovascular complications. We report a rare case of myocarditis associated with SARS-CoV-2 infection. A 61-year-old man was admitted to the hospital with a positive nucleic acid test for SARS-CoV-2. A sudden increase in troponin level (up to .144 ng/mL) was observed on the 8th day after admission. He developed symptoms of heart failure and progressed rapidly to cardiogenic shock. Echocardiography on the same day showed reduced left ventricular ejection fraction, reduced cardiac output, and segmental ventricular wall motion abnormalities. Takotsubo cardiomyopathy associated with SARS-CoV-2 infection was considered based on the typical echocardiography findings. We immediately started veno-arterial extracorporeal membrane oxygenation (VA-ECMO) treatment. The patient was successfully withdrawn from VA-ECMO after 8 days following recovery of ejection fraction to 65% and all indicators qualifying the withdrawal criteria. Echocardiography plays an important role in dynamic monitoring of cardiac changes in such cases and can help determine the timing of extracorporeal membrane oxygenation treatment and withdrawal.

RESUMO

Mitral valve leaflet aneurysm (MVA) is a rare and potentially devastating complication of infective endocarditis. Here, we report the case of a 49-year-old man with mitral endocarditis who had an anterior MVA without aortic morphologic change and only mild regurgitation. By real-time 3D transesophageal echocardiography (TOE), we found two perforations and a hard mass in the aneurysm accompanied with severe regurgitation. The patient underwent valve replacement surgery combined with anti-infection treatment and was successfully discharged. In addition to the case report and literature review related to MVA, we also summarize the application value of RT-3D TOE in these cases.

Semitransparent organic solar cells (ST-OSCs) offer potentially more opportunities in areas of self-powered greenhouses and building-integrated photovoltaic systems. In this work, the effort to use a combination of solution-processable gold nanobipyramids (AuNBPs)-based hole transporting layer and a low/high dielectric constant double layer optical coupling layer (OCL) for improving the performance of ST-OSCs over the two competing indexes of power conversion efficiency (PCE) and average visible transmittance (AVT) is reported. The fabrication and characterization of the ST-OSCs are guided, at design and analyses level, using the theoretical simulation and experimental optimization. The use of a low/high dielectric constant double layer OCL helps enhancing the visible light transparency while reflecting the near-infrared (NIR) photons back into the photoactive layer for light harvesting. NIR absorption enhancement in the ST-OSCs is realized through the AuNBPs-induced localized surface plasmon resonance (LSPR). The weight ratio of the polymer donor to nonfullerene acceptor in the bulk heterojunction is adjusted to realize the maximum NIR absorption enhancement, enabled by the AuNBPs-induced LSPR, achieving the high-performance ST-OSCs with a high PCE of 13.15% and a high AVT of 25.9%.

This study aimed to investigate the effect of dietary tetramethylpyrazine (TMP) on the growth performance, nutrient digestion, blood parameters and immunity of broilers under oxidative stress. Five treatments included negative control, positive control with lipopolysaccharide induction and TMP addition at 50, 100 and 150 mg/kg of diet using 600 male Arbor Acres broiler chicks. Results showed that during 1-14 days of age, body weight gain and feed efficiency in the positive control were worsened (p < .05) compared with the negative control, while with incremental TMP doses from 0 to 150 mg/kg there were linear and quadratic increases (p < .001) in body weight gain and a linear decrease in feed/gain (p = .001). During 12-14 days of age, with incremental TMP doses, crude protein digestibility was linearly increased (p = .001), and gross energy utilization was linearly and quadratically changed (p < .001). At 14 days of age, the TMP beneficially regulated digestive enzymes, blood parameters and immunoglobulins, showing linear and quadratic responses (p ≤ .008) on trypsin, lipase, glutamic pyruvic transaminase, glucose, lipoproteins, albumin, immunoglobulins (M, Y), interleukin 6 and interferon α, and only linear changes (p ≤ .030) on amylase, glutamic oxaloacetic transaminase, immunoglobulin A and interleukin 2. Most parameters in TMP groups reached to the levels of negative control and the effects of TMP at 100 or 150 mg/kg were more pronounced on body weight gain, crude protein digestibility, trypsin and glutamic pyruvic transaminase. It is concluded that TMP can be used as a feed additive capable of improving growth, blood parameter and immunity of broiler chicks under oxidative stress.

OBJECTIVE: This study aimed to investigate the effect of Salvia miltiorrhiza (S.m.) aerial parts as an alternative ingredient on growth performance, nutrient digestibility, and digestive enzymes in growing rabbits. METHODS: Treatments included five tested diets: a control (basal diet), antibiotic (basal diet +enramycin at 5 mg/kg), and S.m. aerial parts powder added at 3.0%, 6.0%, and 9.0% of feed using 300 growing rabbits. RESULTS: The diets with S.m. aerial parts addition at 9.0% decreased (p<0.05) feed/gain compared to the control, but there were no differences in feed intake and body weight gain. In contrast with the control, the addition of antibiotic increased (p<0.05) digestibility of dry matter, crude protein, energy, fiber, and ash. The herb addition did not cause differences in the digestibility of most nutrients compared to the antibiotic, but fiber digestibility of the herb at 6.0% and 9.0% was lower (p<0.05) than that of the antibiotic. Moreover, the antibiotic and the herb also similarly increased (p<0.05) the activities of duodenal α-amylase, maltase, lipase, and trypsin, compared to the control, and the herb at 6.0% and 9.0% showed a greater (p<0.05) activity of elastase than the dose 3.0%. CONCLUSION: The obtained data indicate that S.m. aerial parts can be a potential forage in rabbit's diet at 9.0% with a beneficial regulation on nutrition and digestion.

The present study aimed to investigate the effect of probiotic Lactobacilli addition on Listeria monocytogenes load, inflammatory reaction, and virulence properties in broilers from 1 to 14 D of age. A total of 480 broiler chicks were randomly allocated to 4 treatments of 6 replicates each. All birds were infected with L. monocytogenes on the first day and supplemented an equal amount mixture of Lactobacillus acidophilus and Lactobacillus plantarum at doses of 0 (control), 106, 108, 1010 cfu/kg of diet. The results showed that on 7 and 14 D after administration, Lactobacilli addition at the 3 doses decreased (P < 0.05) L. monocytogenes loads in the cecum, skin, liver, and spleen by 0.065 to 0.933 log10 cfu, and the pathogen linearly reduced (P ≤ 0.015) with the increasing doses of probiotics in the skin. Serum cytokines including IL-1ß, IL-6, tumor necrosis factor-α, and interferon-γ in probiotics treatments were decreased (P < 0.05) by 25.4 to 51.1%. Transcriptional levels of genes related to anti-inflammatory reactions including IL-10, hypoxia inducible factor 1 alpha (HIF1A), prostaglandin E receptor 2, and prostaglandin-endoperoxide synthase 2 in the intestinal mucosa were upregulated (P < 0.05) in Lactobacilli treatments, and linear and quadratic responses (P ≤ 0.019) were found on HIF1A. Furthermore, the probiotics attenuated (P < 0.05) listerial adhesion, pore-forming, and invasion properties by downregulating autolysin Ami, listeriolysin O, internalin A and B, and a linear (P = 0.006) dose response of probiotics was exhibited on flagellin. The findings indicate that dietary coadministration of L. acidophilus and L. plantarum can attenuate L. monocytogenes infection by depressing its intestinal inoculation, translocation, inflammatory reaction, and virulence property in broilers and suggest that the probiotics can be an alternative against listerial infection in broilers.

INTRODUCTION: Porcine epidemic diarrhoea virus (PEDV) infection causes watery diarrhoea, vomiting, anorexia, and weight loss, especially among neonatal piglets, inflicting on them morbidity and mortality potentially reaching 90%-100%. Despite it being known that certain mammalian cell phases are arrested by PEDV, the mechanisms have not been elucidated, and PEDV pathogenesis is poorly understood. This study determined the effect of an epidemic PEDV strain on cell cycle progression. MATERIAL AND METHODS: We observed the effect of the PEDV SHpd/2012 strain on an infected Vero cell cycle through flow cytometry and Western blot, investigating the interrelationships of cell-cycle arrest, the DNA damage-signalling pathway caused by PEDV and the phosphorylation levels of the key molecules Chk.2 and H2A.X involved upstream and downstream in this pathway. RESULTS: PEDV induced Vero cell-cycle arrest at the G1/G0 phase. The phosphorylation levels of Chk.2 and H2A.X increased with the prolongation of PEDV infection, and no significant cell-cycle arrest was observed after treatment with ATM or Chk.2 inhibitors. The proliferation of PEDV was also inhibited by treatment with ATM or Chk.2 inhibitors. CONCLUSION: PEDV-induced cell-cycle arrest is associated with activation of DNA damage-signalling pathways. Our findings elucidate the molecular basis of PEDV replication and provide evidence to support further evaluation of PEDV pathogenesis.
