RESUMO

The development of simple and effective dual-mode analytical methods plays crucial regulatory roles in the discrimination of relevant target species, because of their built-in cross reference correction and high accuracy. In this work, a novel polymer carbon nanodots (PCNDs) prepared from a facile one-pot hydrothermal procedure using readily available l-tryptophan and l-phenylalanine as precursors, showed excellent aqueous solubility and blue fluorescence property with a high quantum yield of 29%. Moreover, the PCNDs was demonstrated to be a robust luminophore with electrochemiluminescence (ECL) efficiency of 43% was achieved by using K2S2O8 as a coreactant. The spooling ECL spectroscopy was employed to take insight into excited states responsible for the potential-dependent ECL emissions. Most importantly, when introduced into construction of the fluorescence and ECL dual mode sensing platform, for the first time, the PCNDs displayed prominent performance for the detection of ferric ions (Fe3+). The ferric ions could be quantified ranging from micromolar to millimolar with a detection limit of 0.22 and 5.3 µM, respectively. Such a dual-functional sensing platform also exhibits excellent selectivity, reproducibility and stability. Results from this work indicate that PCNDs showing great promise as a bright luminophore for the fabrication of low-cost, high-performance dual-signal readout platforms for ferric ions determination.

Exploiting low cost, water-soluble, and near-infrared (NIR) emissive electrochemiluminophores (ECLphores) is significantly important for biological applications. In this study, bright and NIR electrogenerated chemiluminescence (ECL) emissive copper nanoclusters (Cu NCs) were synthesized through a facile one-pot wet chemical reduction method. ECL properties of obtained Cu NCs were examined in the presence of potassium persulfate, resulting in maximum intensity at 735 nm, at least 135 nm red-shifted with respect to all other Cu NCs. Electrochemistry, photoluminescence (PL), and spooling ECL spectroscopies were used to track NIR ECL emission of Cu NCs ascribed to the monomeric excited states. Due to the abundant binding sites of bovine serum albumin (BSA) to anchor target biomolecules, a sandwich-type ECL immunosensor was thus fabricated using such BSA-templated Cu NCs as tags and alpha fetoprotein antigen (AFP) as a model protein for the first time. Without assisting any signal amplification strategies, the proposed NIR ECL biosensor exhibited a wide linear range (1-400 ng mL-1) and low detection limit (0.02 ng mL-1) as well as superior selectivity and reproducibility and was successfully applied in real human serum sample determination. This work sets the stage for the development of novel non-noble metal nanoclusters for large-scale and emerging nanotechnology applications.

In this paper we report the modification with citrulline of the surface of a glassy carbon electrode (GCE) through cyclic voltammetry to prepare a polycitrulline-modified electrode (PCit/GCE). The electrochemical behaviour of ractopamine on this PCit/GCE was then investigated by cyclic voltammetry, to establish a novel electrochemical method for the detection of ractopamine residues in animal tissue. The optimum preparation conditions for the modified electrode were found to include a polymerisation solution pH of 6.8. The cyclic voltammetry required 12 scan cycles at a scan rate of 40 mV/s with a potential range of -1.6 to 2.3 V. The optimum conditions to determine ractopamine were in a solution of pH 6.0 with cyclic voltammetry performed at a scan rate of 60 mV/s and a potential range of 0.4 to 1.0 V. To analyse meat, the tissue sample was extracted with ethanol, filtered, evaporated to dryness and diluted with phosphate buffered saline at pH of 6.0 before being analysed in the electrochemical cell. The limit of detection of the new method for ractopamine was 1.0 ng/g and the correlation coefficient (R) was 0.9991 in the concentration range of 140-5410 ng/g. Ractopamine residue levels in spiked beef and mutton samples were detected by the modified electrode and the recovery was in the range of 96%-101%, giving results consistent with a validated HPLC method. This study provides a new method for detection and quantification of ractopamine residues in animal tissue.

Usnic acid, a major active compound in lichens, was first isolated in 1884. Since then, usnic acid and its sodium salt (sodium usnic acid) have been used in medicine, perfumery, cosmetics, and other industries due to its extensive biological activities. However, its acaricidal activity has not been studied. In this paper, we investigated the acaricidal activity of usnic acid and sodium usnic acid against Psoroptes cuniculi in vitro. After evaluating the acaricidal activity and toxicity of usnic acid and sodium usnic acid in vitro, the results showed that at doses of 250, 125, and 62.5 mg/ml, usnic acid and sodium usnic acid can kill mites with 91.67, 85.00, and 55.00% and 100, 100, and 60.00% mortality after treatment 24 h. The LT50 values were 4.208, 8.249, and 16.950 h and 3.712, 7.339, and 15.773 h for usnic acid and sodium usnic acid, respectively. Sodium usnic acid has a higher acaricidal activity than usnic acid, which may be related to the difference in their structures.

The rice water weevil, Lissorhoptrus oryzophilus Kuschel, has two generations in southern Zhejiang, China. To determine oogenesis in first-generation females (summer females) and its relations to temperature, females were collected from a rice field in early and mid-July and reared on young rice plants at 28, 31 and 34 degrees C in the laboratory. Percentage of females having oocytes, number of oocytes of different stages (stage-I, from early previtellogenesis to middle vitellogenesis; stage-II, late vitellogenesis; and mature-oocyte stage), and length of ovarioles were determined every 10 d of feeding. At each temperature, oogenesis took place in over 40% of females after 20~40 d of feeding, but only 0.0~3.3 stage-I, 0.0~0.8 stage-II and 0.0~1.1 mature oocytes were observed at each observation date. Temperature had significant effect on number of stage-I oocytes but not on number of stage-II and mature oocytes in early July females; temperature had no significant effect on number of oocytes of either stage in mid-July females. Conclusively, in southern Zhejiang, summer L. oryzophilus females have great potential to become reproductive on rice, but their oogenesis activity is very low, with the overall procedures little affected by temperature.
