RESUMO

River water quality is influenced by natural processes and human activities. Multi-scale landscape patterns can affect river water quality by altering the generation and transport processes of pollutants at different spatial scales. Taking Taizi River Basin in Northeast China as an example, we analyzed the relationship between landscape patterns and non-point source pollution in rivers based on water quality monitoring data and land use data by using correlation analysis and redundancy analysis methods. We aimed to determine the key spatial scales for the responses of landscape patterns to non-point source pollution and identify the key landscape indices influencing river non-point source pollution. The results showed that water quality of Taizi River Basin had seasonal differences, with better water quality during the flood season than non-flood season. Spatially, total nitrogen (TN) and total phosphorus (TP) were higher at the confluence points of tributaries and downstream areas. The impact of landscape patterns on non-point source pollution was stronger during the non-flood season than the flood season, while the influence on TN was stronger than on TP. At the spatial scale of within 500 m buffer zone during the flood season and at the sub-watershed scale during the non-flood season, landscape patterns showed the highest explanatory power for the variations of TN and TP. At the type level, built-up land, cropland, and bare land were positively correlated with TN and TP, while forest was negatively correlated with TN and TP, which were the key types influencing non-point source pollution. At the landscape level, patch density, percentage of like adjacencies, and contagion index were key indicators affecting watershed water quality. Lower patch density was associated with better connectivity and aggregation of "sink" landscapes, leading to better purification effects on TN, but more pronounced retention effects on TP. Conversely, higher landscape diversity and denser pattern of multiple types would cause the deterioration of water quality. Our results suggested that rational allocation of landscape types within the watershed and riparian buffer zones, appropriately enriching landscape diversity, and optimizing landscape aggregation and connectivity would be effective measures for improving water quality and achieving sustainable ecological management.

BACKGROUND: Sensitive skin is hypersensitive to various external stimuli and a defective epidermal permeability barrier is an important clinical feature of sensitive skin. Claudin-5 (CLDN5) expression levels decrease in sensitive skin. This study aimed to explore the impact of CLDN5 deficiency on the permeability barrier in sensitive skin and the regulatory role of miRNAs in CLDN5 expression. MATERIALS AND METHODS: A total of 26 patients were retrospectively enrolled, and the CLDN5 expression and permeability barrier dysfunction in vitro were assessed. Then miRNA-224-5p expression was also assessed in sensitive skin. RESULTS: Immunofluorescence and electron microscopy revealed reduced CLDN5 expression, increased miR-224-5p expression, and disrupted intercellular junctions in sensitive skin. CLDN5 knockdown was associated with lower transepithelial electrical resistance (TEER) and Lucifer yellow penetration in keratinocytes and organotypic skin models. The RNA-seq and qRT-PCR results indicated elevated miR-224-5p expression in sensitive skin; MiR-224-5p directly interacted with the 3`UTR of CLDN5, resulting in CLDN5 deficiency in the luciferase reporter assay. Finally, miR-224-5p reduced TEER in keratinocyte cultures. CONCLUSION: These results suggest that the miR-224-5p-induced reduction in CLDN5 expression leads to impaired permeability barrier function, and that miR-224-5p could be a potential therapeutic target for sensitive skin.

Based on data collected from 2054 saplings of Larix gmelinii forest in 55 fixed plots in 2018-2019 in Cuigang Forestry Station, Daxing'anling area, we classified the stand density index (SDI) into four classes, i.e., Class Ⅰ (SDI1<1863 plants·hm-2), Class Ⅱ (1863 plants·hm-2≤SDI2<2155 plants·hm-2), Class Ⅲ (2155 plants·hm-2≤SDI3<2459 plants·hm-2) and Class Ⅳ (SDI4≥2459 plants·hm-2) by using the quartile method. We constructed a dummy variable model and quantile regression model for the height-breast diameter of saplings of L. gmelinii with dummy variable method introduced SDI. The results showed that among the five selected representative non-linear tree height curve models, the Richards model fitted the best, with Ra2, RMSE and MAE of 0.7637, 0.8250 m and 0.5696 m. The dummy variable model including the SDI constructed based on the Richards model showed a 1.3% increase in Ra2 compared with the base model, while RMSE, MAE, and AIC decreased by 2.1%, 1.5%, and 11.2%, respectively. When the quantile τ was 0.5, Ra2 of quantile regression model was the maximum, and RMSE, MAE, AIC was the minimum, being 0.7612, 0.8294 m, 0.5657 m, and -767.19, respectively. Compared with SDI1, sapling height in SDI2-SDI4 was increased by 5.6%, 5.6%, and 11.3%, suggesting reasonable that regulation of stand density was conducive to increase the height growth of saplings in regeneration.

BACKGROUND: The true incidence of acute gastrointestinal illness in China is underrecognized by surveillance systems. The aims of this study were to estimate the incidence and prevalence of self-reported AGI in the community of China, and to investigate sociodemographic and epidemiological determinants of AGI. METHODS: We conducted a 12-months cross-sectional population-based survey in eight provinces of China during 2014-2015. The survey determined the prevalence and incidence of acute gastrointestinal illness (AGI) in the total permanent resident population in China according to the census of the population in 2010. The random multilevel population sample was stratified by geographic, population, and socioeconomic status. We used a recommended case definition of AGI, with diarrhea (three loose or watery stools) and/or any vomiting in a four-week recall. A face-to-face survey was conducted by selecting the member in the household with the most recent birthday. RESULTS: Among 56,704 sampled individuals, 948 (1,134 person-time) fulfilled the case definition; 98.5% reported diarrhea. This corresponds to 2.3% (95% CI:1.9%-2.8%) of an overall standardized four-week prevalence and 0.3 (95% CI: 0.23-0.34) episodes per person-year of annual adjusted incidence rate. There was no significant difference between males and females. The incidence rates were higher among urban residents, and in the spring and summer. In the whole study period, 50% of the cases sought medical care, of which 3.9% were hospitalized and 14.3% provided a biological sample for laboratory identification of the causative agent. Children aged 0-4 and young adults aged 15-24, people living in rural areas and people who traveled frequently had higher prevalence of AGI. CONCLUSION: Results showed that AGI represents a substantial burden in China, and will contribute to the estimation of the global burden of AGI. Complemented with data on the etiologies of AGI, these estimates will form the basis to estimate the burden of foodborne diseases in China.

OBJECTIVE: To explore the anti-inflammatory effects and mechanisms of action of thymol in Aspergillus fumigatus (A. fumigatus) keratitis. METHODS: The minimum inhibitory concentration of thymol against A. fumigatus was detected. To characterize the anti-inflammatory effects of thymol, mouse corneas and human corneal epithelial cells were pretreated with thymol or dimethyl sulfoxide (DMSO) before infection with A. fumigatus spores. Slit-lamp microscopy, immunohistochemistry, myeloperoxidase detection, quantitative real-time polymerase chain reaction, and Western blotting were used to assess infection. Neutrophil and macrophage recruitment, in addition to the secretion of LOX-1 and IL-1ß, were quantified to evaluate the relative contribution of thymol to the inflammatory response. RESULTS: We confirmed that the growth of A. fumigatus was directly inhibited by thymol. In contrast with the DMSO group, there was a lower degree of inflammation in the mouse corneas of the thymol-pretreated group. This was characterized by significantly lower clinical scores, less inflammatory cell infiltration, and lower expression of LOX-1 and IL-1ß. Similarly, in vitro experiments indicated that the production of LOX-1 and IL-1ß was significantly inhibited after thymol treatment, in contrast with the DMSO-pretreated group. CONCLUSION: Our findings demonstrate that thymol exerted a direct fungistatic activity on A. fumigatus. Furthermore, thymol played a protective role in fungal keratitis by inhibiting LOX-1/IL-1ß signaling pathway and reducing the recruitment of neutrophils and macrophages.

AIM: To investigate the expression of lacrimal androgen-binding proteins (ABPs) in mice Pseudomonas aeruginosa (P. aeruginosa) keratitis. METHODS: P. aeruginosa mice model from different gender was developed by intra-stromal injection. The expression of lacrimal ABPs in lacrimal gland specimens from P. aeruginosa keratitis mice was detected by the quantitative polymerase chain reaction (qRT-PCR). Corneal virulence was evaluated based on clinical scores. To study the mechanism of lacrimal ABPs' expression, experimental subjects were pre-treated with 4E-BP1 inhibitor, and were used to evaluate the expression levels by qRT-PCR. RESULTS: Compared with control groups, the expression of ABPα, ABPη and ABPζ in lacrimal gland from P. aeruginosa keratitis mice had no meaningful changes, while ABPε and ABPδ were significantly higher at 1d after infection. The expression of ABPδ in lacrimal gland of male mice was higher than female mice, regardless of whether or not P. aeruginosa keratitis occurred. After 4E-BP1 inhibitor subconjunctival injection or lacrimal injection, the expression of ABPδ and ABPε has no significant change compared with the control group. CONCLUSION: ABPδ and ABPε secreted by mice lacrimal gland may involve in the progress of alleviating the severity of corneal damage in P. aeruginosa keratitis. The expression of ABPδ and ABPε upon P. aeruginosa infection is independent of cap-dependent mRNA translation activated by 4E-BP1.
