RESUMO

Existing deep models for facade parsing often fail in classifying pixels in heavily occluded regions of facade images due to the difficulty in feature representation of these pixels. In this paper, we solve facade parsing with occlusions by progressive feature learning. To this end, we locate the regions contaminated by occlusions via Bayesian uncertainty evaluation on categorizing each pixel in these regions. Then, guided by the uncertainty, we propose an occlusion-immune facade parsing architecture in which we progressively re-express the features of pixels in each contaminated region from easy to hard. Specifically, the outside pixels, which have reliable context from visible areas, are re-expressed at early stages; the inner pixels are processed at late stages when their surroundings have been decontaminated at the earlier stages. In addition, at each stage, instead of using regular square convolution kernels, we design a context enhancement module (CEM) with directional strip kernels, which can aggregate structural context to re-express facade pixels. Extensive experiments on popular facade datasets demonstrate that the proposed method achieves state-of-the-art performance.

Long noncoding RNAs (lncRNAs) have drawn growing attention due to their regulatory roles in various diseases, including tumors. Recently, lncRNA FOXD3 antisense RNA 1 (FOXD3-AS1) was shown to be overexpressed in colon adenocarcinoma and glioma, exerting oncogenic functions. However, its expression and effects in cervical cancer (CC) remained unknown. In this research, our group first reported that the levels of FOXD3-AS1 were distinctly elevated in CC samples and cell lines. The distinct upregulation of FOXD3-AS1 was associated with lymphatic invasion, distant metastasis, and International Federation of Gynecology and Obstetrics stage, and also predicted poor clinical results of CC patients. Next, transcription factor SP1 was demonstrated to resulting in the upregulation of FOXD3-AS1 in CC. Functional assays indicated that knockdown of FOXD3-AS1 distinctly suppressed CC progression via affecting cell proliferation, cell apoptosis, and metastasis. Moreover, mechanistic studies suggested that FOXD3-AS1 acted as an endogenous sponge by directly binding miR-296-5p, resulting in the suppression of miR-296-5p. In addition, we also reported that high mobility group A, a direct target of miR-296-5p, could mediate the tumor-promotive effects that FOXD3-AS1 displayed. Overall, our present study might help to lead a better understanding of the pathogenesis of CC, provide a novel possible tumor biomarker, and probe the feasibility of lncRNA-directed treatments for CC.

OBJECTIVE: The aim of this study is to investigate the role of Sirtuin1 (Sirt1) in the regulation of autophagy for human osteoarthritis (OA) chondrocytes. DESIGN: All cartilage samples were collected from human donors, including young group, aged group, and OA group. Primary chondrocytes were isolated and cultured with Sirt1 activator or inhibitor. Sirt1 expression in cartilage tissue and chondrocytes was evaluated, and the deacetylation activity of Sirt1 was determined. The alteration of autophagy activity after upregulating or downregulating Sirt1 was detected. Chondrocytes were treated with autophagy activator and inhibitor, and then the protein level of Sirt1 was examined. The interactions between Sirt1 and autophagy-related proteins Atg7, microtubule associated protein 1 light chain 3 (LC3), and Beclin-1 were determined by using immunoprecipitation. RESULTS: The assay of articular cartilage revealed that the expression of Sirt1 might be age-related: highly expressed in of younger people, and respectively decreased in the elderly people and OA patients. In vitro study was also validated this result. Further study confirmed that higher levels of Sirt1 significantly increased autophagy in aged chondrocytes, while the lower expression of Sirt1 reduced autophagy in young chondrocytes. Of note, the high levels of Sirt1 reduced autophagy in OA chondrocytes. When the chondrocytes were treated with autophagy activator or inhibitor, we found the expression of Sirt1 was not affected. In addition, we found that Sirt1 could interact with Atg7. CONCLUSION: These results suggest that Sirt1 in human chondrocytes regulates autophagy by interacting with autophagy related Atg7, and Sirt1 may become a more important target in OA treatment.

KEY MESSAGE: The pollen and pistil polygalacturonases in Nicotiana tabacum were identified and found to regulate pollen tube growth and interspecific compatibility. Polygalacturonase (PG) is one of the enzymes catalyzing the hydrolysis of pectin. This process plays important roles in the pollen and pistil. In this research, the pollen and pistil PGs in Nicotiana tabacum (NtPGs) were identified, and their expression, localization and the potential function in the pollen and interspecific stigma incompatibility were explored. The results showed that 118 NtPGs were retrieved from the genome of N. tabacum. The phylogenetic tree and RT-qPCR analysis led to the identification of 10 pollen PGs; among them, two, seven and one showed specifically higher expression levels in the early development of anthers, during pollen maturation and in mature anthers, respectively, indicating their function difference. Immunofluorescence analysis showed that PGs were located in the cytoplasm of (1) mature pollen and (2) in vitro grown pollen tubes, as well as in the wall of in vivo grown pollen tubes. Four NtPGs in clade A were identified as the pistil PGs, and the pistil PGs were not found in clade E. Significantly higher PGs expression was recorded after incompatible pollination in comparison with the compatible stigma, indicating a potential function of PGs in regulating stigma incompatibility. The influence of PGs on pollen tube growth was explored in vitro and partly in vivo, showing that high PGs activity inhibited pollen tube growth. The application of PGs on the otherwise compatible stigma resulted in pollen tube growth inhibition or failure of germination. These results further supported that increased PGs expression in incompatible stigma might be partially responsible for the interspecific stigma incompatibility in Nicotiana.

The preservation of seed viability and quality in storage is an important trait both for commercial and germplasm usage. To better explore potential mechanisms of tobacco seed deterioration, seed packed in cloth bag (C) and vacuum bag (V) were stored under room temperature (RT) and low temperature (LT, 18°C), and sampled periodically for laboratory testing. Seed stored in low temperature with vacuum bag (LT/V) owned the highest seed vigour after 25 months of storage and in room temperature with cloth bag (RT/C) lost seed vigour and germination ability after 20-month storage. Meanwhile, seed in RT/C notably increased about 5-fold endogenous hydrogen peroxide (H2O2), 4-fold malondialdehyde (MDA) contents, 12-fold Lipoxygenases (LOX) activity and 2-fold the expression of NtLOX3 comparing with LT/V at the end of 15-month storage. In addition, regression analysis indicated that LOX activity was strongly negatively correlated with seed vigour as the R 2 value reached 0.970 in RT/C. Furthermore, caffeic acid and catechin, the inhibitors of LOX activity, were applied to tobacco seeds pre-treatment and followed with artificial accelerated aging. Seeds pretreated with inhibitors, especially caffeic acid, reduced LOX activity by 50%, MDA and H2O2 contents by 40% and 20%, respectively, and increased more than 1.2-fold seed vigour and seedling quality comparing with seeds pretreated with H2O after 6-day artificial aging, indicating a better seed storability after artificial accelerated aging. These results suggest that LOX accelerated seed aging, and suppression of LOX activity enhanced seed vigour and viability in accelerated aging tobacco seed, opening new opportunities for effective management of seed germplasm under long-term storage and conservation.

Heavy occlusions in cluttered scenes impose significant challenges to many computer vision applications. Recent light field imaging systems provide new see-through capabilities through synthetic aperture imaging (SAI) to overcome the occlusion problem. Existing synthetic aperture imaging methods, however, emulate focusing at a specific depth layer, but are incapable of producing an all-in-focus see-through image. Alternative in-painting algorithms can generate visually-plausible results, but cannot guarantee the correctness of the results. In this paper, we present a novel depth-free all-in-focus SAI technique based on light field visibility analysis. Specifically, we partition the scene into multiple visibility layers to directly deal with layer-wise occlusion and apply an optimization framework to propagate the visibility information between multiple layers. On each layer, visibility and optimal focus depth estimation is formulated as a multiple-label energy minimization problem. The layer-wise energy integrates all of the visibility masks from its previous layers, multi-view intensity consistency and depth smoothness constraint together. We compare our method with state-of-the-art solutions, and extensive experimental results demonstrate the effectiveness and superiority of our approach.

Near infrared spectroscopy (NIRS) technology developed fast in recent years, due to its rapid speed, less pollution, high-efficiency and other advantages. It has been widely used in many fields such as food, chemical industry, pharmacy, agriculture and so on. The seed is the most basic and important agricultural capital goods, and seed quality is important for agricultural production. Most methods presently used for seed quality detecting were destructive, slow and needed pretreatment, therefore, developing one kind of method that is simple and rapid has great significance for seed quality testing. This article reviewed the application and trends of NIRS technology in testing of seed constituents, vigor, disease and insect pests etc. For moisture, starch, protein, fatty acid and carotene content, the model identification rates were high as their relative contents were high; for trace organic, the identification rates were low as their relative content were low. The heat-damaged seeds with low vigor were discriminated by NIRS, the seeds stored for different time could also been identified. The discrimination of frost-damaged seeds was impossible. The NIRS could be used to identify health and infected disease seeds, and did the classification for the health degree; it could identify parts of the fungal pathogens. The NIRS could identify worm-eaten and health seeds, and further distinguished the insect species, however the identification effects for small larval and low injury level of insect pests was not good enough. Finally, in present paper existing problems and development trends for NIRS in seed quality detection was discussed, especially the single seed detecting technology which was characteristic of the seed industry, the standardization of its spectral acquisition accessories will greatly improve its applicability.

With the wide development of UAV (Unmanned Aerial Vehicle) technology, moving target detection for aerial video has become a popular research topic in the computer field. Most of the existing methods are under the registration-detection framework and can only deal with simple background scenes. They tend to go wrong in the complex multi background scenarios, such as viaducts, buildings and trees. In this paper, we break through the single background constraint and perceive the complex scene accurately by automatic estimation of multiple background models. First, we segment the scene into several color blocks and estimate the dense optical flow. Then, we calculate an affine transformation model for each block with large area and merge the consistent models. Finally, we calculate subordinate degree to multi-background models pixel to pixel for all small area blocks. Moving objects are segmented by means of energy optimization method solved via Graph Cuts. The extensive experimental results on public aerial videos show that, due to multi background models estimation, analyzing each pixel's subordinate relationship to multi models by energy minimization, our method can effectively remove buildings, trees and other false alarms and detect moving objects correctly.

Analgesic treatment with anti­inflammatory drugs may aid the prevention of postoperative pain and the attenuation of the postoperative immune inflammatory response. The current study presents a randomized, double­blind controlled study, which was performed to investigate the levels of Th1, Th2, Th17 and Treg cytokines, including interleukin (IL)­2, interferon (IFN)­Î³, IL­4, IL­10, IL­17, IL­23 and transforming growth factor (TGF)­ß in the peripheral blood of patients with cervical cancer following laparoscopy. The effects of perioperative multi­dose parecoxib on postoperative immune function was evaluated. A total of 80 patients with cervical cancer (stage IB/IIA, ASA I­III, aged 18­65 years) that were scheduled for laparoscopy were randomly assigned into either the parecoxib (I; n=40) or control (II; n=40) groups. Group I received 40 mg parecoxib 30 min prior to surgery and then every 12 h subsequent to surgery for 60 h, and group II received normal saline at the corresponding time points. Intravenous tramadol (100 mg) was prescribed for pain relief as required. The mRNA and protein expression levels of cytokines in the peripheral blood were detected by quantitative polymerase chain reaction and ELISA. Pain visual analog scales (VAS) and incidence, analgesic relief, adverse events and the length of hospital stay were recorded. It was demonstrated that the mRNA and protein levels of IL­2, IFN­Î³ and IL­17 in the two groups were reduced subsequent to surgery, while mRNA and protein expression levels of IL­4, IL­10 and TGF­ß were enhanced. Administration of multi­dose parecoxib may diminish the increase in postoperative IL­2, IFN­Î³ and IL­17 levels, and suppress the excessive production of IL­4, IL­10 and TGF­ß. This effect is accompanied by lower VAS scores, pain incidence, postoperative nausea/vomiting and infections. In conclusion, perioperative multi­dose parecoxib was able to alleviate postoperative pain and ameliorate surgery­induced immune suppression by balancing Th1, Th2, Th17 and Treg cytokines following laparoscopy in patients with cervical cancer. The current study provides support to the hypothesis that parecoxib may be a more effective therapeutic strategy than the currently available options, for postoperative pain and immune function management of patients with cancer.

Drought is one of the most important stress factors limiting the seed industry and crop production. Present study was undertaken to create novel drought-resistant pelleted seeds using the combined materials with superabsorbent polymer, poly(2-acrylamide-2-methyl propane sulfonic acid) (PAMPS) hydrogel, and drought resistance agent, salicylic acid (SA). The optimized PAMPS hydrogel was obtained as the molar ratio of 2-acrylamido-2-methyl-propanesulfonic acid (AMPS) to potassium peroxydisulfate (KPS) and N, N'-methylene-bis-acrylamide (MBA) was 1 : 0.00046 : 0.00134. The hydrogel weight after swelling in deionized water for 24 h reached 4306 times its own dry weight. The water retention ratio (RR) of PAMPS was significantly higher as compared with the control. It could keep as high as 85.3% of original weight after 30 min at 110 °C; even at 25 °C for 40 d, the PAMPS still kept RR at 33.67%. PAMPS disintegration ratio increased gradually and reached around 30% after embedding in soil or activated sludge for 60 d. In addition, there were better seed germination performance and seedling growth in the pelleted treatments with SA-loaded PAMPS hydrogel under drought stress than control. It suggested that SA-loaded PAMPS hydrogel, a nontoxic superabsorbent polymer, could be used as an effective drought resistance material applied to tobacco pelleted seeds.

OBJECTIVE: To explore the effects of Yiweining Recipe (YWNR), a compound Chinese herbal medicine, on expressions of metalloproteinase-2 (MMP-2) and cyclooxygenase-2 (COX-2) mRNAs in rats with endometriosis (EM). METHODS: Operational self-transplantation was applied in establishing the rat models. Detection of MMP-2 and COX-2 mRNAs was conducted with hybridization in situ. RESULTS: There were significant differences in the expressions of MMP-2 and COX-2 mRNAs between the untreated group and the high-dose YWNR-treated group. YWNR could reduce the expressions of MMP-2 and COX-2 mRNAs. CONCLUSION: YWNR can treat EM through reducing the positive expressions of MMP-2 and COX-2 mRNAs.

OBJECTIVE: To observe the clinical efficacy and safety of Yiweining (YWN) and gestrinone (GT) in treating post-operational patients of stage III endometriosis (EM-III). METHODS: Fifty-two patients of EM-III after operation were randomly assigned into three groups, the YWN group (20 patients) was treated through oral intake of YWN 200 ml, twice a day; the GT group (19 patients) treated with gestrinone 2.5 mg, twice every week, with the medication starting from the 7th post-operational day and lasting for 6 months. The control group (13 patients) was untreated. Six months was one therapeutic course, and follow-up study was carried out in the 6 - 30 months after the end of the medication. RESULTS: The recurrence rate in the YWN group and the GT group were 5.0% and 5.3% respectively, showing insignificant difference between the two groups, but they were lower than that in the control group (30.7%, P < 0.05). Besides, the adverse reaction rate in the YWN group was lower than that in the GT group (10.0% vs 31.6%, P < 0.05). CONCLUSION: Application of YWN to prevent the post-operational recurrence of endometriosis is effective and safe, and its efficacy is similar to that of GT.
