RESUMO

A Mimosa pudica var. unijuga-associated toxicity affecting horses occurred in Araguari, Triângulo Mineiro, Southeast Brazil. Affected horses had gradual hair loss of the mane and tail and endocrine dermatosis after grazing for three months during the dry season on a paddock invaded by the plant. The main histological lesions include compact ortho-keratotic hyperkeratosis and numerous flame follicles. Toxicological analysis by HPLC-UV demonstrated 0.8 mg/g of mimosine in the leaves.

RESUMO

We reported thirteen cases of bilateral stringhalt associated with Hypochaeris radicata that occurred in horses in Uruguay during a severe drought in the summer of 2023. All horses were affected chronically and progressively by bilateral hyperflexion of hindlimbs. In two severely affected horses, the main histological lesions included neuronal chromatolysis and axonal spheroids in the ventral gray horn in the lumbar and sacral spinal cord and axonal degeneration and digestion chambers in ventral roots fibers and long peripheral nerves. We suggest that in addition to injuries to peripheral nerves, lesions in the spinal cord play an important role in the clinical signs of stringhalt in horses.

In the summer of 2023, ingestion of Astylus atromaculatus (pollen beetle) was linked to spontaneous fatal disease in grazing cattle and sheep in Argentina and Uruguay. While the disease was experimentally reproduced in sheep and guinea pigs in the 1970's, no experimental reproductions have been attempted in cattle, and controversy exists as to whether this insect is indeed noxious to cattle and at which dose. Here, we demonstrate that A. atromaculatus causes acute fatal disease in Hereford calves at single oral dosages of 2.5, 4.5, 10.0, and 15.0 g of insect/kg body weight. Death or severe disease necessitating euthanasia occurred at 38 to 48 hours postinoculation regardless of the dose, suggesting that the single fatal dosage is likely <2.5 g/kg body weight (this dose representing approximately 850 mL of intact beetles in a 100 kg calf). Clinically, the disease was characterized by acute anorexia, prolonged recumbency, reluctance to move, listlessness/apathy, depression, ruminal hypomotility and tympany, hypothermia, bruxism with frothing at the mouth, and mucoid diarrhea progressing to death. Hematologic and biochemical alterations included hemoconcentration, stress/acute inflammatory leukogram, negative energy balance, and ketosis. The pathological hallmark of this experimental disease is acute necrotizing omaso-reticulo-rumenitis, fibrinohemorrhagic enteritis, and exfoliative colitis with intralesional chitinous insect fragments. While A. atromaculatus might contain a gastrointestinal toxin or pathogen, extensive toxicological testing failed to identify a causative toxin. Other pathomechanisms such as direct physical damage caused by insect fragments on the alimentary tract seem plausible, although further studies are needed to elucidate the pathogenesis of A. atromaculatus-associated disease.

RESUMO

Leucaena leucocephala poisoning is reported in horses in different Brazilian regions. The poisoning occurred one month after the horses were introduced into paddocks invaded by the plant or after 10 days of consuming cut Leucaena administered as the only food. Affected horses showed moderate to severe hair loss on the mane and tail, orthokeratotic hyperkeratosis with marked follicular telogenization, and hyperplasia of thyroid follicular cells. Mimosin concentration in leaves (5.5 mg/g) was determined by a new HLPC-UV method which is also reported.

ABSTRACT Introduction: One of the most critical complications in myelodysplastic syndromes (MDS) is the progression to acute myeloid leukemia (AML). The dynamics of clonal evolution in MDS and how acquired mutations can be used as biomarkers to track disease progression remains under investigation. Objective and method: Herein, we investigated the frequency of common myeloid clonal mutations (FLT3, NPM1, JAK2, IDH1 and IDH2) in 88 patients with MDS and 35 AML patients with myelodysplasia-related changes, followed at a single reference center in northeastern Brazil. Results: Overall, 9/88 (10%) ofthe MDSpatients and 9/35 (26%) of the secondary AML patients had at least one mutation. While the JAK2 V617F mutation was the most frequent in the MDS patients, the FLT3, NPM1, IDH1 and IDH2 mutations were more frequently found in the secondary AML group. Furthermore, there was a higher frequency of FLT3, NPM1, IDH1 and IDH2 mutations in MDS patients classified as high-risk subtypes than in those of lower risk. Conclusion: Despite the limited sample size, our data suggest that mutations in FLT3, NPM1, IDH1 and IDH2 genes could be potential biomarkers to detect early disease progression in MDS.

This study analyzed the effect of thermal stress on erythrocytes of Notothenia rossii and Notothenia coriiceps, abundant notothenioids in Admiralty Bay, Antarctic Peninsula. In both species, the antioxidant defense system enzymes, superoxide dismutase, catalase, glutathione peroxidase, glutathione-S transferase, glutathione reductase were punctually altered (8°C for 1, 3 and 6 days) in erythrocytes, indicating that these markers are not ideal for termal stress. However, under the influence of thermal stress, morphological changes in Notothenia coriiceps erythrocytes were observed at all exposure times (1, 3 and 6 days at 8°C), and in Notothenia rossii occurred in 6 days. These results suggest that Notothenia corriceps presents a lower tolerance to thermal stress at 8°C for up to 6 days, since the cellular and nuclear alterations recorded are pathological and may be deleterious to the cells. Among the morphological markers analyzed in this work, we believe that the shape change and nuclear bubble formation may be good stress biomarkers in erythrocytes of Notothenia rossii and Notothenia coriiceps.

Introdução: de acordo com a legislação brasileira, um dentifrício não pode conter mais que 1.500 ppm de flúor total (FT), e o fabricante deve declarar na embalagem a concentração e o tipo de sal de flúor que o produto contém. No entanto, para que possa exercer um efeito anticárie, o produto deve conter pelo menos 1.000 ppm F (mgF/kg) na forma quimicamente solúvel. Objetivo: avaliar as concentrações de flúor total (FT) e flúor solúvel total (FST) em dentifrícios distribuídos pelo serviço público de saúde da cidade de Salvador, Bahia. Materiais e métodos: três amostras de um dentifrício (MFP/CaCO3, 1.500 ppm F como FT) que estava sendo distribuído pelo serviço público de saúde de Salvador, BA, foram obtidas. Como controle, foi utilizado um dentifrício com a mesma formulação (1.450 ppm de FT) encontrado no comércio da cidade de Piracicaba, SP. As análises foram realizadas com eletrodo específico para fluoreto, empregando metodologia já validada e os resultados expressos em ppm F (mg F/kg). Resultados: em todas as amostras avaliadas, foram encontrados apenas 61,8 ppm F de FT, evidenciando que não foram fluoretadas, fato este nunca antes relatado no Brasil. De outro modo, no dentifrício utilizado como controle, havia 1404,7 ppm F de FT, do qual 1.270,0 ppm estavam solúveis (FST). Conclusão: os cremes dentais avaliados nesta pesquisa não são capazes de exercer um efeito anticárie, uma vez que não foram fluoretados. Esses dados mostram a urgência de modificação da regulamentação brasileira vigente e a necessidade de maior controle de qualidade desses produtos.(AU)


Introduction: according to Brazilian legislation, a toothpaste cannot contain more than 1500 ppm of total fluoride (TF), as well as the manufacturer must provide on the packaging the concentration and type of fluoride salt that the product contains. However, to have an anti-caries effect, it must contain at least 1,000 ppm F (mgF/kg) in its chemically soluble form. Research Aim: To evaluate the total fluoride (TF) and total soluble fluoride (TSF) concentrations in dentifrices distributed by the public health service in the city of Salvador, Bahia. Materials and methods: for the analyses, three samples of a dentifrice (MFP/CaCO3, 1,500 ppm F as TF) that was being distributed by Salvador's public health system were obtained. As a control, it was used a dentifrice of the same formulation (1,450 ppm of TF) found in commerce of the city of Piracicaba- -SP. The analyses were performed with a specific fluoride electrode using a validated methodology and the results were expressed in ppm F (mg F/ kg). Results: In all the samples evaluated, only 61.8 ppm F of TF were found, showing that it was not fluoridated, a fact that was never reported in Brazil before. On the other hand, in the dentifrice used as a control, there were 1404.7 ppm F of TF, of which 1270.0 ppm were soluble (TSF). Conclusion: the samples evaluated in this research are not able to exert an anti-caries effect, since they were not fluoridated. These data show the urgency to change the current Brazilian regulation and the need for a greater quality control of these products.(AU)

Landfill is a public and environmental health problem; establishing and understanding methodologies to decrease its toxicity are thus necessary. Leachate samples were collected, at a sanitary landfill, immediately after the exit from the landfill, i.e. raw leachate (collection point A), after conventional treatment (point B) and after treatment by wetlands (point C). D. parodizi specimens were exposed to 3%, 10% and control (0%) dilutions of leachate from these collection points for 7 days. Markers of antioxidant defences and cell damage were analysed. At point B, the gills of D. parodizi showed higher glutathione-S-transferase (GST) and glutathione reductase (GR) activity; the latter is a supplier of glutathione reductase (GSH). The low GST activity at point A was associated with the hormesis effect. Higher levels of superoxide dismutase (SOD), ethoxyresorufin-O-deethylase (EROD) and glutathione peroxidase (GPx) occurred at point A. Glucose-6-phosphate dehydrogenase (G6PDH) was inhibited at the points with the highest pollutant load and at the highest leachate dilutions. Higher levels of markers at point A may be related to the high pollutant charge and specific compounds present in the untreated leachate. The multi-xenobiotic resistance mechanism (MXR), metallothionein-like proteins (MT) and lipid peroxidation (LPO) did not vary among treatments. The biomarker responses showed negative effects of the leachate on the freshwater bivalve and simultaneously showed that the wetland treatment employed at the Caximba sanitary landfill is effective.

Resumo Introdução: a reabilitação funcional (RF) se apresenta como um elo entre os cuidados de saúde e as demandas do contexto laboral e envolve, necessariamente, intervenções no contexto de trabalho. Objetivo: apresentar a implantação de ações de RF em uma instituição federal de ensino superior (Ifes) da região Sudeste. Métodos: são descritos o processo de definição teórico-metodológico, a problematização dos princípios versados na legislação vigente, os critérios e procedimentos aplicados na RF, as possibilidades e os limites do contexto institucional. São analisados os resultados do programa no primeiro ano de implantação da RF, por meio de avaliação de casos de trabalhadores reinseridos. Resultados: as ações de RF foram oferecidas para nove casos-piloto direcionados para a equipe e distribuídas entre seus membros. No primeiro ano do programa, 2016, após a implantação de ações de RF, foi observada redução de 37% no percentual médio do número de dias de afastamento do trabalho, comparada aos afastamentos do ano anterior. Conclusão: é importante elaborar diretrizes políticas que melhor definam o escopo conceitual e epistemológico da RF, refletidas na legislação, e que apresentem possibilidades teórico-metodológicas para viabilizar a implantação de serviços de RF nas Ifes.


Abstract Introduction: functional rehabilitation (FR) is a link between healthcare and work demands, and necessarily leads to intervention in the working context. Objective: to present the implementation of FR measures in a federal university from the Southeast region of Brazil. Methods: we described the FR theoretical-methodological definition, the principles included in the current legislation, the criteria and procedures applied to FR as well as its institutional possibilities and limitations. We evaluated the results of the first year of FR implementation, 2016, by analyzing cases of returning to work. Results: FR measures were provided to nine pilot cases. After the first year, there was a reduction of 37% in sick leave, compared to the previous year. Conclusion: it is important to formulate policies and legislation that better define the FR conceptual/epistemological scope and present theoretical/methodological framework for enabling the implementation of FR services in federal universities.

The thermal stability of the Antarctic Ocean raises questions concerning the metabolic plasticity of Antarctic notothenioids to changes in the environmental temperature. In this study, Notothenia rossii survived 90 days at 8 °C, and their condition factor level was maintained. However, their hepatosomatic (0.29×) index decreased, indicating a decrease in nutrient storage as a result of changes in the energy demands to support survival. At 8 °C, the plasma calcium, magnesium, cholesterol, and triglyceride concentrations decreased, whereas the glucose (1.91×) and albumin (1.26×) concentrations increased. The main energy substrate of the fish changed from lipids to glucose due to a marked increase in lactate dehydrogenase activity, as demonstrated by an increase in anaerobic metabolism. Moreover, malate dehydrogenase activity increased in all tissues, suggesting that fish acclimated at 8 °C exhibit enhanced gluconeogenesis. The aerobic demand increased only in the liver due to an increase (2.23×) in citrate synthase activity. Decreases in the activities of superoxide dismutase, catalase, and glutathione-S-transferase to levels that are most likely sufficient at 8 °C were observed, establishing a new physiological activity range for antioxidant defense. Our findings indicate that N. rossii has some compensatory mechanisms that enabled its long-term survival at 8 °C.

Antarctic Nototheniidae is an attractive group for studying metabolic and physiological responses at high temperatures. The present work investigated the metabolic responses of the carbohydrate metabolism and antioxidant system to thermal stress at 8 °C (for 2-144 h) in the brains of Notothenia rossii and Notothenia coriiceps. In N. coriiceps, glycogenolysis was essential in the first hours of exposure (2 h) at 8 °C and, in addition to inhibiting glucose-6-phosphatase activity, was important for activating the pentose phosphate pathway. In N. rossii, anaerobic metabolism was reduced in the first hours of exposure (2 and 6 h) at 8 °C, followed by reduced hexokinase activity, suggesting energy regulation between neurons and astrocytes. The antioxidant system results indicated the importance of the actions of the glutathione-dependent antioxidant enzymes glutathione-S-transferase and glutathione peroxidase as well as those of catalase in N. coriiceps and the action of glutathione-S-transferase, glutathione peroxidase and glutathione reductase in N. rossii, especially during the first 12 h of thermal stress exposure. These results indicate tissue-specific patterns and species-specific responses to this stress.

Carbohydrate metabolism and the antioxidant defence system of heart and muscle of the Antarctic notothenioids Notothenia rossii and Notothenia coriiceps were evaluated in response to heat stress (8 °C) over 144 h. N. rossii heart exhibited decreased glycolysis and aerobic metabolism after up to 12 h of exposure to 8 °C, and anaerobiosis was inhibited within 24 h. However, these pathways were stimulated after 72 h at 8 °C. The consumption of glucose-6-phosphate, derived from hexokinase (HK), by glucose-6-phosphate dehydrogenase (G6PDH) decreased in N. rossii heart within 6 h at 8 °C, with a subsequent increase at 72 h. In N. rossii muscle at 8 °C, glycolysis was stimulated within 2 h by an increase in pyruvate kinase (PK), and aerobic metabolism was stimulated at 144 h, together with anaerobiosis. In N. coriiceps heart at 8 °C, glucose break down by HK decreased within 2 h and subsequently increased at 12 and 24 h. Increased glucose-6-phosphate consumption by G6PDH occurred within 6 h at 8 °C. In N. coriiceps muscle at 8 °C, glycolysis was stimulated at 2 and 6 h, with subsequent inhibition within 24 h, as indicated by HK activity. Aerobic metabolism was inhibited at 72 and 144 h at 8 °C through the inhibition of citrate synthase (CS). Heat stress caused responses were only occasional and transient in antioxidant defence system of both species in the heart and muscle, leading to increased glutathione (GSH) and decreased levels of lipoperoxidation in the heart of both species. The results obtained in this study in the heart and muscles indicate that under heat stress at 8 °C, N. rossii is more responsive than N. coriiceps with respect to carbohydrate metabolism.

The objective of the present study was to assess the effect of short-term (2-144 h) heat stress (8 °C) on energy production processes and antioxidant defense systems in the kidneys and gills of Notothenia rossii and Notothenia coriiceps. Heat stress affected energy metabolism and oxidative stress parameters in a time-, tissue-, and species-dependent manner, and gills were more sensitive than kidneys to heat stress. N. rossii kidneys were able to stabilize carbohydrate metabolism after 12 h of heat stress, whereas the glycogen levels in N. coriiceps kidneys fluctuated in response to varying glucose-6-phosphatase (G6Pase) levels. The gills of N. rossii were able to stabilize their energy demand and aerobic metabolism under heat stress, whereas in the gills of N. coriiceps, changes in carbohydrate metabolic pathways depended on the exposure time: initially, anaerobiosis was activated after 6 h; the energy demand, characterized by glycogen consumption, increased after 72 h, and aerobic metabolism was activated within 144 h. With regard to the antioxidant defenses of the N. rossii kidney, it was found that levels of antioxidant enzymes were reduced during the first hours of heat stress, contributing to increased lipid peroxidation, whereas N. coriiceps kidneys did not show signs of oxidative damage. The gills of N. rossii exhibited more pronounced oxidative damage in response to heat stress than those of N. coriiceps despite the presence of increasing levels of antioxidants, likely due to tissue hypoxia.

Embora incomum, a impacção de incisivos centrais superiores é geralmente diagnosticada precocemente, devido à importância destes dentes na função mastigatória e na aparência do indivíduo. O objetivo deste trabalho foi relatar dois casos clínicos nos quais uma abordagem cirúrgica e ortodôntica foi realizada para tratar incisivos centrais impactados. Em ambos os casos, foi realizada a expansão rápida da maxila com o auxílio de um aparelho disjuntor de Haas. Da mesma forma, foi efetuada a exposição cirúrgica e o tracionamento ortodôntico do incisivo central superior impactado. Forças ortodônticas leves foram utilizadas durante todo o tracionamento ortodôntico, as quais foram aplicadas em arcos rígidos. Although uncommon, the impaction of maxillary central incisors is usually early diagnosed, due to the importance of these teeth in masticatory function and appearance of the individual. The aim of this paper was to report two clinical cases, in which surgical and orthodontic approach were performed, in order to treat impacted central incisors. In both cases, we have done the rapid disjunction of the midpalatal suture, as well as exposure and orthodontic traction of the impacted maxillary central incisor. Light forces were used during the all the orthodontic traction, which were applied on rigid archs. It was noted that the teeth presented adequate clinical crown height, satisfactory gingival contours and pulp vitality. It can be concluded that a well-planned surgical exposure of the impacted teeth in association with orthodontic traction contributed to periodontal and aesthetic optimal treatment outcomes...

Objective: To establish cultures of cells from the pulp of permanent teeth by the explant method assessing parameters usually presented by stem cells, such as the expression of certain markers and the differentiation ability into osteogenic, adipogenic, and chondrogenic lineages. This study also aimed to assess the expression of ALDH1 (aldehyde dehydrogenase 1) enzyme activity on the isolated cells. Materials and method: The pulp tissue, obtained from wisdom teeth, was placed in a 6-well plate containing proper culture medium, and stored at 37 °C and 5% CO2 for cell proliferation and plastic adherence. Cells were tested for the expression of surface markers and for ALDH1 enzyme activity, by flow cytometry. In addition, cells were assessedfor multi-differentiation potential. Results: The isolated cells showed high expression of CD44 (98.8%), CD73 (100%), and CD90 (97.2%), and moderate expression of STRO-1 (18.4%) and ALDH1 (16.2%), by flow cytometry. Similarly, the cells showed differentiation ability into all three lineages of cells tested. Conclusion: Our results suggest that the explant method - or cell proliferation method - is suitable for the isolation and cultureof stem cells from dental pulp of permanent teeth. The isolated cells may be considered stem cells, based on the current criteria for their characterization, such as plastic adherence, expression of certain markers, and the absence of others, as well as multi-differentiation potential, which showed to be promising for the application in tissue regeneration.
