RESUMO

Lipid peroxidation and mineral metabolism were studied in patients with III degree of obesity and bad standing of sodium restricted diet before and after treatment with including in diet of monosodium glutamate (MSG). It was established well being of MSG and absence of negative effect on loss of body mass in course of dietary treatment. MSG caused normalization of level of diene conjugates and some minerals in serum of patients.

RESUMO

The levels of diene conjugates (DC) and TBA-reactive substances (TBA-RS) were studied in serum blood of patients with gastritis before and after including in diet of monosodium glutamate (MSG). For investigation 15 patients were selected who had unacid or subacid signs determined by histamine test. It was shown that levels of DC and TBA-RS were increased in serum of patients. MSG increased the gastric secretion in patients but had no effect of levels of DC and TBA-RS in serum.

RESUMO

The effect of fat composition with a different ratio of polyunsaturated fatty acids of families omega 3 and omega 6 on expression of experimental dietary anaphylaxis, liver monooxygenase system and metabolism of 17-oxycorticosteroides (17-OCS) in guinea-pig have been investigated. Fat components in experimental diets were presented by sunflower, ichthyene oils and mix of these oils in the ratio 1:1. A ratio of PUFA of a family omega 6 to omega 3 in diets were 64,0; 0,13 and 2,64. It was shown that a burden of dietary anaphylaxis were reduced significantly at the introduction in diets of sunflower oil.ulakcuu is authentically reduced. Simultaneously a marked increase of a ratios P-450B to P-450L and b5 to P-450 and reduction of a hexenal sleep duration were determined. The amount of excreted polar 17-OCS was also decreased in animals fed sunflower oil. Probable mechanisms of hypoallergenic action of dietary sunflower oil were conditioned by reorganization of liver cytochrome P-450 system and respective alterations of 17-OCS metabolism are discussed.

RESUMO

Adult guinea pigs were fed for 10-14 days with synthetic diets, fat constituting 11% of its total energy. Dietary fat was composed of coconut, corn, dairy and soybean oils mixtures with ratio of polyunsaturated fatty acids (PUFA) omega-6 to PUFA omega-3 equal to 24.2 (K1) or 5.53 (K2). The animals were sensitized orally by pasteurized cow milk (PCM) or epicutaneously by formaldehyde (F) during these diets feeding. The degree of the sensitization was assessed in the reaction of active anaphylactic shock (AAS) in PCM-sensitized animals and in the reaction of leukocytes specific lysis (LSL) in F-sensitized guinea pigs. In the latter pigs the concentration of serum antibodies (Ab) against dietary soya protein was measured by ELISA. Animals fed by K1 and K2 were also tested for histamine mean lethal dose resistance. The lowest lethality in AAS, number of convulsions, of positive LSL cases and Ab level were found in animals fed by K1 compared to both K2 and to animals fed by common animal chew. Resistance to histamine was similar in K1 and K2 groups, but was significantly higher compared to control (chew) group. In convulsion, the changes in PUFA omega-6/PUFA omega-3 ratio have marked effect on different indices of allergic sensitivity.

Noninbred male guinea-pigs (b. mass 250-300 g) were kept for 35 days on diets in which 50% of standard proteins were replaced for cow milk protein (CMP) 20/80 and its enzymatic hydrolyzate (EH) 20/80 obtained by ultrafiltration. CMP 20/80 and EH 20/80 have been examined for the effect on proteolytic activity of gastric mucosa, small intestinal activity of trypsin and chymotrypsin, hepatic cytochrome P-450 and metabolism of excreted 17-OCS. The animals given EH 20/80 exhibited enhanced total proteolytic activity of trypsin and chymotrypsin, higher total levels of cytochromes P-450 and P-450B in the liver, increased urine excretion of 17-OCS, reduced amount of polar forms in all the glucocorticoid fractions. Potential mechanisms securing the action of enzymatic hydrolyzates on the systems responsible for nonspecific resistance to food allergens are suggested for discussion.

Noninbred guinea-pig experiments were performed to investigate specific potential allergenicity of cow milk proteins (CMP) consisting of casein (80%) and serum proteins (20%), of low-molecular peptides (LMP 20/80) produced from CMP 20/80 by enzymatic hydrolysis and ultrafiltration; to evaluate nonspecific effect of test diet CMP 20/80 and LMP 20/80 on anaphylactic response of the animals to ovalbumin and histamine LD50. LMP 20/80 versus CMP 20/80 had a hypoallergenic effect. Potential mechanisms of low anaphylactic sensitivity induced by LMP 20/80 are discussed.

Two groups of noninbred male guinea pigs (initial bw 250-300 g) received test rations in which 50% of standard ration protein were substituted for milk protein (P 20/80) and its enzymatic hydrolysate (H 20/80). The influence of P 20/80 and H 20/80 on some parameters of the gastro-intestinal tract (GIT) barrier function and 17-OCS metabolism was studied. The action of H 20/80 as compared to P 20/80 was expressed in the intensified excretion of free fraction of 17-OCS and glucuronides. The content of hydroxylated glucocorticoids increased in both fractions. At the same time elevated total proteolytic activity in the gastric mucosa and a tendency to decreased GIT permeability for chicken ovalbumin macromolecules were observed in animals that received H 20/80. It has been suggested that enzymatic hydrolysates of food proteins influence endocrine regulation of metabolic processes.

The complex evaluation was carried out of potential allergenicity of three soya protein specimens - IPS, IPS-1 and IPS-2 manufactured by Ardex R, USA, which were attended for employment as protein component of specialized infant formulae. The content of total protein, amino-nitrogen, peptide fractions of molecular weight below 9000 D, precipitating protein antigens, trypsin inhibitor, was studied in the specimens. The apparent digestibility was measured by a multienzymatic system in vitro. The allergenicity of isolates was compared by means of histamine LD50 resistance test in guinea pigs and also by the reaction of active anaphylactic shock (AAS). The evidences have been obtained that IPS specimen containing the lowest levels of trypsin inhibitor and small peptide levels is characterized by the lowest sensitizing activity in AAS and simultaneously decreases the animals' resistance to histamine in the highest degree. Apparent digestibility of all the three specimens did not differ significantly. A conclusion has been made that the evaluation is necessary of infant hypoallergenic formulae components by a complex of independent experimental data.

Male guinea pigs, intact and orally sensitized with chick ovalbumin (OVA), having initial bw 250-300 g, received glycine intraperitoneally or orally in a single dose of 100 or 200 mg/kg/day, during 3 days. The specific volume of cytochromes P450 and b5 in the microsomal fraction of the liver, activity of aminopyrine N-demethylase and aniline-p-hydroxylase, severity of food anaphylaxis to OVA and the animals' sensitivity to intravenous histamine LD50 were studied. The inducing effect of glycine on the cytochrome P450 system in the liver was more pronounced in the presensitized animals, that was attended by a decreased severity of food anaphylaxis and by a protective action in respect to histamine LD50. Possible mechanisms of glycine action are considered regarding its participation in metabolic processes.

In experiments on mice, the effect of cyclic analogue of enkephalin (CAE) on the processes of learning and memory was studied in control animals and in animals with changed functional state of monoaminergic brain systems. Administration of the peptide to intact animals significantly accelerated the acquisition of conditioned reflexes of two-way avoidance and did not significantly affect the retention of these reflexes and their subsequent reproduction. Retention and reproduction of conditioned reflexes elaborated in one combination, was disturbed. Administration of iprazid did not eliminate the "accelerating effect" of CAE on the formation of conditioned reflexes of the two-way avoidance but sharply disturbed their retention. In such conditions, the amnesing iprazid effect increased still more. Besides, under CAE effect, the activity of acetylcholinesterase in the motor and especially in the visual cortex of the mice increased. The obtained data testify to an important role of the monoaminergic and cholinergic brain mechanisms in realization of CAE effects on the processes of learning and memory.
