RESUMO

Anti-Trypanosoma cruzi activity of compounds from fruits of Schinus terebinthifolius Raddi (pink pepper) were evaluated, using sustainable techniques such as steam distillation (SD) and supercritical fluid extraction (SFE). SD was optimised using a design of experiment and SFE was carried out using supercritical CO2 solvent (300 bar and 60 °C). Results of the anti-T. cruzi activity showed that the essential oil presented high activity (IC50 = 4.5 ± 0.3 µg/mL), whereas the supercritical extract had a moderate effect (IC50 = 19.7 ± 2.9 µg/mL). The differences in the anti-T. cruzi activity can be attributed to the extraction of non-volatile compounds in the SFE, such as moronic and (Z)-masticadienoic acids. In contrast, SD extracted only volatile compounds such as monoterpenes and sesquiterpenes. Therefore, these results suggest that the volatile compounds from pink pepper are involved with the anti-T. cruzi activity.

Optical sensors can be used to detect a variety of substances ranging from diagnostics on biological samples to the detection of hazardous substances. This type of sensor can be a valuable alternative to more complex analytical techniques, being fast and requiring little to no sample preparation at the expense of the reusability of the device. Here, we show the construction of a colorimetric nanoantenna sensor using gold nanoparticles (AuNPs) embedded in poly(vinyl alcohol) (PVA) and decorated with the methyl orange (MO) azo dye (AuNP@PVA@MO) that is potentially reusable. As a proof of concept, we apply this sensor to detect H2O2 both visually and using a smartphone-based app for colorimetric measurements. Furthermore, through chemometric modeling of the app data, we can reach a detection limit of 0.0058% (1.70 mmolL-1) of H2O2 while being able to visually detect changes on the sensor. Our results reinforce the combination of nanoantenna sensors with chemometric tools as guidelines for sensor design. Finally, this approach can lead to novel sensors allowing for the visual detection of analytes in complex samples and their quantification using colorimetry.

Intracellular modifications caused by two metal-based antitumor compounds were assessed by confocal Raman imaging assisted by multivariate curve resolution method, a very powerful deconvolution tool that can be used to extract the characteristic spectral profile of the individual or "purest" components from an image dataset. The use of this Raman methodology has the advantage of being non-invasive and totally label-free. Four main different intracellular processes were observed under the Raman imaging and multivariate approach combination, and even, significant differences could be identified between the treatments with both metallodrugs. Leakage of the nucleus and nucleolus content into the cytoplasm, along with releasing of cytochrome c were observed for the treatment with the Cu-based complex. At the same time, changes of hydrogen-bonding network were also evidenced, indicating an apoptotic cellular death process, consistent with complementary Total Reflection X-Ray fluorescence (TXRF) and fluorescence experiments attesting mitochondria and DNA as main targets after uptake of the complex by cells. For treatment with the Zn-based complex, changes associated with cytochrome c were not detected, neither a rapid leakage of nucleus content upon 24 h treatment. The hydrogen-bonding network also followed a quite different pattern, suggesting that with this metallodrug, the cellular death follows a different mechanism.
