OBJECTIVES: Alzheimer's disease (AD) is considered the most common cause of dementia in older people. Recently, blood-based markers (BBM) Aß1-42, Aß1-40, and phospho Tau181 (p-Tau181) have demonstrated the potential to transform the diagnosis and prognostic assessment of AD. Our aim was to investigate the effect of different storage conditions on the quantification of these BBM and to evaluate the interchangeability of plasma and serum samples. METHODS: Forty-two individuals with some degree of cognitive impairment were studied. Thirty further patients were retrospectively selected. Aß1-42, Aß1-40, and p-Tau181 were quantified using the LUMIPULSE-G600II automated platform. To assess interchangeability between conditions, correction factors for magnitudes that showed strong correlations were calculated, followed by classification consistency studies. RESULTS: Storing samples at 4 °C for 8-9 days was associated with a decrease in Aß fractions but not when stored for 1-2 days. Using the ratio partially attenuated the pre-analytical effects. For p-Tau181, samples stored at 4 °C presented lower concentrations, whereas frozen samples presented higher ones. Concerning classification consistency in comparisons that revealed strong correlations (p-Tau181), the percentage of total agreement was greater than 90 % in a large number of the tested cut-offs values. CONCLUSIONS: Our findings provide relevant information for the standardization of sample collection and storage in the analysis of AD BBM in an automated platform. This knowledge is crucial to ensure their introduction into clinical settings.

The extraordinary occurrence of COVID-19 by the fast expansion of viral infections has propelled particular interest in developing novel antiviral and virucidal agents to guarantee personal security. The main objective of this work is to propose novel formulations able to optimize the use of personal protection elements. In recent years, chitosan (CH) has attracted attention for being an interesting multifunctional, biodegradable, non-antigenic, non-toxic, and biocompatible natural polymer with antimicrobial properties. In this work, formulations based on a CH matrix containing silver, and Copper based nanoparticles have been developed. The novelty of this proposal is that almost liquid formulations have been reached, possessing verified properties to inhibit evolved virus such as herpes simplex type 1 (HSV-1) and bovine betacoronavirus (BCoV), the latter belonging to the same family of the well-known the well-known SARS-CoV-2. Besides antibacterial bioactivity; as well as the ability of these formulations to be easily sprayed on various surfaces, including conventional face masks, have been verified and discussed. The results presented in this contribution provide strong evidence on CH films as an ideal biosafe surface-protective for several daily used materials including the conventional face masks.

Advances in tissue engineering require the development of new biomaterials with adequate properties of cell attachment and growth. The properties of biomaterials can be improved by incorporation of bioactive molecules to enhance in vitro and/or in vivo functions. In this work, we study the role of a wheat germin-like protease inhibitor (GLPI), free or immobilized in biocompatible matrices to improve cell-attachment ability on different mammalian cell lines. The phylogenetic relationships and functional diversity of the GLPI were analyzed among diverse genera to get insights into sequence motif conservations. The cytocompatibility effect of free GLPI on C2C12 premyoblastic cells and B16 cells as tumoral model has been tested. GLPI promoted proliferation and metabolic activity of both cell types on in vitro models, not showing cytotoxic effects. Furthermore, GLPI was immobilized in chitin microparticles and in chitosan films; we demonstrated an accelerated cell adhesion process in both biomaterials.

Agrobiotechnology challenges involve the generation of new sustainable bioactives with emerging properties as plant biostimulants with reduced environment impact. We analyzed the potential use of recently developed chitosan microparticles (CS-MP) as growth promoters of tomato which constitutes one of the most consumed vegetable crops worldwide. Treatments of tomato seeds with CS-MP improved germination and vigor index. In addition, CS-MP sustained application triggered an improvement in root and shoot biomass reinforcing tomato performance before transplanting. The level of reactive oxygen species (ROS), antioxidant enzyme activities and defense protein markers were modulated by CS-MP treatment in tomato plantlets. Analyses of ARR5:GUS and DR5:GUS transgenic reporter tomato lines highlighted the participation of cytokinin and auxin signaling pathways during tomato root promotion mediated by CS-MP. Our findings claim a high commercial potential of CS-MP to be incorporated as a sustainable input for tomato production.

Clay-based nanocomposites (nanoclays) are interesting systems to hold a wide type of active substances with a wide field of industrial applications. Bentonite-chitosan nanoclay was obtained via cationic exchange of natural bentonite (Bent) with an aqueous solution of chitosan (CS). Their physicochemical and morphological properties were discussed under the light of Fourier transform infrared spectroscopy, X-ray diffraction, thermogravimetric analysis, and scanning electron microscopy. Bent-CS characterization indicated that CS was intercalated in 10% (w/w). This polycationic polymer was oriented mostly in a monolayer arrangement, interacting by electrostatic forces between Bent sheets. The antimicrobial action of Bent-CS nanoclay was assayed onto phytopathogens, the bacterium model Pseudomonas syringe pv. tomato DC3000 ( Psy) and the necrotrophic fungus Fusarium solani f. sp. eumartii ( F. eumartii). In addition to demonstrating cell death on both microorganisms, Bent-CS exerted elicitor property on tomato plantlets. The biological actions of this natural nanomaterial might make it proper to be used in crops.

RESUMEN El objetivo del estudio fue conocer el estado nutricional y la duración habitual de sueño en días de semana y fin de semana de escolares entre 6 a 15 años de la ciudad de Punta Arenas. Sujetos y métodos: Se obtuvo una muestra aleatoria simple de 481 escolares de 2 establecimientos educacionales (uno particular-subvencionado y uno municipalizado). Para diagnosticar el estado nutricional, se evaluó peso, talla y pliegue cutáneo tricipital. Para conocer la duración habitual del sueño se aplicó la versión española del Pediatric Sleep Questionnaire y los resultados se compararon con la recomendación entregada por la National Sleep Foundation. Resultados: El 60,4% de los escolares presentó malnutrición por exceso, durmiendo en promedio 8,70±1,07 horas los días de semana y 9,85±1,55 horas los fines de semana. Conclusión: Los escolares diagnosticados con obesidad fueron los que presentan un menor cumplimiento de la recomendación, sin embargo, no hubo diferencia significativa entre ambas variables.


ABSTRACT The aim of the study was to study the nutritional status and the average number of sleep hours on weekdays and weekend among 6 to 15-year-old schoolchildren in the city of Punta Arenas, Chile. Subjects and methods: A simple random sample of 481 schoolchildren from two schools (one state-subsidized and one public) was obtained. To evaluate nutritional status, we measured weight, height and triceps skinfold. In order to determine usual sleep duration, the Spanish version of the Pediatric Sleep Questionnaire was used and the results were compared with the recommendation given by the National Sleep Foundation. Results: 60.4% of schoolchildren showed malnutrition due to excess, slept on average 8.70±1.07 hours on weekdays and 9.85±1.55 hours on weekends. Conclusion: Obese children had less compliance with recommendations, however differences were not statistically significant.

BACKGROUND: The genus Fusarium comprises a heterogeneous group of fungi important for agriculture. Fusarium solani f. sp. eumartii (F. eumartii), historically considered to be a fungal pathogen of potato, has also been associated with tomato disease. Currently, chitosan and its derivatives have been receiving more attention as environmentally friendly antimicrobial compounds in sustainable practices. The aim of the present work was to characterize downstream events associated with the mode of action of chitosan, including nitrosative reactive species, in order to identify new biomarkers of its cytotoxic action. RESULTS: Data indicated that chitosan-mediated nitric oxide (NO) production might lead to conidial death, concomitant with the strong reduction in fungal pathogenicity in tomato plants. Following chitosan applications, a notably dose-dependent reduction in conidial viability was demonstrated in F. eumartii. Thereafter, the infectivity of chitosan-treated spores was tested by a bioassay using tomato seedlings. CONCLUSION: All these data highlight NO valuable properties as a quantitative and qualitative biomarker of cytotoxic action of chitosan in conidial cells. In addition, these findings place the chitosan assayed here as a fungicide with a high potential of application in sustainable horticultural practices.
