Soil salinization has emerged as a major factor with an adverse influence on agricultural green development worldwide. It is necessary to develop high-efficiency and ecologically beneficial management measures for alleviating soil salinization. The experiment of application for cow manure (CM), biochar (BC), and bio-organic fertilizer (BIO) in soil with light salinity was conducted to investigate the remediation of organic materials on soil salinization with melon (Cucumis melo L.) by reducing the availability of saline ions and shifting the soil microbial community. Results showed that BC treatment significantly decreased the EC values of the soil and soil solution by 19.23% and 27.02% and the concentrations of Na+, K+, and Cl- by 13.28%, 13.08%, and 15.21%, respectively, followed by CM and BIO treatments. High-throughput sequencing identified that organic amendments significantly improved the richness of the soil bacterial community and increased the relative abundances of Acidobacteria and Firmicutes by 33.11% and 111.2%, respectively, and the beneficial salt-tolerant bacterial genera Flavobacterium, Bacillus and Arthrobacter by 32.04%, 38.92% and 35.67%, respectively. Additionally, soil Na+, Ca2+, K+ and Cl- were significantly negatively correlated with Acidobacteria and Flavobacterium and were also the most important factors driving the changes in the structure of the soil bacterial communities. The bacterial networks were more complex in the organic amendments treatments than in CK, reflecting through more nodes and links and a higher average clustering coefficient, density and modularity. This study provided a comprehensive understanding of the application of organic amendments in alleviating soil salinization and improving soil bacterial and fungal communities and provides scientific support for agriculture green development.

Bacillus megaterium is well known as a plant growth-promoting rhizobacterium, but the relevant molecular mechanisms remain unclear. This study aimed to elucidate the effects of B. megaterium HT517 on the growth and development of and the control of disease in greenhouse tomato and its mechanism of action. A pot experiment was conducted to determine the effect of B. megaterium on tomato growth, and this experiment included the HT517 group (3.2 × 108 cfu/pot) and the control group (inoculated with the same amount of sterilized suspension). An antagonistic experiment and a plate confrontation experiment were conducted to study the antagonistic effect of B. megaterium and Fusarium oxysporum f.sp. lycopersici. Liquid chromatography-mass spectrometry was used to determine the metabolite composition and metabolic pathway of HT517. PacBio+Illumina HiSeq sequencing was utilized for map sequencing of the samples. An in-depth analysis of the functional genes related to the secretion of these substances by functional bacteria was conducted. HT517 could secrete organic acids that solubilize phosphorus, promote root growth, secrete auxin, which that promotes early flowering and fruiting, and alkaloids, which control disease, and reduce the incidence of crown rot by 51.0%. The complete genome sequence indicated that the strain comprised one circular chromosome with a length of 5,510,339 bp (including four plasmids in the genome), and the GC content accounted for 37.95%. Seven genes (pyk, aceB, pyc, ackA, gltA, buk, and aroK) related to phosphate solubilization, five genes (trpA, trpB, trpS, TDO2, and idi) related to growth promotion, eight genes (hpaB, pheS, pheT, ileS, pepA, iucD, paaG, and kamA) related to disease control, and one gene cluster of synthetic surfactin were identified in this research. The identification of molecular biological mechanisms for extracellular secretion by the HT517 strain clarified that its organic acids solubilized phosphorus, that auxin promoted growth, and that alkaloids controlled tomato diseases.

Soil salinization is a global problem that limits agricultural development and impacts human life. This study aimed to understand the dynamic changes in water and salinity in saline-alkali soil based on an indoor soil column simulation. We studied the changes in the water and salt contents of soils with different degrees of salinization under various irrigation conditions. The results showed that after seven irrigations, the pH, conductivity and total soluble salt content of the percolation samples after irrigation generally increased initially then decreased with repeated irrigation. The soil moisture did not change significantly after irrigation. The pH, conductivity, and total soluble salt content of each layer of the soil profile exhibited general declining trends. In the soil profile from Changguo Township (CG), the pH decreased from 8.21-8.35 to 7.71-7.88, the conductivity decreased from 0.95-1.14 ms/cm to 0.45-0.68 ms/cm, and the total soluble salt content decreased from 2.63-2.81 g/kg to 2.28-2.51 g/kg. In the soil profile from Zhongjie Industrial Park (ZJ), the pH decreased from 8.36-8.54 to 7.73-7.96, the conductivity decreased from 1.58-1.68 ms/cm to 1.45-1.54 ms/cm, and the total soluble salt decreased from 2.81-4.03 g/kg to 2.56-3.28 g/kg. The transported salt ions were primarily K+, Na+ and Cl-. After several irrigations, a representative desalination effect was achieved. The results of this study can provide technical guidance for the comprehensive management of saline-alkali soils.
