Growing concerns about environmental pollution have highlighted the need for efficient and sustainable methods to remove dye contamination from various ecosystems. In this context, computational methods such as molecular dynamics (MD), Monte Carlo (MC) simulations, quantum mechanics (QM) calculations, and machine learning (ML) methods are powerful tools used to study and predict the adsorption processes of dyes on various adsorbents. These methods provide detailed insights into the molecular interactions and mechanisms involved, which can be crucial for designing efficient adsorption systems. MD simulations, detailing molecular arrangements, predict dyes' adsorption behaviour and interaction energies with adsorbents. They simulate the entire adsorption process, including surface diffusion, solvent layer penetration, and physisorption. QM calculations, especially density functional theory (DFT), determine molecular structures and reactivity descriptors, aiding in understanding adsorption mechanisms. They identify stable adsorption configurations and interactions like hydrogen bonding and electrostatic forces. MC simulations predict equilibrium properties and adsorption energies by sampling molecular configurations. ML methods have proven highly effective in predicting and optimizing dye adsorption processes. These models offer significant advantages over traditional methods, including higher accuracy and the ability to handle complex datasets. These methods optimize adsorption conditions, clarify adsorbent functionalization roles, and predict dye removal efficiency under various conditions. This research explores MD, MC, QM, and ML approaches to connect molecular interactions with macroscopic adsorption phenomena. Probing these techniques provides insights into the dynamics and energetics of dye pollutants on adsorption surfaces. The findings will aid in developing and optimizing new materials for dye removal. This review has significant implications for environmental remediation, offering a comprehensive understanding of adsorption at various scales. Merging microscopic data with macroscopic observations enhances knowledge of dye pollutant adsorption, laying the groundwork for efficient, sustainable removal technologies. Addressing the growing challenges of ecosystem protection, this study contributes to a cleaner, more sustainable future.

Annelid biodiversity studies in the Red Sea are limited and integrative taxonomy is needed to accurately improve reference libraries in the region. As part of the bioblitz effort in Saudi Arabia to assess the invertebrate biodiversity in the northern Red Sea and Gulf of Aqaba, Perinereis specimens from intertidal marine and lagoon-like rocky environments were selected for an independent assessment, given the known taxonomic ambiguities in this genus. This study used an integrative approach, combining molecular with morphological and geographic data. Our results demonstrate that specimens found mainly in the Gulf of Aqaba are not only morphologically different from other five similar Perinereis Group I species reported in the region, but phylogenetic analysis using available COI sequences from GenBank revealed different molecular operational taxonomic units, suggesting an undescribed species, P.kaustianasp. nov. The new species is genetically close and shares a similar paragnath pattern to the Indo-Pacific distributed P.helleri, in particular in Area III and Areas VII-VIII. Therefore, we suggest it may belong to the same species complex. However, P.kaustianasp. nov. differs from the latter mainly in the shorter length of the postero-dorsal tentacular cirri, median parapodia with much longer dorsal Tentacular cirri, posteriormost parapodia with much wider and greatly expanded dorsal ligules. Additionally, two new records are reported for the Saudi Neom area belonging to P.damietta and P.suezensis, previously described only for the Egyptian coast (Suez Canal) and are distributed sympatrically with the new species, but apparently not sympatric with each other.

The advent of polymer-based dielectrics marked a significant breakthrough in dielectric materials. However, despite their many advantages, they pose serious environmental threats. Therefore, in recent years, there has been growing interest in bio-based polymers as a sustainable alternative to traditional petroleum-based polymers. Their renewable nature and reduced environmental impact can fulfil the rising demand for eco-friendly substitutes. Beyond their ecological benefits, bio-based polymers also possess distinctive electrical properties that make them extremely attractive in a variety of applications. Considering these, herein, we present recent advancements in bio-based dielectric polymers and nanocomposites. First, the fundamental concepts of dielectric and polymer-based dielectric materials are covered. Then, we will delve into the discussion of recent advancements in the dielectric properties and thermal stability of bio-based polymers, including polylactic acid, polyhydroxyalkanoates, polybutylene succinate, starch, cellulose, chitosan, chitins, and alginates, and their nanocomposites. Other novel bio-based dielectric polymers and their distinct dielectric characteristics have also been pointed out. In an additional section, the piezoelectric properties of these polymers and their recent biomedical applications have been highlighted and discussed thoroughly. In conclusion, this paper thoroughly discusses the recent advances in bio-based dielectric polymers and their potential to revolutionize the biomedical industry while cultivating a more sustainable and greener future.

The contamination of wastewater with antibiotics has emerged as a critical global challenge, with profound implications for environmental integrity and human well-being. Adsorption techniques have been meticulously investigated and developed to mitigate and alleviate their effects. In this study, we have investigated the adsorption behaviour of Erythromycin (ERY), Gentamicin (GEN), Levofloxacin (LEVO), and Metronidazole (MET) antibiotics as pharmaceutical contaminants (PHCs) on amide-functionalized (RC (=O)NH2)/MIL-53 (Al) (AMD/ML53A), using molecular simulations and density functional theory (DFT) calculations. Based on our DFT calculations, it becomes apparent that the adsorption tendencies of antibiotics are predominantly governed by the presence of AMD functional groups on the adsorbent surface. Specifically, hydrogen bonding (HB) and van der Waals (vdW) interactions between antibiotics and AMD groups serve as the primary mechanisms facilitating adsorption. Furthermore, we have observed that the adsorption behaviors of these antibiotics are influenced by their respective functional groups, molecular shapes, and sizes. Our molecular simulations delved into how the AMD/ML53A surfaces interact with antibiotics as PHCs. Moreover, various chemical quantum descriptors based on Frontier Molecular Orbitals (FMO) were explored to elucidate the extent of AMD/ML53A adsorption and to assess potential alterations in their electronic properties throughout the adsorption process. Monte Carlo simulation showed that ERY molecules adsorb stronger to the adsorbent in acidic and basic conditions than other contaminants, with high energies: -404.47 kcal/mol in acidic and -6375.26 kcal/mol in basic environments. Molecular dynamics (MD) simulations revealed parallel orientation for the ERY molecule's adsorption on AMD/ML53A with 80% rejection rate. In conclusion, our study highlighted the importance of modeling in developing practical solutions for removing antibiotics as PHCs from wastewater. The insights gained from our calculations can facilitate the design of more effective adsorption materials, ultimately leading to a more hygienic and sustainable ecosystem.

This work introduces an easy method for producing Bi2O3, ZnO, ZnO-Bi2O3 nanoparticles (NPs) by Biebersteinia Multifida extract. Our products have been characterized through the outcomes which recorded with using powder X-ray diffractometry (PXRD), Raman, energy dispersive X-ray (EDX), field emission-scanning electron microscopy (FE-SEM), and Fourier-transform infrared (FT-IR) techniques. The finding of SEM presented porous structure and spherical morphology for Bi2O3 and ZnO NPs, respectively. While FE-SEM image of bimetallic nanoparticles showed both porous and spherical morphologies for them; so that spherical particles of ZnO have sat on the porous structure of Bi2O3 NPs. According to the PXRD results, the crystallite sizes of Bi2O3, ZnO and ZnO-Bi2O3 NPs have been obtained 57.69, 21.93, and 43.42 nm, respectively. Antibacterial performance of NPs has been studied on Staphylococcus epidermidis and Pseudomonas aeruginosa bacteria, to distinguish the minimum microbial inhibitory concentration (MIC). Antimicrobial outcomes have showed a better effect for ZnO-Bi2O3 NPs. Besides, wondering about the cytotoxic action against cancer cell lines, the MTT results have verified the intense cytotoxic function versus breast cancer cells (MCF-7). According to these observations, obtained products can prosper medical and biological applications.

Dental decay is known in the world as the most common human infectious disease. Ascending process of dental caries index in the world shows the failure of oral disease prevention. Streptococcus mutans bacteria cause acid damage and tooth decay by producing acid over time. Nanomaterials with suitable functionality, high permeability, extremely large surface area, significant reactivity, unique mechanical features, and non-bacterial resistance can be considered as promising agents for antimicrobial and antiviral applications. In this study, nickel oxide (NiO) nanoparticles with size range from 2 to 16 nm containing Stevia natural sweetener were eco-friendly synthesized via a simple method. Additionally, their various concentrations were evaluated on S. mutans bacteria by applying the broth dilution method. The results demonstrated that these spherical NiO nanoparticles had efficient bacteriostatic activity on this gram-positive coccus.

Follicle-stimulating hormone (FSH) contributes to the acquisition of oocyte competence by modulating signalling pathways in cumulus cells (CCs), albeit much less is known about transcription factors (TFs) that orchestrate the downstream transcriptional changes. This work allowed to prospect TFs involved in FSH-mediated signalling during oocyte in vitro maturation (IVM). Bovine cumulus-oocyte complexes underwent IVM with FSH (FSH+) or without FSH (control/CTL) for 22 h, and CCs were subjected to gene expression profiling. Five software identified reference genes for RT-qPCR (ATP1A1, UBB, and YWHAZ). The transcript levels of FSH-responsive genes HAS2 and PTGS2 (COX2) validated the experimental design. Among candidate TFs, MYC was down-regulated (0.35-fold; P < 0.0001), and THAP11 (RONIN) was up-regulated (1.47-fold; P = 0.016) under FSH+ conditions. In silico analyses predicted binding motifs at MYC and THAP11 genes for previously known FSH-responsive TFs. Signalling pathways (EGFR, ERK, GSK3, PKA, and P38) may execute post-translational regulation due to potential phosphorylation sites in MYC and THAP11 proteins. Prediction of protein-protein interaction networks showed MYC as a core component of FSH signalling, albeit THAP11 acts independently. Hence, MYC integrates FSH signalling networks and may assist in exploring genome-wide transcriptional changes associated with the acquisition of oocyte competence.

By allowing for the creation of embryo banks, reproductive biotechnologies play an essential role in the preservation of endangered goat breeds' genetic diversity. This study focused on comparing both available embryo collection methods [laparotomy (LAP) vs. nonsurgical embryo recovery (NSER)] in Canindé goats to create an embryo bank for later use in a breed conservation program. Twelve females were superovulated and subjected to either the LAP or NSER technique for embryo recovery. The recovery rate was similar (p > 0.05) between NSER (86.8% ± 5.6%) and LAP (92.8% ± 4.0%). Moreover, there were no differences (p > 0.05) in the number of structures recovered, the viable embryos, and the freezable embryos per goat, respectively, for NSER (11.7 ± 1.3, 11.2 ± 1.5, and 10.2 ± 1.1) and LAP (10.3 ± 1.0, 8.7 ± 0.7, and 8.0 ± 0.8). Overall, 132 structures were collected out of 151 ovulations (∼12.6 ± 1.2 corpora lutea per goat). Finally, the procedure duration time was also similar (p > 0.05) for NSER versus LAP, respectively: 32.3 ± 3.3 versus 30.8 ± 3.9 minutes. In conclusion, the NSER method results proved to be similar to the LAP technique in small-sized Canindé goats. It was noticeable, however, that the NSER technique is simpler and provides the possibility for successive procedures with few health risks and sequels for females. This study may hopefully boost in vivo embryo production programs in the Canindé breed, facilitating the formation of embryo banks and so assuring the availability of genetic diversity before any decline becomes irreversible.

BACKGROUND AND AIM: Oocyte in vitro maturation (IVM) is an appealing approach for several assisted reproductive technologies and dissecting oocyte maturation. Nonetheless, IVM leads to lower developmental competence and usually relies on undefined, serum-containing media. Therefore, biochemical profiling aimed to explore fluctuations in IVM media content during the acquisition of oocyte developmental competence. MATERIALS AND METHODS: Bovine cumulus-oocyte complexes (COCs) underwent IVM in TCM199 medium with Earle's salts, supplemented with 2.0 mM L-glutamine, 10% fetal bovine serum, antibiotics, and 0.05 IU/mL porcine follicle-stimulating hormone (FSH+) or vehicle control (CTL) medium for 22 h. RESULTS: FSH withdrawal (CTL) diminished several processes associated with the acquisition of oocyte developmental competence, such as reduced cumulus cell expansion, diminished estradiol synthesis (FSH+: 116.0±0.0 pg/mL vs. CTL: 97.6±18.0 pg/mL), and lower oocyte nuclear maturation rate (FSH+: 96.47% vs. CTL: 88.76%). Fresh media formulations (i.e., TCM199 with FSH or vehicle) were indistinguishable under biochemical profiling threshold conditions. Biochemical profiling showed similar total protein and lipid concentrations between groups. Further, total sugar concentrations diminished from fresh media to their post-IVM counterparts, albeit in an FSH-independent manner. Glycogen concentrations remained unaltered after IVM within CTL media, albeit were substantially lower after IVM under FSH+ conditions. CONCLUSION: FSH mediates the consumption of serum-derived glycogen by bovine COCs during IVM and implies that serum-free media should contain increased glucose concentrations to facilitate the acquisition of oocyte developmental competence.

The polychaete genus Laeonereis (Annelida, Nereididae) occurs over a broad geographic range and extends nearly across the entire Atlantic coast of America, from the USA to Uruguay. Despite the research efforts to clarify its diversity and systematics, mostly by morphological and ecological evidence, there is still uncertainty, mainly concerning the species Laeonereis culveri, which constitutes an old and notorious case of taxonomic ambiguity. Here, we revised the molecular diversity and distribution of Laeonereis species based on a multi-locus approach, including DNA sequence analyses of partial segments of the cytochrome c oxidase subunit I (COI), 16S rRNA, and 28S rRNA genes. We examined Laeonereis specimens collected from 26 sites along the American Atlantic coast from Massachusetts (USA) to Mar del Plata (Argentina). Although no comprehensive morphological examination was performed between different populations, the COI barcodes revealed seven highly divergent MOTUs, with a mean K2P genetic distance of 16.9% (from 6.8% to 21.9%), which was confirmed through four clustering algorithms. All MOTUs were geographically segregated, except for MOTUs 6 and 7 from southeastern Brazil, which presented partially overlapping ranges between Rio de Janeiro and São Paulo coast. Sequence data obtained from 16S rRNA and 28S rRNA markers supported the same MOTU delimitation and geographic segregation as those of COI, providing further evidence for the existence of seven deeply divergent lineages within the genus. The extent of genetic divergence between MOTUs observed in our study fits comfortably within the range reported for species of polychaetes, including Nereididae, thus providing a strong indication that they might constitute separate species. These results may therefore pave the way for integrative taxonomic studies, aiming to clarify the taxonomic status of the Laeonereis MOTUs herein reported.

Certain species of parasitic flies belonging to the Phoridae are known to attack Atta spp. workers foraging along trails, near nest openings used by the ants to supply the colony with plant material, and in the areas where the ants are actively cutting plant material. However, there have been no previous studies of phorid parasitism of non-foraging worker ants, for example excavators and soldiers. Excavators can be found on the surface around specialized nest openings, carrying and dumping soil on characteristic mounds. Soldiers can be found on the trails protecting foragers or guarding the different types of nest openings. The current study was performed to investigate the differential parasitism rates of Atta laevigata (Smith, 1858) worker castes by four species of phorids. Ants of all castes on trails and at nest entrances were collect from 18 mature colonies in the field. A total of 21,254 ants were collected from trails and 14,649 collected from the mounds of loose soil near nest openings. The captured workers were maintained under controlled laboratory conditions to evaluate the rate of parasitism. Of the ants collected from trails, 1,112 (5.23%) were found to have been parasitized, of which 1,102 were foragers and only 10 were soldiers. Of the ants collected from the soil mounds near the nest openings, only 27 (0.18%) were found to have been parasitized, of those 25 were excavators and 2 were soldiers. When evaluating parasitism of ants on the trails, 46.2% were attacked by Apocephalus attophilus Borgmeier, 1928, 22.6% by Myrmosicarius grandicornis Borgmeier, 1928, 16.6% by Eibesfeldtphora erthali (Brown, 2001) and 14.6% by Apocephalus vicosae Disney, 2000. Only two species of phorid, M. grandicornis and E. erthali, were observed parasitizing excavators, whilst only E. erthali parasitized soldiers. This is the first time that Atta spp. excavators and soldiers have been shown to be parasitized by phorids. The low rates of parasitism and specificity of certain phorid species for excavators and soldiers is discussed in relation to the behavioral interactions of hosts and their parasitoids, as well as the relationship between host and parasitoid size.

The extent to which mammalian cells share similar transcriptomes remains unclear. Notwithstanding, such cross-species gene expression inquiries have been scarce for defined cell types and most lack the dissection of gene regulatory landscapes. Therefore, the work was aimed to determine C-MYC relative expression across mammalian fibroblasts (Ovis aries and Bos taurus) via cross-species RT-qPCR and comprehensively explore its regulatory landscape by in silico tools. The prediction of transcription factor binding sites in C-MYC and its 2.5 kb upstream sequence revealed substantial variation, thus indicating evolutionary-driven re-wiring of cis-regulatory elements. C-MYC and its downstream target TBX3 were up-regulated in Bos taurus fibroblasts. The relative expression of C-MYC regulators [RONIN (also known as THAP11), RXRß, and TCF3] and the C-MYC-associated transcript elongation factor CDK9 did not differ between species. Additional in silico analyses suggested Bos taurus-specific C-MYC exonization, alternative splicing, and binding sites for non-coding RNAs. C-MYC protein orthologs were highly conserved, while variation was in the transactivation domain and the leucine zipper motif. Altogether, mammalian fibroblasts display evolutionary-driven C-MYC relative expression that should be instructive for understanding cellular physiology, cellular reprogramming, and C-MYC-related diseases.

Abstract: Polychaetes are common in coastal and estuarine environments worldwide and constitute one of the most complex groups of marine invertebrates. The morpho-physiology of the female reproductive system (FRS) can be understood by using histological tools to describe reproductive cycle and gametogenesis paths and, among other purposes, aiming to identify and differentiate polychaete species. However, this histology-based approach is rarely combined with molecular tools, which is known to accurately delimitate species. In the same way, the description and understanding of oogenesis and vitellogenesis paths within polychaetes are lacking for most families, narrowing the range of its utility. Therefore, the present study aims to describe the oogenesis in three polychaete species common and abundant on the South American Atlantic coast (Laeonereis culveri, Scolelepis goodbodyi and Capitella biota) and investigate the utility of reproductive features and gametogenesis as a relevant associate knowledge to discriminate species, particularly useful for putative cryptic species, integrated with morphological and molecular data. In a first attempt, the results obtained herein allow the authors to describe two new subtypes of oogenesis, dividing it in extraovarian oogenesis type I and II and intraovarian type I and II. The results also demonstrate that the following histological characters of the FRS can be relevant for the separation of related species: a) oogenesis type, b) occurrence or absence of a true ovary, c) ovary tissue organization, d) type of accessory cells present, and e) oocyte morphology. Additionally, these histological features of FRS, when compared with correlated species studied under this scope, converge with the genetic data. The analysis of cytochrome oxidase I (COI) barcode sequences differentiates between North and South American Atlantic populations of L. culveri (16.78% genetic distance), while in S. goodbodyi and C. biota it discriminates them from their congeneric species. These results highlight the importance of multi-tool approach and shows that both FRS histology and histo-physiology, and DNA barcoding can be used to identify and discriminate cryptic species, which is usually not possible when using morphological characters. Besides, these characters may also be useful in differentiating related species, and/or geographically distinct populations among polychaetes.


Resumo: Os poliquetas são comuns em ambientes costeiros e estuarinos em todo o mundo e constituem um dos grupos mais complexos de invertebrados marinhos. A morfo-fisiologia do sistema reprodutor feminino (FRS) pode ser compreendida por meio de ferramentas histológicas para identificar e diferenciar estes anelídeos. No entanto, essa abordagem histológica raramente é combinada com ferramentas moleculares, amplamente conhecidas por delimitar espécies congenéricas ou crípticas com maior precisão. Do mesmo modo, a descrição e o entendimento da oogênese e vitelogênese dentre os poliquetas, para a maioria das famílias, é ainda limitado. Portanto, o presente estudo tem como objetivo descrever a oogênese em três espécies de poliquetas comuns e abundantes na costa sul-americana (Laeonereis culveri, Scolelepis goodbodyi e Capitella biota) e investigar a utilidade das características reprodutivas e da gametogênese como um conhecimento associado relevante para discriminar espécies, particularmente útil para espécies crípticas putativas, integradas a dados morfológicos e moleculares. Os resultados aqui obtidos permitiram descrever dois novos subtipos de oogênese, dividindo-a em oogênese extra-ovariana dos tipos I e II e intra-ovariana dos tipos I e II. Os resultados também demonstram que os seguintes caracteres histológicos do FRS podem ser relevantes para a separação de espécies relacionadas: a) tipo de oogênese, b) presença ou ausência de um ovário verdadeiro, c) organização tissular ovariana, d) tipo de células acessórias presentes e, e) morfologia do ovócito. Além disso, essas características histológicas do FRS, quando comparadas às espécies correlatas estudadas sob esse escopo, convergem com os dados genéticos separando espécies putativas e congenéricas. As análises com DNA barcode demonstraram que em L. culveri é possível diferenciar as populações atlânticas Norte e Sul-americanas (16,78% de distância genética), enquanto para S. goodbodyi e C. biota fica evidente sua distinção com espécies congenéricas. Esses resultados destacam a importância da abordagem com múltiplas ferramentas e mostram que tanto a histologia quanto a histo-fisiologia do FRS e o DNA barcode podem ser usados para identificar e discriminar espécies crípticas e potencialmente crípticas, o que geralmente não é possível quando se utilizam apenas caracteres morfológicos. Além disso, esses caracteres também podem ser úteis na diferenciação de espécies relacionadas e / ou populações geograficamente distintas desses poliquetas.

Abstract Background: Proper timing for embryo collection and transfer in horses -which is critical for the success of this biotechnology- is still debated. Additionally, there is little information on this technology under tropical conditions. Objective: To determine the best day for collection and transfer of embryos in Mangalarga Marchador mares under Brazilian northeast's conditions. Methods: Donors (n= 30) and recipients (n= 76) in diestrus phase were selected based on both clinical and gynecology examinations. Estrus was induced on both donor and recipient mares by intramuscular injection of 5 mg Dinoprost, aiming to obtain an ovulation interval of -1 to +3 between recipient and donor. Ovulation was induced with buserelin acetate when the largest follicle reached at least 35 mm in diameter. At this time, mares were subjected to artificial insemination at 48-hour intervals until ovulation. The embryos were collected on days 7, 8, and 9 after ovulation. Results: The embryo collection on day 8 was more efficient (p<0.05) than on day 7, but it was not more effective (p>0.05) than day 9, which presented the same efficiency (p>0.05) as day 7. From a total of 76 embryos transferred to the recipients, that were between days 4 and 9 after ovulation, there was no influence (p>0.05) of the day of transfer on pregnancy rate. Conclusions: The embryo collection must be performed on day 8 after ovulation, and transfer can be performed on any day of that interval (4-9) without affecting the pregnancy rate.


Resumen Antecedentes: El momento mas apropiado para la recolección y transferencia de embriones en equinos -que es fundamental para el éxito de esta biotecnología- continua siendo sujeto de estudio. Además, es escasa la información sobre esta tecnología en condiciones tropicales. Objetivo: Determinar el momento mas adecuado para la recolecta y transferencia de embriones en yeguas Mangalarga Marchador, en las condiciones del nordeste Brasileño. Métodos: Donadoras (n= 30) y receptoras (n= 76) en la fase de diestro se seleccionaron con base en los exámenes clínicos y ginecológicos. El estro de las yeguas donadoras y receptoras fue inducido con 5 mg de Dinoprost, vía intramuscular, intentando obtener un intervalo de ovulación de -1 a +3 entre la receptora y la donadora. La ovulación fue inducida con acetato de buserelina cuando el folículo mayor alcanzó 35 mm de diámetro. En ese momento, las yeguas fueron sometidas a inseminación artificial en intervalos de 48 horas hasta la ovulación. Los embriones fueron recolectados en los días 7, 8 y 9 después de la ovulación. Resultados: La recolecta de embriones en el día 8 fue más eficiente (p<0,05) que en el día 7, pero no fue más efectivo (p>0,05) que en el día 9, el cuál presentó la misma eficiencia (p>0,05) que en el día 7. De un total de 76 embriones transferidos a las receptoras, que se encontraban entre el día 4 y 9 después de la ovulación, no se registró influencia (p>0,05) del día de la transferencia en la tasa de preñez. Conclusiones: La recolecta embrionaria debe ser realizada el día 8 después de la ovulación, y la transferencia puede ser realizada en cualquier día de este intervalo (4 a 9) sin que se afecte la tasa de preñez.

Adicionalmente, existe pouca informação sobre essa tecnologia em condições tropicais. Objetivo: Determinar o melhor dia para colheita e para transferência de embriões em eguas manga larga marchador nas condições do nordeste brasileiro. Métodos: Doadoras (n = 30) e receptoras (n = 76) na fase de diestro foram selecionadas com base nos exames clínico e ginecológicos. O estro das éguas doadoras e receptoras foi induzido com 5 mg de Dinoprost administrado por via intramuscular, buscando obter um intervalo de ovulação de -1 a +3 entre a receptora e a doadora. A ovulação foi induzida com acetato de buserelina quando o foliculo maior alcançou o tamanho de 35 mm de diâmetro. Nesse momento, as éguas foram submetidas a inseminação artificial em intervalos de 48 horas até a ovulação. Os embriões foram colhidos nos dias 7, 8 e 9 depois da ovulação. Resultados: A colheita de embriões no dia 8 foi mais eficiente (p<0,05) do que no dia 7, porem não foi mais efetivo (p>0,05) do que o dia 9, o qual apresentou a mesma eficiência (p>0,05) que o dia 7. De um total de 76 embriões transferidos para as receptoras que se encontravam entre os dias 4 e 9 depois da ovulação, não se registrou influência (p>0,05) do dia da transferência sobre a taxa de prenhez. Conclusões: A colheita embrionária deve ser realizada no dia 8 depois da ovulação, e a transferência pode ser realizada em qualquer dia desse intervalo (4-9) sem que a taxa de prenhez seja afetada.

Quantitative reverse transcription PCR (RT-qPCR) remains as an accurate approach for gene expression analysis but requires labor-intensive validation of reference genes using species-specific primers. To ease such demand, the aim was to design and test a multi-species primer set to validate reference genes for inter-genus RT-qPCR gene expression analysis. Primers were designed for ten housekeeping genes using transcript sequences of various livestock species. All ten gene transcripts were detected by RT-PCR in Bos taurus (cattle), Bubalus bubalis (buffaloes), Capra hircus (goats), and Ovis aries (sheep) cDNA. Primer efficiency was attained for eight reference genes using B. taurus-O. aries fibroblast cDNA (95.54-98.39%). The RT-qPCR data normalization was carried out for B. taurus vs. O. aries relative gene expression using Bestkeeper, GeNorm, Norm-finder, Delta CT method, and RefFinder algorithms. Validation of inter-genus RT-qPCR showed up-regulation of TLR4 and ZFX gene transcripts in B. taurus fibroblasts, irrespectively of normalization conditions (two, three, or four reference genes). In silico search in mammalian transcriptomes showed that the multi-species primer set is expected to amplify transcripts of at least two distinct loci in 114 species, and 79 species would be covered by six or more primers. Hence, a multi-species primer set allows for inter-genus gene expression analysis between O. aries and B. taurus fibroblasts and further reveals species-specific gene transcript abundance of key transcription factors.

Effective identification of species using short DNA fragments (DNA barcoding and DNA metabarcoding) requires reliable sequence reference libraries of known taxa. Both taxonomically comprehensive coverage and content quality are important for sufficient accuracy. For aquatic ecosystems in Europe, reliable barcode reference libraries are particularly important if molecular identification tools are to be implemented in biomonitoring and reports in the context of the EU Water Framework Directive (WFD) and the Marine Strategy Framework Directive (MSFD). We analysed gaps in the two most important reference databases, Barcode of Life Data Systems (BOLD) and NCBI GenBank, with a focus on the taxa most frequently used in WFD and MSFD. Our analyses show that coverage varies strongly among taxonomic groups, and among geographic regions. In general, groups that were actively targeted in barcode projects (e.g. fish, true bugs, caddisflies and vascular plants) are well represented in the barcode libraries, while others have fewer records (e.g. marine molluscs, ascidians, and freshwater diatoms). We also found that species monitored in several countries often are represented by barcodes in reference libraries, while species monitored in a single country frequently lack sequence records. A large proportion of species (up to 50%) in several taxonomic groups are only represented by private data in BOLD. Our results have implications for the future strategy to fill existing gaps in barcode libraries, especially if DNA metabarcoding is to be used in the monitoring of European aquatic biota under the WFD and MSFD. For example, missing species relevant to monitoring in multiple countries should be prioritized for future collaborative programs. We also discuss why a strategy for quality control and quality assurance of barcode reference libraries is needed and recommend future steps to ensure full utilisation of metabarcoding in aquatic biomonitoring.

Innovative composites based on an amorphous-carbon matrix containing a second phase ZnO oxide and/or highly dispersed Zn metallic were synthesized via a modified Pechini route, in which a partial pyrolysis method was reached. Studies of adsorption in the dark and the photocatalytic activity for the cationic azo-dye, basic blue 41, and degradation were carried out. X-ray diffraction patterns for the carbon matrix and its composite with Zn show characteristics of the amorphous carbon. The infrared in the mid region of the composite prepared with ZnO and Zn exhibit vibrational bands related to bonds zinc oxide. The surface pH of the material is the main factor responsible for the adsorption of the azo-dye, but the contribution of mesopores favored the diffusion of molecules from the bulk of solution to the pore framework. Esters-like functional groups on the surface of carbons hinder the adsorption of the azo-dye. When Zn is embedded within amorphous carbon the photocatalytic activity of the composites showed up to 2.4 higher than neat ZnO. The enhancement in the photocatalytic activity and stability of C/ZnO/Zn and C/Zn composites is discussed in terms of a protector effect by the carbon layers inserted in composites. Carbon layers are responsible to inhibit the lixiviation of ZnO particles along irradiation.

This research focuses on the microstructural characterization of nickel matrix composites reinforced by carbon nanotubes (CNTs). The nanocomposites were produced by a conventional powder metallurgy process and the dispersion of CNTs and mixture with nickel powders was performed in a single step by ultrasonication. Microstructural characterization of Ni-CNT nanocomposites was performed by scanning and transmission electron microscopy, electron backscattered diffraction, high-resolution transmission electron microscopy, selected area electron diffraction, and fast Fourier transform analyses. This characterization revealed CNTs embedded in the nickel grains and mainly presented as clusters at the grain boundaries. CNTs hinder recrystallization during sintering, and dislocation cells and subgrains form as a result of the recovery process.

The use of lignocellulosic remnants of the açaí agro-business will benefit the environment with a precursor material for biomedical applications. Nanocellulose (NC) allows the biomimetic growth of biphasic ceramics on its surface, with characteristics compatible with bone tissue, including bioactive properties and biocompatibility. In this study, the composites were obtained from açaí tegument (Euterpe Oleracea Mart.) NC using acid hydrolysis. The characterization performed by scanning electron microscopy showed the characteristic crystals of hydroxyapatite (HA) and calcium triphosphate (ß-TCP) based on the results of X-ray diffraction, with the peak at 22°, showing the NC nucleation of HA and peak at 17° showing tricalcium phosphate (ß-TCP). Fourier transform infrared spectroscopy confirmed the presence of O-H at 3400 cm-1 and C-H at 2900 cm-1, which is characteristic of cellulose; peaks were also observed at 1609 cm-1, verifying the reduction in lignin content. Groups PO4-3 at approximately 1070 cm-1, P-OH at 910⁻1040 cm-1, and HCO3- at 2450 cm-1 confirmed the formation of HA and ß-TCP. The zeta potential had a range of -11 ± 23.8 mV related to particle size, which had a range of 164.2 × 10-9⁻4748 × 10-9 m.
