RESUMO

Peyote, a cactus containing the hallucinogen mescaline, is used to induce altered states of consciousness in religious ceremonies or for recreational purpose. This study reports a case of an underage boy suspected of mescaline abuse. For this purpose, we analyzed a dark green liquid sample found in the bedroom of the boy whose urine and hair samples were collected shortly after the drink was found. A method by gas chromatography-mass spectrometry tandem mass spectrometry (GC-MS/MS) in positive chemical ionization mode was developed and validated in terms of linearity, specificity, accuracy, and sensitivity for mescaline determination at the low concentrations present in hair. GC-MS analysis of the liquid identified mescaline, while urine was negative; GC-MS/MS segmental hair analysis identified mescaline in the proximal segment (root to 2 cm), while the distal segments were negative. Although peyote was uncommonly encountered, its use was confirmed by segmental hair analysis that can provide long-term information about drugs use.

The aim of this study was to determine airborne polycyclic aromatic hydrocarbons (PAHs) and biphenyl at an airport by gas chromatography-mass spectrometry and to evaluate occupational exposure by environmental monitoring. A total of 12 samplings were carried out in three areas: (1) a handling area where baggage was unloaded manually from vehicles onto conveyor belts (n=5); (2) the runway with plane and motor vehicle traffic (n=5) and (3) a departure lounge (n=2). PAHs levels were in most cases low. The higher levels found refer to naphthalene (130-13,050 ng/m3) and to its methyl-substitutes 2-methylnaphthalene (64-28,500 ng/m3) and 1-methylnaphthalene (24-35,300 ng/m3), and biphenyl (24-1610 ng/m3). A method was used to quantify twenty-four airborne PAHs, and biphenyl, and to detect a variety of other chemical compounds by means of the deconvolution program AMDIS. After sampling air on quartz filter and PUF and XAD-2 sorbents; extraction with dichloromethane, and concentration and purification on silica cartridges, analyses were carried out by gas chromatography-ion trap mass spectrometry. We used 20 deuterated PAHs to quantify both the 24 native PAHs and biphenyl. The native substances had been subdivided into small groups and in this way, their volatility was adequately reflected by the D-PAH present in each group. The limit of detection was 0.1 ng/m3 for all the PAHs, and a linear range of at least about three-fold the maximum level studied (naphthalene) was obtained both for D-PAHs and the native PAHs. A good recovery pattern was obtained for D-PAHs on quartz filters, PUF and XAD-2.

RESUMO

A headspace solid-phase microextraction and gas chromatography-mass spectrometry (HS-SPME-GC-MS) procedure for the simultaneous detection of methylen-dioxyamphetamine (MDA), methylen-dioxymethamphetamine (MDMA), methylen-dioxyethamphetamine (MDE) and N-methyl-1-(1,3-benzodioxol-5-yl)-2-butanamine (MBDB) in hair has been developed. This method is suitable for the separation of primary and secondary amines, is reproducible, is not time consuming, requires small quantities of sample and does not require any derivatization. It provides sufficient sensitivity and specificity, with limits of detection (LOD) and limits of quantitation (LOQ) for each substance of <0.7 and 1.90 ng/mg, respectively. Intra- and inter-day precision were within 2 and 10%, respectively. This method is suitable for routine clinical, epidemiological and forensic purposes and can be used for the preliminary screening of many other substances (amphetamine, methamphetamine, ketamine, ephedrine, nicotine, phencyclidine, methadone) in hair and other biological matrices such as saliva, urine and blood. We also describe the first application of this HS-SPME-GC-MS procedure to the analysis of hair and saliva samples from young people attending a disco in the Rome area. All positive hair samples were confirmed by the gas chromatography-mass-mass (GC-MS(2)) technique in positive chemical ionization (PCI) mode. Some examples of the use of the method in detecting different drugs are reported.

The analysis of solvent residues trapped into crystals of illicit drugs provides useful evidence for monitoring current use trend in the chemical underground, and is also a suitable tool to achieve the complete chemical characterisation of street drugs for comparative examination of separate specimens. This paper describes a method developed in order to perform simultaneous qualitative and quantitative analysis of solvent residues in cocaine samples. The method is based on GC-MS analysis of solvents after their extraction/concentration from drug matrices accomplished by solid-phase microextraction (SPME) in static head space. The proposed method has been used to detect residues of solvents in 47 illicit street cocaine samples. Quantitative analyses were carried out only for the solvents identified at concentration values higher than 1 ppm. Statistical evaluation of our results allowed us to group the illicit samples into various classes according to different kinds of residual solvent, in connection with different clandestine manufacturing processes used to prepare illicit cocaine.
