This study evaluates the pediculicidal activity of nanoformulations containing different binary essential oil component mixtures (eugenol:linalool, 1,8 -cineole:linalool, and eugenol:thymol) using immersion bioassays. These have allowed us to evaluate the knockdown time affecting 50% of the individuals (KT50). In addition, the type of interaction between the components in each mixture was established in terms of the combination index (IC). The KT50 values were 6.07; 8.83; 7.17 and 27.23 h for linalool, 1,8 -cineole, eugenol, and thymol, respectively. For the eugenol:linalool mixtures, the efficacy was lower or equal to that obtained for the nanoformulations of the pure compounds, with values of KT50 about 13.33, 8.16 and 6.71 h for mixtures with ratios 3:1, 1:1 and 1:3, respectively. These mixtures present IC > 1, evidencing antagonistic interaction, which is enhanced with eugenol content. In the case of the binary mixtures of 1,8 -cineole: linalool, KT50 values were similar to those obtained for eugenol:linalool mixtures with similar ratios. In this case, IC assumes values close to unity, suggesting additive interactions independently of the mixture composition. On the other side, mixtures of eugenol:thymol with 1:1 and 1:3 ratios showed values of 9.40 and 32.93 h, while the mixture with a 3:1 ratio showed the greatest effectiveness (KT50 of 4.42 h). Eugenol:thymol mixtures show synergistic interaction (IC < 1) for combinations 3:1 and 1:1, while no interaction was observed for 1:3 combination. This indicates that eugenol enhances thymol activity. These results must be considered an important step forward to the development of effective pediculicidal nanoformulations based on botanical compounds.

Drosophila suzukii Matsumura, known as spotted wing drosophila (SWD), is an Asiatic invasive fruit pest that has spread over the world in the last 15 years, due to its high reproductive rate, its tolerance to different environmental conditions, the international fruit trade, and its wide range of host plants. In Buenos Aires, Argentina, blueberry is a major susceptible crop, although other cultivated and non-cultivated fruit species are frequent. The aim of this study was to evaluate the host suitability of commercial and non-cultivated fruit species (blueberries, plums, mulberries, and cherries) at two stages of maturity by estimating an index that takes into account biological and biometric parameters. The development and survival of SWD cohorts reared on different fruits were followed from egg to adult emergence. Then, adults were sexed and some biometric traits were measured. The indices: Wing loading, Wing aspect, and the Relative Performance Index (RPI) were estimated. The shortest developmental time and the maximum egg to adult survival were observed in the specimens developed in mulberry, in both stages of maturity. Only the length of the thorax showed significant differences between treatments in both sexes, and the largest adults were those reared in the ripe mulberries. The RPI, which relates performance and biometric variables, was the best index to evaluate the host suitability of SWD. So, it could be used as an indicator of the nutritional quality of fruits available in a region and to evaluate the importance of alternative hosts in the population dynamic of SWD.

To investigate the presence of Theileria equi in an endemic area of equine piroplasmosis 42 horses (Equus caballus) from Corrientes City, Argentina were sampled. Eighty-one percent (34 blood samples) of the analyzed horses were tested positive to the presence of piroplasmid 18S rDNA. All these samples could be identified as T. equi by amplifying the specific EMA-1 (merozoite antigen 1) gene of this species. Phylogenetic analysis of an obtained 18S rDNA complete sequence from one strain resulted in the identification of this sample as T. equi sensu stricto (genotype A). This study presents the first molecular detection and characterization of T. equi by the complete 18S rDNA sequence in Argentina. Based on these results further studies should be carried out to investigate the distribution and heterogeneity of presented genotypes of T. equi in Argentina, which is essential for the diagnosis, prevention and treatment of equine piroplasmosis.

Since the potential anticancer activity of auranofin was discovered, gold compounds have attracted interest with a view to developing anticancer agents that follow cytotoxic mechanisms other than cisplatin. Two benzimidazole gold(I) derivatives containing triphenylphosphine (Au(pben)(PPh3)) (1) or triethylphosphine (Au(pben)(PEt3)) (2) were prepared and characterized by standard techniques. X-ray crystal structures for 1 and 2 were solved. The cytotoxicity of 1 and 2 was tested in human neuroblastoma SH-SY5Y cells. Cells were incubated with compounds for 24 h with concentrations ranging from 10 µM to 1 nM, and the half-maximal inhibitory concentration (IC50) was determined. 1 and 2 showed an IC50 of 2.7 and 1.6 µM, respectively. In order to better understand the type of cell death induced by compounds, neuroblastoma cells were stained with Annexin-FITC and propidium iodide. The fluorescence analysis revealed that compounds were inducing apoptosis; however, pre-treatment with the caspase inhibitor Z-VAD did not reduce cell death. Analysis of compound effects on caspase-3 activity and reactive oxygen species (ROS) production in SH-SY5Y cells revealed an antiproliferative ability mediated through oxidative stress and both caspase-dependent and caspase-independent mechanisms.

Introdução: Uma das técnicas cirúrgicas utilizadas como uma alternativa à amputação no tratamento de sarcomas ósseos é a substituição do segmento acometido por endopróteses não convencionais. Embora a endoprótese melhore a qualidade de vida dos indivíduos existe uma necessidade de reabilitação intensiva para se obter melhores resultados funcionais. Objetivo: Realizar uma revisão de literatura sobre as intervenções fisioterapêuticas no pós-operatório de endoprótese não convencional de tíbia proximal, para o tratamento de sarcomas ósseos. Métodos: Revisão de literatura, direcionada a protocolos fisioterapêuticos na reabilitação de pacientes submetidos à cirurgia de preservação de membros de tíbia proximal. Resultados: Dentre os artigos estudados, três foram selecionados para comparação de protocolos fisioterapêuticos de tratamento no pós-operatório de cirurgia de preservação de membros de tíbia proximal. Conclusão: Concluiu-se que é necessário um maior incentivo no desenvolvimento de pesquisas relacionadas a protocolos de reabilitação fisioterapêutica no pós-operatório de LSS em tíbia proximal, para que sejam obtidas mais alternativas de tratamento que visam melhorar a qualidade de vida e sobrevida destes pacientes. Introduction: One of the surgical treatment techniques used as an alternative to amputation is limb salvage surgery (LSS) with endoprothesis. Although the LSS can improve quality of life, causes a variety of functional deficiencies, and creates a need to integrate physical therapy rehabilitation into the team for LSS. Objective: To carry out a review on the physiotherapeutic interventions in the postoperative LSS in patients with proximal tibial osteosarcoma. Methods: Literature review, aimed at physiotherapeutic protocols in the rehabilitation of patients submitted to preservation surgery of limbs of proximal tibia. Results: Among the articles studied, three were selected for comparison of physiotherapeutic treatment protocols in the postoperative of preservation surgery of limbs of proximal tibia. Conclusion: We concluded that a greater incentive is needed in the development of research related to physiotherapeutic rehabilitation protocols in the LSS postoperative period in the proximal tibia, in order to obtain more treatment alternatives that aim to improve the quality of life and survival of these patients. OBJECTIVE: Hand-held dynamometry is a quantitative and accessible means of determining the isometric force of muscle groups. METHODS: A total of 52 women aged 20-29 years with no complaints of hip pain who were sedentary or sporadically active and had a body mass index of 18.5-24.99 kg/m2 were included. All participants underwent bilateral assessments using hand-held dynamometry of the flexor, extensor, adductor, and abductor muscles as well as the internal and external rotator hip muscles. All hip movements were measured. All contraction data collected by the dynamometer are expressed in kilograms, normalized according to body weight, and expressed as percentages. RESULTS: The flexor muscles exhibited an isometric muscle force of 38.54% of body weight versus a muscle force of 27.04% for the extensor muscles, 16.89% for the adductors, 16.85% for the abductors, and 17.09% for the external rotators, and 23.82% for the internal rotators. CONCLUSION: Standardization of isometric strength values according to body weight proved feasible. This result is important for clinical practice since it allows the establishment of patterns of normality and criteria for discharge, return to sports, or assessment of the impact of injuries in terms of loss of muscle strength. MÉTODOS: Incluídas 52 mulheres entre 20 e 29 anos, sem queixas algicas no quadril, consideradas sedentárias ou irregularmente ativas e com IMC entre 18,50 e 24,99 kg/m2. Todas participantes foram submetidas a avaliação bilateral por dinamometria manual para músculos flexores, extensores, adutores, abdutores, rotadores internos e externos do quadril, os valores foram expressos em forma de porcentagem do peso corporal. RESULTADOS: Não foi encontrada diferença estatística na força entre lados dominante e não dominante. Os músculos flexores apresentaram força muscular isométrica correspondente a 38.54% do peso corporal, ja extensores apresentaram 27.04%. Os adutores com 16.89% e abdutores com 16,85% do peso corporal, não apresentando diferença estatística entre eles. Os rotadores externos atingiram 17.09% do peso corporal e rotadores internos chegaram a 23,82%. CONCLUSÃO: Mostrou-se possível a padronização e obtenção dos valores de força isométrica em relação ao peso corporal. Para prática clínica este resultado é importante por possibilitar conhecimento de padrões de normalidade e utilizar estes dados como critérios de alta, retorno ao esporte e avaliação da repercussão de diversas lesões na perda de força muscular. Nível de evidência III, Estudo de pacientes não consecutivos sem padrão de referência "ouro" aplicado uniformemente.

The aim of this work was to generate knowledge on ecological aspects of Amblyomma sculptum in Argentina, such as seasonal dynamics, geographical range size, hosts, genetic diversity and phylogeography. Adult and immature A. sculptum ticks were collected in different localities of Argentina to know the geographical range size and hosts. The genetic diversity of this tick was studied through analyses of 16S rDNA sequences. To describe the seasonal dynamics, free-living ticks were monthly collected from October 2013 to October 2015. A. sculptum shows a marked ecological preference for Chaco Húmedo eco-region and "Albardones" forest of the great rivers in the wetlands in the Chaco Biogeographical Province, and for Selvas Pedemontanas and Selva Montana in the Yungas Biogeographical Province. This species has low host specificity, and it has large wild and domestic mammals as principal hosts to both immature and adult stages. Amblyomma sculptum is characterized by a one-year life cycle. Larvae peak in early winter, nymphs peaked during mid-spring, and adults during late summer and mid-summer. The genetic divergence was low and the total genetic variability was attributable to differences among populations. This fact could be associated to stochastics process linked to micro-habitat variations that could produce a partial restriction to gene flow among populations. The geographic regions do not contribute much to explain the A. sculptum population genetic structure, with an ancestral haplotype present in most populations, which gives rise to the rest of the haplotypes denoting a rapid population expansion.

The effect and the role played by dietary α-lactalbumin (α-LAC) on hepatic fat metabolism are yet to be fully elucidated. We reported previously that α-LAC intake induced atherogenic dyslipidaemia in Balb/c mice. The aim of the present study was to investigate if this atherogenic effect could be due to a possible α-LAC-induced hepatic steatosis. We examine the ability of dietary α-LAC to induce liver steatosis, identifying the molecular mechanisms underlying hepatic lipid metabolism in association with the lipid profile, peripheral insulin resistance (IR) and changes in the hepatic oxidative environment. Male Balb/c mice (n 6) were fed with diets containing either chow or 14 % α-LAC for 4 weeks. The α-LAC-fed mice developed abdominal adiposity and IR. Moderate liver steatosis with increased TAG and NEFA contents was correlated with atherogenic dyslipidaemia. There was increased nuclear expression of liver X receptor αß (LXRαß), sterol regulatory element-binding protein-1c (SREBP-1c) and PPARγ transcription factors and of the cytosolic enzymes acetyl-CoA carboxylase 1 (ACC1) and fatty acid synthase involved in the hepatic de novo lipogenesis. The opposite was found for the nuclear receptor PPARα and the mitochondrial enzyme carnitine palmitoyltransferase-1 (CPT-1), leading to reduced fatty acid ß-oxidation (FAO). These changes were associated with a significant decrease in both p-Thr172-AMP-activated protein kinase α (AMPKα) (inactivation) and p-Ser79-ACC1 (activation) and with a more oxidative liver environment increasing lipid peroxidation and protein oxidation and reducing GSH:GSSG ratio in the α-LAC-fed mice. In conclusion, 4 weeks of 14 % α-LAC feeding induced liver steatosis associated with atherogenic dyslipidaemia, IR and oxidative stress by enhancing nuclear LXRαß/SREBP-1c/PPARγ expression and diminishing PPARα/CPT-1 expression and AMPKα phosphorylation shifting the hepatic FAO toward fatty acid synthesis in Balb/c mice.

The development and optimization of preantral follicle culture methods are crucial in fertility preservation strategies. As preantral follicle dynamics are usually assessed by various invasive techniques, the need for alternative noninvasive evaluation tools exists. Recently, neutral red (NR) was put forward to visualize preantral follicles in situ within ovarian cortical fragments. However, intense light exposure of NR-stained tissues can lead to cell death because of increased reactive oxygen species production, which is also associated with elevated oxygen tension. Therefore, we hypothesize that after repeated NR staining, follicle viability and dynamics can be altered by changes in oxygen tension. In the present study, we aim (1) to determine whether NR can be used to repeatedly assess follicular growth, activation, and viability and (2) to assess the effect of a low (5% O2) or high (20% O2) oxygen tension on the viability, growth, and stage transition of preantral follicles cultured in vitro by means of repeated NR staining. Cortical slices (n = 132; six replicates) from bovine ovaries were incubated for 3 hours at 37 °C in a Leibovitz medium with 50 µg/mL NR. NR-stained follicles were evaluated in situ for follicle diameter and morphology. Next, cortical fragments were individually cultured in McCoy's 5A medium for 6 days at 37 °C, 5% CO2, and 5% or 20% O2. On Days 4 and 6, the fragments were restained by adding NR to the McCoy's medium and follicles were reassessed. In both low and high oxygen tension treatment groups, approximately 70% of the initial follicles survived a 6-day in vitro culture, but no significant difference in follicle survival on Day 4 or 6 could be observed compared with Day 0 (P > 0.05). A significant decrease in the number of primordial and increase in primary and secondary follicles was observed within 4 days of culture (P < 0.001). In addition, a significant increase of the mean follicle diameter in NR-stained follicles was observed (P < 0.001), resulting in an average growth of 11.82 ± 0.81 µm (5% O2) and 11.78 ± 1.06 µm (20% O2) on Day 4 and 20.94 ± 1.24 µm (5% O2) and 19.12 ± 1.36 µm (20% O2) on Day 6 compared with Day 0. No significant differences in follicle growth rate or stage transition could be observed between 5% and 20% O2 (P > 0.05). In conclusion, after repeated NR staining, we could not find a difference between low and high oxygen tension in terms of follicle viability, stage transition, or growth. Therefore, under our culture conditions follicle dynamics are not determined by the oxygen tension in combination with quality assessment protocols using repeated NR staining.

El hígado graso no alcohólico (HGNA) es una enfermedad que se define como un espectro continuo que oscila entre una esteatosis macrovesicular, de curso clínico favorable, hasta un cuadro de esteatohepatitis no alcohólica (EHNA), que da lugar a daños irreversibles y que se ha convertido en los últimos años en un problema sanitario de primera magnitud. El HGNA se caracteriza por una infiltración grasa de los hepatocitos que se asocia con un estado de resistencia a la insulina y por ello ligado, como factor de riesgo, al síndrome metabólico. Los factores de transcripción SREBP-1c (proteína de unión al elemento regulador del esterol), ChREBP (proteína de unión al elemento de respuesta a carbohidratos) y LXR (receptor X hepático) son reguladores fundamentales de la homeostasis lipídica y glucídica y de la inflamación, cuya activación regula al alza genes implicados en la síntesis de novo de los ácidos grasos en respuesta a insulina, glucosa y oxiesteroles, tanto en condiciones fisiológicas como patológicas. En esta revisión se describen datos recientes sobre la biología, la regulación y la coordinación funcional entre SREBP-1c, ChREBP y LXR y su relación con el HGNA. El desarrollo de agonistas selectivos de estos factores les hacen ser prometedoras dianas en el tratamiento del HGNA y de la EHNA


Nonalcoholic fatty liver disease (NAFLD) has become a major public health issue that comprises a disease spectrum which ranges from benign hepatic steatosis to non-alcoholic steatohepatitis (NASH), leading to irreversible liver damages. Deposition of excess triglycerides within liver cells is the hallmark of NAFLD, which is associated with a loss of insulin sensitivity. A growing body of evidence implicates the lipogenic transcription factors SREBP-1c (sterol regulatory element-binding protein), ChREBP (carbohydrate responsive element-binding protein) and LXR (liver X receptor) in the pathogenesis of NAFLD. These factors have emerged as central regulators of the de novo fatty acid synthesis, the glucose homeostasis and the inflammation in response to insulin, glucose and oxysterols, under both physiological and physiopathological conditions. In this review we describe recent findings in the biology, the function and the cross-regulation between SREBP-1c, ChREBP and LXR on the control of lipid and glucose metabolism and their link to NAFLD. Specific pharmacologic ligands are available, making them attractive therapeutic targets for NAFLD and NASH

Grape antioxidant dietary fibre (GADF) is a grape product rich in dietary fibre and natural antioxidants. We reported previously that GADF intake reduced apoptosis and induced a pro-reducing shift in the glutathione (GSH) redox status of the rat proximal colonic mucosa. The aim of the study was to elucidate the molecular mechanisms responsible for the anti-apoptotic effect of GADF and their association with the oxidative environment of the distal colonic mucosa. The ability of GADF to modify colonic crypt cell proliferation was also investigated. Male Wistar rats (n 20) were fed with diets containing either cellulose (control group) or GADF (GADF group) as fibre for 4 weeks. GADF did not modify cell proliferation but induced a significant reduction of colonic apoptosis. The anti-apoptotic proteins Bcl-2 (B-cell lymphoma-2) and Bcl-xL (B-cell lymphoma extra large) were up-regulated in the mitochondria and down-regulated in the cytosol of the GADF mucosa, whereas the opposite was found for the pro-apoptotic protein Bax (Bcl-2-associated X protein), leading to an anti-apoptotic shift in the pattern of expression of the Bcl-2 family. Cytosolic cytochrome c and cleaved caspase-3 levels and caspase-3 activity were reduced by GADF. The modulation of the antioxidant enzyme system and the increase of the cytosolic GSH:glutathione disulfide (GSSG) ratio elicited by GADF helped to reduce oxidative damage. The cytosolic GSH:GSSG ratio was negatively related to apoptosis. These results indicate that GADF acts on the expression of the pro- and anti- apoptotic Bcl-2 proteins, attenuating the mitochondrial apoptotic pathway in the distal colonic mucosa. This effect appears to be associated with the antioxidant properties of GADF.

UNLABELLED: The digesta is a highly active biological system where epithelial cells, microbiota, nondigestible dietary components, and a large number of metabolic products interact. The gut microbiota can be modulated by both endogenous and exogenous substrates. Undigested dietary residues are substrates for colonic microbiota and may influence gut microbial ecology. The objective of this work was to study the capacity of grape antioxidant dietary fiber (GADF), which is rich in polyphenols, to modify the bacterial profile in the cecum of rats. Male adult Wistar rats were fed for 4 wk with diets containing either cellulose or GADF as dietary fiber. The effect of GADF on bacterial growth was evaluated in vitro and on the cecal microbiota of rats using quantitative real time polymerase chain reaction (RT-PCR). The results showed that GADF intake stimulates proliferation of Lactobacillus and slightly affects the composition of Bifidobacterium species. GADF was also found to have a stimulative effect on Lactobacillus reuteri and Lactobacillus acidophilus in vitro. These findings suggest that the consumption of a diet rich in plant foods with high dietary fiber and polyphenol content may enhance the gastrointestinal health of the host through microbiota modulation. PRACTICAL APPLICATION: Grape antioxidant fiber combines nutritional and physiological properties of dietary fiber and natural antioxidants from grapes. Grape antioxidant fiber could be used as an ingredient for functional foods and as a dietary supplement to increase the intake of dietary fiber and bioactive compounds.

The compound [2bpytmH](2)[I(3)](2)[I(2)], which contains protonated 2bpytm, and four neutral monomeric complexes [CoCl(2)(2bpytm)]·H(2)O (1), [CoBr(2)(2bpytm)] (2), [CoI(2)(2bpytm)]·1/2H(2)O (3) and [NiBr(2)(2bpytm)]·H(2)O (4) have been obtained during a study into the reactivity of the bis(2-pyridylthio)methane (2bpytm) ligand towards cobalt(II) and nickel(II) halides. Furthermore, a cyclic dimer [CuBr(2)(2bpytm)](2) (5) and a 1D polymer [CuBr(2)(2bpytm)](n)·CH(3)CN (6) have been obtained from copper(II)/(I) bromide salts. An unprecedented S-CH(2)-S activation and cleavage in 2bpytm has been observed on using copper(II) salts with organic and voluminous inorganic anions. The cleavage of 2bpytm enabled the isolation of copper(II) complexes containing the in situ generated ligands 2-pyridinethiolate, 2-pyridinesulfenate or 2-pyridinesulfonate.

The versatile coordination chemistry of bis(pyrimidin-2-ylthio)methane (bptm) allows the synthesis of a series of different metal complexes with diverse structures. We describe here the synthesis and structural characterization of three kinds of compounds: four cyclic dimers, [Co(2)Cl(4)(bptm)(2)] (1), [Zn(2)I(4)(bptm)(2)] (2), [Cd(2)I(4)(bptm)(2)] (3), [Cu(2)(NO(3))(4)(bptm)(2)] (4); two acyclic dimers, [Cu(2)Br(4)(bptm)(3)] (5), [Cu(2)(AcO)(4)(bptm)(2)] (6); and two 2D coordination polymers, [CuBr(bptm)] (7) and [CuI(bptm)] (8). Different structural models are obtained on making small changes in the building units under different synthetic conditions. The effects of the weak interactions in the crystal packing were also analyzed.

Grape antioxidant dietary fibre (GADF) is a grape product rich in dietary fibre and natural antioxidants. We showed previously that the GADF intake induced an epithelial hypoplasia in the rat colonic mucosa. In the present study, we propose that the antioxidant effect of GADF could modulate mucosal apoptosis via modulation of the cellular redox environment. Male Wistar rats (n 20) were fed with diets containing either cellulose (control diet group) or GADF (GADF diet group) as fibre for 4 weeks. The GSH:GSSG ratio, the redox state of the GSSG/2GSH couple (Ehc), the mitochondrial and/or cytosolic antioxidant enzyme activities (superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx)), lipid peroxidation (LPO) and apoptosis were evaluated. GADF enhanced the cytosolic GSH:GSSG ratio, shifting the redox potential (Ehc) to a more pro-reducing status. Decreased Cu,ZnSOD:CAT, Cu,ZnSOD:GPx and MnSOD:GPx ratios could indicate an enhanced capacity for reducing H2O2, contributing to decreased cytosolic LPO. Reduced apoptosis in GADF-treated mucosa was inversely related to MnSOD activity. Furthermore, apoptosis increased directly as GSSG content increased. These results suggest that the reduction in apoptosis associated with GADF intake may be due to a modulation of the glutathione redox system and endogenous antioxidant enzymes.
