RESUMO

The concentrations of free and peptide-bound amino acids aspartate (Asp), glutamate (Glu), glycine (Gly) and gamma-aminobutyric acid (GABA) were studied following ischemia and recirculation in the ventral and dorsal gray matter of the rabbit spinal cord. No changes in the concentration of amino acids following ten minutes (min) of ischemia and four days of recovery were found. The most significant change after 20 min, and especially 40 min, of ischemia was a decrease in free Asp and GABA levels in both parts of the gray matter. The relatively greater decrease in the concentration of free amino acids in the ventral horns corresponds with the greater morphological damage to this part of spinal cord following ischemia. Following 40 min of ischemia and recirculation decrease in peptide-bound Glu in the ventral horns and Asp and Glu in the dorsal horns was found.

The authors studied the metabolic activity of rat embryonic cerebral cortex grafts (ED 15-16) implanted into rat brains immediately (TR0) and 14 days (TR14) after cavity formation. Over a period of two months, the ATP, lactate and glucose concentration in TR0 transplants remained at the same level as observed in the intact cortex, whereas in TR14 transplants the ATP and glucose concentration fell significantly and the lactate concentration rose. The DNA concentration rose in both types of transplants, but the increase was more pronounced in TR0 grafts. Choline acetyltransferase activity (a neuron marker) fell significantly in both cases, but the decrease was greater in TR14 transplants. The results indicate that grafts implanted into the brain immediately after cavities had been formed have better metabolic activity and are capable of longer survival than grafts implanted 14 days after cavitation.

Concentrations of Asp, Glu, Gly, GABA and Gln were studied in the ventral and dorsal horns of the rabbit spinal cord after ligation of the abdominal aorta. The most significant changes observed after 10, 20 and 40 min ischaemia were an increase in the Asp and GABA concentration in the ventral horns and an increase in the Asp, Gly and GABA concentration in the dorsal horns. These changes correspond to shifts in the relevant reactions under conditions of the altered redox equilibrium in the tissue during ischaemia. Four days after 10 min ischaemia, amino acid concentrations in the spinal cord were at the control levels. Four days after 20 and 40 min ischaemia Asp, Gly and GABA concentrations were decreased in the ventral horns and Asp, Gly, GABA and Glu concentrations in the dorsal horns. The percentually greater decrease in the concentration in the ventral horns may be associated with the greater morphological damage to these structures.

Choline acetyltransferase (ChAT) and acetylcholinesterase (AChE) activity measured in the ventral and dorsal part of the dog spinal cord (L6-S2) and in the stumps of the sciatic nerve 5, 10, 15 and 21 days after its transection were compared with the corresponding activities in the intact contralateral nerve and in sham-operated animals. AChE was also examined histochemically. Changes in the enzyme activities in the central nerve stump were correlated with activity changes in the spinal cord. In the central nerve stump, a marked (25%) increase in AChE activity was found on the fifth day after transection, but by the 21st day it fell below control value levels; up to the 15th day it showed good correlation with AChE activity in the ventral spinal cord. Histochemically, pronounced reduction of enzymatic activity was found in the ipsilateral part of the spinal cord. On the 15th day, ChAT activity in the ventral spinal cord was also significantly decreased and the accumulation of the enzyme in the central nerve stump was negligible. On the contrary, at the last 21-day interval examined, a significant increase in ChAT activity and a nonsignificant increase in AChE activity was found in the spinal cord, but their activities in the central nerve stump were decreased. In the degenerated peripheral nerve stump ChAT activity dropped by an average of 99% and AChE activity by 48% during the first 15 days after transection but, on the 21st day, AChE activity was 22% higher than at the preceding interval.

Activities of choline acetyltransferase (ChAT) and acetylcholinesterase (AChE) in the ventral spinal cord, ventral spinal roots and in the central and peripheral stumps of the sciatic nerve transected under conditions of partial ischemia (produced by aortic ligation just below the renal arteries) were compared to those obtained under intact blood supply in time intervals 5, 10, or 15 days after surgery. The significant increase of ChAT activity in the central part of the sciatic nerve following 15 days of partial ischemia correlated with less significant elevation of ChAT in the ventral spinal cord. The changes of AChE activity were not significant during partial ischemia. ChAT in the peripheral stump of the sciatic nerve following 5 days of partial ischemia was preserved by 40% and AChE by 20% more than under normal blood supply. On the contrary, in the next 5 days interval losses of enzymes activity in the degenerating nerve were greater. ChAT was almost totally inactivated whereas 50% of AChE activity was preserved until the end of period examined.

The authors studied the effect of 40 min and 6 days occlusion of the abdominal aorta on the aspartate [Asp], glutamic acid [Glu], glutamine [Gln], glycine [Gly] and alanine [Ala] concentration in both parts of the grey matter of the lumbosacral cord, in the spinal ganglia and along the dorsal fasciculi. After 40 min ischaemia, an increase was found in the Glu, Gln, Gly and Ala concentration in the dorsal part of the grey matter and in the Glu, Gly and Ala concentration in the spinal ganglia. In the ventral part of the grey matter only the Ala concentration was increased. After 6-day ligation, the Asp and Gly concentration fell in the ventral horns, while the Gly concentration rose in the dorsal horns. The Ala and Asp concentration in the spinal ganglia rose. After this interval the Asp and Glu concentration also rose in the fasciculus gracilis. The Ala/Glu concentration ratio showed the most pronounced increase in the central horns of the grey matter.

The effect of occlusion of the abdominal aorta for 10, 20 and 40 minutes on the concentration of aspartic and glutamic acids, glutamine, glycine, alanine and gamma-amino butyric acid in the anterior and posterior horns of the lumbosacral spinal cord was studied in the dog, further, concentration of amino acids (except GABA) in lumbosacral spinal ganglia and in the ischiadic nerve following 40 minutes of occlusion. The changes were most marked after 40 minutes of occlusion with a rise in concentration of alanine, glutamine and glutamic acid in the dorsal part of grey matter. Striking was also the simultaneous elevated concentration of Glu and Gln in spinal ganglia. The significance of these changes is discussed from the aspect of metabolism and function of nerve cells. Under physiological conditions the free amino acid pool in the central nervous system remains essentially constant. Under pathological conditions, however, like ischemic-hypoxic states, various changes occur.

The authors studied the effect of 40 and 80 minutes' occlusion of the abdominal aorta on the aspartic acid, glutamic acid, glutamine, glycine and gamma-aminobutyric acid concentration in the sacral, lumbar, thoracic and cervical segments of the dog spinal cord. The most important changes were displayed by the glutamine concentration, which rose significantly after ischaemia in all the regions studied. The alanine and GABA concentration rose in the sacral and lumbar segments respectively, while the glutamic and aspartic acid concentrations in the sacral region fell. At the same time, enzyme activities degrading the synthetic substrates Leu-p-nitroanilide and Gly-p-nitroanilide increased. The significance of the findings is discussed with reference to nerve cell catabolism and function.

Levels of free amino acids were analysed in the cervical, thoracic, lumbar and sacral spinal cord of dogs. The highest content was in the sacral segment (20.2 mmol/kg wet tissue), next in the cervical (17.0) and in the lumbal segment (16.2). It was lowest in the thoracic segment (15.4). There was no difference in the content of glutamine and alanine in grey matter in comparison to white matter of posterior funicles. In the ventral horns the content of glycine and aspartic acid was markedly higher than in dorsal horns, on the other hand, the dorsal grey matter had a higher concentration of GABA than the ventral part.
