RESUMO

Unraveling the role of reactive oxygen species and associated oxidative stress (OS) in male reproduction is one of the key areas of male reproductive research. This article illustrates the scientific landscape of OS in male reproductive research over the past several decades (1941-2018) using a scientometric approach. Scientometric data (articles per year, authors, affiliations, journals, and countries) on OS related to male reproduction were retrieved from the Scopus database and analyzed for each decade. Our analysis revealed an increasing trend in OS-based male reproductive research from 1941 to 2018 with a steep raise in publications and research collaborations starting from the period 1991-2000 (R2 = 0.81). Semen abnormalities and varicocele were the major areas investigated in relation to OS with the highest positive trend in publications from the time interval 1981-1990 to 2011-2018. Analysis of publications based on OS assessment techniques revealed chemiluminescence (n = 180) and evaluation of antioxidants (n = 300) as the most widely used direct and indirect tests, respectively. Furthermore, prognostic/diagnostic studies on OS evaluation increased significantly over the time. Our analysis highlights the evolution of OS in male reproductive research and its emergence as an important prognostic and diagnostic tool in the evaluation of male infertility.

Significance: Male reproductive research at molecular level has gained more attention as it offers the cellular mechanisms and biological pathways implicated in the reproductive physiology. Several researchers across the world have used global proteomic approach in conjunction with advanced bioinformatics software to identify putative biomarkers for various male infertility conditions. Recent Advances: Introduction of advance proteomic platforms has made it easier to generate enormous amount of data in a short period of time. In this article, we have reviewed the functional and comparative proteomic studies in the area of male reproductive research. We have discussed the key proteins and associated cellular pathways such as oxidative phosphorylation and mitochondrial dysfunction implicated in the various male infertility conditions. Furthermore, for the first time scientometric approach was used to analyze the publication trends and hot topics in proteomics of male reproductive research. Critical Issues: Analysis of publication trends revealed that majority of the published studies were focused on varicocele and asthenozoospermia, while very limited research has been conducted on assisted reproductive technology (ART). This area of research requires more attention as it would facilitate identification of novel biomarkers to catalogue proteomic characteristics of spermatozoa for achieving better results in ART. Future Directions: Future research should be focused on the development and validation of a biomarker panel for specific male infertility scenarios based on etiology. Translation of validated proteomic biomarkers into tests or assays for male infertility conditions would enable the physician to provide better management for the patients.

Circular RNAs (circRNAs) are being hailed as a newly rediscovered class of covalently closed transcripts that are produced via alternative, noncanonical pre-mRNA back-splicing events. These single-stranded RNA molecules have been identified in organisms ranging from the worm (Cortés-López et al. 2018. BMC Genomics, 19: 8; Ivanov et al. 2015. Cell Rep. 10: 170-177) to higher eukaryotes (Yang et al. 2017. Cell Res. 27: 626-641) to plants (Li et al. 2017. Biochem. Biophys. Res. Commun. 488: 382-386). At present, research on circRNAs is an active area because of their diverse roles in development, health, and diseases. Partly because their circularity makes them resistant to degradation, they hold great promise as unique biomarkers for ocular and central nervous system (CNS) disorders. We believe that further work on their applications could help in developing them as "first-in-class" diagnostics, therapeutics, and prognostic targets for numerous eye conditions. Interestingly, many circRNAs play key roles in transcriptional regulation by acting as miRNAs sponges, meaning that they serve as master regulators of RNA and protein expression. Since the retina is an extension of the brain and is part of the CNS, we highlight the current state of circRNA biogenesis, properties, and function and we review the crucial roles that they play in the eye and the brain. We also discuss their regulatory roles as miRNA sponges, regulation of their parental genes or linear mRNAs, translation into micropeptides or proteins, and responses to cellular stress. We posit that future advances will provide newer insights into the fields of RNA metabolism in general and diseases of the aging eye and brain in particular. Furthermore, in keeping pace with the rapidly evolving discipline of RNA"omics"-centered metabolism and to achieve uniformity among researchers, we recently introduced the term "cromics" (circular ribonucleic acids based omics) (Singh et al. 2018. Exp. Eye Res. 174: 80-92).

OBJECTIVE: To test the ability of the smartphone-based YO Home Sperm Test to accurately and precisely measure motile sperm concentration (MSC) versus the SQA-Vision, an automated laboratory semen analyzer. DESIGN: MSC compared for the YO device on Galaxy and iPhone smartphones versus the SQA-Vision in a double-blind manner. SETTING: Academic medical center. PATIENT(S): Donor semen samples from 24 men in 144 aliquots. INTERVENTION(S): None. MAIN OUTCOME MEASURE(S): Accuracy, precision, and agreement assessed between the YO device and the SQA-Vision for MSC results. RESULT(S): The YO device demonstrated good correlation and good to moderate agreement with the SQA-Vision for MSC results up to a range of 94 × 106/mL with Pearson and concordance correlation coefficient above 0.92. The YO also showed a very high level of accuracy (97.8%) with positive and negative percent agreement above 94%. The difference in coefficient of variation between the YO and the SQA-Vision was low (between 9.4% and 11.2%) and not statistically significant. The precision among the YO phone devices was lower (16.0%) than the manufacturer's claim of ≤20%. CONCLUSION(S): The smartphone-based device has a high level of accuracy and precision when compared with the SQA-Vision. It can detect samples with abnormally "low" MSC (below 6 × 106/mL cutoff), which supports its use as an effective home sperm test for screening "low" and "moderate/normal" MSC cases. In addition, the device effectively identifies varying levels of normal MSC in a precise manner over a wide range of normal MSC. Thus, the YO Score can improve patient satisfaction and empowerment.
