RESUMO

The present study was conducted to record the prevalence of gastrointestinal parasites of pigs by faecal sample examination. Faecal samples were collected in three different seasons viz; monsoon, post monsoon and pre monsoon, from in and around Meerut district of Western Uttar Pradesh. A total of 642 faecal samples were collected for a period of one year (2020-21). The overall prevalence rate recorded for gastrointestinal parasites in pigs was 64.4%.The findings of the present work showed that seasons have a significant role on the prevalence of gastrointestinal parasites infection in pigs, with highest prevalence in monsoon (78.34%) followed by pre-monsoon (60.19%) and lowest in post- monsoon seasons (54.67%). Thus, the present study will be helpful in designing the effective control strategy of parasitic infections for optimum production in porcine industry as well as preventive measures against zoonotic diseases spread by pigs.

The present study reports the circulating oxidative stress associated with Psoroptes natalensis infestation in Indian water buffaloes. Three non-descriptive water buffaloes, age ranging between 4 and 9 years, presented to Referral Veterinary Polyclinic, IVRI, for treatment served as clinical subject. The infested animals were treated with Ivermectin subcutaneously and Amitraz topically along with antioxidant like ascorbic acid, Vitamin E and selenium. The level of lipid peroxidase was significantly higher (3.94 ± 0.34) in Psoroptes infested buffalo and was reduced significantly (P ≤ 0.05) after treatment (1.56 ± 0.40). The significantly higher levels of MDA before treatment signify the role of lipid peroxide mediated skin lesions in P. natalensis infested buffaloes. Similarly the activities of the body antioxidant like GSH and CAT were significantly higher (P ≤ 0.05) after treatment. The less level of the body antioxidant (GSH) and reduced activities of the antioxidant enzymes like CAT and SOD before treatment imply that Psoroptes mite-infested buffaloes were in a state of significant oxidative stress. The study provides information on oxidative stress indices in P. natalensis infested buffaloes and gives additional insight regarding the pathogenesis of the disease and its management.

Cryptosporidium spp. are recognized as one of the most important enteric pathogens causing enteritis and severe diarrhoea in calves up to 1 month of age. Although the infection may be responsible for some mortality, its impact is mainly associated with the impairment of gastrointestinal functions and lower performance of animals. A female buffalo calf of 25 days old was presented to OPD section, College of Veterinary and Animal Sciences, SVPUA&T, Meerut, with the symptoms of severe voluminous watery cholera like diarrhea with mucous and blood tinge since 4-5 days. On physical examination, calf was dehydrated, weak, and emaciated with normal temperature. Parasitological examination of the faeces by the direct smear and modified Ziehl-Neelsen staining technique revealed presence of high number of Cryptosporidium spp. oocysts. The affected female buffalo calf was treated with azithromycin and provided supportive care. Diarrohoeal symptoms were stopped from 3rd day and animal returned to normal condition by 7th day post treatment.

Beta-tubulin is an important multifunctional protein of eukaryotes abundant in the cytoskeleton and responsible for the formation of tubulin, structures responsible for cell morphology and which aid in motility and intracellular transportation. It has been used as a genotypic marker for studying the evolutionary history and phylogenetic relationships between eukaryotic organisms. Internal transcribed spacers of the ribosomal RNA genes have been widely used for typing inter-species and intra-species variation. An Indian isolate of Eimeria tenella was genotyped following the cloning and sequencing of beta-tubulin and internal transcribed spacer-2 (ITS-2) and compared with other reference isolates of E. tenella. The ß-tubulin has 99 % intra-species similarity at the gene level and the functional homology of the protein is very high with more than 95 % amino-acid similarity across the Apicomplexa. The ITS-2 sequence had a similar pattern of nucleotide base arrangement with 99 % homology to Houghton and Nippon strains of E. tenella.

Studies on the prevalence of gastrointestinal parasites of chicken reared under backyard and intensive systems were carried out in two north Indian states viz., Uttar Pradesh and Uttarakhand. Out of 58 poultry farms screened for gastrointestinal parasites, 81.03 % were positive for Eimeria spp., 15.52 % for Ascaridia galli, 3.45 % for Hetarakis gallinarum, 1.72 % for Syngamus trachea, 5.17 % for Capillaria spp, 1.72 % for Raillietina spp., 1.72 % for Trichostrongylus tenuis, 1.72 % for Choanotaenia infundibulum and 1.72 % for Strongyloides avium. In broiler farms, the prevalence of Eimeria spp. was higher (88.24 %) as compared to layer farms (71.43 %) and backyard poultry (70 %). Identification of Eimeria spp. using COCCIMORPH software revealed prevalence of E. acervulina, E. tenella, E. necatrix, E. mitis and E. praecox in 94.3, 17.14, 31.44, 85.7 and 2.86 % farms, respectively. However, E. maxima and E. brunetti could not be identified in any of the farms using this software. The prevalence of helminthic infections was higher in poultry farms of Uttarakhand (40.0 %) as compared to Uttar Pradesh (11.62 %) with higher prevalence in backyard poultry (36.4 %), followed by layer farms (28.6 %) and lowest in broiler farms (9.1 %). A. galli was the most common G.I. helminth and it was recorded in free-range (backyard poultry) as well as intensive systems (broiler and layer farms).

Oestrus ovis is a well-known parasite of the nasal cavities and adjoining sinuses in sheep and goats. A 3-year-old female Jamunapari goat was presented with a history of anorexia, weight loss, in-coordinated movements, nasal discharge since 10 days and two nasal bots expelled during sneezing 3 days back. Upon clinical examination the animal was dull, dysphonic, unsteady gait and having blocked nostrils with thick mucoid discharge along with an increased respiratory rate. Parasitiological examination revealed it as an O. ovis (nasal bots). The occurrence of nasal bots in a goat and its successful therapeutic management and public health significance have been reported and discussed in the present communication.

RESUMO

A 11 month old female goat was presented to Polyclinic, IVRI with the history of swelling in the left hind leg on the inner aspect of thigh and inguinal region. Physical and radiological examination revealed a large fluctuating fluid filled bladder with distinct dimension. The cyst was successfully removed along with its membrane and parasitological examination revealed it as a Coenurus gaigeri, the intermediate stage of T. multiceps gaigeri. The animal recovered completely by 10th postoperative day without any complications. This is being described in detail as under.

Molecular modelling studies were performed to identify the essential structural requirements of quinoline-based derivatives for improving their antimalarial activity. The developed CoMFA, CoMSIA and HQSAR models for a training set comprising 37 derivatives showed good statistical significance in terms of internal cross validation (q(2)) 0.70, 0.69 and 0.80 and non-cross validation (r(2)) 0.80, 0.79 and 0.80. Also, the predicted r(2) values (r(2)pred) of 0.63, 0.61 and 0.72 for a test set consisting of 12 compounds suggested significant predicting ability of the models. Structural features were correlated in terms of steric, electrostatic, hydrophobic, hydrogen bond donor and hydrogen bond acceptor interactions. Furthermore, the bioactive conformation was explored and explained by docking compounds #28, 32 and 40 into the active binding site of lactate dehydrogenase of Plasmodium falciparum. The QSAR models, contour map and docking binding affinity obtained could be successfully utilized as a guiding tool for the design and discovery of novel quinoline-based derivatives with potent antimalarial activity.

Toxocara canis is one of the most common parasitic helminth worm of dogs and also a causative agent of zoonotic disease in humans. This pilot study was conducted to determine the presence of T. canis infection in dog population in and around Bareilly, Uttar Pradesh, India. A total of 558 faecal samples both from stray and owned dogs were screened and overall 24.3 % dogs were found positive for T. canis. A comparison between owned and stray dogs suggests that the higher prevalence was observed in the latter group. The age of the dogs had a considerable influence on prevalence, with a much higher proportion of younger dogs being infected. Among the stray dogs, the infection rate is much higher (62.79 %) in pups, as compared to 7.8 % in adult. Similarly, of the owned dogs screened 41.74 % pups were infected while the infection rate in adults was only 3.38 %. The higher rate of prevalence of this parasite in dogs could be the source of soil contamination for transmission of Toxocariasis which is of public health importance in this region.

BACKGROUND: Dengue is commonly associated with myalgia, but there is paucity of studies on the frequency, severity, and basis of muscle involvement. The aim of this study was to document the clinical, electromyographic, and histological changes in dengue-associated muscle dysfunction. MATERIALS AND METHODS: Seropositive dengue patients admitted to the neurology ward during 2010 were enrolled in this study. Detailed medical history, including bleeding diathesis and organomegaly, were noted. Muscle power on a 0-5 scale, muscle tone, reflex, sensations and coordination were tested. Blood counts, hemoglobin, and serum chemistry, including creatine kinase (CK) evaluations, were carried out. Concentric needle electromyography (EMG) and muscle biopsy were performed when clinical conditions were suitable. RESULTS: The study cohort comprised 39 patients with dengue, with a median age of 28 years. Of these, 31 patients showed evidence of muscle involvement-16 with clinical and 15 with subclinical muscle involvement. Eight of these patients had severe weakness and five had hyporeflexia. Thrombocytopenia was present in 26 patients, elevated serum creatinine in three patients and liver dysfunction in 31 patients. The median CK level was 837 (range 194-3,832) U/L. The EMG revealed polyphasic normal to short duration motor unit potentials, but spontaneous activity was absent. Muscle biopsy in three patients revealed interstitial hemorrhage with occasional necrosis and myophagocytosis. There was no vasculitis, but subtle inflammatory changes were present in one patient. The severity of muscle weakness correlated with the platelet count and CK level. All patients improved by 15 days of treatment initiation. CONCLUSION: Dengue commonly results in benign and self-limiting transient muscle dysfunction.

For the management of status epilepticus (SE), lorazepam (LOR) is recommended as the first and phenytoin or fosphenytoin as the second choice. Both these drugs have significant toxicity. Intravenous levetiracetam (LEV) has become available, but its efficacy and safety has not been reported in comparison to LOR. We report a randomized, open labeled pilot study comparing the efficacy and safety of LEV and LOR in SE. Consecutive patients with convulsive or subtle convulsive SE were randomized to LEV 20 mg/kg IV over 15 min or LOR 0.1 mg/kg over 2-4 min. Failure to control SE within 10 min of administration of one study drug was treated by the other study drug. The primary endpoint was clinical seizure cessation and secondary endpoints were 24 h freedom from seizure, hospital mortality, and adverse events. Our results are based on 79 patients. Both LEV and LOR were equally effective. In the first instance, the SE was controlled by LEV in 76.3% (29/38) and by LOR in 75.6% (31/41) of patients. In those resistant to the above regimen, LEV controlled SE in 70.0% (7/10) and LOR in 88.9% (8/9) patients. The 24-h freedom from seizure was also comparable: by LEV in 79.3% (23/29) and LOR in 67.7% (21/31). LOR was associated with significantly higher need of artificial ventilation and insignificantly higher frequency of hypotension. For the treatment of SE, LEV is an alternative to LOR and may be preferred in patients with respiratory compromise and hypotension.

Clinico-radiological features of two patients with cerebrospinal fluid polymerase chain reaction-positive Epstein Barr virus (EBV) encephalitis have been reported. Both the patients presented with fever and altered sensorium, one had visual hallucination, decerebration followed by visual loss and the other had downward ocular deviation and orofacial and upper limb choreiform movement. Magnetic resonance imaging (MRI) revealed parieto-occipital involvement in both the patients. Follow-up MRI at one month was normal in one and revealed regression of lesion in the other. Both the patients, however, had severe neurologic sequelae at 18 months' follow-up. EBV encephalitis may have diverse clinical presentation with characteristic parieto-occipital involvement.

We report three children with pseudohypoparathyroidism aged 13-16 years who presented with seizures and tetany. CT scan revealed striatopallidal calcification in two. MRI revealed wide-spread involvement showing T1 hyperintensity in striatopallidodentate distribution in all three and midbrain in one patient. T2 and FLAIR images were normal. T1 hyperintensity could represent early stage of calcification in whom MRI is more sensitive.

There is paucity of studies regarding the utility of various conventional MRI sequences in the diagnosis of viral encephalitis. The present study evaluates the usefulness of various MRI sequences in acute viral encephalitis. 88 consecutive viral encephalitis patients, aged 2-72 years were subjected to clinical evaluation. Consciousness was assessed by Glasgow Coma Scale (GCS). Serum or cerebrospinal fluid (CSF) was analyzed for dengue, Japanese encephalitis (JE), herpes, measles, echo, coxsackie and polio viruses using ELISA or PCR. Cranial MRI was done and T1, T2, FLAIR and DW images were obtained. The MRI changes were correlated with type of encephalitis and duration of illness. All the patients had altered sensorium and 37 had seizures. 22 patients had JE, 9 had dengue, 8 had herpes simplex encephalitis (HSE), 2 had Epstein-Barr virus encephalitis (EBVE) and 47 had non-specific encephalitis. The median duration of MRI study from onset was 10 days. In JE (20/22), HSE (8/8), and EBVE (2/2), MRI abnormalities were more common compared to dengue (2/9) and non-specific (20/47) encephalitis. The MRI abnormalities were more common in FLAIR (57.1%) compared to T2 (52.9%), DWI (38.1%) and T1 (19.3%) sequences. The mean ADC value in JE patients was lower (974.0±110.85×10⁻6 mm²/s) than HSE (1024.33±485.76×10⁻6 mm²/s). Additional MRI lesions were seen in 12.6% cases on FLAIR sequence. FLAIR and T2 sequences were more sensitive in revealing abnormalities in viral encephalitis.

Carabeef samples were sliced, pressed, cured and divided into 6 groups. Starter cultures (Micrococcus varians M483 (MV), Staphylococcus carnosus (SC), Lactobacillus sakei (LS), M. varians M483+ Lb. sakei and Staph. carnosus + Lb. sakei) were inoculated at the dose of 10(6)-0(7)cfu/g and stored at 10 ± 1°C for 7 days. Uninoculated samples were maintained as control. Samples were then divided into 2 treatment groups. Samples of treatment 1 (T1) were smeared with a paste of turmeric followed by application of a thick layer of the paste of garlic, cumin, black pepper and red pepper whereas, samples of treatment 2 (T2) were applied with a thick layer of spices as above without turmeric. With the gradual fall in pH there was a reduction in water-holding capacity (WHC) of samples. The WHC of samples treated with SC+LS of T1 reduced to 6.3 ± 0.03 cm(2) and those inoculated with MV+LS of T2 to 6.2 ± 0.03 cm(2). The extract release volume (ERV) increased in all samples during storage. The least ERV of 11.7 and 11.6 ml were recorded in samples inoculated with MV of T1 and T2, respectively. The tyrosine (TV) and thiobarbituric acid (TBA) number of turmeric treated samples were significantly lower than non turmeric treated samples. The samples inoculated with LS had the least TV of 30.9 mg tyrosine/100 g of meat and TBA number of 0.06 mg manoladehyde/kg of meat. Samples inoculated with MV and LS of both T1 and T2 were better in physico-chemical qualities.
