RESUMO

Cystic echinococcosis (CE) is a life-threatening helminthic disease caused by the Echinococcus granulosus sensulato complex. Previous evidence indicates that the host's innate immune responses against CE can combat and regulate the growth rate and mortality of hydatid cyst in the host's internal organs. However, the survival mechanisms of CE are not yet fully elucidated in the human body. In the present study, the apoptotic effects of fertile and infertile hydatid fluid (HF) were tested on murine peritoneal cells in vivo mice model. Mice were divided into five groups including; control group, fertile HF-treated peritoneal cells, infertile HF-treated peritoneal cells, protoscolices (PSCs)-treated peritoneal cells and HF+PSCs-treated peritoneal cells group. Mice groups were intraperitoneally inoculated with PBS, HF, and/or PSCs. Afterwards, peritoneal cells were isolated and mRNA expression of STAT3, caspase-3, p73 and Smac genes were evaluated by quantitative Real-time PCR. After 48 hours of exposure, the protein levels of Smac and STAT3 was determined by western blotting technique. After 6 hours of exposure, Caspase-3 activity was also measured by fluorometric assay. The intracellular reactive oxygen species (ROS) production was examined in all groups. The mRNA expression levels of p73, caspase-3 and also Caspase-3 activity in HF+PSCs-treated peritoneal cells were higher than in the test and control groups (Pv<0.05), while the mRNA expression level of anti-apoptotic STAT3 and Smac genes in HF+PSC-treated peritoneal cells were lower than in the other groups (Pv<0.05). As well, the level of intracellular ROS in the fertile HCF-treated peritoneal cells, infertile HCF-treated peritoneal cells, PSC-treated peritoneal cells and HF+PSC-treated peritoneal cells groups were significantly higher than in the control group (Pv<0.05).Current findings indicates that oxidative stress and p73 can trigger the apoptosis of murine peritoneal cells through modulator of HF-treated PSCs that is likely one of the hydatid cyst survival mechanisms in vivo mice model.

PURPOSE: Doxorubicin (DOX) is a common chemotherapeutic agent, with toxic side effects, and chemoresistance. Combination chemotherapy is a successful approach to overcome these limitations. Here, we investigated the effects of pioglitazone (PGZ), a PPARγ agonist, and/or DOX on the viability, cell cycle, apoptosis on THP-1 cells and normal human monocytes (NHMs). METHODS: MTT assay was used to evaluate the cytotoxicity of DOX and/or PGZ. Cell cycle progression and apoptosis induction were examined by PI or Annexin V-PI double staining, and analyzed by flow cytometry. Quantitative RT-PCR was used to evaluate the changes in the mRNA expression of cell cycle progression or apoptosis-associated genes including P27, P21, CDK2, P53, BCL2 and FasR. RESULTS: DOX, PGZ and DOX + PGZ exerted their cytotoxic effects in a dose- and time-dependent manner with low toxicity on NHMs. The cell growth inhibitory effects of DOX were in association with G2/M arrest, while PGZ executed S phase arrest. PGZ treatment enhanced G2/M among DOX-treated combinations with moderate elevation in the S phase. DOX, PGZ and combined treatments induced apoptosis (mostly late phase) in a dose-dependent manner. All treatments resulted in the significant overexpression of p21, p27, p53 and FasR genes and downregulation of CDK2. DOX + PGZ combined treatments exhibited the most significant changes in mRNA expression. CONCLUSION: We demonstrated that the antiproliferative, cell cycle regulation and apoptosis-inducing capacity of DOX was enhanced by PGZ in THP-1 leukemia cells in a dose-dependent manner. Therefore, the combination of DOX + PGZ could be used as a novel combination to target AML.

Toxocarosis is an important emerging zoonotic disease, which is caused by larvae of Toxocara canis and Toxocara cati, the most widely distributed nematode parasites of dogs and cats. The soil, where Toxocara eggs become infective, is the natural source of infection. The aim of this work was to determine the levels of soil contamination with Toxocara spp. eggs in Sari district, north of Iran, in order to estimate the risk of infection of the population living in this region. A total of 141 soil samples were collected from 12 parks, 12 primary schools and 45 backyards from three regions. Soil samples were investigated for the presence of Toxocara eggs by flotation technique using sucrose solution and examined under light microscope using 10× and 40× objective. Out of 141 soil samples, 67 (47.5%) were found to contain Toxocara spp. eggs. The contamination rate in public parks, primary schools and backyards were observed 50%, 58.3% and 33.3%, respectively. The comparison of soil contamination rate from different areas, public parks, primary schools and backyards, was statistically significant (χ2=6.00, d.f.=2 and P=0.049). The public parks, backyards and primary schools, which are main places that children play, are suitable for defecation of dogs and cats. The knowledge of the soil contamination with Toxocara eggs indicates the need to take an appropriate and effective measures to prevent the infection.

INTRODUCTION: The 2019 coronavirus disease (COVID-19) is a major global health concern. Joint efforts for effective surveillance of COVID-19 require immediate transmission of reliable data. In this regard, a standardized and interoperable reporting framework is essential in a consistent and timely manner. Thus, this research aimed at to determine data requirements towards interoperability. MATERIALS AND METHODS: In this cross-sectional and descriptive study, a combination of literature study and expert consensus approach was used to design COVID-19 Minimum Data Set (MDS). A MDS checklist was extracted and validated. The definitive data elements of the MDS were determined by applying the Delphi technique. Then, the existing messaging and data standard templates (Health Level Seven-Clinical Document Architecture [HL7-CDA] and SNOMED-CT) were used to design the surveillance interoperable framework. RESULTS: The proposed MDS was divided into administrative and clinical sections with three and eight data classes and 29 and 40 data fields, respectively. Then, for each data field, structured data values along with SNOMED-CT codes were defined and structured according HL7-CDA standard. DISCUSSION AND CONCLUSION: The absence of effective and integrated system for COVID-19 surveillance can delay critical public health measures, leading to increased disease prevalence and mortality. The heterogeneity of reporting templates and lack of uniform data sets hamper the optimal information exchange among multiple systems. Thus, developing a unified and interoperable reporting framework is more effective to prompt reaction to the COVID-19 outbreak.

Human toxocarosis is a chronic zoonotic disease, which creates various syndromes in humans such as visceral larva migrans, ocular larva migrans, and neurological toxocarosis. This disease is commonly contracted through ingestion of eggs from the Toxocara species. The eggs are transmitted to humans through contact with soil that contaminated with feces of infected animals by Toxocara species worm. This study, investigated the soil from various parks and public areas of Khorramshahr city southwest of Iran for Toxocara egg contamination. One-hundred and fifty soil samples were collected from 21 parks and green public areas from April to July 2018 and examined by the modified floatation method using saturated sucrose solution. The Toxocara eggs were isolated in 27 (18%) out of 150 samples collected. Based on Chi-square test, none of the factors: location of sampling (in city and the suburb), the presence of dogs and cats and the size of the sampling location; was not significant difference by soil contamination by Toxocara eggs. The presence of Toxocara eggs in Khorramshahr parks soil and green public areas is one of the main risk factors for catch the toxocarosis. This is important to develop preventive measures in the city and suburb areas to reduce the Environmental contaminations and thus reduce the incidence of disease in humans.

BACKGROUND: Toxocariasis is one of the most important zoonotic diseases caused by Toxocara larva stage in humans. One of the major transmission routes of infection, especially in children is pica. The aim of this topic was study the contamination of Abadan public parks with Toxocara eggs. MATERIALS AND METHODS: Two hundred and ninety one samples of soil were collected from 31 parks. The samples were examined for Toxocara spp. eggs by modified floatation method using saturated sucrose. The results were analyzed using SPSS version 19 and Chi-square test. RESULTS: Eighty five (29.2%) out of 291 samples were infected with Toxocara spp. eggs, means19 (61.2%) of the 31 parks were contaminated. There was no significant difference between the urban and suburb parks contamination (p = 0.208) but there was significant relation between contamination with Toxocara spp. eggs and traces of cats and dogs presence in the parks (p = 0.001). CONCLUSION: As the contamination of Abadan public parks soil with Toxocara spp. eggs is relatively high, the people and specially children might get the contamination during stay in the parks and measures should be taken to control the stray cats and dogs.
