RESUMO

Following Afghanistan's fall in August 2021, many refugees were settled in the United States as part of Operation Allies Welcome. They were flown from Kabul to the Middle East and Europe before continuing to the U.S. By late September Philadelphia was the sole destination. From there refugees were transported to Safe Haven military bases around the country. Philadelphia International Airport became the site of a months-long operation involving city, state, federal, and private agencies engaged in processing, medical screening, and COVID-testing of arriving refugees. The Philadelphia Fire Department played an integral role. Minor medical conditions were treated onsite. Higher acuity patients were transported to nearby hospitals. The goal was to maintain flow of refugees to their next destination while addressing acute medical issues. Between August 28, 2021, and March 1, 2022, the airport processed 29,713 refugees. Philadelphia's experience may serve as a guide for planning future such refugee operations.

RESUMO

Recently, scalar coupled-wave theory has been employed to analyze a medium with periodic time-varying permittivity, providing simple expressions and, consequently, straightforward insights into the parametric amplification mechanism. Here, we combine such an approach with the Möbius transformation method to investigate the dispersion and optical response of a finite "time-slab" of the aforementioned medium. We demonstrate the temporal analog of a Bragg grating, discuss the differences with its spatial counterpart, and examine nontrivial scenarios of the permittivity's time-modulation, such as chirping and apodization. Furthermore, we propose a highly selective and, moreover, single-spatial-interface optical sensor, based on phase delineation.

A new mechanism of Bragg phenomenon is theoretically identified that, remarkably, occurs in a uniform medium and relies on resonant tuning of the medium parameters rather than on wavelength-matching. Due to the uniformity, reflection ensues over a broad wavelength range, much like a metal, but is polarization dependent: one circular state is reflected, whereas the other is transmitted. Such a medium can thus provide a broadband, low-loss polarization divider/combiner. Assessing a realistic embodiment with loss and material dispersion, we discuss practical realizations within range of current meta-media technology at terahertz and optical frequencies.

The prognosis of older patients with acute myelogenous leukemia is generally poor. The interleukin-3 receptor α-chain (CD123) is highly expressed on the surface of acute leukemia cells compared with normal hematopoietic stem cells. CSL362 is a fully humanized, CD123-neutralizing monoclonal antibody containing a modified Fc structure, which enhances human natural killer cell antibody-dependent cell-mediated cytotoxicity. Six continuous acute myelogenous leukemia xenografts established from patient explants and characterized by cell and molecular criteria, produced progressively lethal disease 42-202 days after transplantation. CSL362 alone reduced engraftment of one of four and three of four acute myelogenous leukemia xenografts in the bone marrow and peripheral organs, respectively. A cytarabine and daunorubicin regimen was optimized using this model to identify potentially synergistic interactions with CSL362. Cytarabine/daunorubicin improved the survival of mice engrafted with four of four acute myelogenous leukemia xenografts by 31-41 days. Moreover, CSL362 extended the survival of cytarabine/daunorubicin-treated mice for two of two acute myelogenous leukemia xenografts, while augmentation of natural killer cell-deficient NSG mice with adoptively transferred human natural killer cells improved survival against a single xenograft. Interestingly, this enhanced CSL362 efficacy was lost in the absence of chemotherapy. This study shows that acute myelogenous leukemia xenografts provide a platform for the evaluation of new therapeutics, simulating complex in vivo interactions, and that the in vivo efficacy of CSL362 supports continued clinical development of this drug.

We discuss the use of Mueller matrices for characterizing the reflecting properties of beetles, including matching patterns of significant elements to specific cuticular architectures. In the case of illumination by natural light, the parameters of the reflected light are related to the elements of the first column of the matrix. The green and red beetle Stephanorrhina guttata is shown to be a narrowband polarization-preserving reflector apart from depolarizing white patches, the green Calloodes grayanus a narrowband left-circular reflector, and the gold Anoplognathus parvulus behaves as a broadband left-circular reflector. Comparison of experimental and simulated matrices confirms that the beetle reflectors are natural analogs of all-dielectric thin-film reflectors. However, the gold Chrysina resplendens, which was formerly known as Plusiotis resplendens and reflects both right-handed and left-handed light, is represented by an ensemble of laterally incoherent chiral thin-film reflectors.

RESUMO

Physically valid electromagnetic continuity equations can be generated from either the usual form of the Poynting vector E x H or the alternative E x B form. However, the continuity equations are not identical, which means that quantities following from E x H cannot always be compared directly to those from E x B. In particular, the work done on the bound current densities are attributed differently in the two representations.We also comment on the negative refraction condition used.

RESUMO

The success of engineered monoclonal antibodies as biopharmaceuticals has generated considerable interest in strategies designed to accelerate development of antibody expressing cell lines. Stable mammalian cell lines that express therapeutic antibodies at high levels typically take 6-12 months to develop. Here we describe a novel method to accelerate selection of cells expressing recombinant proteins (e.g., antibodies) using multiparameter fluorescence activated cell sorting (FACS) in association with dual intracellular autofluorescent reporter proteins. The method is co-factor-independent and does not require complex sample preparation. Chinese hamster ovary (CHO) clones expressing high levels of recombinant antibody were selected on the basis of a two-color FACS sorting strategy using heavy and light chain-specific fluorescent reporter proteins. We were able to establish within 12 weeks of transfection cell lines with greater than a 38-fold increase in antibody production when compared to the pool from which they were isolated, following a single round of FACS. The method provides a robust strategy to accelerate selection and characterization of clones and builds a foundation for a predictive model of specific productivity based upon on two-color fluorescence.

We appraise recent claims that classical gravitation can induce negative refraction of electromagnetic radiation in vacuum. By recasting the previous literature in covariant notation, we show that the criterion used hitherto for determining negative refraction in vacuum is inappropriate, and can even be satisfied by parametrized transformations in Minkowski spacetime. Using instead a covariantly acceptable definition, we find that in classical vacuum the power flux of a plane electromagnetic wave points in the direction of phase advance.

The structure of an inorganic chiral medium represented as a stack of identical form-birefringent layers that twist steadily with increasing thickness is perturbed by realigning a fraction of each layer to a fixed direction. Experimental results show that the resulting chiral-birefringent composite medium exhibits Bragg resonance with elliptically polarized light, and simulations indicate that Bragg reflectors can be designed for any polarization including linear.

Alterations in Ca2+ signaling may contribute to tumorigenesis and the mechanism of action of some anti-cancer drugs. The plasma membrane calcium-ATPase (PMCA) is a crucial controller of intracellular Ca2+ signaling. Altered PMCA expression occurs in the mammary gland during lactation and in breast cancer cell lines. Despite this, the consequences of PMCA inhibition in breast cancer cell lines have not been investigated. In this work, we used Tet-off PMCA antisense-expressing MCF-7 cells to assess the effects of PMCA inhibition in a human breast cancer cell line. At a level of PMCA inhibition that did not completely prevent PMCA-mediated Ca2+ efflux and did not induce cell death, a dramatic inhibition of cellular proliferation was observed. Fluorescence-activated cell sorting analysis indicated that PMCA antisense involves changes in cell cycle kinetics but not cell cycle arrest. We concluded that modulation of PMCA has important effects in regulating the proliferation of human breast cancer MCF-7 cells.

We consider the circularly polarized localized modes of chiral photonic structures with combined central twist and isotropic layer defects. The general filter is shown to suffer from anomalous remittance and saturation of linewidth as the thickness of the structure is increased. However, by choosing parameters that phase match the elements of the round-trip matrix of the isotropic layer defect, we demonstrate the existence of supermodes that maintain exceptional purity of polarization state and exponential decrease in linewidth as the thickness is increased.

Diffraction loss in radiometry has gained in importance recently because of an increased interest in longer wavelengths and the continuous improvement in experimental accuracy. The deviation from geometrical optics now contributes significantly to the errors of experiments. Previous research has concentrated on geometries classified as F1 and F2, leaving an intermediate case yet to be investigated. This intermediate case has some interesting behavior, as it is in this envelope of geometries that it is possible to have zero diffraction loss. We designate this intermediate geometric regime as F3. We introduce a numerical regime to calculate diffraction loss for intermediate geometries, which is also highly efficient for the F1 and F2 regimes.
