RESUMO

The characteristics of the cells epigenotypes Dunaliella viridis Teod. in the process of chronological and replicative aging were investigated. By 40th day of accumulative cultivation (which coincided with the stationary growth phase) DNA content in the cells of Dunaliella viridis increased 2 times, triacylglycerides 3 times, beta-carotene and carbonyl proteins 2 times, RNA content decreased in comparison with cells in exponential growth phase, i. e., the 40th day of growth of culture forms the age-related epigenotype. 4 received subcultures were being transplanted during 2 years in mid-logarithmic growth phase (subculture-10), early stationary phase of growth (subculture-20), in the mid-stationary growth phase (subculture-30), and late stationary growth phase (subculture-40). It is shown that epigenotype of subculture-10 remained unchanged over 2 years of cultivation, i. e., it does not manifest replicative aging. At the same time, the subculture-20, although long enough (at least 40 passages), maintained epigenotype characteristic of young cultures, and showed age-related changes. Pronounced age-dependent changes of epigenotype in the course of cultivation were identified for subculture-30, and subculture-40 was characterized by unstable epigenotype. Thus, cultivation conditions determine the intensity of replicative aging in Dunaliella viridis.

RESUMO

Two fractions of hepatocytes were isolated from the rat liver by nonfermentative method. These fractions were different for mechanical stability to the action of perfusion factors. It has been shown that cells of these fractions were differently separated in the linear density gradient of sucrose. It was connected with structural and functional heterogeneity of hepatocytes in the liver. Results obtained allow us to confirm that hepatocytes in the liver form at least two classes of cells with different lipid content. The plasmatic membranes of hepatocytes with high content of lipids were inclined to damage in the process of liver perfusion as compared to cells with low lipid content.

RESUMO

The authors have improved the method for the gradient isolation of monocytes from human whole blood: an expensive reagent, percoll, is replaced by verograffin, an x-ray contrast agent. Monocytes were isolated by this method from the blood of 8 normal subjects. The isolated monocytes were identified and their population composition studied electronmicroscopically, cytochemically, and immunocytochemically. The monocyte population of healthy donors was found morphologically and cytochemically heterogeneous, consisting of 4 types of cells, forming on the monocytogram stable (in their per cent shares) classes. The improved method for monocyte isolation and the results of morphometric analysis may be used to assess the system of human mononuclear phagocytes in various diseases.

RESUMO

Population kinetics of monocytes (MC) was studied to prove their participation in atherogenesis. Increased number of cells containing lipids (CCL) in patients with severe stable angina pectoris is demonstrated (42.5 versus 7.4% in healthy persons). This is followed by MC esterase activity enhancement. In patients with angina of effort new types of MC appear (with high peroxidase and low esterase activity) and there is a drastic increase of CCL (more than 56%). The correlation between the number of CCL and indices of blood lipid metabolism in anginal patients is lacking. The results obtained may serve as a criterion for evaluating the degree and exacerbation of coronary atherosclerosis in humans.

RESUMO

The lipid composition and synthesis rate in liver microsomes of linear and hybrid rats are investigated. It is found that microsomes of hybrid rats had less amount and synthesis rate of phosphatidylcholine and phosphatidylethanolamine as well as nonesterified fatty acids. Linear rats differed from each other in phosphatidylinositol exchange rate. These genetic peculiarities of the microsomal lipid composition and synthesis rate can explain the experimentally determined absence of correlation between glucose-6-phosphatase activity and lipid maintenance in membranes of different genetic forms.
