Wallachian and Sumava sheep are autochthonous breeds that have undergone a significant bottleneck effect and subsequent restoration efforts. The first objective of this study was to evaluate the degree of genetic variability of both breeds and, therefore, the current management of the breeding. The second was to determine whether these two breeds still retain their genetic uniqueness in relation to each other and other breeds, despite regenerative interventions. Our data consisted of 48 individuals of Sumava and 37 individuals of Wallachian sheep. The comparison data contained 25 other breeds (primarily European) from the HapMap dataset generated by the International Sheep Genomics Consortium. When comparing all 27 breeds, the Czech breeds clustered with 15 other breeds and formed a single branch with them according to Nei's distances. At the same time, however, the clusters of both breeds were integral and easily distinguishable from the others when displayed with principal component analysis (PCA). Population substructure analysis did not show any common genetic ancestry of the Czech national breeds and breeds used for regeneration or, eventually, breeds whose ancestral population was used for regeneration. The average values of FST were higher in Wallachian sheep (FST = 0.14) than in Sumava sheep (FST = 0.08). The linkage disequilibrium (LD) extension per autosome was higher in Wallachian than in Sumava sheep. Consequently, the Ne estimates five generations ago were 68 for Sumava versus 34 for Wallachian sheep. Both native Czech breeds exhibit a wide range of inbreeding based on the excess of homozygosity (FHOM) among individuals, from -0.04 to 0.16 in Sumava and from -0.13 to 0.12 in Wallachian. Average inbreeding based on runs of homozygosity was 0.21 in Sumava and 0.27 in Wallachian. Most detected runs of homozygosity (ROH) were less than 5 Mb long for both breeds. ROH segments longer than 15 Mb were absent in Wallachian sheep. Concerning putative selection signatures, a total of 471 candidate genes in Wallachian sheep within 11 hotspots and 653 genes within 13 hotspots in Sumava sheep were identified. Czech breeds appear to be well differentiated from each other and other European breeds. Their genetic diversity is low, especially in the case of the Wallachian breed. Sumava is not so threatened by low diversity but has a larger share of the non-native gene pool.

The aim of the present study was to compare the chemical composition and fatty acid (FA) content of the muscle tissues of lambs and adult sheep bred for meat production. Throughout the research period, the animals were managed in a confinement indoor system under uniform environmental conditions. After slaughter, meat samples collected from the musculus biceps femoris were used to determine the basic chemical composition and fatty acid content in the intramuscular fat. The analyses showed that, compared to lambs, meat from adult animals exhibited a more beneficial ratio of n-6 to n-3 FAs, as well as a favorable percentage of OFAs (dietary fatty acids having an undesirable hypercholesterolemic effect on humans). The results of this study support the inclusion of dietitian-recommended mutton into the human diet and the promotion of the production of meat from adult animals as a high-value product. The sheep industry could benefit economically, particularly in countries where this meat type has not enjoyed a high standing in consumer preferences.

This study determined effects of milk production, milk components, or fatty acids (FA) profile on live weight of suckling lambs till their weaning. Live weight (LW, kg) of 42 purebred Wallachian lambs (from 33 ewes) was recorded during four control days with approximately 30-day intervals during rearing. At the same time, their mothers were examined for milk production (kg), milk fat (g), proteins (g), lactose (g), and fatty acids (%) contents. Results investigated using linear regression analysis showed 5.93 kg (p < 0.05) increase of lambs LW corresponded with 1 kg of ewe's daily milk production increase during the observed period. Similarly, significant 0.13 kg or 0.11 kg increases of lambs live weight corresponded with 1 g increase of milk protein or milk lactose at this time. Milk with higher prevalence of trans-palmitoleic acid, trans-vaccenic acid, cis-vaccenic acid, linolelaidic acid, linoleic acid, or conjugated linolenic acid (CLA) significantly improved lambs LW. Moreover, significantly positive Pearson partial correlation between LW and trans-vaccenic acid (r = 0.305) or CLA (r = 0.347) indicated on genetic correlation between these traits. Therefore, milk (natural or artificially supplied) with higher distribution of these specified FAs could improve lambs' LW.

In this Research Communication we evaluate the fatty acid (FA) profile of Wallachian sheep milk. The study was performed on 38 ewes in Beskydy Mountains. Samples were collected 4 times during the lactation, at monthly intervals. FA profile as well as groups of saturated, monounsaturated (MUFA), and polyunsaturated (PUFA) FAs were investigated. Considerable increase over the lactation was detected for lauric, myristic, myristoleic and palmitic acids, while stearic acid showed the opposed tendency. Variability, supported by significant differences among particular days of milk collection, was demonstrated for oleic acid; its highest distribution occurred at the beginning and at the end of the trial. The highest distribution of CLA was at the second sample day. Milk of Wallachian sheep naturally grazed at permanent pasture areas showed higher content of PUFA and MUFA in contrast with intensive or semi-intensive sheep breeds reported in the literature.
