This study evaluated the inhibitory impacts of phytic acid on the growth of T. roseum both in vitro and in apple fruit, as well as elucidated the potential mechanisms underlying its action. Results showed that phytic acid suppressed the lesion diameter caused by T. roseum inoculation in apples, as well as spore germination and mycelial growth of T. roseum in vitro. Phytic acid reduced intracellular conductivity and soluble sugar content, while increasing malondialdehyde and soluble protein contents. Phytic acid treatment inhibited the activities of pectin lyase, pectin methyl polygalacturonase, ß-glucosidase, cellulase, xylanase, pectin methyl trans-eliminase, polygalacturonase, and polygalacturonase both in vitro and in apples. In contrast, inoculation of control and phytic acid-treated fruit with T. roseum resulted in increased enzyme activity. These findings suggest that phytic acid decrease the occurrence of heart rot in apples through inducing disruption of the cell membrane of T. roseum and mediating cell wall metabolism.

Eliminating "sulfur starvation" caused by competition for sulfate transporters between chromate and sulfate is crucial to enhance the content of sulfur-containing compounds and improve the tolerance and reduction capability of Cr(VI) in bacteria. In this study, the effects of sulfur salts on the Cr(VI) bioremediation and the possible mechanism were investigated in Rhodobacter sphaeroides SC01 by cell imaging, spectroscopy, and biochemical measurements. The results showed that, when the concentration of metabisulfite was 2.0 g L-1, and the initial OD600 was 0.33, the reduction rate of R. sphaeroides SC01 reached up to 91.3% for 500 mg L-1 Cr(VI) exposure at 96 h. Moreover, thiosulfate and sulfite also markedly increased the concentration of reduced Cr(VI) in R. sphaeroides SC01. Furthermore, the characterization results revealed that -OH, -CONH, -COOH, -SO3, -PO3, and -S-S- played a major role in the adsorption of Cr, and Cr(III) reduced by bacteria was bioprecipitated in the production of Cr2P3S9 and CrPS4. In addition, R. sphaeroids SC01 combined with metabisulfite significantly increased the activity of glutathione peroxidase and the content of glutathione (GSH) and total sulfhydryl while decreasing reactive oxygen species (ROS) accumulation and cell death induced by Cr(VI) toxic. Overall, the results of this research revealed a highly efficient and reliable strategy for Cr(VI) removal by photosynthetic bacteria combined with sulfur salts in high-concentration Cr(VI)-contaminated wastewater.

RESUMO

Paper substrate room temperature phosphorescence (RTP) of theobromine (TB), caffeine (CF) and theophylline (TP) were investigated. The method is based on fast speed quantitative filter paper as substrate and KI-NaAc as heavy atom perturber. Various factors affecting their RTP were discussed in detail. Under the optimum experimental conditions, the linear dynamic range, limit of detection (LOD), and relative standard deviation (R.S.D.) were 14.41 approximately 576.54 ng per spot, 1.14 ng per spot, 4.8% for TB, 5.44 approximately 699.08 ng per spot, 0.78 ng per spot, 1.56% for CF, 7.21 approximately 360.34 ng per spot, 1.80 ng per spot, 3.80% for TP, respectively. The first analytical application for the determination of these compounds was developed. The recovery of standard samples added to commercial products chocolate, tea, coffee and aminophylline is in the range 92.80-106.08%. The proposed method was successfully applied to real sample analysis without separation.
