RESUMO

A convenient and mild synthesis of 5-bromo-N4-substituted-1-(beta-D-arabinofuranosyl)cytosine and 5-bromo-O4-methyl-1-(beta-D-arabinofuranosyl)pyrimidin-2(1H)-one derivatives by selective oxyfunctionalization of the corresponding 4-thionucleosides with 3,3-dimethyldioxirane is reported. The cytotoxicity and the antiviral activity against parainfluenza 1 (Sendai virus) of all new synthesized products are also reported.

RESUMO

Several new 6-oxiranyl-, 6-methyloxiranyluracils, and pyrimidinone derivatives, synthesized by the lithiation-alkylation sequence of 1,3,6-trimethyluracil, 1,3-dimethyl-6-chloromethyluracil, and 2-alkoxy-6-methyl-4(3H)-pyrimidinones, showed a potent and selective antiviral activity against the parainfluenza 1(Sendai) virus replication.

RESUMO

We describe the reaction of formamide with 2'-deoxycytidine to give pyrimidine ring opening by nucleophilic addition on the electrophilic C(6) and C(4) positions. This information is confirmed by the analysis of the products of formamide attack on 2'-deoxycytidine, 5-methyl-2'-deoxycytidine, and 5-bromo-2'-deoxycytidine, residues when the latter are incorporated into oligonucleotides by DNA polymerase-driven polymerization and solid-phase phosphoramidite procedure. The increased sensitivity of 5-bromo-2'-deoxycytidine relative to that of 2'-deoxycytidine is pivotal for the improvement of the one-lane chemical DNA sequencing procedure based on the base-selective reaction of formamide with DNA. In many DNA sequencing cases it will in fact be possible to incorporate this base analogue into the DNA to be sequenced, thus providing a complete discrimination between its UV absorption signal and that of the thymidine residues. The wide spectrum of different sensitivities to formamide displayed by the 2'-deoxycytidine analogues solves, in the DNA single-lane chemical sequencing procedure, the possible source of errors due to low discrimination between C and T residues.

From a selective enrichment culture prepared with different soil samples on starch-containing polyethylene we isolated four microaerophilic microbial communities able to grow on this kind of plastic with no additional carbon source. One consortium, designated community 3S, was tested with pure isotactic polypropylene to determine whether the consortium was able to degrade this polymer. Polypropylene strips were incubated for 5 months in a mineral medium containing sodium lactate and glucose in screw-cap bottles. Dichloromethane crude extracts of the cultures revealed that the weight of extracted materials increased with incubation time, while the polypropylene sample weight decreased. The extracted materials were characterized by performing chromatographic and spectral analyses (thin-layer chromatography, liquid chromatography, gas chromatography-mass spectrometry, infrared spectrometry, nuclear magnetic resonance). Three main fractions were detected and analyzed; a mixture of hydrocarbons at different degrees of functionalization was found together with a mixture of aromatic esters, as the plasticizers usually added to polyolefinic structures.

A biochemical study of dental pulp of calves has been performed concerning: a) peroxydability b) A, E, C vitamins content c) glutation (GSH) content d) presence of paramagnetic compounds e) phosphorylation ratio The dental pulp from incisors of 5-months-old calves has been preserved. Immediately after decapitation the pulp was immersed in liquid nitrogen. Chromatographic (HPLC) and spectroscopic (NMR-ESR) techniques have been used. GSH in dental pulp are present and dosable (4.56 +/- 0.08 n moles/mg prot.) and GSSG (1.05 +/- 0.01 n moles/mg prot.). Because of blood traces in the extracted pulps, the AA. have determined the hemoglobin (Hb) dosage and GSH of erythrocytic derivation (Fig. 1). After deduction of GSH of erythrocytic derivation, the GSH really present in the pulp was 4.41 n moles/mg prot. and the GSSG was 0.90 n moles/mg prot. Peroxydability of the dental pulp has been evaluated with Lowry method with dental pulp homogenate and rat liver homogenate (see Table 1). The ESR spectre shows 4 resonances with the following values: g. 2.24-2.04-2.00-1.97; there are some free intermediary radicals (gr.-2.00) (Fig.2). The NMR spectre shows the presence of ATP (0.22 n moles/g) of inorganic phosphate (16.58 n moles g) (Fig.3). The pulp seems to have a lot of antioxidant factors. The next researches will be to study E, A and C vitamins concentrations. This high presence of GSH and GSSG may be an embryonic peculiarity.

Morphological researches have been carried out through histological studies with fine and semifine sections at the electronic scan microscope on the dental pulp of calves. The general aim was to identify the antioxidant properties of pulp and study cellular density and microfibrillar architecture. The dental pulps of calves of 5-6 months taken immediately after slaughtering were used with immersions into the fixing liquid. The scan microscope showed fibroblasts in the deepest regions of the pulp in a stroma of collagenic fibres, not organised but scattered among the cells of the connective tissue. The odontoblasts presented well ordered one beside the others with well visible details such as the swollen basal portion, the nucleus and a very tight villosity. No clear interodontoblastic connections were evident. The electron transmission microscope revealed typical cells with histiocyte appearance with microfilaments evident in the cytoplasma (myofibroblasts) presenting a wide variety of cytoplasmatic interconnections with interdigitations.
