Pistacia integerrima leaf galls are used in several traditional medicines to cure many diseases such as diarrhea, asthma, fever, cough, vomiting, and hepatitis. The main goal of the present investigation was to assess the antidiarrheal effect of the Pistacia integerrima extracts/fractions and four isolated flavonoid compounds (1-4) on mice. An in vivo assay involving castor-oil-induced diarrhea was used to evaluate the antidiarrheal potential of extracts/fractions at 100, 200, and 400 mg/kg p.o., as well as isolated compounds at 5, 10, and 20 mg/kg p.o. Pretreatment of mice with extracts/fractions significantly attenuated castor-oil-induced diarrhea in a dose-dependent manner. Among all crude extracts and fractions, the ethyl acetate extract was the most effective with 100% protection against diarrhea followed by chloroform (75% protection) at 400 mg/kg p.o. Although all the isolated compounds exhibited strong antidiarrheal activity, isolated compounds 1 and 4 demonstrated 100% protection against diarrhea. Moreover, docking models were performed using the Molecular Operating Environment (MOE) and AutoDock software and suggested that the extracts and isolated compounds exert antidiarrheal activity by inhibiting mu-opioid and delta-opioid receptors. Therefore, our finding affords a strong pharmacological basis for the traditional use of P. integerrima galls in the treatment of diarrhea.

Urinary tract infections (UTIs) are considered to be the most common infections worldwide, having an incidence rate of 40-60% in women. Moreover, the prevalence of this disorder in adult women is 30 times more than in men. UTIs are usually found in many hospitals and clinical practice; as disorders, they are complicated and uncomplicated; in uncomplicated cases, there is no structural or functional abnormality in the urogenital tract. However, obstruction, retention of urine flow and use of catheters increase the complexity. There are several bacteria (e.g., E. coli, Klebsiella pneumoniae, Proteus vulgaris, etc.) successfully residing in the tract. The diagnosis must not only be accurate but rapid, so early detection is an important step in the control of UTIs caused by uropathogens. The treatment of UTIs includes appropriate antimicrobial therapy to control the infection and kill the causal microbes inside the body. A long-time usage of antibiotics has resulted in multidrug resistance causing an impediment in treatment. Thus, alternative, combinatorial medication approaches have given some hope. Available treatments considered Homeopathic, Ayurvedic, Unani, and other herbal-based drugs. There are new upcoming roles of nanoparticles in combating UTIs which needs further validation. The role of medicinal plant-based nanotechnology approaches has shown promising results. Therefore, there must be active research in phyto-based therapies of UTIs, such as Ayurvedic Biology.

Drug development is a key point in the research of new therapeutic treatments for increasing maximum drug loading and prolonged drug effect. Encapsulation of drugs into multivesicular liposomes (DepoFoam) is a nanotechnology that allow delivery of the active constituent at a sufficient concentration during the entire treatment period. This guarantees the reduction of drug administration frequency, a very important factor in a prolonged treatment. Currently, diverse DepoFoam drugs are approved for clinical use against neurological diseases and for post-surgical pain management while other are under development for reducing surgical bleeding and for post-surgical analgesia. Also, on pre-clinical trials on cancer DepoFoam can improve bioavailability and stability of the drug molecules minimizing side effects by site-specific targeted delivery. In the current work, available literature on structure, preparation and pharmacokinetics of DepoFoam are reviewed. Moreover, we investigated approved DepoFoam formulations and preclinical studies with this nanotechnology.

Fruits of Terminalia chebula Retz. (Combretaceae) are widely used as crude drugs in various traditional medicine systems. The aim of this article is to review the available scientific information regarding the traditional uses, bioactive chemical constituents and the pharmacological activities of T. chebula. Numerous researches conducted on T. chebula have confirmed the presence of wide range of the phytochemicals such as flavonoids, tannins, phenolic acids and other bioactive compounds. T. chebula is also widely studied regarding its pharmacological activities such as antioxidant, hepatoprotective, neuroprotective, cytotoxic, antidiabetic, anti-inflammatory activities among others. However, more in vivo and clinical studies for mechanism-based pharmacological evaluation should be conducted in future to provide stronger scientific evidences for their traditional uses.

The present study investigated antioxidant and antibacterial activities of 5 different extracts and derived fractions from the S. nepalense tubers. Identification of the most active fractions, their phytochemical characterization, total phenolic and flavonoid contents, and biological activities were also evaluated. Petroleum ether, chloroform, ethyl acetate, methanol, water extracts and methanol fractions were screened for their antibacterial activity at 10, 50 and 100 mg/mL doses against ten Gram-negative and Gram-positive bacterial strains by disc diffusion method. Their total antioxidant activity was measured by DPPH and ABTS assays. Identification of the main compounds was performed by LC-MS/MS. Methanol extract exhibited the highest antioxidant (IC50= 30.79 µg/mL and 24.53 µg/mL for DPPH and ABTS, respectively) and antibacterial (MIC 71.5 to >100 µg/mL) activities in comparison with the other extracts. Levels of phenolics and flavonoids were also the highest in the same extract, i.e. 19.2 mg GAE/g and 11.20 mg QE/g, respectively. Phytochemical investigation of the active fractions of the methanol extract led to the isolation of gallic acid (19.04 mg/g) and quercetin (23.4 mg/g). Therefore, methanol extract showed an interesting potential for both antioxidant and antibacterial activities, thus deserving attention for future applications in the fields of medicinal plants and food supplements.

Uttarakhand Himalayan region holds Asteraceae or Compositae as the largest family of flowering, medicinal and aromatic plants. Species belonging to this family rises from low altitude to the alpine region. Among Asteraceae, Saussurea obvallata (DC.) Edgew. is widely used in several indigenous systems of medicine. Flowers, leaves and rhizomes of S. obvallata are used for several traditional, religious, therapeutic and ornamental purposes. Aim of this study was to determine the chemical composition and antibacterial efficacy of petroleum ether extract (PEE) of S. obvallata. Gas chromatography-mass spectrometry (GC-MS) analysis was used for identifying phytochemicals present in the plant extract. Furthermore, the PEE was assessed for in-vitro antibacterial activity against selected Gram positive and negative strains. Structure of squalene and α-linolenic acid methyl ester were identified in PEE by GC-MS analysis, by comparing the results obtained with NIST library and literature reports. PEE exhibited significant activity against Staphylococcus aureus, Escherichia coli, Bacillus cereus, Bacillus subtilis with IC50 value of 87.2 ± 1.6, 98.4 ± 1.1 and 90.2 ± 1.8 µg/ml, respectively. These results showed that squalene and α-linolenic acid derivative identified in S. obvallata may be responsible for the observed antibacterial activity. To the best of our knowledge, this is the first report focused on the chemical composition and antibacterial activity of S. obvallata.

Programmed cell death (PCD) is probably the most widely discussed subject among the topics of cancer therapy. Over the last 2 decades an astonishing boost in our perception of cell death has been seen, and its role in cancer and cancer therapy has been thoroughly investigated. A number of discoveries have clarified the molecular mechanism of PCD, thus expounding the link between PCD and therapeutic tools. Even though PCD is assumed to play a major role in anticancer therapy, the clinical relevance of its induction remains uncertain. Since PCD involves multiple death programs including programmed necrosis and autophagic cell death, it has contributed to our better understanding of cancer pathogenesis and therapeutics. In this review, we discuss a brief outline of PCD types as well as their role in cancer therapeutics. Since irregularities in the cell death process are frequently found in various cancers, key proteins governing cell death type could be used as therapeutic targets for a wide range of cancer.
