RESUMO
We have performed screening tests for imidazo[1,2-a]azepine derivatives by studying their antispasmodic activity on the isolated strips of urinary bladder of female rats. Test compounds showed the ability to reduce the basal tone of bladder stripes on the background of hyperpotassium constriction (40 mM KCl).

The effects of four new fluorine-containing pinacidyl analogs (FPs) on the bladder contractile function in vitro and in vivo, and on the vessel tone in vitro were examined. All the four drugs produced a concentration-dependent relaxation of isolated rat detrusor strips and isolated aorta rings. The contractility inhibition effect of compound FP-5 was decreased by preliminary glibenclamide perfusion. This compound also effectively inhibited bladder contractility function in vivo.

The literature data on the effect of histamine on the heart and vessels in different species of animals and man were analyzed. Nonheterogeneity of histamine effect can be presumably explained by a strong representation of various types of receptors to which this biogenic amine is bound (H1, H2, H3) in the organs and tissues, their unequal location on the pre- and postsynaptic membrane, the differences in their physiological functions.

The present day data concerning biosynthesis, storage, release and inactivation of histamine in the brain of mammals are given. The possibility to regulate histamine of the action of physiologically active substances is discussed.

Histamine was found to suppress the myocardial contractile function, to decrease systemic arterial blood pressure and to exert a positive chronotropic effect in unanesthetized dogs. A preliminary blockade of H1-receptors prevented the hypotensive effect of histamine, blockade of H2-receptors prevented the development of the positive chronotropic effect.

In experiments on rats with different models of pathology it was demonstrated that a new non-steroidal anti-inflammatory drug pyrimidant exerts pronounced anti-inflammatory, analgesic and antipyretic effects. In some cases its activity exceeds that of indomethacin, brufen, butadione, mefenamic acid, sodium salicylate.
