RESUMO

South Africa has a rich history of medicinal plant species and their documented uses as traditional medicines, and is also home to three well-known, blue-flowered sage species of ethnobotanical importance. The Namaqualand bloublomsalie (Salvia dentata) has so far remained unstudied and apparently overlooked. Our study is the first to report on the essential oil chemistry of this medicinally relevant species and provide a comparison with the other two (well-studied) closely related Cape bloublomsalies (Salvia africana and S. chamelaeagnea). The data, generated from three geographically isolated populations comprised of 13 individual plants of S. dentata, revealed diagnostically high levels of camphor (14.37%), α-pinene (11.43%), camphene (10.18%), 1,8-cineole (eucalyptol) (9.42%) and bornyl acetate (8.56%) which provide a distinct chemical profile from the other two species.

Vineyard exposure to wildfire smoke can taint grapes and wine. To understand the impact of this taint, it is imperative that the analytical methods used are accurate and precise. This study compared the variance across nine commercial and research laboratories following quantitative analysis of the same set of smoke-tainted wines. In parallel, correlations between the interlaboratory consensus values for smoke-taint markers and sensory analyses of the same smoke-tainted wines were evaluated. For free guaiacol, the mean accuracy was 94 ± 11% in model wine, while the free cresols and 4-methylguaiacol showed a negative bias and/or decreased precision relative to guaiacol. Similar trends were observed in smoke-tainted wines, with the cresols and glycosidically bound markers demonstrating high variance. Collectively, the interlaboratory results show that data from a single laboratory can be used quantitatively to understand smoke-taint. Results from different laboratories, however, should not be directly compared due to the high variance between study participants. Correlations between consensus compositional data and sensory evaluations suggest the risk of perceivable smoke-taint can be predicted from free cresol concentrations, overcoming limitations associated with the occurrence of some volatile phenols, guaiacol in particular, as natural constituents of some grape cultivars and of the oak used for barrel maturation.

Variation in plant species and extraction solvents play a crucial role in the recovery of their bioactive compounds and antifungal efficacy. Thus, in this study, a comparative investigation was carried out using extraction solvents: 70% acetone and 95% ethanol to obtain crude aqueous extracts from Helichrysum odoratissimum and H. patulum. Crude aqueous extracts were screened using gas chromatography-mass spectrometry (GC-MS), to gain insight into their chemical composition. Phytochemical properties (total polyphenols (TP) and radical scavenging capacity via 2,2-diphenyl-1-picrylhydrazyl (DPPH)), and antifungal activity against Botrytis cinerea of the crude extracts were evaluated. Fungicide (Rovral® WP) and extraction solvents were used as controls. Variation in Helichrysum spp. and extraction solvent had influence on the chemical composition, phytochemicals, and antifungal activities. Metabolites such as γ-terpinene (≈0.1%), α-amorphene (≈0.6%) α-gurjunene (≈1.4%), ß-selinene (2.2-3.2%), γ-gurjunene (≈3.3%), and methyl cinnamate (≈20%) were detected only in extracts of H. patulum. Crude extract of H. odoratissimum using 70% acetone had the highest TP (19.3 ± 0.76 g GA 100 g-1), and DPPH capacity (13,251.5 ± 700.55 µmol Trolox g-1) compared to H. patulum (p ≤ 0.05). Ethanolic extracts of H. patulum showed highest antifungal efficacy (≈65%) against B. cinerea (p ≤ 0.05) compared to other crude extracts. This study showed that Helichrysum spp. differ in their potential as a source for bioactive compounds and antifungal treatments/formulations.

Knowledge regarding the potential impacts of crude oil on endocrine signaling in freshwater aquatic vertebrates is limited. The expression of selected genes as biomarkers for altered endocrine signaling was studied in African clawed frog, Xenopus laevis, tadpoles and juvenile Mozambique tilapia, Oreochromis mossambicus, exposed to weathered bunker and unweathered refinery crude oil water accommodated fractions (WAFs). In addition, the expression of the aforementioned genes was quantified in X. laevis tadpoles exposed to surface water collected from the proximity of an underground oil bunker. The (anti)estrogenicity and (anti)androgenicity of crude oil, crude oil WAFs, and surface water were furthermore evaluated using recombinant yeast. Thyroid hormone receptor beta expression was significantly down-regulated in X. laevis in response to both oil WAF types, whereas a further thyroid linked gene, type 2 deiodinase, was up-regulated in O. mossambicus exposed to a high concentration of bunker oil WAF. In addition, both WAFs altered the expression of the adipogenesis-linked peroxisome proliferator-activated receptor gamma in X. laevis. The crude oil and WAFs exhibited antiestrogenic and antiandrogenic activity in vitro. However, O. mossambicus androgen receptor 2 was the only gene, representing the reproductive system, significantly affected by WAF exposure. Estrogenicity, antiestrogenicity, and antiandrogenicity were detected in surface water samples; however, no significant changes were observed in the expression of any of the genes evaluated in X. laevis exposed to surface water. The responses varied among the 2 model organisms used, as well as among the 2 types of crude oil. Nonetheless, the data provide evidence that crude oil pollution may lead to adverse health effects in freshwater fish and amphibians as a result of altered endocrine signaling.
