RESUMO

Single-Photon Avalanche Diode (SPAD) direct Time-of-Flight (dToF) sensors provide depth imaging over long distances, enabling the detection of objects even in the absence of contrast in colour or texture. However, distant objects are represented by just a few pixels and are subject to noise from solar interference, limiting the applicability of existing computer vision techniques for high-level scene interpretation. We present a new SPAD-based vision system for human activity recognition, based on convolutional and recurrent neural networks, which is trained entirely on synthetic data. In tests using real data from a 64×32 pixel SPAD, captured over a distance of 40 m, the scheme successfully overcomes the limited transverse resolution (in which human limbs are approximately one pixel across), achieving an average accuracy of 89% in distinguishing between seven different activities. The approach analyses continuous streams of video-rate depth data at a maximal rate of 66 FPS when executed on a GPU, making it well-suited for real-time applications such as surveillance or situational awareness in autonomous systems.

RESUMO

Differentiation of stem and progenitor cells is a highly regulated process that involves the coordinated action of multiple layers of regulation. Here we show how the post-transcriptional regulatory layer instructs the level of chromatin regulation via miR-144 and its targets to orchestrate chromatin condensation during erythropoiesis. The loss of miR-144 leads to impaired chromatin condensation during erythrocyte maturation. Among the several targets of miR-144 that influence chromatin organization, the miR-144-dependent regulation of Hmgn2 is conserved from fish to humans. Our genetic probing of the miR-144/Hmgn2 regulatory axis establish that intact miR-144 target sites in the Hmgn2 3'UTR are necessary for the proper maturation of erythrocytes in both zebrafish and human iPSC-derived erythroid cells while loss of Hmgn2 rescues in part the miR-144 null phenotype. Altogether, our results uncover miR-144 and its target Hmgn2 as the backbone of the genetic regulatory circuit that controls the terminal differentiation of erythrocytes in vertebrates.

RESUMO

The proto-oncogene c-MYC is a key representative of the MYC transcription factor network regulating growth and metabolism. MML-1 (Myc- and Mondo-like) is its homolog in C. elegans. The functional and molecular cooperation between c-MYC and H3 lysine 79 methyltransferase DOT1L was demonstrated in several human cancer types, and we have earlier discovered the connection between C. elegans MML-1 and DOT-1.1. Here, we demonstrate the critical role of DOT1L/DOT-1.1 in regulating c-MYC/MML-1 target genes genome-wide by ensuring the removal of "spent" transcription factors from chromatin by the nuclear proteasome. Moreover, we uncover a previously unrecognized proteolytic activity of DOT1L, which may facilitate c-MYC turnover. This new mechanism of c-MYC regulation by DOT1L may lead to the development of new approaches for cancer treatment.

RESUMO

3D time-of-flight (ToF) image sensors are used widely in applications such as self-driving cars, augmented reality (AR), and robotics. When implemented with single-photon avalanche diodes (SPADs), compact, array format sensors can be made that offer accurate depth maps over long distances, without the need for mechanical scanning. However, array sizes tend to be small, leading to low lateral resolution, which combined with low signal-to-background ratio (SBR) levels under high ambient illumination, may lead to difficulties in scene interpretation. In this paper, we use synthetic depth sequences to train a 3D convolutional neural network (CNN) for denoising and upscaling (×4) depth data. Experimental results, based on synthetic as well as real ToF data, are used to demonstrate the effectiveness of the scheme. With GPU acceleration, frames are processed at >30 frames per second, making the approach suitable for low-latency imaging, as required for obstacle avoidance.

Single-Photon Avalanche Detector (SPAD) arrays are a rapidly emerging technology. These multi-pixel sensors have single-photon sensitivities and pico-second temporal resolutions thus they can rapidly generate depth images with millimeter precision. Such sensors are a key enabling technology for future autonomous systems as they provide guidance and situational awareness. However, to fully exploit the capabilities of SPAD array sensors, it is crucial to establish the quality of depth images they are able to generate in a wide range of scenarios. Given a particular optical system and a finite image acquisition time, what is the best-case depth resolution and what are realistic images generated by SPAD arrays? In this work, we establish a robust yet simple numerical procedure that rapidly establishes the fundamental limits to depth imaging with SPAD arrays under real world conditions. Our approach accurately generates realistic depth images in a wide range of scenarios, allowing the performance of an optical depth imaging system to be established without the need for costly and laborious field testing. This procedure has applications in object detection and tracking for autonomous systems and could be easily extended to systems for underwater imaging or for imaging around corners.

RESUMO

Objective: The present study aimed to evaluate the effect of global postural re-education (GPR) on sleep quality and stress in female health science lecturers. Methods: A total of 24 female university lecturers were allocated in this randomized controlled trial into intervention (n = 12) and control (n = 12) groups. The intervention group underwent familiarization and training on the therapy to execute an 8-week treatment with GPR. Data collected on sleep quality were analyzed using 24-h accelerometry (Actigraphy GT3X+) in addition to the Pittsburgh Sleep Quality Index (PSQI) questionnaire score as well as the State-Trait Anxiety Inventory (STAI) anxiety questionnaire. Data on stress were collected by measuring salivary cortisol. Results: After comparing the GPR of the groups, there was a main effect of the group (F = 5.278, p = 0.044) for PSQI. The post-hoc analysis revealed that both groups decreased scores between pre- and post-treatment. Additionally, post-treatment, there were differences between groups. For sleep latency, there were main effects of group (F = 6.118, p = 0.031) and score × group (F = 9.343, p = 0.011) interactions. The post-hoc analysis revealed that treatment groups decreased scores between pre- and post-treatment, and there were differences between groups (all p < 0.050). Conclusion: The self-administered GPR improves sleep quality in female university lecturers, providing a valuable self-regulation tool for enhanced sleep quality and enhanced academic performance. Further study may help to develop this as a potential tool to help university lecturers' job performance.

RESUMO

Single-photon-sensitive depth sensors are being increasingly used in next-generation electronics for human pose and gesture recognition. However, cost-effective sensors typically have a low spatial resolution, restricting their use to basic motion identification and simple object detection. Here, we perform a temporal to spatial mapping that drastically increases the resolution of a simple time-of-flight sensor, i.e., an initial resolution of 4 × 4 pixels to depth images of resolution 32 × 32 pixels. The output depth maps can then be used for accurate three-dimensional human pose estimation of multiple people. We develop a new explainable framework that provides intuition to how our network uses its input data and provides key information about the relevant parameters. Our work greatly expands the use cases of simple single-photon avalanche detector time-of-flight sensors and opens up promising possibilities for future super-resolution techniques applied to other types of sensors with similar data types, i.e., radar and sonar.

ABSTRACT The building that was built with the name of Nicolás Salvador Acea de los Ríos in the province of Las Villas is currently the physical space of "Paquito González Cueto" University Pediatric Hospital of Cienfuegos territory. That is why, we carried out a literature review using induction-deduction methods; analysis and synthesis and historical-logical method with the aim of enunciating the contribution of Nicolás Acea as benefactor of Cienfuegos in the construction of the elder's home of the same name after he died. Although there are no longer free spaces between the building and the sea walk, nor the breeze of the nearby sea that impacted fully on its façade, benefits that endorsed the construction; after the revolutionary triumph on 1st. of January 1959 changes its social purpose, but it is maintained as a vocation the health care, now of infants and adolescents of the territory.

RESUMO

3D time-of-flight (ToF) imaging is used in a variety of applications such as augmented reality (AR), computer interfaces, robotics and autonomous systems. Single-photon avalanche diodes (SPADs) are one of the enabling technologies providing accurate depth data even over long ranges. By developing SPADs in array format with integrated processing combined with pulsed, flood-type illumination, high-speed 3D capture is possible. However, array sizes tend to be relatively small, limiting the lateral resolution of the resulting depth maps and, consequently, the information that can be extracted from the image for applications such as object detection. In this paper, we demonstrate that these limitations can be overcome through the use of convolutional neural networks (CNNs) for high-performance object detection. We present outdoor results from a portable SPAD camera system that outputs 16-bin photon timing histograms with 64×32 spatial resolution, with each histogram containing thousands of photons. The results, obtained with exposure times down to 2 ms (equivalent to 500 FPS) and in signal-to-background (SBR) ratios as low as 0.05, point to the advantages of providing the CNN with full histogram data rather than point clouds alone. Alternatively, a combination of point cloud and active intensity data may be used as input, for a similar level of performance. In either case, the GPU-accelerated processing time is less than 1 ms per frame, leading to an overall latency (image acquisition plus processing) in the millisecond range, making the results relevant for safety-critical computer vision applications which would benefit from faster than human reaction times.

RESUMO

AIMS: The aim of the LAICA study was to evaluate the long-term effectiveness and safety of intermittent levosimendan infusion in patients with advanced heart failure (AdHF). METHODS AND RESULTS: This was a multicentre, randomized, double-blind, placebo-controlled clinical trial of intermittent levosimendan 0.1 µg/kg/min as a continuous 24-h intravenous infusion administered once monthly for 1 year in patients with AdHF. The primary endpoint [incidence of rehospitalization (admission to the emergency department or hospital ward for >12 h) for acute decompensated HF or clinical deterioration of the underlying HF] occurred in 23/70 (33%) of the levosimendan group (Group I) and 12/27 (44%) of the placebo group (Group II) (P = 0.286). The incidence of hospital readmissions for acute decompensated HF (Group I vs. Group II) at 1, 3, 6, and 12 months was 4.2% vs. 18.2% (P = 0.036); 12.8% vs. 33.3% (P = 0.02); 25.7% vs. 40.7% (P = 0.147); 32.8% vs. 44.4% (P = 0.28), respectively. In a secondary pre-specified time-to-event analysis no differences were observed in admission for acute decompensated HF between patients treated with levosimendan compared with placebo (hazard ratio 0.66; 95% CI, 0.32-1.32; P = 0.24). Cumulative incidence for the aggregated endpoint of acute decompensation of HF and/or death at 1 and 3 months were significatively lower in the levosimendan group than in placebo group [5.7% vs. 25.9% (P = 0.004) and 17.1% vs. 48.1% (P = 0.001), respectively], but not at 6 and 12 months [34.2% vs. 59.2% (P = 0.025); 41.4% vs. 66.6% (P = 0.022), respectively]. Survival probability was significantly higher in patients who received levosimendan compared with those who received placebo (log rank: 4.06; P = 0.044). There were no clinically relevant differences in tolerability between levosimendan and placebo and no new safety signals were observed. CONCLUSIONS: In our study, intermittent levosimendan in patients with AdHF produced a statistically non-significant reduction in the incidence of hospital readmissions for acute decompensated HF, a significantly lower cumulative incidence of acute decompensation of HF and/or death at 1 and 3 month of treatment and a significant improvement in survival during 12 months of treatment.

RESUMO

RNA provides the framework for the assembly of some of the most intricate macromolecular complexes within the cell, including the spliceosome and the mature ribosome. The assembly of these complexes relies on the coordinated association of RNA with hundreds of trans-acting protein factors. While some of these trans-acting factors are RNA-binding proteins (RBPs), others are adaptor proteins, and others still, function as both. Defects in the assembly of these complexes results in a number of human pathologies including neurodegeneration and cancer. Here, we demonstrate that Silencing Defective 2 (SDE2) is both an RNA binding protein and also a trans-acting adaptor protein that functions to regulate RNA splicing and ribosome biogenesis. SDE2 depletion leads to widespread changes in alternative splicing, defects in ribosome biogenesis and ultimately complete loss of cell viability. Our data highlight SDE2 as a previously uncharacterized essential gene required for the assembly and maturation of the complexes that carry out two of the most fundamental processes in mammalian cells.

RESUMO

Human transmission of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), causative pathogen of the COVID-19 pandemic, exerts a massive health and socioeconomic crisis. The virus infects alveolar epithelial type 2 cells (AT2s), leading to lung injury and impaired gas exchange, but the mechanisms driving infection and pathology are unclear. We performed a quantitative phosphoproteomic survey of induced pluripotent stem cell-derived AT2s (iAT2s) infected with SARS-CoV-2 at air-liquid interface (ALI). Time course analysis revealed rapid remodeling of diverse host systems, including signaling, RNA processing, translation, metabolism, nuclear integrity, protein trafficking, and cytoskeletal-microtubule organization, leading to cell cycle arrest, genotoxic stress, and innate immunity. Comparison to analogous data from transformed cell lines revealed respiratory-specific processes hijacked by SARS-CoV-2, highlighting potential novel therapeutic avenues that were validated by a high hit rate in a targeted small molecule screen in our iAT2 ALI system.
