OBJECTIVES: This pilot study aims to explore the potential of a positive psychology intervention (PPI) in enhancing the subjective well-being of older individuals with amnestic mild cognitive impairment (MCI), a precursor to dementia. DESIGN AND SETTING: A randomized trial was conducted, initially recruiting 51 participants aged 65 and above from the Greek Association of Alzheimer's Disease and Related Disorders in Thessaloniki, Greece. The study employed a control-experimental group setup. To ensure randomization, each participant was assigned a unique number, and a random number generator was used for group allocation. PARTICIPANTS: A total of 41 eligible participants with amnestic mild cognitive impairment (MCI) were included in the study after screening. INTERVENTION: The intervention consisted of a 3-week positive psychology program (PPI) where the PERMA Profiler questionnaire was administered at three intervals: pre-intervention, post-intervention, and one month after completion. MAIN OUTCOME MEASURES: The subjective well-being of participants. RESULTS: The analysis, conducted mainly through mixed-measures ANOVAs, supported the study's hypotheses, revealing that the 3-week PPI led to increased PERMA model scores and overall well-being, which persisted even after one month. Conversely, non-participants experienced declines in most domains except for Positive Emotion and Meaning, which demonstrated improvement and recovery during follow-up. CONCLUSIONS: These findings suggest the potential of PPI in enhancing the subjective well-being of older adults with amnestic MCI, with implications for addressing dementia-related challenges. Further investigation is warranted to pinpoint PPI effects on MCI and tailor interventions for improved subjective well-being.

Alzheimer's disease (AD) is a rapidly growing disease that affects millions of people worldwide, therefore there is an urgent need for its early diagnosis and treatment. A huge amount of research studies are performed on possible accurate and reliable diagnostic biomarkers of AD. Due to its direct contact with extracellular space of the brain, cerebrospinal fluid (CSF) is the most useful biological fluid reflecting molecular events in the brain. Proteins and molecules that reflect the pathogenesis of the disease, e.g., neurodegeneration, accumulation of Abeta, hyperphosphorylation of tau protein and apoptosis may be used as biomarkers. The aim of the current manuscript is to present the most commonly used CSF biomarkers for AD as well as novel biomarkers. Three CSF biomarkers, namely total tau, phospho-tau and Abeta42, are believed to have the highest diagnostic accuracy for early AD diagnosis and the ability to predict AD development in mild cognitive impairment (MCI) patients. Moreover, other biomarkers such as soluble amyloid precursor protein (APP), apoptotic proteins, secretases and inflammatory and oxidation markers are believed to have increased future prospects.

BACKGROUND: Destination memory defined as the ability to remember to whom we addressed a piece of information is found to be impaired in normal aging. Theories of affect development and research findings have shown that emotional charging improves performance on memory tasks, and also that Mu rhythm is desynchronized as an index of mirror neuron activation during such tasks. OBJECTIVE: In this paper, we sought to investigate the differences in Mu rhythm during an emotional destination memory task, between younger and older adults. METHODS: 16 cognitively normal older adults, recruited from Alzheimer's disease day center and 16 young adults, recruited via advertisements, participated in this experimental study. We investigated destination memory of emotionally charged faces (Emotional Destination Memory, EDM) while applying electroencephalograph (EEG) in real time in young versus older adults. We measured Mu rhythm in frontal, fronto-temporal and central areas. EEG data has been pre-processed, segmented in non-overlapping epochs, and independent component analysis (ICA) has been conducted to reject artifacts. RESULTS: Results showed that young adults performed better than older adults in remembering facts associated with angry faces. Also, different neurophysiological activation was found, with older adults showing Mu suppression in frontal and fronto-temporal regions, specifically in F3, F7 and F8 electrodes, in contrast with young adults who showed Mu enhancement. Regarding within group differences, it was found that in the older adults group, electrodes F8 and central C3 were the most activated, while in the young adults group, C3 was the most activated electrode. CONCLUSION: The findings suggest better behavioral performance of young adults as a result of better cognitive state and adaptive bias. On a neurophysiological level, it is suggested that older adults employ Mu suppression, thus possible activation of mirror neurons, as a compensatory mechanism while mirroring properties are not spontaneously activated in young adults.

BACKGROUND: Cognitive disorders such as dementia are common among older adults admitted to general hospitals. They can complicate treatment leading to longer hospitalization and worse outcomes. They often remain underdiagnosed as the busy routine of the hospital does not enable efficient screening and available screening instruments are not suitable for the hospital environment. Computerized cognitive testing (CCT) has been proposed as an efficient screening method as it can be employed by nonspecialists, such as nurses while featuring automatic scoring and interpretation of results. OBJECTIVE: This study validated a newly developed questionnaire for measuring the attitudes of Greek nurses towards computerized dementia screening. MATERIAL AND METHODS: The questionnaire was validated in a sample of 212 undergraduate psychology students and subsequently administered to a sample of 19 nurses working in a general hospital. Reliability of the questionnaire was calculated using Cronbach's alpha (= 0.762). Factor analysis revealed the existence of a single factor (acceptability-feasibility) that accounted for 33.73% of variance with an eigenvalue of 3.036. RESULTS: The total score of all the items loading on the single factor (acceptability-feasibility) was calculated. Scores ranged between 10 and 40 with the average score for the validation group being 29.33 (SD = 4.89) and the average score for the nurses' group being 29.50 (SD = 3.20). DISCUSSION: The questionnaire has acceptable reliability. Results indicate that acceptability-feasibility is high in both groups and there were no statistically significant differences between the two groups.
