RESUMO

Objective.Measurement of the time-of-flight (TOF) difference of each coincident pair of photons increases the effective sensitivity of positron emission tomography (PET). Many authors have analyzed the benefit of TOF for quantification and hot spot detection in the reconstructed activity images. However, TOF not only improves the effective sensitivity, it also enables the joint reconstruction of the tracer concentration and attenuation images. This can be used to correct for errors in CT- or MR-derived attenuation maps, or to apply attenuation correction without the help of a second modality. This paper presents an analysis of the effect of TOF on the variance of the jointly reconstructed attenuation and (attenuation corrected) tracer concentration images.Approach.The analysis is performed for PET systems that have a distribution of possibly non-Gaussian TOF-kernels, and includes the conventional Gaussian TOF-kernel as a special case. Non-Gaussian TOF-kernels are often observed in novel detector designs, which make use of two (or more) different mechanisms to convert the incoming 511 keV photon to optical photons. The analytical result is validated with a simple 2D simulation.Main results.We show that if two different TOF-kernels are equivalent for image reconstruction with known attenuation, then they are also equivalent for joint reconstruction of the activity and the attenuation images. The variance increase in the activity, caused by also jointly reconstructing the attenuation image, vanishes when the TOF-resolution approaches perfection.Significance.These results are of interest for PET detector development and for the development of stand-alone PET systems.

We recently proposed a new approach for the real-time monitoring of particle therapy treatments with the goal of achieving high sensitivities on the particle range measurement already at limited counting statistics. This method extends the Prompt Gamma (PG) timing technique to obtain the PG vertex distribution from the exclusive measurement of particle Time-Of-Flight (TOF). It was previously shown, through Monte Carlo simulation, that an original data reconstruction algorithm (Prompt Gamma Time Imaging) allows to combine the response of multiple detectors placed around the target. The sensitivity of this technique depends on both the system time resolution and the beam intensity. At reduced intensities (Single Proton Regime-SPR), a millimetric proton range sensitivity can be achieved, provided the overall PG plus proton TOF can be measured with a 235 ps (FWHM) time resolution. At nominal beam intensities, a sensitivity of a few mm can still be obtained by increasing the number of incident protons included in the monitoring procedure. In this work we focus on the experimental feasibility of PGTI in SPR through the development of a multi-channel, Cherenkov-based PG detector with a targeted time resolution of 235 ps (FWHM): the TOF Imaging ARrAy (TIARA). Since PG emission is a rare phenomenon, TIARA design is led by the concomitant optimisation of its detection efficiency and Signal to Noise Ratio (SNR). The PG module that we developed is composed of a small PbF[Formula: see text] crystal coupled to a silicon photoMultiplier to provide the time stamp of the PG. This module is currently read in time coincidence with a diamond-based beam monitor placed upstream the target/patient to measure the proton time of arrival. TIARA will be eventually composed of 30 identical modules uniformly arranged around the target. The absence of a collimation system and the use of Cherenkov radiators are both crucial to increase the detection efficiency and the SNR, respectively. A first prototype of the TIARA block detector was tested with 63 MeV protons delivered from a cyclotron: a time resolution of 276 ps (FWHM) was obtained, resulting in a proton range sensitivity of 4 mm at 2[Formula: see text] with the acquisition of only 600 PGs. A second prototype was also evaluated with 148 MeV protons delivered from a synchro-cyclotron obtaining a time resolution below 167 ps (FWHM) for the gamma detector. Moreover, using two identical PG modules, it was shown that a uniform sensitivity on the PG profiles would be achievable by combining the response of gamma detectors uniformly distributed around the target. This work provides the experimental proof-of-concept for the development of a high sensitivity detector that can be used to monitor particle therapy treatments and potentially act in real-time if the irradiation does not comply to treatment plan.

Post-anthesis phosphorus (P) uptake and the remobilization of the previously acquired P are the principal sources of grain P nutrition in wheat. However, how the acquired P reaches the grains and its partitioning at the whole plant level remain poorly understood. Here, the temporal dynamics of the newly acquired P in durum wheat organs and its allocation to grain were examined using pulse-chase 32P-labeling experiments at 5 and 14 days after anthesis. Durum wheat plants were grown hydroponically under high and low P supplies. Each labeling experiment lasted for 24 h. Plants were harvested 24, 48, and 96 h after labeling. Low and high P treatments significantly affected the allocation of the newly acquired P at the whole plant level. Three days (96 h) after the first 32P-labeling, 8% and 4% of the newly acquired P from exogenous solution were allocated to grains, 73% and 55% to the remainder aboveground organs, and 19% and 41% to the roots at low and high P supplies, respectively. Three days after the second labeling, the corresponding values were 48% and 20% in grains, 44% and 53% in the remainder aboveground organs, and 8% and 27% in roots at low and high P supplies, respectively. These results reveal that the dynamics of P allocation to grain was faster in plants grown under low P supply than under high supply. However, the obtained results also indicate that the origin of P accumulated in durum wheat grains was mainly from P remobilization with little contribution from post-anthesis P uptake. The present study emphasizes the role of vegetative organs as temporary storage of P taken up during the grain filling period before its final allocation to grains.

We propose a novel prompt-gamma (PG) imaging modality for real-time monitoring in proton therapy: PG time imaging (PGTI). By measuring the time-of-flight (TOF) between a beam monitor and a PG detector, our goal is to reconstruct the PG vertex distribution in 3D. In this paper, a dedicated, non-iterative reconstruction strategy is proposed (PGTI reconstruction). Here, it was resolved under a 1D approximation to measure a proton range shift along the beam direction. In order to show the potential of PGTI in the transverse plane, a second method, based on the calculation of the centre of gravity (COG) of the TIARA pixel detectors' counts was also explored. The feasibility of PGTI was evaluated in two different scenarios. Under the assumption of a 100 ps (rms) time resolution (achievable in single proton regime), MC simulations showed that a millimetric proton range shift is detectable at 2σwith 108incident protons in simplified simulation settings. With the same proton statistics, a potential 2 mm sensitivity (at 2σwith 108incident protons) to beam displacements in the transverse plane was found using the COG method. This level of precision would allow to act in real-time if the treatment does not conform to the treatment plan. A worst case scenario of a 1 ns (rms) TOF resolution was also considered to demonstrate that a degraded timing information can be compensated by increasing the acquisition statistics: in this case, a 2 mm range shift would be detectable at 2σwith 109incident protons. By showing the feasibility of a time-based algorithm for the reconstruction of the PG vertex distribution for a simplified anatomy, this work poses a theoretical basis for the future development of a PG imaging detector based on the measurement of particle TOF.

RESUMO

Built on top of the Geant4 toolkit, GATE is collaboratively developed for more than 15 years to design Monte Carlo simulations of nuclear-based imaging systems. It is, in particular, used by researchers and industrials to design, optimize, understand and create innovative emission tomography systems. In this paper, we reviewed the recent developments that have been proposed to simulate modern detectors and provide a comprehensive report on imaging systems that have been simulated and evaluated in GATE. Additionally, some methodological developments that are not specific for imaging but that can improve detector modeling and provide computation time gains, such as Variance Reduction Techniques and Artificial Intelligence integration, are described and discussed.

Phosphorus (P) in durum wheat grains after anthesis originates from either the external P source or the internal remobilization of P from different plant organs. The supply of P and its use by the plant are important factors that can affect the contribution of each source to grain P nutrition. Thus, this experiment aimed to quantify the origin of P in grains of durum wheat plants with different P nutritional status. Wheat plants were grown from juvenile stages to maturity in complete nutrient solutions with either high (0.125 mM) or low (0.025 mM) P concentrations in greenhouse conditions. Phosphorus in nutrient solutions was spiked by introducing 32P after anthesis to quantify the external P uptake and its partitioning within plant organs (spikelets, leaves, stems, roots, and post-anthesis tillers) and grains. Phosphorus use efficiency in durum wheat plants was also determined. The low and high P supply resulted in two highly different plant nutritional P status. Plants with low P status remobilized most of their stored P in all organs and allocated more than 72% of post-anthesis P uptake to grain P nutrition, whereas in the high P plants this was only 56%. Enhanced remobilization of P and the efficient allocation of newly acquired P to grains were crucial for durum wheat grain P nutrition grown under low P supply. The remobilization of P represented 81% of grain P in low P plants while it represented 65% for high P plants. Organs that contributed the most to P remobilization in low P plants were spikelets (43%) and leaves (35%). The post-anthesis tiller development was reduced in low P plants suggesting a preferential allocation of P to grains under this treatment. We concluded that P loading into grains in durum wheat is mainly derived from the remobilization of internal P sources stored before anthesis, even at high external P supply during grain filling.

Since the seventies, positron emission tomography (PET) has become an invaluable medical molecular imaging modality with an unprecedented sensitivity at the picomolar level, especially for cancer diagnosis and the monitoring of its response to therapy. More recently, its combination with x-ray computed tomography (CT) or magnetic resonance (MR) has added high precision anatomic information in fused PET/CT and PET/MR images, thus compensating for the modest intrinsic spatial resolution of PET. Nevertheless, a number of medical challenges call for further improvements in PET sensitivity. These concern in particular new treatment opportunities in the context personalized (also called precision) medicine, such as the need to dynamically track a small number of cells in cancer immunotherapy or stem cells for tissue repair procedures. A better signal-to-noise ratio (SNR) in the image would allow detecting smaller size tumours together with a better staging of the patients, thus increasing the chances of putting cancer in complete remission. Moreover, there is an increasing demand for reducing the radioactive doses injected to the patients without impairing image quality. There are three ways to improve PET scanner sensitivity: improving detector efficiency, increasing geometrical acceptance of the imaging device and pushing the timing performance of the detectors. Currently, some pre-localization of the electron-positron annihilation along a line-of-response (LOR) given by the detection of a pair of annihilation photons is provided by the detection of the time difference between the two photons, also known as the time-of-flight (TOF) difference of the photons, whose accuracy is given by the coincidence time resolution (CTR). A CTR of about 10 picoseconds FWHM will ultimately allow to obtain a direct 3D volume representation of the activity distribution of a positron emitting radiopharmaceutical, at the millimetre level, thus introducing a quantum leap in PET imaging and quantification and fostering more frequent use of 11C radiopharmaceuticals. The present roadmap article toward the advent of 10 ps TOF-PET addresses the status and current/future challenges along the development of TOF-PET with the objective to reach this mythic 10 ps frontier that will open the door to real-time volume imaging virtually without tomographic inversion. The medical impact and prospects to achieve this technological revolution from the detection and image reconstruction point-of-views, together with a few perspectives beyond the TOF-PET application are discussed.

OBJECTIVE: Cochlear obliteration after vestibular schwannoma excision has been noted, with implications on cochlear implantation. Early postoperative cochlear enhancement with gadolinium on magnetic resonance imaging (MRI) has also been observed. Timing of enhancement and association with obliteration is described here. STUDY DESIGN: Retrospective case review. SETTING: Tertiary referral center, ambulatory. PATIENTS: Patients receiving vestibular schwannoma excision surgery by the senior author performed at one institution between January 2015 and July 2017 with postoperative MRIs INTERVENTION:: Diagnostic. MAIN OUTCOME MEASURE(S): The imaging characteristics on postoperative MRIs examined were loss of fluid signal on postoperative T2 images and cochlear enhancement on gadolinium enhanced T1 images. In the patients receiving labyrinthine sparing procedures, presence of postoperative hearing was evaluated. RESULTS: Of the 42 patients evaluated, 24 received the translabyrinthine approach and 18 received a labyrinth sparing surgery. Twenty-nine had evidence of cochlear enhancement on T1 with gadolinium contrast, and 27 had evidence of cochlear obliteration on T2 images. The odds ratio of patients with cochlear enhancement having obliteration was 30.0:1 (p < 0.0001). Intense cochlear enhancement (n = 21) appeared a median of 163 days after surgery, and complete or near complete obliteration (n = 18) appeared a median of 480 days after surgery, a statistically significant difference (p < 0.001). Within the labyrinth sparing group, there was no statistically significant association between hearing loss and cochlear obliteration or enhancement. CONCLUSIONS: Cochlear enhancement is correlated with cochlear obliteration and may precede it.

Computed tomography is a powerful medical imaging modality for longitudinal studies in cancer to follow neoplasia progression and evaluate anticancer therapies. Here, we report the generation of a photon-counting micro-computed tomography (PC-CT) method based on hybrid pixel detectors with enhanced sensitivity and precision of tumor imaging. We then applied PC-CT for longitudinal imaging in a clinically relevant liver cancer model, the Alb-R26Met mice, and found a remarkable heterogeneity in the dynamics for tumors at the initiation phases. Instead, the growth curve of evolving tumors exhibited a comparable exponential growth, with a constant doubling time. Furthermore, longitudinal PC-CT imaging in mice treated with a combination of MEK and BCL-XL inhibitors revealed a drastic tumor regression accompanied by a striking remodeling of macrophages in the tumor microenvironment. Thus, PC-CT is a powerful system to detect cancer initiation and progression, and to monitor its evolution during treatment.

Fixation reactions reduce the concentration of soluble phosphorus (P) and affect crop growth in alkaline calcareous soils. In lab and greenhouse studies, phosphoric acid (PA) or diammonium phosphate (DAP) were evaluated at various P rates (0, 18, 36 and 54 mg kg-1 soil), either as non-mix (designated as NM-PA and NM-DAP, respectively) or after premixing with farmyard manure (FYM) at 400 mg kg-1 soil (designated as PM-PA and PM-DAP, respectively). The amended soil was incubated at 25 °C and 70% water holding capacity for 7 weeks; thereafter, 32P dynamics were measured using the Freundlich kinetic model. A greenhouse study was also conducted using the same thirteen treatments (as used in incubation experiment) and wheat cultivar (Galaxy 2013) was grown following standard agronomic practices. The results showed that application of PM-PA at the highest rate, which caused maximum change in Pr (ΔPr = 59%) in laboratory condition, also produced maximum P uptake by grain (190.3 mg pot-1) and grain yield (44.1 g pot-1) of wheat in greenhouse experiment. Similarly, regression analysis showed that an increase in Pr values caused a corresponding increase in crop parameters. The results suggested that pre-mixing P fertilizer with FYM could be a viable technique to increase P supply and enhance productivity of wheat in alkaline calcareous soils.

During land-aquatic transfer, carbon (C) and inorganic nutrients (IN) are transformed in soils, groundwater, and at the groundwater-surface water interface as well as in stream channels and stream sediments. However, processes and factors controlling these transfers and transformations are not well constrained, particularly with respect to land use effect. We compared C and IN concentrations in shallow groundwater and first-order streams of a sandy lowland catchment dominated by two types of land use: pine forest and maize cropland. Contrary to forest groundwater, crop groundwater exhibited oxic conditions all-year round as a result of higher evapotranspiration and better lateral drainage that decreased the water table below the organic-rich soil horizon, prevented the leaching of soil-generated dissolved organic carbon (DOC) in groundwater, and thus limited consumption of dissolved oxygen (O2). In crop groundwater, oxic conditions inhibited denitrification and methanogenesis resulting in high nitrate (NO3-; on average 1140 ±â€¯485 µmol L-1) and low methane (CH4; 40 ±â€¯25 nmol L-1) concentrations. Conversely, anoxic conditions in forest groundwater led to lower NO3- (25 ±â€¯40 µmol L-1) and higher CH4 (1770 ±â€¯1830 nmol L-1) concentrations. The partial pressure of carbon dioxide (pCO2; 30,650 ±â€¯11,590 ppmv) in crop groundwater was significantly lower than in forest groundwater (50,630 ±â€¯26,070 ppmv), and was apparently caused by the deeper water table delaying downward diffusion of soil CO2 to the water table. In contrast, pCO2 was not significantly different in crop (4480 ±â€¯2680 ppmv) and forest (4900 ±â€¯4500 ppmv) streams, suggesting faster degassing in forest streams resulting from greater water turbulence. Although NO3-concentrations indicated that denitrification occurred in riparian-forest groundwater, crop streams nevertheless exhibited important signs of spring and summer eutrophication such as the development of macrophytes. Stream eutrophication favored development of anaerobic conditions in crop stream sediments, as evidenced by increased ammonia (NH4+) and CH4 in stream waters and concomitant decreased in NO3- concentrations as a result of sediment denitrification. In crop streams, dredging and erosion of streambed sediments during winter sustained high concentration of particulate organic C, NH4+ and CH4. In forest streams, dissolved iron (Fe2+), NH4+ and CH4 were negatively correlated with O2 reflecting the gradual oxygenation of stream water and associated oxidations of Fe2+, NH4+ and CH4. The results overall showed that forest groundwater behaved as source of CO2 and CH4 to streams, the intensity depending on the hydrological connectivity among soils, groundwater, and streams. CH4 production was prevented in cropland in soils and groundwater, however crop groundwater acted as a source of CO2 to streams (but less so than forest groundwater). Conversely, in streams, pCO2 was not significantly affected by land use while CH4 production was enhanced by cropland. At the catchment scale, this study found substantial biogeochemical heterogeneity in C and IN concentrations between forest and crop waters, demonstrating the importance of including the full vegetation-groundwater-stream continuum when estimating land-water fluxes of C (and nitrogen) and attempting to understand their spatial and temporal dynamics.

Urban sewage sludge is a potential source of phosphorus (P) for agriculture and represents an alternative way to recycle P as fertilizer. However, the use of thermally conditioned sewage sludge (TCSS) required an accurate assessment of its value as P-fertilizer. This work aimed at assessing the plant-availability of P from TCSS. Uptake of P by a mixture of ryegrass and fescue from TCSS and triple super phosphate (TSP) fertilizers was studied using (32)P-labeling technique in a greenhouse experiment. Phosphorus was applied at the rate of 50 mg P kg(-1).We also conducted incubation experiments considering the same treatments to assess soil microbial respiration. Applications of TCSS and TSP increased plant P uptake that is related to the root P acquisition. The P taken up by plant from soil plant-available P was lower for control compared to TSP or TCSS that was attributed to the increase of root interception of soil P. The contribution of TSP to ryegrass nutrition (Pdff%) was 55% with 22% of the applied P which was taken up by plants (CPU%). The Pdff value for TCSS was 56% with 14% of fertilizer P recovery (CPU%). Shoot biomass and total P uptake from TCSS were lower than those from TSP. As a result, the agronomic effectiveness of TCSS calculated from Pdff value (in comparison with TSP treatment) was 102%, while the AE of TCSS estimated from CPU value (in % TSP) was 64%, which is attributed to microbial activity stimulation inducing P immobilization onto soil constituents and microbial biomass during plant growth. The high C/N ratio of TCSS stimulated soil microbial biomass that competes with plant roots to acquire nutrients, such as P. As a consequence, the P taken up from either native soil or TCSS decreased in similar proportions. The AE value calculated with Pdff% took into account these interactions between soil, plant, and microbial biomass, and is less dependent on operational conditions than the AE value calculated with %Precovery.

OBJECTIVE: Can magnetic resonance imaging (MRI) diagnose abnormally thin and dehiscent superior semicircular canals (SSCs) that traditionally rely on evaluation by computed tomography (CT) imaging? STUDY DESIGN: Retrospective clinical study. SETTING: Tertiary referral center. PATIENTS: Adults who underwent both MRI and CT of the temporal bones over the past 3 years. INTERVENTIONS: CT and MR images of SSCs were separately reviewed, in a blinded fashion by three neuroradiologists at our institution. CT diagnosis of abnormally thin or dehiscent SSC was used as the "gold" standard. MAIN OUTCOME MEASURES: 1) Dehiscent SSC. 2) Abnormally thin SSC. 3) Normal SSC. RESULTS: One hundred temporal bones with evaluable superior semicircular canals from 51 patients were eligible for review on CT and MR imaging. There were 26 patients of thin SSC and 17 patients of SSC dehiscence on CT imaging, of which 13 and 15 respectively were also found on MRI. There were nine false-positive dehiscent SSC patients and four thin SSC patients observed on MR imaging while not observed on CT. For thin SSCs, MRI sensitivity was 61.9% and specificity of 94.3% with a positive predictive value of 81.3% and a negative predictive value of 86.2%. For dehiscent SSCs, sensitivity was 88.2% and specificity of 89.2% with a positive predictive value of 62.5% and a negative predictive value of 97.4%. CONCLUSION: In this series, MRI in the axial and coronal plane had a high negative predicative value for thin SSC (86%) and dehiscent SSC (97%). However, MRI cannot conclusively diagnose thin or dehiscent SSCs.

The aim of this study was to demonstrate the potential of a wireless pixelated ß+-sensitive intracerebral probe (PIXSIC) for in vivo positron emission tomographic (PET) radiopharmacology in awake and freely moving rodents. The binding of [(11)C]raclopride to D2 dopamine receptors was measured in anesthetized and awake rats following injection of the radiotracer. Competitive binding was assessed with a cold raclopride injection 20 minutes later. The device can accurately monitor binding of PET ligands in freely moving rodents with a high spatiotemporal resolution. Reproducible time-activity curves were obtained for pixels throughout the striatum and cerebellum. A significantly lower [(11)C]raclopride tracer-specific binding was observed in awake animals. These first results pave the way for PET tracer pharmacokinetics measurements in freely moving rodents.

We analyzed the linearity of relationships between soil test P (STP) and cumulative phosphorus (P) budget using data from six long-term fertilized grassland sites in four countries: France (Ercé and Gramond), Switzerland (Les Verrières), Canada (Lévis), and Finland (Maaninka and Siikajoki). STP was determined according to existing national guidelines. A linear-plateau model was used to determine the presence of deflection points in the relationships. Deflection points with (x, y) coordinates were observed everywhere but Maaninka. Above the deflection point, a significant linear relationship was obtained (0.33 < r (2) < 0.72) at four sites, while below the deflection point, the relationship was not significant, with a negligible rate of STP decrease. The relationship was not linear over the range of STP encountered at most sites, suggesting a need for caution when using the P budget approach to predict STP changes in grasslands, particularly in situations of very low P fertilization. Our study provides insights and description of a tool to improve global P strategies aimed at maintaining STP at levels adequate for grassland production while reducing the risk of P pollution of water.

Monte Carlo simulation (MCS) plays a key role in medical applications, especially for emission tomography and radiotherapy. However MCS is also associated with long calculation times that prevent its use in routine clinical practice. Recently, graphics processing units (GPU) became in many domains a low cost alternative for the acquisition of high computational power. The objective of this work was to develop an efficient framework for the implementation of MCS on GPU architectures. Geant4 was chosen as the MCS engine given the large variety of physics processes available for targeting different medical imaging and radiotherapy applications. In addition, Geant4 is the MCS engine behind GATE which is actually the most popular medical applications' simulation platform. We propose the definition of a global strategy and associated structures for such a GPU based simulation implementation. Different photon and electron physics effects are resolved on the fly directly on GPU without any approximations with respect to Geant4. Validations have shown equivalence in the underlying photon and electron physics processes between the Geant4 and the GPU codes with a speedup factor of 80-90. More clinically realistic simulations in emission and transmission imaging led to acceleration factors of 400-800 respectively compared to corresponding GATE simulations.

To increase the phosphorus recycling potential from swine slurry, mineral phosphorus products which could be used as fertilizers should be obtained and new processes need to be investigated. A routine method is needed to better evaluate the dissolved and solid mineral phosphorus in swine slurry. Cold perchloric acid extraction method previously developed for wastewater or sludge analysis was adapted. Ionic chromatography was used to measure orthophosphate in extracts. Only one extraction step was needed to distinguish between mineral and organic phosphorus in slurry. Reproducibility of the method was high (less than 5% of variation on the measured fractions). Selectivity was assessed by adding several organic and mineral phosphorus sources in the slurry. Cold perchloric extraction followed by ionic chromatography was very selective in quantifying both the mineral and organic forms of phosphorus in swine slurry.

A new beta-microprobe (betaP) has been used to locally measure the time-concentration curve of a radiolabeled substance. The betaP, analogous to positron emission tomography methodology, is useful for in vivo animal studies because it can acquire time-concentration curves with high temporal and spatial resolution. Using [18F]fluoro-2-deoxy-D-glucose and betaP, we evaluated the reliability of the biologic parameters and we compared this method with the [14C]2-deoxy-D-glucose autoradiography. BetaP time-concentration curves in three regions of the brain were obtained from 24 rats. Four kinetic parameters (K1-k4) were estimated from 60-minute experimental periods using a three-compartment model. Best fits were obtained when the vascular fraction (Fv) was estimated simultaneously with the four kinetic parameters (K1-k4). The mean estimated Fv values were about 5.5% for the frontal cortex regions and 8.0% for the cerebellum. Correlation coefficients higher than 0.830 were observed between regional cerebral metabolic rates for glucose (rCMRglc) values obtained by betaP and autoradiography. In addition, the betaP-derived input function was similar to that obtained by manual sampling. Our findings show that reliable rCMRglc values can be obtained using betaP.
