This study aimed to evaluate the discrepancies between genetic and visual coat color assignment in the Sarcidano Horse and to elucidate potential reasons. Individual DNA from 90 Sarcidano Horses was used for genetic assignment of coat color to explore the correspondence with individual forms containing phenotypical traits. The MC1R exon 1 and ASIP exon 3 have been genotyped and sequenced to obtain a picture of the coat color distribution in this breed. Surprisingly, once we compared the genetic results with the individual forms reporting the phenotypic data for each subject, a certain degree of non-correspondence between the phenotypic and genetic data in relation to coat color emerged. From the genetic analysis, Chestnuts (n = 58) resulted the most common Sarcidano Horse (n = 58), followed by a quite large number of Blacks (n = 28) and a very small number of Bays (n = 4), whereas phenotypic distribution resulted in 38 Chestnuts, 40 Bays, only 2 Blacks, and 10 Grays (without the possibility of recognizing the true color they carried). Chestnut resulted a very representative coat color, while many horses that visually identified as Bays were genetically Blacks. This discrepancy, that could be due to a variety of individual and external factors, including age, time of year, living situation and dietary condition, suggesting the importance of accurate coat color identification to ensure adequate features registration and reliable prediction of offspring's coat color.

Melatonin receptor MT1, encoded by the MTNR1A gene, is the main one involved in the seasonal regulation of reproductive activity. A correlation between this gene polymorphism and reproductive performance has been demonstrated in sheep. To date, no data about MTNR1A gene polymorphism are available regarding Italian goat breeds other than the Sarda goat. This study aimed to detect any PCR-RFLP polymorphic sites of MTNR1A using MnlI and RsaI enzymes in Northern Italian goat breeds, which are characterized by a pronounced reproductive seasonality. One-hundred-eight adult female goats belonging to four different breeds were included in the study (i.e., Frisa Valtellinese, n = 29; Orobica, n = 23; Lariana, n = 29; Camosciata delle Alpi, n = 27). Blood was sampled from each goat. Genomic DNA was extracted from each sample and the main part of exon II of MTNR1A gene was amplified by PCR and digested with MnlI and RsaI enzymes. Unexpectedly, none of the fragments were found to be polymorphic. The absence of polymorphism may be linked to the macro group of goat breeds that evolved during human migrations. Breeds of the Alpine-European strain would appear to show no polymorphism, as confirmed by our study, whereas breeds belonging to the Mediterranean-African or Asian-Middle Eastern strains do.

The aim of the present research was to evaluate the association between the SNPs rs430181568 and rs407388227 located in the MTNR1A gene with the reproductive recovery of Sarda sheep in different months of ram introduction in the flock (February, March, April and May). In order to address this research gap, we selected two farms, each of which consisted of approximately 1000 animals; a total of 800 ewes (400 for each farm) were genotyped for the two single nucleotide polymorphisms rs430181568 and rs407388227 located in the exon 2 of the MTNR1A. These SNPs are completely linked; thus, each genotype of rs430181568 corresponded to the same genotype for rs407388227. Among the genotyped animals, 240 individuals were selected and divided into four homogeneous groups (A, B, C and D) of 60 subjects, each group based on age (range 3−6 years old), body condition score (BCS) (range 2.0−4.0) and genotype (20 ewes carrying CC/CC, 20 CT/CT and 20 TT/TT genotype). The dates of the ram introduction in each group were 15 February, 15 March, 15 April and 15 May, respectively. In all groups, the lambing date and the number of lambs born from 150 to 220 days after the ram introduction were recorded. In all the groups, the genotypes CC/CC and CT/CT of the polymorphism (rs430181568 and rs407388227) showed the greatest fertility (the ratio between the number of lambed ewes and the ewes exposed to the rams) (p < 0.01) and the shortest distance between ram introduction to lambing (p < 0.01), compared with the TT/TT genotype. In conclusion, we determined that the polymorphisms rs430181568 and rs407388227 were associated with reproductive recovery, after ram introduction, even in February and March, months subsequent to the photorefractoriness period.

The goal of this study was to contribute to the general knowledge of the Sarcidano Horse, both by the identification of the genetic basis of the coat color and by updating the exact locations of the genotyping sites, based on the current EquCab3.0 genome assembly version. One-hundred Sarcidano Horses, living in semi-feral condition, have been captured to perform health and biometric checks. From that total number, 70 individual samples of whole blood were used for DNA extraction, aimed to characterize the genetic basis of the coat color. By genotyping and sequencing analyses of the MC1R Exon 1 and ASIP Exon 3, a real image of the coat color distribution in the studied population has been obtained. Chestnut and Black resulted in the most representative coat colors both from a phenotypic and genotypic point of view, that is suggestive of no human domestication or crossbreeding with domestic breed. Due to its ancient origin and genetic isolation, an active regional plan for the conservation of this breed would be desirable, focused on maintenance of resident genotypes and genetic resources. Collection and management of DNA, sperm, embryos, with the involvement of research centers and Universities, could be a valid enhancing strategy.

Stakeholders place great emphasis upon rationalizing the management and rearing techniques which are utilized within sheep farms. The present study aimed to investigate factors which may improve the reproductive performance of melatonin-treated Sardinian sheep via a series of three trials. The first trial (n = 100) investigated the effect of melatonin treatment alongside body condition score (BCS), the second trial (n = 150) investigated the effect of treatment alongside the date of treatment (treatment period) and the third trial (n = 150) investigated the effect of treatment alongside the previous lambing of the ewes. The findings indicated that melatonin is an effective tool for anticipating and improving the reproductive activity of in Sarda breed sheep during the springtime. Furthermore, to obtain optional results, melatonin implantation should be conducted in April, in ewes that have a BCS of >2.5 and that have passed their third month of lactation.

This study aims to find reliable strategies for advancing first mating and shortening the period from weaning to first lambing in ewe lambs. Sarda ewe lambs (n = 400) were selected from two farms and allocated into four separated groups of 100, all of which were exposed to fertile, adult rams over the course of a 50-day breeding period. The first treatment group (RMR) was exposed to four melatonin-treated rams which were replaced every ten days, whilst the second treatment group (RM) was exposed to four melatonin-treated rams which were not replaced. Alternatively, the first control group (RCR) was exposed to four untreated rams which were replaced every ten days, whilst the second control group (RC) was exposed to four untreated rams which were not replaced. In each group, lambing dates, fertility rate, litter size, and distance in days from ram introduction to lambing (DRIL) were recorded. The highest fertility rate was recorded in the RMR group (p ≤ 0.05). Shorter DRIL (p ≤ 0.01) and higher lambing concentrations were recorded in the RM and RMR groups as compared to the controls. The findings indicate that melatonin treatment of rams and their replacement at 10-day intervals results in earlier onset of first mating, increased fertility rate in ewe lambs, and a higher number of ewes that lambs in a shorter time frame.

The economy in Mediterranean areas is tightly linked to the evolution of the sheep-farming system; therefore, improvement in ewe's reproductive performance is essential in the developing countries of this area. MTNR1A is the gene coding for Melatonin receptor 1 (MT1), and it is considered to be involved in the reproductive activity in sheep. The aims of this study were: (1) identifying the polymorphisms from the entire MTNR1A coding region and promoter in Lebanese Awassi sheep flocks, and (2) investigating the association between the found polymorphisms and the reproductive performance, assessed as lambing rate, litter size, and days to lambing (DTL). The study was conducted in two districts of Lebanon, where 165 lactating ewes, aged 5.2 ± 1.5 years, with body condition score (BCS) 3.3 ± 0.4, were chosen and exposed to adult and fertile rams. From 150 to 220 days after ram introduction, lambing dates and litter sizes were registered. This study provided the entire coding region of the MTNR1A receptor gene in the Awassi sheep breed. Thirty-one single nucleotide polymorphisms (SNPs) were detected, five of which were missense mutations. The H2, H3, and H4 haplotypes were associated with lower DTL (p < 0.05), as well as the SNPs rs430181568 and rs40738822721, named from now on SNP20 and SNP21, respectively. These SNPs were totally linked and can be considered as a single marker. The ewes carrying the C allele at both these polymorphic sites advanced their reproductive recovery (p < 0.05). These results are essential for improving reproductive management and obtaining advanced lambing in Awassi ewes.

The aim of this study was to characterize the MTNR1A locus in Sarda sheep breed, in order to identify potential single nucleotide polymorphisms (SNPs) associated with reproductive resumption. The reproductive performance of 200 lactating ewes, aged 3-5 years, with body condition score (BCS) 2.5-4.0, at least at their third lambing were monitored for two consecutive years. In both year the enrolled ewes were exposed for 100 days to 10 adult, fertile rams. Mating, pregnancy and lambing for each ewe were recorded in order to evaluate differences in reproductive performance according to the analysed genotypes. From individual blood samples, DNA was extracted to amplify and to sequence promotor, the coding region, a part of intron and of 3' Untranslated region (3' UTR) of the MTNR1A gene. A total number of 29 SNPs were found (named SNP1 to SNP29), five of which caused also amino acid changes. The polymorphic sites found at positions g.17355452C > T (SNP16, rs430181568) and g.17355358C > T (SNP17, rs407388227) were linked (D' = 1 and r2 = 1) and showed a significant association to DRIL trait (distance in days from ram introduction to lambing). In both years, the ewes carrying C/C genotype in both these polymorphic sites showed the lowest DRIL compared to the other genotypes (P < 0.05). The ewes carrying C/C and T/C genotype exhibited the lambing peak at 170 days, and approximately 60% of the total lambing at 180 days from the ram introduction. Instead, ewes carrying T/T genotype showed the lambing peak around 200 days after ram introduction. Six haplotypes have been identified and the most frequent haplotype was also associated with lower DRIL (P < 0.05). Litter size displayed no statistical significance either among genotypes or among haplotypes. This study provided the major part of the MTNR1A gene in Sarda sheep breed and evidenced that SNP17 is associated with a shorter DRIL. The obtained results underlined the role of this polymorphism in improving reproductive efficiency in Sarda sheep and provides a suitable information for improving genetic selection.

The aims were to evaluate the reproductive response to ram placement with Sarda ewes with different allelic variants at position g.15099485 A > G of the MTNR1 A gene. Ram placements occurred between the early and late spring and there was analysis of whether this polymorphism is associated with other nucleotide substitutions. In each of the eight farms where the study was conducted (named F1-F8), 150 ewes (50 with A/A, A/G and G/G genotypes) were selected. In each group of 150 ewes, eight males were joined with ewes on the following dates: 25 March (T1) for F1-F2, 15 April (T2) for F3-F4, 5 May (T3) for F5-F6, and 1 June (T4) for F7-F8. The lambing dates and number of new born lambs were recorded until 220 days after joining rams with ewes. The ewes with G/G or A/G genotypes had a greater fertility rate compared with those with A/A in T1, T2 (P < 0.01), and in T3 and T4 (P < 0.05). The duration of time in days from the time of ram joining with ewes to lambing was less in the ewes with G/G and A/G compared with those with A/A (P < 0.01). The g.15099485A>G variation was always associated with that at position g.15099391G>A. Results indicate there is a positive effect of the g.15099485A>G variant on reproduction when males were joined with ewes in March or April. The association that was ascertained in this study between the g.15099391G>A and g.15099485A>G polymorphisms for the MTNR1 A gene, could provide new insights to clarify the mechanism of action of melatonin.

This study aimed to evaluate the effect of melatonin treatment carried out between late winter and early spring on reproductive response in Sarda breed sheep and whether the photo-refractoriness can influence this reproductive response. The study was conducted on 3200 adult ewes, aged 3-6years old, with body condition score (BCS) 2.5-4.0, from 16 commercial sheep farms in Northern Sardinia. In each farm 200 animals were enrolled and subdivided into 2 groups (n=100 each): Group M (treated with one 18mg melatonin implant), and group C (untreated). The melatonin treatments were performed on 10th February; 10th March; 10th April and on 10th May each time in 4 different randomly selected farms. Adult males, treated with 3 melatonin implants 1 week before females, were introduced in each flock 35days after ewes' treatment, and removed after 45days of cohabitation with females. Pregnancy was determined by transabdominal ultrasonography examination between 45th and 90th day after ram introduction. Data showed that melatonin treatment increased the fertility rate significantly (P<0.05), with the higher fertility rate in the ewes treated in April and May. The average time in days from male introduction to lambing was shorter in treated than in control ewes (P<0.05). Further, at the 160th and 170th day after male introduction the group M showed a higher number of lambed ewes compared to C (P<0.01). This effect was observed at 180th and 190th days after ram introduction, also, but with lower significance (P<0.05). In conclusion, melatonin treatment improved reproductive efficiency and advanced breeding season in Sarda sheep, especially when ewes were treated in spring.

The aim was to characterize the INSIG-2 gene in Sarda sheep and to highlight associations between polymorphisms and milk traits. Two-hundred ewes, in their third or fourth lactation who lambed a single lamb between 20th and 30th of November, were chosen. Monthly individual milk yield was recorded and from each ewe a sample of milk was taken to analyze fat and protein content. PCR-RFLP and DNA sequencing were carried out to detect polymorphisms. Five exons have been characterized and five mutations have been found G88A, 436TCAGdel, A471G, C1071T and T1737G all in the intronic regions. The ovine sequence and related variations were deposited in GenBank with accession number JX843812.1. The animals carrying AA genotype at position 88 showed a lower milk fat concentration than those with the AG or GG genotype (P < 0.05). A lower milk fat concentration was registered also in the animals with the TCAG deletion in position 436 (P < 0.05) and in the animals carrying AA genotype at position 471 compared to those with the AG or GG genotype (P < 0.05). Moreover, the animals carrying CC genotype at position 1071 had a greater milk yield than those with CT or TT genotype (P < 0.05) while ewes with TT genotype showed a higher milk protein concentration compared to the others (P < 0.05). A total of 11 haplotypes were detected but no significant associations with milk traits were found. In conclusion for the first time the complete coding sequence of INSIG-2 gene and its association with milk trait has been reported in this study.

The aim of this research was to investigate variability in each of the five exons of the caprine growth hormone (gGH) gene, in order to establish the possible relationships with milk traits in Sarda breed goat. The general linear model procedure was used to analyse the effects of the single strand conformation (SSCP) profiles on milk traits of 100 lactating goats. Analysis of conformational polymorphism at exons 1-5 revealed a total of 25 differing banding patterns. Sequencing revealed 21 nucleotide changes (compared with GenBank D00476): 14 were polymorphic and 7 monomorphic; 19 in exonic regions, 5 of which were nonsynonymous. A SNP upstream of the transcription initiation codon (c.-3A>G) and an indel (c.*29_30insC) in the 3'UTR, were detected. Alignment of 4 cloned sequences including the entire gGH gene led to the identification of 22 nucleotide variations within the intron regions, including two indels. Association analysis revealed that each exon, except exon-1, affected milk yield, exons 1 and 3 influenced milk fat percentage, and all exons, except exon-2, had an effect on protein percentage, supporting previous results in livestock. The variability detected at the caprine GH gene might provide useful information for the phylogeny of ruminants and, more importantly, have implications on the biological function of the growth hormone and on those traits resulting from its physiological action, including milk production and composition. The caprine GH gene may become a useful molecular marker for a more effective genetic selection for milk production traits in goats.

The effect of CSN1S1 genotype and lactation stage on milk yield and composition were investigated in 80 extensively reared goats. Milk yield was recorded in early, mid and late lactation and individual milk samples were collected to determine: fat, protein, lactose and casein content, pH, freezing point, somatic cell count (SCC) and total microbic mesophilic count (TMC). Relative casein composition and amino acid profile were quantified by HPLC. Fatty acid profile was measured by gas-chromatography. Genotype did not affect milk yield, while this trait was significantly affected by lactation stage (P < 0.01). CSN1S1 BB goats produced significantly higher protein and casein percentages (P < 0.05). αs1-casein (CN) was significantly higher in BB and AB goats than AF and BF, showing intermediate values in AA goats (P < 0.01). The protein percentage and the αs1 and αs2-CN fractions were not affected by lactation stage, while the casein content and the ß and κ-CN significantly increased throughout lactation (P < 0.01). C4 : 0 and C6 : 0 were not affected by genotype, while C8 : 0 and C10 : 0 were higher in the AA goats than BB; most of the long chain FA were higher in BB than AA goats. MUFA and PUFA increased in late lactation. In addition, BB goats showed higher essential amino acids, resulting in an optimal composition from the nutritional point of view, when compared with AA goats. The increase of MUFA, PUFA, essential and cis-FA in late lactation indicate that the lipid composition of goat's milk, with the progress of lactation, tends to improve its nutritional value.

The aim of the study was to detect polymorphism in the POU1F1 gene in Sarda breed goat, as well as to establish if SNPs could be associated with milk productive traits. The research was conducted on 129 Sarda breed goats from 4 to 5 years old, multiparous, lactating and in their third to fifth lactation. We report nine exonic and seven non-coding regions SNPs within the Sarda goat POU1F1 gene, namely, Ex 1 61 G>C; Ex 1 108 G>A; Ex 3 C>T; Ex 3 92 C>T; Ex 4 110 A>G; Ex 5 34 G>A resulting in Arg213Lys change; IVS4 641 G>A, IVS4 643 A>C, IVS4 659 G>A, IVS4 677 A>C, IVS4 G699Del, IVS4 709 C>G, Ex 6 17 G>A resulting in Arg228Ser change, Ex 6 58 G>T, Ex 6 172 T>C, 3'UTR 110 T>C. A statistically significant association was found between genotype TT, in position 17 of the exon 6 (3.1 % of frequency), and increased milk yield (P < 0.01) while genotype GT (25.6 % of frequency) was associated with a higher fat content. Genotype TT in position 58 of the exon 6 (3.9 % of frequency) was found to be associated with a higher fat (P < 0.01) and protein content (P < 0.05). Twenty-eight haplotypes were detected, but no significant association between the haplotypes and the milk production traits have been found. Our data, as well as providing new SNPs extending the POU1F1 gene characterization, evidence a relationship between polymorphism and milk production traits in Sarda goat breed.

The aim of the study was to develop a reliable method for the RNA extraction from milk of Sarda sheep breed and to highlight if the extracted RNA can be used for expression study on mammary genes involved in milk fat synthesis using RT-qPCR. The main result is that a sample of 150 ml of milk provides an optimal amount of RNA (73.5 µg/ml). The highest RNA concentration has been found in the samples analysed within 4 h after collection. The RNA extracted was positively correlated to the number of somatic cells (P < 0.001). The efficiency of the extraction method was confirmed by the results obtained from qPCR which showed a Ct value, for SREBPF1 gene of 26.8 ± 0.15. This research demonstrated that the high-quality of the RNA obtained is suited to use for studies of mammary genes expression in sheep, avoiding any damage caused by mammary gland biopsy.

The aim of this study was to examine the expression patterns of SREBP-1 gene in milk somatic cells and its association with milk fat yield during early lactation in Sarda breed sheep. A sample of 20 Sarda ewes, aged between 4 and 5 years, in their third to fourth lactation were chosen. From each ewe 28 days after lambing milk yield was measured, and a 160 ml milk sample for the RNA extraction and to test somatic cells count and lactose, fat and protein contents were collected. From the obtained RNA, total cDNA was synthesized and the quantitative PCR was performed. The fat, proteins and lactose content showed many differences among the animals, but these variations were no correlated with the milk yield. The SREBP-1 gene expression resulted higher in the high milk fat producing ewes. The correlation analysis showed that the SREBP-1 expression level is directly related to the amount of milk fat (g/die) (P < 0.001), while the total RNA obtained from each sample was found to be related to the somatic cells number (P < 0.001). Instead the expression of this gene showed no relations with the concentration of fat in milk. Our data highlight that in sheep SREBP-1 gene is expressed in the mammary gland during early lactation. Moreover, the positive relationship between SREBP-1 gene expression and the milk fat yield suggests that SREBP-1 gene is required for the lipid synthesis in the sheep mammary gland.

The ovine POU1F1 gene is localized on chromosome 1 and it contains five introns and six exons. In different mammalian species some mutations in different exons are associated with different production traits. The aim of our research was to study the POU1F1 gene nucleotide sequence to detect possible polymorphisms and their relationships with milk productive traits in Sarda breed sheep. The study had been conducted on 140 ewes, 4 or 5 years old coming from a farm located in Sardinia. All the animals were multiparous, lactating and in their third to fifth lactation. Individual milk yield had been recorded monthly and for each sample fat, protein, casein, lactose, and somatic cell count values were analysed. A jugular blood sample was collected from each ewe to perform genomic DNA extraction. PCR, SSCP and sequencing analysis were carried out to examine the six exons to highlight possible SNPs. One-way ANOVA was used to analyse association of variants with milk yield and/or its composition. Two novel SNP were found: 121 C>T in the 5'UTR of the fourth intron fragment and 249 G>A in the 3'UTR of the sixth exon fragment. The statistical analysis did not shown association between milk productive traits and the found polymorphisms. However, further investigations about the promoter region or the prophet genes, like the PROP-1, could clarify its exact role in regulating the productive traits in sheep.

This study evaluated the effect of storage on renneting properties of goat milk investigated using the Formagraph method. Milk samples from 169 goats in three farms (F1, F2 and F3) were analysed during an entire lactation (45, 75, 105, 135 and 165 days in milking DIM), to obtain renneting parameters, both from fresh milk and after storage with Bronopol and freezing at -20°C and -80°C. As regards fresh milk, mean values of clotting time were between 12·51 (45 DIM) and 13·29 min (105 DIM and F2), the curd firming time between 1·77 (45 DIM) and 2·15 min (F1) and curd firmness between 42·09 (165 DIM) and 49·55 mm (45 DIM). No statistical difference was recorded after storage. After regression analysis, all prediction models showed significance value at P<0·001 with the highest R2 value for clotting time, 0·710 (fresh vs. frozen milk at -20°C), and the lowest for clot firmness, 0·281 (fresh vs. frozen milk at -80°C). Results demonstrated that assessment of goat milk coagulation properties using the Formagraph method is also achievable after freezing or Bronopol addition.

The effect of MTNR1A gene polymorphisms on the fertility rate after AI in Sarda sheep was evaluated in 600 lactating adult ewes. Genomic DNA was subjected to amplification of the MTNR1A gene exon II. Amplicons were digested with restriction endonuclease MnlI. Ten samples from each genotype were sequenced. A polymorphism was detected (A612G) and ewes were determined to be +/+, +/- or -/- for the allele. Allelic frequency was 0.77 for the+allele and 0.23 for the - allele. The frequency of the +/+, +/- and -/- genotypes was 68, 19 and 13%, respectively. On 16 May 2009, 60 ewes from each genotype group were synchronised using intravaginal sponges containing 40 mg fluorogestone acetate for 14 days. At sponge removal, the ewes were administered 350 IU pregnant mare's serum gonadotropin and were then inseminated, 54-56 h later, with 400 × 10(6) spermatozoa. Pregnancies were confirmed 50 days after AI using transabdominal ultrasonography. Lambing dates and the number of newborn lambs were recorded within 155 days after AI. Conception and lambing rate were higher for ewes with the +/+ and +/- genotypes compared with those with the -/- genotype (P<0.01). In conclusion, there was a positive correlation between MTNR1A allele polymorphisms the reproductive response following synchronisation and AI in the spring.
