RESUMO

Mercury cycling in coastal metropolitan areas on the west coast of India becomes complex due to the combined effects of both intensive domestic anthropogenic emissions and marine air masses. The present study is based on yearlong data of continuous measurements of gaseous elemental mercury (GEM) concentration concurrent with meteorological parameters and some air pollutants at a coastal urban site in Mumbai, on the west coast of India, for the first time. The concentration of GEM was found in a range between 2.2 and 12.3 ng/m3, with a mean of 3.1 ± 1.1 ng/m3, which was significantly higher than the continental background values in the Northern Hemisphere (~ 1.5 ng/m3). Unlike particulates, GEM starts increasing post-winter to peak during the monsoon and decrease towards winter. July had the highest concentration of GEM followed by October, and a minimum in January. GEM exhibited a distinct diurnal cycle, mainly with a broad peak in the early morning, a narrow one by nightfall, and a minimum in the afternoon. The peaks and their timing suggest the origin of urban mobility and the start of local activities. A positive correlation between SO2, PM2.5, temperature, relative humidity, and GEM indicates that emissions from local industrial plants in the Mumbai coastal area. Principal component analysis (PCA) and cluster analysis (CA) confirm this fact. Monthly back trajectory analysis showed that air mass flows are predominantly from the Arabian Sea and local human activities. Assessment of human health risks by USEPA model reveals that the hazardous quotient, HQ < 1, implies negligible carcinogenic risk. GEM observations in Mumbai during the study period are below the World Health Organization's (WHO) safe limit (200 ng/m3) for long-term inhalation.

Source apportionment study of PM2.5 using positive matrix factorization was performed to identify the emission characteristic from different sectors (sub-urban residential, industrial and rapidly urbanizing) of Delhi during winter. Chemical characterization of PM2.5 included metals (Ca, Cd, Cr, Cu, Fe, K, Mg, Mn, Na, Ni, Pb and Zn), water soluble ionic compounds (WSICs) (Cl-, NO3-, SO42- and NH4+) and Carbon partitions (OC, EC). Particulates (PM2.5) were collected on filter twice daily for stable and unstable atmospheric conditions, at the locations with specific characteristics, viz. Ayanagar, Noida and Okhla. Ions solely occupied 50% of the total PM2.5 concentration. Irrespective of location, high correlation between OC and EC (0.871-0.891) at p ≤ 0.1 is observed. Relatively lower ratio of NO3/SO4 at Ayanagar (0.696) and Okhla (0.84) denotes predominance of emission from stationary sources rather than mobile sources like that observed at Noida (1.038). Using EPA PMF5.0, optimum factors for each location are fixed based on error estimation (EE). Crustal dust, vehicular emission, biomass burning and secondary aerosol are the major contributing sources in all the three locations. Incineration contributes about 19% at Ayanagar and 18% at Okhla. Metal industries in Okhla contribute about 19% to PM2.5. These specific local emissions with considerable potency are to be targeted for long-term policymaking. Considerable secondary aerosol contribution (15%-24%) indicates that gaseous emissions also need to be reduced to improve air quality.

Subpopulations of neurons display increased activity during memory encoding and manipulating the activity of these neurons can induce artificial formation or erasure of memories. Thus, these neurons are thought to be cellular engrams. Moreover, correlated activity between pre- and postsynaptic engram neurons is thought to lead to strengthening of their synaptic connections, thus increasing the probability of neural activity patterns occurring during encoding to reoccur at recall. Therefore, synapses between engram neurons can also be considered as a substrate of memory, or a synaptic engram. One can label synaptic engrams by targeting two complementary, non-fluorescent, synapse-targeted GFP fragments separately to the pre- and postsynaptic compartment of engram neurons; the two GFP fragments reconstitute a fluorescent GFP at the synaptic cleft between the engram neurons, thereby highlighting synaptic engrams. In this work we explored a transsynaptic GFP reconstitution system (mGRASP) to label synaptic engrams between hippocampal CA1 and CA3 engram neurons identified by different Immediate-Early Genes: cFos and Arc. We characterized the expression of the cellular and synaptic labels of the mGRASP system upon exposure to a novel environment or learning of a hippocampal-dependent memory task. We found that mGRASP under the control of transgenic ArcCreERT2 labeled synaptic engrams more efficiently than when controlled by viral cFostTA, possibly due to differences in the genetic systems rather than the specific IEG promoters.

RESUMO

A new species of natricine snake of the genus Rhabdophis is described from Northeast India. Morphologically the new species is similar to Rhabdophis himalayanus but it can be distinguished from it by lacking a nuchal groove and nuchal glands, being relatively smaller, and having a distinct neck, ventral, and subcaudal coloration. In overall coloration it resembles R. chrysargos, from which it is distinguished by several aspects of its morphology and genetic differences. The discovery of Rhabdophis bindi sp. nov. reveals that forested regions in the northeastern part of India needs further biological exploration.

AIM: To examine in a laboratory setting the efficacy of moderate to high strength magnetic fields, as a potential bacteriostatic stimulus, against Enterococcus faecalis, one of the causative agents for infection during root canal treatments. METHODOLOGY: Four different strengths (1, 2, 3 and 4 T) of the pulsed magnetic field (PMF) were applied in thirty repetitions to bacterial suspension. A pickup coil setup was used to measure the electromotive force induced inside the bacterial suspensions. The optical density (OD) was monitored over time (for 16 h 40 min) during the post-treatment period to assess bacterial growth. Along with the change in OD values, live/dead assay, membrane depolarization study, atomic force microscopy (AFM), scanning electron microscopy (SEM) and reactive oxygen species (ROS) assay on selected samples were studied to evaluate the effect of PMFs. All results were analysed using one-way ANOVA followed by post hoc Tukey test and considered significant at p < .05. Regression analysis (at a confidence of 95%, α = 0.05) was performed on the bacterial growth and membrane depolarization studies to determine progressive changes of the outcomes. RESULTS: The peak value of the induced electromotive force was recorded as 0.25 V, for the 4 T magnetic field pulse with a pulse width of 16 ms. There was a significant arrest of bacterial cell growth after an exposure to PMFs of 1 T, 3 T and 4 T (ANOVA score: F (4, 495) =395.180 at p = .05). The image-based qualitative results of the live/dead assay using fluorescence microscopy techniques indicated that an exposure to higher PMFs (3 T/ 4 T) induced a bacteriostatic effect in a longer post-exposure timescale. Evidence of altered membrane potential within the 2 h of exposure to 4 T PMF was supported by the incidence of elevated ROS. For the ROS assay, a significant difference occurred for 4 T exposed samples (ANOVA score: calculated F (1, 3) =20.2749 at p = .05). SEM and AFM observations corroborated with the outcomes, by portraying significant membrane damage. CONCLUSION: In a laboratory setting, PMFs with higher magnitudes (3 T and 4 T) were capable of inducing bacteriostatic effects on E. faecalis.

In this growing era, a massive amount of digital electronic health records (EHRs) are transferred through the open network. EHRs are at risk of a myriad of security threats, to overcome such threats, encryption is a reliable technique to secure data. This paper addresses an encryption algorithm based on integer wavelet transform (IWT) blended with deoxyribo nucleic acid (DNA) and chaos to secure the digital medical images. The proposed work comprises of two phases, i.e. a two-stage shuffling phase and diffusion phase. The first stage of shuffling starts with initial block confusion followed by row and column shuffling of pixels as the second stage. The pixels of the shuffled image are circularly shifted bitwise at the first stage of diffusion to enhance the security of the system against differential attack. The second stage of diffusion operation is based on DNA coding and DNA XOR operations. The experimental analyses have been carried out with 100 DICOM test images of 16-bit depth to evaluate the strength of the algorithm against statistical and differential attacks. By the results, the maximum entropy has been obtained an average of 15.79, NPCR of 99.99, UACI of 33.31, and larger keyspace of 10140, which infer that our technique overwhelms various other state-of-the-art techniques.

During simultaneous liver-kidney transplantation (SLK) in highly sensitized patients, donor specific anti-human leukocyte antigen antibodies (DSA, HLA) can be present prior to transplant leading to positive crossmatch, yet these recipients have relatively low incidences of acute rejection. The mechanisms and timing underlying immunologic changes that occur intra-operatively remain largely unknown. Therefore, we measured the intra- and peri-operative kinetics of anti-HLA antibodies in highly sensitized SLK recipients. In this study, pre- and post-operative blood samples were obtained from sensitized SLK candidates with documented DSA. Intra-operative samples were obtained from a sub-group of SLK recipients. Pretransplant anti-HLA antibody profiles were created and flow cytometry and anti-human globulin complement-dependent cytotoxic crossmatches were performed. Significant reductions in anti-HLA class I and II DSA were seen intra-operatively shortly after reperfusion of the liver allograft. This effect was most pronounced for anti-HLA class I DSA (mean change, -85%, p < 0.05); changes to anti-HLA class II DSA were less robust (mean change, -47%, p = 0.15). Importantly, non-DSA anti-HLA antibodies remained unchanged throughout the perioperative period, suggesting the mechanism(s) by which the liver lowers DSA levels are specific to the DSA. These data demonstrate the immunologic benefit of performing SLK is lasting and occurs very shortly after liver reperfusion.

While local anthropogenic emission sources contribute largely to deteriorate metro air quality, long range transport can also play a significant role in influencing levels of pollutants, particularly carbon monoxide (CO) that has a relatively long life span. A nationwide lockdown of two months imposed across India amid COVID-19 led to a dramatic decline in major sources of emissions except for household, mainly from cooking. This initially led to declined levels of CO in two of the largest megacities of India, Delhi and Mumbai under stable weather conditions, followed by a distinctly different variability under the influence of prevailing mesoscale circulation. We hereby trace the sources of CO from local emissions to transport pathways and interpret the observed variability in CO using the interactive WRF-Chem model and back trajectory analysis. For this purpose, COVID-19 emission inventory of CO has been estimated. Model results indicate a significant contribution from externally generated CO in Delhi from surrounding regions and an unusual peak on 17th May amid lockdown due to long range transport from the source region of biofuel emissions in central India. However, the oceanic winds played a larger role in keeping CO levels in check in a coastal megacity Mumbai which otherwise has high CO emissions from household sources due to a larger share of urban slums. Keeping track of evolving carbon-intensive pathways can help inform government responses to the COVID-19 pandemic to prioritize controls of emissions sources.

The COVID-19 pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), is rapidly spreading across the globe due to its contagion nature. We hereby report the baseline permanent levels of two most toxic air pollutants in top ranked mega cities of India. This could be made possible for the first time due to the unprecedented COVID-19 lockdown emission scenario. The study also unfolds the association of COVID-19 with different environmental and weather markers. Although there are numerous confounding factors for the pandemic, we find a strong association of COVID-19 mortality with baseline PM2.5 levels (80% correlation) to which the population is chronically exposed and may be considered as one of the critical factors. The COVID-19 morbidity is found to be moderately anti-correlated with maximum temperature during the pandemic period (-56%). Findings although preliminary but provide a first line of information for epidemiologists and may be useful for the development of effective health risk management policies.

OBJECTIVE: The aim of the present study was to compare the effect of transdermal diclofenac patch against oral diclofenac for post-endodontic pain control. MATERIALS AND METHODS: Thirty-two patients with symptomatic irreversible pulpitis in single-rooted premolar teeth of either arch were treated endodontically in a single visit by a single endodontist. Oral diclofenac (50 mg twice daily) for group I and transdermal diclofenac patch (100 mg once daily) for group II were administered as post-endodontic analgesics for two days. Visual analogue scale (VAS) chart was used to record pain intensity scores preoperatively and at intervals of 4, 8, 12 and 24 h postoperatively for 2 days. Paracetamol 650 mg tablets were provided as rescue medication. RESULTS: There was a significant decrease in the postoperative pain intensity scores for both groups. The postoperative scores gradually decreased from day 1 to day 2 in both groups. Twelve out of sixteen patients who had received diclofenac tablets complained of gastric discomfort. CONCLUSION: Transdermal diclofenac patch was as effective as an oral diclofenac tablet and can be used as an alternative and effective analgesic for post-endodontic pain management, especially in patients with gastric discomfort.

Crop residue burning during post monsoon season in the neighboring provinces leads to frequent episodes of extreme pollution events, associated with premature morbidity and mortality. A synergistic use of multiple satellite measurements in conjunction with actual field incidences data at the ground led us to develop the realistic high-resolution emission inventory of the hazardous pollutant PM2.5 due to stubble burning. We quantify the share of biomass burning in deteriorating Delhi's air quality during 2018 using the SAFAR chemical transport model that has been validated with dense observational network of Delhi. The impact of biomass burning on Delhi's PM2.5 is found to vary on day-to day basis (peaking at 58%) as it is highly dependent on transportation pathway of air mass, controlled by meteorological parameters from source to target region. Comprehending the multi-scale nature of such events is crucial to plan air quality improvement strategies.

The monotypic genus Dravidogecko, represented by its type-species D. anamallensis, is singular amongst peninsular Indian gekkonid lineages in its endemism to the Western Ghats. Molecular species delimitation approaches reveal at least seven species-level lineages within the genus from its distribution range across the mid-high elevations of the southern Western Ghats of India. These lineages, albeit superficially cryptic, are patently diagnosable from each other by employing a limited but precise set of morphological characters. Six of these lineages that were obscured under the nomen D. anamallensis are herein recognized as distinct species. A reappraisal of the genus Dravidogecko is provided based on external morphology and osteological characters, along with a detailed redescription of the holotype of D. anamallensis. A key to the species based on diagnostic characters is presented. Gene-trees based on mitochondrial and nuclear DNA data recovered marginally disparate topologies and were consequently coalesced into a species-tree for phylogenetic inference. Timetree analysis reveals late Miocene cladogenesis in this group and establishes late Palaeocene divergence from its sister genus, Hemidactylus, making Dravidogecko one of the earliest, extant lizard lineages to have colonized peninsular India.

Black carbon (BC) aerosol emitted in incomplete combustion processes is known for causing warming in the climate system also poses serious health issues. Identification of the sources of BC is essential for the development of mitigation strategies to regulate their effects in changing climate. Among different observational and analytical techniques currently available, source apportionment methods based on optical measurements are relatively simple. For example, 'Aethalometer model' was developed based on Aethalometer observations. However, there are a few limitations with this model arising from assumption of wavelength and angstrom exponent pairs. We have developed an empirical method which also relies on Aethalometer observations named as 'Two alpha method' which assumes angstrom exponent from fossil fuel as 1 and estimates bio-mass fraction and angstrom exponent for bio-mass burning. This method has been applied to Aethalometer observations from five different locations (rural, semi-urban and urban) over Indian sub-continent to quantify sources of BC. Fossil fuel is found to be the major source of BC (∼70%) irrespective of the location. Collocated measurements of Carbon Monoxide (CO) over rural site correlated well with derived bio-mass fraction. Results from this study demonstrated the capabilities of empirical method and shall provide spatio-temporal variability in sources of BC if applied to more locations.

RESUMO

The American Society of Anesthesiologists (ASA) physical status is a tool commonly used to classify a patient's physical fitness before surgery. Since its introduction in 1941 it has undergone very few modifications to improve its reliability and to eliminate subjectivity, despite vast changes in both surgical and anaesthetic technique. We present the history of the ASA physical status and review the literature on its applicability to contemporary anaesthetic practice.

BACKGROUND: Streptococcus mutans is well-known causative microorganism in the development of dental caries because they drop the plaque pH and produce acids from carbohydrates and survive in the acidic environment. It is now evident that knowledge of the bacteria enforces empirical approach to therapy, then specific antimicrobial therapy that might allow more conservative treatment options. Over the past few decades, there has been a remarkable increase in the prevalence rate of dental caries among children and the elders. Genotypic methods help in the detection and manipulation of nucleic acids which allows microbial genes to be examined directly. AIM: The aim of this study is to isolate and characterize S. mutans from rural population and to obtain genomic DNA and screen DNA band pattern. METHODOLOGY: A total of 80 plaque samples were collected from the buccal surfaces of maxillary and lingual surfaces of mandibular first molar with carious teeth in patients at a rural outreach center in Chikkaballapur district, Karnataka. Among these, 48 clinical isolates of S. mutans were recovered. Further, genomic DNA was extracted from all the positively isolated strains including the standard strain (microbial type culture collection 497), and stored at 4°C in tris EDTA buffer (TE). To analyze the molecular heterogeneity of the clinical strains, polymerase chain reaction (PCR), and restriction fragment length polymorphism was performed using restriction enzymes Hind III and Hae III. Using agarose gel electrophoresis, genomic DNA band pattern was analyzed. RESULTS: Statistically significant difference was seen in the "dex" gene collected from sample DNA and standard DNA in three different parameters (S. mutans 497). CONCLUSION: Genomic DNA of S. mutans was successfully isolated from the rural population. Dex gene was successfully amplified using PCR. Hae III enzymes successfully digested PCR amplicons and the fragments exhibited visible heterogeneity.

Bleeding during and after surgery ranges from trivial to fatal. Bleeding is in part determined by the patient's coagulation status. The UK National Institute for Health and Care Excellence recommends a pre-operative clotting test for patients with a history of abnormal bleeding. Anaesthetists are familiar with the prothrombin time assay, used to monitor warfarin effect, but anaesthetists may be less familiar with the activated partial thromboplastin time (APTT), which tests the function of the 'intrinsic' clotting pathway. The activated partial thromboplastin time may be prolonged due to contamination, anticoagulant therapy, clotting factor deficiencies, lupus anticoagulant or acquired inhibitors of specific clotting factors. A prolonged activated partial thromboplastin time should lead to: further testing to exclude heparin contamination or therapy, mixing studies to identify factor deficiencies and if necessary dynamic studies, such as the dilute Russell's viper venom time and the Actin FS-activated partial thromboplastin time, to identify direct factor inhibitors. These tests identify abnormalities and their implications for bleeding, helping anaesthetists and haematologists to manage haemostasis for individual patients.

BACKGROUND: Intraradicular procedures remove radicular dentin and jeopardizing longevity of a tooth. Traditional in vitro methods used to determine the remaining dentin thickness (RDT) have limitations. AIM: The aim is to evaluate, using cone-beam computed tomography (CBCT), RDT following rotary instrumentation and post space preparation in buccal and palatal roots of maxillary first premolars. MATERIALS AND METHODS: Twenty-three maxillary first premolars with two roots were selected. CBCT images were taken preoperatively, after instrumentation and Parapost 3 and 4 preparation (CBCT 1, 2, 3, and 4). RDT was measured 5 mm above apex, 1 mm below furcation, and 1 mm above furcation (Levels 1, 2, and 3). Data were analyzed using ANOVA and post hoc Tukey's test (P = 0.05). RESULTS: Rotary instrumentation and post space preparation reduced RDT in all walls of buccal and palatal roots. Post space preparation with Parapost 3 and 4 reduced RDT in the palatal wall of buccal root to <1 mm and Parapost 4 reduced all walls to 1 mm of dentin. CONCLUSION: Post space preparation in maxillary first premolars should be performed cautiously. It is safer to place a post in the palatal root of this tooth and limit preparation to Parapost 3.

OBJECTIVE: The objective of the present study is to understand the role of yttria stabilized zirconia (YSZ) in achieving the desired spectrum of clinically relevant mechanical properties (hardness, elastic modulus, fracture toughness and brittleness index) and chemical solubility of mica glass ceramics. METHODS: The glass-zirconia mixtures with varying amounts of YSZ (0, 5, 10, 15 and 20wt.%) were ball milled, compacted and sintered to obtain pellets of glass ceramic-YSZ composites. Phase analysis was carried out using X-ray diffraction and microstructural characterization with SEM revealed the crystal morphology of the composites. Mechanical properties such as Vickers hardness, elastic modulus, indentation fracture toughness and chemical solubility were assessed. RESULTS: Phase analysis of sintered pellets of glass ceramic-YSZ composites revealed the characteristic peaks of fluorophlogopite (FPP) and tetragonal zirconia. Microstructural investigation showed plate and lath-like interlocking mica crystals with embedded zirconia. Vickers hardness of 9.2GPa, elastic modulus of 125GPa, indentation toughness of 3.6MPa·m1/2, and chemical solubility of 30µg/cm2 (well below the permissible limit) were recorded with mica glass ceramics containing 20wt.% YSZ. SIGNIFICANCE: An increase in hardness and toughness of the glass ceramic-YSZ composites with no compromise on their brittleness index and chemical solubility has been observed. Such spectrum of properties can be utilised for developing a machinable ceramic for low stress bearing inlays, onlays and veneers.

Importance of atmospheric aerosols through direct and indirect effects on hydrological cycle is highlighted through multiple studies. This study tries to find how much the aerosols can affect evapo-transpiration (ET), a key component of the hydrological cycle over high NDVI (normalized difference vegetation index)/dense canopy, over Dibrugarh, known for vast tea plantation. The radiative effects of aerosols are calculated using satellite (Terra-MODIS) and reanalysis data on daily and monthly scales. Aerosol optical depth (AOD) obtained from satellite and ground observations compares well. Aerosol radiative forcing (ARF), calculated using MERRA data sets of 'clean-clear radiation' and 'clear-radiation' at the surface, shows a lower forcing efficiency, 35 Wm-zs, that is about half of that of ground observations. As vegetation controls ET over high NDVI area to the maximum and that gets modified through ARF, a regression equation is fitted between ET, AOD and NDVI for this station as ET = 0.25 + (-84.27) × AOD + (131.51) × NDVI that explains 82% of 'daily' ET variation using easily available satellite data. ET is found to follow net radiation closely and the direct relation between soil moisture and ET is weak on daily scale over this station as it may be acting through NDVI.
