RESUMO

Accumulation of peroxidases and their mRNAs was localized in colonies of Phanerochaete chrysosporium sandwiched between perforated polycarbonate membranes. Northern (RNA) blot analyses of colonial rings and in situ hybridizations with specific probes for manganese(II)-dependent peroxidase (MnP-1) and lignin peroxidase (LiP H8) mRNAs indicated that the expression of MnP-1 and Lip H8 genes started simultaneously in the central area of 3-day-old colonies. With time the signals for both transcripts spread to more-peripheral areas while decreasing in intensity. Furthermore, the appearance of MnP protein, as detected with specific immune serum, immediately followed accumulation of the MnP-1 mRNA transcript. However, LiP protein could be detected only some time after accumulation of LiP H8 mRNA.

Urine concentrates from workers in a Söderberg potroom and an anode paste plant were tested for mutagenicity by the Salmonella reversion assay. The study is aimed at group exposure as an indicator of the effect of the work atmosphere. Urine from exposed smokers showed mutagenic activity, whereas urine from exposed non-smokers did not. The mutagenicity of exposed smokers' urine was not significantly different from the urine from non-exposed smokers. Significant mutagenicity of smokers' urine was evident only in the presence of a rat-liver metabolic system. Since an earlier expectorate analysis has shown that mutagens from the work atmosphere are deposited in the workers' respiration system and the urine analysis does not show any effect of occupational exposure, it is likely that the mutagens are eliminated from the body via other routes than renal excretion.

According to a method (the AM-test) previously developed by us, the counting of alveolar macrophages (AM) from expectorates provides an estimation of the potential pulmonary insult from occupational air pollution +/- smoking. In this article we demonstrate a relationship between the number of AM and self-registered pulmonary symptoms and complaints. Employees from different workplaces in a primary aluminum reduction plant gave expectorate samples for the AM-test. A gradient in the number of AM was observed, with increasing AM numbers from the controls to the administration employees to the potroom workers, who had the highest numbers. Pulmonary symptoms and complaints were recorded by means of a questionnaire. A clear covariation between AM numbers and respiratory symptoms and complaints was observed and was most pronounced at the highest pollution levels (in the potrooms). We therefore suggest that the AM-test may serve as an effect indicator as well as a dose indicator.

The major proportion of the cellular components found in most of the sputa from persons working in polluted atmospheres are alveolar macrophages, and counts of the alveolar macrophages present in smears made from sputa from exposed workers probably reflect the lung reaction to air pollution. An investigation of this phenomenon was undertaken using the sputa of workers in different types of industries: a coke plant, two different aluminum reduction plants, a ship-building yard and asphalt work on a road. The results showed that the alveolar macrophage count increases with a higher level of particulate pollution in the workplace. A synergistic effect of occupational air pollution and smoking habits was also recorded.

Filter extracts of airborne particles from a Söderberg potroom and an anode paste plant were tested for mutagenicity by the Salmonella reversion assay. The extracts were mutagenic to strains TA100 and TA98, mainly after metabolic activation, but positive results were obtained also without S9 mix in strain TA98. These findings indicate that the particulate phase of air from the potroom and the anode paste plant of aluminum plants contain mostly indirect mutagens of both the base-pair substitution and frameshift type, and--to a less degree--frameshift mutagens. The relationship between concentration and mutagenicity was more positive for the potroom extract than for the anode paste plant extract.

Methanol extracts of hydrolyzed expectorate samples from workers in a Söderberg potroom were evaluated by the Salmonella reversion assay. The expectorates from exposed workers--mostly from smokers, but also to a certain extent from nonsmokers--were mutagenic; however, the control samples from both smokers and nonsmokers were not. The positive results produced by the expectorate samples from the exposed smokers suggest a synergistic relationship between exposure to air pollution of the working atmosphere and smoking.

RESUMO

The first ultrastructural description of the human bulbo-urethral gland (Cowper's gland) is presented. In light microscopy the secretory cells are cuboidal or cylindrical with a clear cytoplasm and a peripherally located round, rather dense nucleus. Myoepithelial cells are demonstrated in the secretory tissue. The electron microscopy reveals numerous membrane bound secretory granules with dense cored matrix in the cytoplasm. This special composition of the secretory granules corresponds to the analogue glands in ram and bull. The secretory activities seem to be correlated to the development of the gland as a function of sexual maturity of the individual. Secretory granules are found also in surrounding capillaries, which suggests that this gland also has an endocrine function.

The concentration of lead in five lung segments and in regional lymph nodes was determined post mortem in ten subjects by atomic absorption technique. In each case there was a considerable variation in the concentration of lead in different lung segments, and, in general, the apical segment in the upper lobe tended to be more contaminated than segments in the lower lobes. With one exception, the lead levels in the regional lymph nodes exceeded those of the lung segments. Individual differences in the distribution of lead in the lungs may be due to differences both in exposure and physiological response. When comparative analyses are made, we suggest that samples be taken from the same and preferably the basal segments of the lung.

RESUMO

The direct-atomization technique of flameless atomic absorption spectroscopy has shown to be a useful and accurate method when applied to determination of the concentration of lead in human lung tissue. The analytical results are highly reproducible and agree with those obtained by the conventional flame atomic absorption technique.
