RESUMO

The anti-inflammatory actions of glucosamine (GlcN) on arthritic disorders involve the suppression of inflammatory mediator production from synovial cells. GlcN has also been reported to inhibit the activation of the p38 mitogen-activated protein kinase (MAPK) pathway. The present study aimed to determine the cooperative and anti­inflammatory actions of functional food materials and evaluated the production of interleukin (IL)­8 and phosphorylation of p38 MAPK in IL-1ß-activated synovial cells, incubated with the combination of GlcN and various functional food materials containing L­methionine (Met), undenatured type II collagen (UC­II), chondroitin sulfate (CS), methylsulfonylmethane (MSM) and agaro-oligosaccharide (AO). The results indicated that Met, UC­II, CS, MSM and AO slightly or moderately suppressed the IL-1ß-stimulated IL­8 production by human synovial MH7A cells. The same compounds further decreased the IL­8 level lowered by GlcN. Similarly, they slightly suppressed the phosphorylation level of p38 MAPK and further reduced the phosphorylation level lowered by GlcN. These observations suggest a possibility that these functional food materials exert an anti­inflammatory action (inhibition of IL­8 production) in combination with GlcN by cooperatively suppressing the p38 MAPK signaling (phosphorylation).

A double-blind parallel group comparison design clinical study was conducted in Japanese patients with mild to moderate erectile dysfunction to investigate the efficacy of a supplement containing Pycnogenol® and L-arginine. Subjects were instructed to take a supplement (Pycnogenol® 60 mg/day, L-arginine 690 mg/day and aspartic acid 552 mg/day) or an identical placebo for 8 weeks, and the results were assessed using the five-item erectile domain (IIEF-5) of the International Index of Erectile Function. Additionally, blood biochemistry, urinalysis and salivary testosterone were measured. Eight weeks of supplement intake improved the total score of the IIEF-5. In particular, a marked improvement was observed in 'hardness of erection' and 'satisfaction with sexual intercourse'. A decrease in blood pressure, aspartate transaminase and γ-glutamyl transpeptidase (γ-GTP), and a slight increase in salivary testosterone were observed in the supplement group. No adverse reactions were observed during the study period. In conclusion, Pycnogenol® in combination with L-arginine as a dietary supplement is effective and safe in Japanese patients with mild to moderate erectile dysfunction.

RESUMO

Continuous-flow isotope ratio mass spectrometry (CF-IRMS) of deuterium, carbon, nitrogen, oxygen and sulfur has been used to analyse samples of pure chondroitin sulfates from known animal sources (shark, squid, salmon, pig and bovine). There is a need to control the origin of this dietary supplement, which is extracted from several types of animals: for traditional, ethical, or economic reasons, a given source of natural products of animal origin can be preferred to another, and can therefore have a different price. Twenty-three samples collected in Europe and Asia were analysed by IRMS. The results, especially the isotopic deviations of sulphur, oxygen and deuterium, show a significant discrimination between marine and terrestrial origins of this compound which will provide a convenient and efficient way to control the declared sources in the market. The differences observed between origins are further discussed.
