RESUMO

Frequent vaccination failures have occurred in the broiler farms in Eurasian countries during Newcastle disease outbreaks. The disease is enzootic in many countries of the region, especially in southwest Asia. I-2 vaccine has been used successfully in village chickens in many Asian and African countries. Our preliminary study showed good efficacy of the vaccine in broiler chickens. Therefore the current experimental study was conducted to compare viral shedding period of heat resistance I-2 vaccine with B1 commercial vaccine following challenge with Herts'33. For this purpose three hundred commercial broilers were randomly allocated into four groups; 1) Thermostable I-2 vaccine, 2) Hitchner B1 vaccine, 3) Challenge group with no vaccine, and 4) Negative control group. Experimental chicks were vaccinated on days 19 and 26 by the eye drop route and then the birds were challenged via intra ocular route on day 40 with a suspension containing 106 EID50/ml challenge virus. Experimental chickens were monitored by collecting buccal and cloacal swabs at different times. Collected swabs were submitted to PCR test. The results showed that vaccination can protect the birds against mortality and also decrease virus shedding; also there was not a significant difference between vaccination with I-2 and B1 vaccines.

RESUMO

We discuss a mechanism for electron capture in fast ion-atom collisions which has not been considered so far. Similarly as in the radiative capture, where the electron transfer occurs due to photon emission, within the mechanism under consideration the electron capture takes place due to the emission of an additional electron. This first-order capture mechanism leads to the so-called transfer ionization and has a number of interesting features; in particular, in the target frame it results in electron emission mainly into the backward semisphere.

RESUMO

AIMS: To evaluate the esterase phenotype in Lactococcus lactis strains isolated from traditional Tunisian dairy products. METHODS AND RESULTS: A collection of 55 L. lactis strains isolated from traditional fermented milk products and three reference strains were identified at species and subspecies level using molecular methods targeted to the 16S rRNA gene and to the histidine operon. The genotypic data obtained allowed the identification of the strains as L. lactis ssp. lactis and L. lactis ssp. cremoris with the prevalence of the ssp. lactis. The phenotypic identification based on arginine hydrolysis, the growth at 40 degrees C and in presence of 4% NaCl showed several discrepancy with the identification data based on genotypic analysis. Additional experiments carried out evaluating the esterase electrophoretic patterns revealed four classes of esterases identified on the basis of their electrophoretic mobility and specific activity on alpha- and beta-naphthyl ester of acetate and propionate. Esterase profiles discriminated the strains in two main groups corresponding to the subspecies cremoris and lactis according to a DNA-based identification. CONCLUSIONS: The evaluation of esterase activity represents a novel phenotype for the taxonomic discrimination of the L. lactis ssp. lactis and cremoris. SIGNIFICANCE AND IMPACT OF THE STUDY: Besides the DNA-based techniques that allow the rapid and accurate species/subspecies identification, the electrophoretic esterase profiles of L. lactis strains represents: (i) a new phenotypic tool to understand the physiology and the ecology of this species; and (ii) a new test for the potential selection of flavour producing strains.
