The effect of temperature on photoluminescence (PL) due to the6Pj→8S7/2(j= 5/2, 7/2) transitions of Gd3+ions was examined between 200 and 500 K for a sol-gel-derived silica-(Gd,Pr)PO4transparent glass-ceramic phosphor with negligible concentration quenching under excitation into the 5d-4f transition of Pr3+ions at 220 nm. The intensity of the narrow-band ultraviolet B (UVB) PL at ∼313 nm associated with the6P7/2→8S7/2transition slightly increased between 200 and 300 K, but was decreased to ∼86% and ∼62% of that at 300 K when temperature was raised to 400 and 500 K, respectively. The observed magnitude of the thermal quenching of the UVB PL intensity was agreed well with that recorded in a prototype narrow-band UVB lamp consisting of another silica-(Gd,Pr)PO4transparent glass-ceramic window and a KrCl excimer lamp as a light source at 222 nm. The silica-(Gd,Pr)PO4transparent glass-ceramics was more resistant to the thermal quenching of the narrow-band UVB PL than a conventional powder phosphor, probably because of the encapsulation of (Gd,Pr)PO4nanocrystals with low quenching centre concentrations in inert silica glass matrix.

Cell therapy with adipose tissue-derived stem cells (ASCs) is expected to be a candidate for the treatment of fulminant hepatic failure (FHF), which is caused by excessive immune responses. In order to evaluate the therapeutic effects of ASCs on FHF, the in vitro and in vivo immunomodulatory effects of ASCs were examined in detail in the mouse model. The in vitro effects of ASCs were examined by assessing their influence on the proliferation of lymphomononuclear cells (LMCs) stimulated with three kinds of mitogens: phorbol 12-myristate 13-acetate (PMA) plus ionomycin, concanavalin A (ConA), and lipopolysaccharide (LPS). The proliferation of LMCs was efficiently suppressed in a dose-dependent manner by ASCs in the cases of PMA plus ionomycin stimulation and ConA stimulation, but not in the case of LPS stimulation. The in vivo effects of transplanted ASCs were examined in the murine FHF model induced by ConA administration. The ALT levels and histological inflammatory changes in the ConA-administered mice were apparently relieved by the transplantation of ASCs. The analysis of mRNA expression patterns in the livers indicated that the expressions of the cytokines such as Il-6, Il-10, Ifn-γ, and Tnf-α, and the cell surface markers such as Cd3γ, Cd4, Cd8α, Cd11b, and Cd11c were downregulated in the ASC-transplanted mice. The immunomodulatory and therapeutic effects of ASCs were confirmed in the mouse model both in vitro and in vivo. These suggest that the cell therapy with ASCs is beneficial for the treatment of FHF.

Stem cell transplantation has been expected to have various applications for regenerative medicine. However, in order to detect and trace the transplanted stem cells in the body, non-invasive and widely clinically available cell imaging technologies are required. In this paper, we focused on magnetic resonance (MR) imaging technology, and investigated whether the trimethylamino dextran-coated magnetic iron oxide nanoparticle -03 (TMADM-03), which was newly developed by our group, could be used for labeling adipose tissue-derived stem cells (ASCs) as a contrast agent. No cytotoxicity was observed in ASCs transduced with less than 100 µg-Fe/mL of TMADM-03 after a one hour transduction time. The transduction efficiency of TMADM-03 into ASCs was about four-fold more efficient than that of the alkali-treated dextran-coated magnetic iron oxide nanoparticle (ATDM), which is a major component of commercially available contrast agents such as ferucarbotran (Resovist), and the level of labeling was maintained for at least two weeks. In addition, the differentiation ability of ASCs labeled with TMADM-03 and their ability to produce cytokines such as hepatocyte growth factor (HGF), vascular endothelial growth factor (VEGF) and prostaglandin E2 (PGE2), were confirmed to be maintained. The ASCs labeled with TMADM-03 were transplanted into the left kidney capsule of a mouse. The labeled ASCs could be imaged with good contrast using a 1T MR imaging system. These data suggest that TMADM-03 can therefore be utilized as a contrast agent for the MR imaging of stem cells.

The purpose of this study is to clarify the effectiveness of the use of ß-blocker in coronary computed tomography angiography (CCTA). In 1783 patients, heart rate was controlled by propranolol injection to patients with heart rates of 61 bpm or more. As a result, the scan heart rate (58.8±6.5 bpm) decreased significantly compared with the initial heart rate (72.7±9.4 bpm). Prospective gating method was used by 61.9% including 64.3% of the intravenous ß-blocker injection group. Moreover, daily use of oral ß-blocker had influence on reduction of the scan heart rate (daily use group: 60.1±6.5 bpm vs. unuse group: 58.5±6.3 bpm p<0.01). When we evaluated the image quality of CCTA by the score, the improvement of the score was obviously admitted by 65 bpm or less of the scan heart rate. The ratio of scan heart rate that was controlled by 65 bpm or less was decreased in the initial heart rate groups that were 81 bpm or more. The incidence of adverse reactions by the propranolol injection was few, and these instances only involved slight symptoms. Therefore, heart rate control with the use of ß-blocker is useful for the image quality improvement of CCTA. This form of treatment can be safely enforced.

RESUMO

Recently, novel taurine-containing uridine derivatives were discovered in mammalian mitochondrial tRNAs, and these modified ribonucleosides existed at the first position of the anti-codon. This paper describes the chemical synthesis of these novel uridine derivatives, 5-taurinomethyluridine (tau m5U) and 5-taurinomethyl-2-thiouridine (tau m5s2U). These taurine-containing uridine derivatives were synthesized in the good yields by the reaction of the corresponding 5-hydroxymethyluridine derivatives with taurine under basic conditions.
