RESUMO

An Eulerian passive tracer model coupled with a quadruple-nested 3D circulation model was used to assess the coastal dispersal of treated wastewater effluent from a sewage treatment plant and the associated impacts on an adjacent seaweed farm using three different operational scenarios. When the discharged volume and source effluent fluxes were decreased by ~16.7%, the accumulated effluent in the farm was reduced by ~25.4%. A tracer flux budget analysis revealed the apparent predominance of the transient component that accounts for the nonlinear interactions primarily from tidal currents and eddies. The transient flux promoted the effluent influx to impede effluent accumulation in the farm, whereas the mean flux contributed to the outgoing flux. A source flux reduction caused a remarkable decrease in the transient flux and thus an even greater effluent accumulation reduction. In turn, a modified source density scenario without total effluent volume change did not work as expected.

A high-resolution 3-D model was developed to assess the impact of a diversion outfall at the Tarumi Sewage Treatment Plant (TSTP) on an adjacent seaweed farm in Osaka Bay, Japan. The model was extensively validated to ensure a reasonable agreement with in situ observations. The western part of the farm is largely influenced by tidal currents, whereas the eastern area is mainly affected by subtidal residual currents that are primarily due to surface wind stress. The released effluent is transported by counterclockwise residual circulation formed off the TSTP. The model reveals that the diversion adequately suppresses the influence on the farm. While the instantaneous effluent concentration is diminished by about 50%, the effluent accumulated on the farm decreased from 2.83 × 104 m3 to 2.01 × 104 m3 due to the diversion, demonstrating an approximately 28% reduction of the effluent from the TSTP by the diversion outfall.

To examine the occurrence in other deep-sea bacteria of two amino acid substitutions (Ala-180 and His-229) in malate dehydrogenase (MDH) found previously in the deep-sea piezophilic Moritella sp. strain 2D2, we cloned and sequenced MDH genes of deep-sea piezophilic Moritella and Shewanella strains isolated from intestinal contents of deep-sea fishes, as well as other Moritella species from deep-sea water and sediments: M. marina, M. japonica, and M. yayanosii. The piezophilic Moritella strains had a Val residue or an Ala residue at position 180 and all the Moritella strains except for one had a His residue at position 229. However, four piezophilic-strain-specific substitutions at positions 103, 111, 229, and 283 were found to be completely conserved in the MDH of the intestinal Moritella strains of deep-sea fishes, indicating the substitutions may be habitat-specific. The piezophilic Shewanella strains had a Val residue and a Gln residue at positions 180 and 229, respectively. However, the MDHs of the Shewanella strains had five piezophilic-strain-specific substitutions at positions 61, 65, 107, 161, and 202. Therefore, the enzymatic strategies for responding to deep-sea high pressure environments of the MDHs between the genera Moritella and Shewanella are potentially different. Moreover, homology modeling shows these substitutions found in the MDHs of both genera except for position 229 in the subunit interface are located on the exposed region of the MDH molecules, indicating the substitutions may be related to the hydration state of the molecules.

The gene encoding malate dehydrogenase (MDH) of the obligately piezophilic deep-sea bacterium Moritella sp. strain 2D2 was cloned and sequenced. There were two positions [close to the active site (Ala-180) and in the subunit interaction site (His-229)] with 2D2-specific substitutions. The MDH genes of strain 2D2 and a psychrophilic bacterium Moritella sp. strain 5710 exhibiting the highest sequence similarity were overexpressed in Escherichia coli. The 2D2 MDH was more heat-stable than the 5710 MDH. The apparent Km value at 62.1 MPa for NADH of the 2D2 MDH was higher than that of the 5710 MDH. The 2D2 MDH in which a His-Gln substitution was introduced at position 229 decreased the thermal stability and Km value at 62.1 MPa. The 5710 MDH that was substituted Gln-229 with His increased the thermal stability and Km value at 62.1 MPa. These results indicate that the His residue at position 229 of the 2D2 MDH may play a role in the thermal stability and the MDH function at high pressure.

From human neuroblastoma-derived SILA cells we have established a rho-0 cell line that is deficient in both respiration and mitochondrial DNA. Lactate dehydrogenase activity, lactate production, and growth in the medium without glucose indicate that these cells shift from aerobic to anaerobic metabolism. Electron microscopic observations revealed abnormal mitochondria with unique cristae structures. Staining with MitoTracker dye showed that the mitochondrial transmembrane potential was reduced by 30-40% from the parent cell levels. These cells were markedly susceptible to H(2)O(2) and died apparently by a necrotic mechanism, a process blocked by deferoxamine in the parent cells but not rho-0 cells. Analysis by inductively coupled plasma-mass spectrometry revealed an approximately 3-fold accumulation of iron in the rho-0 cells at confluence (n = 4-6, three clones, *p < 0.05). Iron and four other metals were all elevated in the cells of one of the rho-0 clones and were similar to control levels in the control cybrid cells, which were replenished with normal mitochondrial DNA. Their sensitivity to H(2)O(2) was also similar to that of the parent cells. These results indicate that a newly established neuronal related rho-0 cell line is highly susceptible to active oxygen species and that these toxicity effects appear to be related to an accumulation of transition metals, which probably occurs through the respiratory impairment.
