RESUMO

To delineate the structural determinants involved in the constitutive activation of the D1 receptor subtypes, we have constructed chimeras between the D1A and D1B receptors. These chimeras harbored a cognate domain corresponding to transmembrane regions 6 and 7 as well as the third extracellular loop (EL3) and cytoplasmic tail, a domain referred herein to as the terminal receptor locus (TRL). A chimeric D1A receptor harboring the D1B-TRL (chimera 1) displays an increased affinity for dopamine that is indistinguishable from the wild-type D1B receptor. Likewise, a chimeric D1B receptor containing the D1A-TRL cassette (chimera 2) binds dopamine with a reduced affinity that is highly reminiscent of the dopamine affinity for the wild-type D1A receptor. Furthermore, we show that the agonist independent activity of chimera 1 is identical to the wild-type D1B receptor whereas the chimera 2 displays a low agonist independent activity that is indistinguishable from the wild-type D1A receptor. Dopamine potencies for the wild-type D1A and D1B receptor were recapitulated in cells expressing the chimera 2 or chimera 1, respectively. However, the differences observed in agonist-mediated maximal activation of adenylyl cyclase elicited by the D1A and D1B receptors remain unchanged in cells expressing the chimeric receptors. To gain further mechanistic insights into the structural determinants of the TRL involved in the activation properties of the D1 receptor subtypes, we have engineered two additional chimeric D1 receptors that contain the EL3 region of their respective cognate wild-type counterparts (hD1A-EL3B and hD1B-EL3A). In marked contrast to chimera 1 and 2, dopamine affinity and constitutive activation were partially modulated by the exchange of the EL3. Meanwhile, hD1A-EL3B and hD1B-EL3A mutant receptors display a full switch in the agonist-mediated maximal activation, which is reminiscent of their cognate wild-type counterparts. Overall, our studies suggest a fundamental role for the TRL in shaping the intramolecular interactions implicated in the constitutive activation and coupling properties of the dopamine D1 receptor subtypes.

RESUMO

A simple new three-wave interferometric technique is used to measure, for what is believed to be the first time, the wave front of femtosecond ultrahigh-peak-power pulses carrying a strong B integral (B = 5.26+/-0.15) in a single shot. Wave-front distortions of a terawatt-class laser system are measured with good accuracy (lambda/50) and discussed. These distortions can significantly reduce the focused peak intensity, emphasizing the necessity of implementing adaptive optics in ultrahigh-intensity chirped-pulse amplification lasers.

RESUMO

Wave-front correction and focal spot improvement of femtosecond laser beams have been achieved, for the first time to our knowledge, with a deformable mirror with an on-line single-shot three-wave lateral shearing interferometer diagnostic. Wave-front distortions of a 100-fs laser that are due to third-order nonlinear effects have been compensated for. This technique, which permits correction in a straightforward process that requires no feedback loop, is also used on a 10-TW Ti:sapphire-Nd:phosphate glass laser in the subpicosecond regime. We also demonstrate that having a focal spot close to the diffraction limit does not constitute a good criterion for the quality of the laser in terms of peak intensity.

RESUMO

An x-ray imaging system with a bent focusing crystal was used for time-resolved one-dimensional imaging of a long plasma column of highly ionized aluminum. This scheme uses a focusing geometry with a spherically bent crystal and a slit on the Rowland circle. Alternative schemes of x-ray monochromatic imaging are briefly discussed. The homogeneity of the up to 40-mm-long laser-generated plasma column was studied with a temporal resolution of 100 ps. The potential spatial resolution of the instrument is 90 microm or better. Monochromatic images taken on the resonance He alpha line of Al XII characterize the homogeneity of a plasma generated to study a recombination x-ray laser scheme, giving an amplified spontaneous emission in Al XI.

RESUMO

The dynamic range of a picosecond visible streak camera has been improved by the combination of a plasma shutter and multishot averaging performed with a photoconductive switch sweep circuit. We use this technique to measure the contrast of a 100-fs laser pulse over 2 ns with a dynamic range of 7 orders of magnitude.
