Skin is the largest organ in body which has important functions. Therefore, to have a healthy skin is very essential, and wound dressings are specifically designed to promote the wound healing process. The aim of this study is to prepare and characterize a fiber-hydrogel wound dressing based on eggshell membrane (ESM) enriched with postbiotic compounds extracted from Lactobacillus plantarum NIMBB003 bacteria. For this purpose, ESM was effectively separated from eggshells through acidic treatment. Then, ultrasound was used for an optimal duration of 1.89 min at 95 % of device's power to expand the pore size of ESM from 6.89 to 10.84 µm to enhance hydrogel infiltration into ESM. The hydrogel (alginate and oxidized alginate) was then infiltrated into the ESM. ATR, SEM, and weight measurement of samples showed the proper infiltration of the hydrogel within the ESM structure. However, biostability analysis revealed that alginate hydrogel was more stable in the hybrid structure compared to oxidase alginate hydrogel. Alginate infiltration into ESM, improved the ultimate strength of the ESM to 1.89 ± 0.17 MPa and water uptake degree to 368.05 % ± 24.34 %. The water vapor transmission rate of the designed dressing was 34.14 ± 1.05 mg/cm2 after 72 h, which means the proper moist management in wound bed. Finally, addition of postbiotics at a concentration of 10 mg/ml into the hydrogel improved cell proliferation in five days. Furthermore, human dermal fibroblast cells adhered to the wound dressings properly and spread along the fibers of the ESM. In general, the developed wound dressing composed of natural biomaterials with extracellular matrix-like structure, can be used effectively to assist the wound healing process.

The different lactobacillus strains isolated from dairy resources may have unique functional properties on human health. Thus, the present study aimed to evaluate the health properties of the lactobacilli isolated from a traditional dairy product in vitro. A total of seven isolated lactobacilli abilities were evaluated for environmental pH reduction, antibacterial activity, cholesterol reduction, and antioxidant activity. Based on the results, the highest decrease in the environment pH was observed in Lactobacillusfermentum B166 with 57%. The antipathogen activity test showed the best results for inhibiting Salmonella typhimurium and Pseudomonas aeruginosa to be obtained by Lact. fermentum 10-18 and Lact. brevis SKB1021 strains, respectively. However, Lact. plantarum H1 and Lact. plantarum PS7319 represented the maximum activity in preventing Escherichia coli; also, Lact. fermentum APBSMLB166 inhibited Staphylococcus aureus more than other strains. Additionally, Lact. crustorum B481 and fermentum 10-18 strains significantly led to a higher reduction in the medium cholesterol than the other strains. The results of antioxidant tests demonstrated that Lact. brevis SKB1021 and Lact. fermentum B166 inhabited radical substrate significantly more than the other lactobacilli. Therefore, four lactobacilli isolated from a traditional dairy product improved some safety indexes positively; thus, they are suggested to be used in manufacturing probiotic supplements.

Nowadays, there is an increasing tendency toward using probiotics in different food systems. In this work, probiotic survival, texture features and sensory properties of synbiotic chewing gum containing encapsulated probiotic organisms (Lactobacillus reuteri) were studied. Probiotics were encapsulated using alginate, inulin (0-1%) and lecithin (0-1%). Storage trials showed that, unlike control, the viability of the probiotic in encapsulated samples was retained after 21 days. Probiotic survival was increased by increasing of inulin and lecithin in cell walls. Samples containing encapsulated organisms had different texture parameters compared to the control. Sensory panelists liked the chewing gum with encapsulated lactobacilli. Thus, chewing gum has been shown to be an excellent food for delivery of probiotic lactobacilli. Principal component analysis (PCA) allowed discriminating among probiotics survival and chewing gum specialties. Partial least squares regression (PLSR) models were applied to find out the relationships between sensory and instrumental data.

OBJECTIVES: Sublingual allergen-specific immunotherapy is a safe and effective method for treatment of IgE-mediated respiratory allergies; however, the underlying mechanisms are not fully understood. This study was planned to test whether sublingual immunotherapy (SLIT) can exert epigenetic mechanisms through which the airway allergic responses can be extinguished. MATERIALS AND METHODS: BALB/c mice were sensitized intraperitoneally and challenged intranasally. Then, they received sublingual treatment with recombinant Che a 2 (rChe a 2), a major allergen of Chenopodium album. After SLIT, allergen-specific antibodies in sera, cytokine profiles of spleen cell cultures, mRNA and protein expression of lung-derived IL-33, IL-25, and TSLP (thymic stromal lymphopoietin), and histone modifications of these three genes were assessed. RESULTS: Following Immunotherapy, systemic immune responses shifted from Th2 to Th1 profile as demonstrated by significant decrease in IgE and IL-4 and substantial increase in IgG2a and IFN-γ. At local site, mRNA and protein levels of lung-derived pro-inflammatory cytokines IL-33 and TSLP were markedly down-regulated following SLIT that was associated with marked enrichment of trimethylated lysine 27 of histone H3 at promoter regions of these two cytokines. CONCLUSION: In our study, sublingual immunotherapy with recombinant allergen effectively attenuated allergic immune responses, at least partly, by induction of distinct histone modifications at specific loci. Additionally, the lung-derived pro-allergic cytokines IL-33 and TSLP could be promising mucosal candidates for either monitoring allergic conditions or therapeutic approaches.
