RESUMO

The exploration of aluminum (Al3+) ion substituted nickel-zinc-cobalt (Ni-Zn-Co) nanoferrites is still at the infancy stage, although the structural, electrical, and magnetic properties have been widely investigated. Single-phase cubic nanospinel ferrites of Ni0.4Zn0.35Co0.25Fe2-x Al x O4 (0 ≤ x ≤ 0.12) with space group Fd 3m were confirmed by the Rietveld refinement X-ray diffraction (XRD) data. Lattice constants displayed a declining trend with compositions x. The average particle size was found to range from 29 to 25 nm. Selected area electron diffraction (SAED) patterns were indexed according to space group Fd 3 m , indicating that nanoparticles are well crystallized. Samples' modes of vibrations swung between redshift and blueshifts as detected in the Raman spectra. The saturation magnetizations (M s) were in the range of 59.85-86.39 emu/g. Frequency-dependent dielectric constants (ε') and ac resistivity (ρ) measurement suggested that samples were highly resistive. These resistive nanoferrites with high saturation magnetizations may function effectively for multifaceted electronic devices.

RESUMO

A multifunctional drug carrier with dual targeting (magnetic and folate-receptor) and pH sensitive core-shell hybrid nanomaterial has been developed to carry an anticancer drug doxorubicin.Superparamagnetic iron oxide nanoparticles (IONPs) were used as core of the carrier and cross-linked folate conjugated chitosan (FA-CS) was acted as shell in which doxorubicin was physically entrapped. Transmission electron microscopy (TEM) analysis confirmed the average particle size of IONPs and FA-CS coated IONPs 8.2 and 15.4 nm respectively. Magnetic measurement indicated that both the IONPs and FA-CS coated IONPs were superparamagnetic at room temperature with a magnetization value 57.72 and 37.44 emu/g respectively. At pH 5.8 (malignant tissue) showed a burst release of 30.05% of the doxorubicin in the first 4 h followed by a sustained release of 88.26% of drug over 72 h. From these results it is expected that doxorubicin loaded nanoparticles can be a promising drug carrier for the treatment of solid tumors with the ability to reduce toxic side effects of drugs by selective targeting and sustained release.

RESUMO

The aim of the present study was to determine whether the combination of two modalities of immunotherapy, targeting two different tumor antigens, may be feasible and non-toxic, yet enhance the killing of a human breast cancer cell line. The first modality was tumor growth factor α-Pseudomonas exotoxin 38 (TGFα-PE38), which specifically targets and kills tumor cells that express the epidermal growth factor receptor. The second modality was mucin-1 (MUC1)-specific cytotoxic T lymphocytes (CTLs), generated by MUC1 stimulation of peripheral blood mononuclear cells, to target the human breast cancer cell line, MCF7. TGFα-PE38 exhibited specific lysis of the MCF7 cells in a concentration- and time-dependent manner. TGFα-PE38 did not kill the normal hematopoietic stem cells or CTLs. Furthermore, TGFα-PE38 was not inhibitory for the growth or differentiation of the normal human hematopoietic stem cells into erythroid and myeloid colonies. In addition, TGFα-PE38 did not inhibit the killing function of CTLs, either when preincubated or co-incubated with CTLs. Finally, therapeutic enhancement was observed, in that TGFα-PE38 and CTLs were additive in the specific lysis of the MCF7 cells. These two modalities of immunotherapy may be beneficial for humans with breast cancer with or without other therapies, including autologous hematopoietic stem cell transplantation, specifically for purging cancer cells from hematopoietic stem cells prior to transplantation.

RESUMO

Interaction between antigen presenting cells and T lymphocytes, through receptors and co-stimulatory molecules present on the surface of these cells, is one of the key means to modulate the adaptive immune system. Tucaresol, a Schiff-base-forming chemical, can be used as a substitute for the physiological donor of carbonyl groups of antigen presenting cells, which can interact with the amine groups of T lymphocytes to modulate the adaptive immune system. This study was done to determine whether tucaresol can enhance killing of cancer cells in vitro as well as protect non-obese diabetic severe combined immunodeficient mice from tumor development by mucin 1 stimulated human mononuclear cells through the adaptive immune system. The expected hypothesis was not supported. Percent specific lysis of MCF-7 tumor cells by mucin 1 stimulated human mononuclear cells was reduced by tucaresol. Furthermore, tucaresol abolished the protective effect of mucin 1 stimulated human mononuclear cells against MCF-7 breast cancer cell growth in non-obese diabetic severe combined immunodeficient mice. This study implies that tucaresol may be of use as an immunosuppressive agent.

RESUMO

CpG-ODNs activate various immune cell subsets and induce the production of numerous cytokines. To determine whether a CpG-ODN-activated innate immune system, without the adaptive immune system, was capable of protecting against cancer cell growth, NOD/SCID mice, which do not have T or B cell function but have a functional innate immune system, were used as a model system. NOD/SCID mice were injected subcutaneously with human prostate cancer cells followed by subcutaneous injection of incremental doses of CpG-ODNs. CpG-ODNs displayed a dose-related antitumoral effect leading to the prevention of tumor growth. These results indicate that ODNs are capable of activating the innate immune system and destroying human cancer cells in the absence of the adaptive immune system.

RESUMO

BACKGROUND: Seasonality has been recognized as a key determinant of human energy balance, especially in low-income countries. OBJECTIVE: The objective was to test the hypothesis that, in rural Bangladesh, different age and sex groups adapt similarly to seasonal changes in energy intake (EI). DESIGN: A prospective study was carried out in 2 rural Bangladeshi villages in the lean and peak seasons. Data on anthropometric measures (weight, height, and midupper arm circumference) and dietary food intake (collected with the use of a 24-h food weighing method) were obtained from all subjects from 304 households. RESULTS: The average EI increased from a least-squares mean (+/- SE) of 7.87 +/- 0.10 MJ x person(-1) x d(-1) in the lean season to 9.47 +/- 0.13 MJ x person(-1) x d(-1) in the peak season. In children and adolescents aged < 18 y of age, the prevalence of underweight (weight-for-age) was not significantly different (56%) in the 2 seasons. Among adults, a significantly higher prevalence of chronic energy deficiency [body mass index (in kg/m(2)) < 18.5] was observed in the peak season (67%) than in the lean season (61%), despite a higher EI in the peak season. Other determinants of seasonal nutritional status are presented. CONCLUSIONS: Seasonal fluctuations in EI were substantial in all age and sex groups. Children and adolescents showed no significant seasonal changes in the prevalence of underweight, which indicated that they adapted to changes in EI. In adults, the season in which EI was high coincided with average weight loss, which indicated that adults did not adapt fully to seasonal fluctuations in EI and that seasonal energy expenditure is probably a major determinant of nutritional status.

RESUMO

Suppurative keratitis is an important preventable cause of blindness, particularly in the developing world. This study analyses 142 cases of suppurative keratitis referred to Chittagong Eye Infirmary Bangladesh. Some 53.5% of cases were bacterial and 35.9% were fungal. The five most common pathogens were: Pseudomonas sp. 24%, Streptococcus pneumoniae 17%, Aspergillus sp. 13%, Fusarium sp. 7% and Curvularia sp. 6%. Gram stain and culture results were consistent in 62.6% of cases. Previous antibiotic treatment was a significant factor for failure of culture isolation and less so for Gram stain failure. On Gram stain, 55.9% of pseudomonal cases were missed, but only 2% of fungal cases were missed. Over all, Gram stain had a sensitivity of 62% and positive predictive value of 84% for bacterial cases, and 98% and 94% for fungal cases, respectively. Fungal ulcers were typically filamentous, but an antecedent history of trauma was not common. The most frequent injury was due to rice grains, but the inoculum appeared to be introduced during eye washing with contaminated water. Pseudomonal ulcers occurred most frequently in the monsoon season, and Fusarium cases were seen only in the hot, dry season.

RESUMO

A case of Dichotomophthoropsis nymphaearum, a dematiaceous hyphomycete, is described from Bangladesh. This species has not previously been reported as a human pathogen. Hyphae were observed in Gram stained corneal scrapings and the species was grown from cultured corneal material. Treatment with topical econazole and subconjunctival injections of miconazole was successful in the short term but long-term outcome could not be determined. The results of sensitivity tests for six antifungal drugs are reported for the isolate.
