RESUMO

BACKGROUND: The different clinical presentations of fibromyalgia (FMS) may play independent roles in the unclear etiology of cognitive impairments and depressive symptoms seen in this population. Understanding how these clinical presentations are associated with FMS's clinical and neurophysiological aspects is important when developing effective treatments. AIM: To explore the relationship between memory complaints and depressive symptoms, and the different clinical and neurophysiological characteristics of FMS. METHODS: Cross-sectional data analysis from a randomized clinical trial. Baseline demographics, physical fitness, sleep, anxiety, depression, cortical excitability, and pain (clinical and mechanistic) data from 63 FMS subjects were used. Multiple linear and logistic association models were constructed. RESULTS: Final regression models including different sets of predictions were statistically significant (p < 0.001), explaining approximately 50% of the variability in cognitive complaints and depression status. Older subjects had higher levels of anxiety, poor sleep quality, lower motor threshold, and higher relative theta power in the central area, are more likely to have clinical depression. Higher anxiety, pain and theta power were associated with an increase memory complaint. CONCLUSION: Depression symptoms seem to be associated with TMS-indexed motor threshold and psychosocial variables, while memory complaints are associated with pain intensity and higher theta oscillations. These mechanisms may be catalyzed and/or triggered by some behavioral and clinical features such as older age, sleep disruption, and anxiety. The correlation with clinical variables suggests the increasing of theta oscillations is a compensatory response in patients with FMS, which can be explored in future studies to improve the treatment for FMS.

RESUMO

INTRODUCTION: In general, two characteristics are ever present in NMR-based metabolomics studies: (1) they are assays aiming to classify the samples in different groups, and (2) the number of samples is smaller than the feature (chemical shift) number. It is also common to observe imbalanced datasets due to the sampling method and/or inclusion criteria. These situations can cause overfitting. However, appropriate feature selection and classification methods can be useful to solve this issue. OBJECTIVES: Investigate the performance of metabolomics models built from the association between feature selectors, the absence of feature selection, and classification algorithms, as well as use the best performance model as an NMR-based metabolomic method for prostate cancer diagnosis. METHODS: We evaluated the performance of NMR-based metabolomics models for prostate cancer diagnosis using seven feature selectors and five classification formalisms. We also obtained metabolomics models without feature selection. In this study, thirty-eight volunteers with a positive diagnosis of prostate cancer and twenty-three healthy volunteers were enrolled. RESULTS: Thirty-eight models obtained were evaluated using AUROC, accuracy, sensitivity, specificity, and kappa's index values. The best result was obtained when Genetic Algorithm was used with Linear Discriminant Analysis with 0.92 sensitivity, 0.83 specificity, and 0.88 accuracy. CONCLUSION: The results show that the pick of a proper feature selection method and classification model, and a resampling method can avoid overfitting in a small metabolomic dataset. Furthermore, this approach would decrease the number of biopsies and optimize patient follow-up. 1H NMR-based metabolomics promises to be a non-invasive tool in prostate cancer diagnosis.

RESUMO

BACKGROUND: Clinical predictors of sleep quality in patients with fibromyalgia syndrome (FMS) are still unknown. By identifying these factors, we could raise new mechanistic hypotheses and guide management approaches. We aimed to describe the sleep quality of FMS patients and to explore the clinical and quantitative sensory testing (QST) predictors of poor sleep quality and its subcomponents. METHODS: This study is a cross-sectional analysis of an ongoing clinical trial. We performed linear regression models between sleep quality (Pittsburgh Sleep Quality Index [PSQI]) and demographic, clinical, and QST variables, controlling for age and gender. Predictors for the total PSQI score and its seven subcomponents were found using a sequential modeling approach. RESULTS: We included 65 patients. The PSQI score was 12.78 ± 4.39, with 95.39% classified as poor sleepers. Sleep disturbance, use of sleep medications, and subjective sleep quality were the worst subdomains. We found poor PSQI scores were highly associated with symptom severity (FIQR score and PROMIS fatigue), pain severity, and higher depression levels, explaining up to 31% of the variance. Fatigue and depression scores also predicted the subjective sleep quality and daytime dysfunction subcomponents. Heart rate changes (surrogate of physical conditioning) predicted the sleep disturbance subcomponent. QST variables were not associated with sleep quality or its subcomponents. CONCLUSIONS: Symptom severity, fatigue, pain, and depression (but no central sensitization) are the main predictors of poor sleep quality. Heart rate changes independently predicted the sleep disturbance subdomain (the most affected one in our sample), suggesting an essential role of physical conditioning in modulating sleep quality in FMS patients. This underscores the need for multidimensional treatments targeting depression and physical activity to improve the sleep quality of FMS patients.

RESUMO

Field experiments testing the effect of phosphate-solubilizing rhizobacteria (PSRB) should consider the cropping history and the method used to inoculate the strains. We evaluated the hypothesis that PSRB previously isolated from soybean seedlings could be effective in promoting growth in this oilseed crop in soils with different cultivation periods. We also evaluated whether this growth promotion could be influenced by cultivation histories or the inoculation method (via seeds or soil). Thus, we conducted an experiment in five fields cultivating Glycine max during two seasons (2019/2020 and 2020/2021), to test the effectiveness of PSRB (SAF9-Brevibacillus sp., SAF11-Brevibacillus sp., and SAC36-Bacillus velezensis) compared with results observed for the inoculant BiomaPhos (mix of Bacillus subtilis and Bacillus megaterium). The present study was based on the evaluation of vegetative growth, nutritional and yield parameters, and microbial biomass carbon (MBC). PSRB were more effective than, or showed similar effectiveness to, BiomaPhos for most of the evaluated vegetative, nutritional, and yield characteristics. In the fields tested in the summer 2019/2020 crop, SAC36 and SAF9 strains stood out as growth promoters, whereas in the 2020/2021 crop, SAF11, SAC36, and BiomaPhos were notable. There did not seem to be a direct relationship between long histories of soybean cultivation as a monoculture and low yield in the field. However, yield seems to be associated with soil nutritional characters such as Ca, Mg, K, P, cation exchange capacity, and organic matter levels. PSRB inoculation positively affected nodulation (NN) and nodule dry mass (NDM) in the evaluated fields in the 2019/2020 crop, and the aerial part dry mass (APDM), NN, NDM, yield, and MBC of the evaluated fields in the 2020/2021 crop. In contrast, the inoculation method was observed to have a strong effect on APDM, NN, root dry mass, and MBC, as the plants inoculated via seed showed higher mean values than those in the plants inoculated via soil. This study demonstrated the growth-promoting potential of new phosphate-solubilizing strains, which may eventually be incorporated by the biostimulants market to freely compete with BiomaPhos.

RESUMO

The cardiac evaluation of wild animals is still a wide and largely unknown field for several species. Therefore, through complimentary examinations such as radiography, echocardiography and serum troponin levels, this study aimed at describing the values observed in 12 crab-eating foxes (Cerdocyon thous) anesthetized with a combination of intramuscular ketamine and midazolam. Thus, through complementary exams such as radiography, echocardiography and serum troponin levels, the aim of this study was to describe the cardiac values in 12 wild foxes (C. thous) anesthetized with an intramuscular injection of ketamine and midazolam. After anaesthetization, the radiographic, echocardiographic and immunoenzymatic reference values for the 12 males in the sample group were determined. Compared with those in domestic canids, there was a decrease in the sizes of the septum, wall and left ventricular cavity as well as decreases in the transmitral blood flow velocity indices, correlated with preserved serum cardiac troponin (cTnI) levels. Thus, M-mode echocardiography proved to be safer, with results that were comparative to those for other species of wild canids with indexed values. In addition, when evaluating the systolic function and segmentary contractions, the anesthetic combination did not have any effects on the results of complementary examinations performed in crab-eating foxes (C. thous) included in this study.

RESUMO

ABSTRACT Objective: To analyze reformatted sagittal sternal tomography images and classify sternal body curvature types, and compare different types of pectus populations with one another and with normal individuals. Methods: In total, 50 controls and 167 pectus patients were selected for chest CT to analyze the median sagittal plane, of whom 89 had pectus carinatum (mean age, 12 ± 10 years) and 78 pectus excavatum (mean age, 14 ± 10 years). Clinical types of pectus were classified as inferior, superior, or lateral pectus carinatum, and localized or broad pectus excavatum. The following types of sternal patterns were defined: gradual vertical curve, gradual posterior curve, gradual anterior curve, proximal third curve, middle third curve, distal third curve, anterior rectilinear, vertical rectilinear, and posterior rectilinear. Statistical analyses were performed to compare the different types of pectus with one another and with the control group. Results: Patients with different thoracic deformities, but with similar sternal curvature patterns, were observed. Some types of sternal curvature were significantly more frequent in certain types of pectus (p < 0,05). The gradual vertical curve and anterior rectilinear types prevailed in controls (p < 0,05). Conclusion: Some sternal curvature patterns were more frequent than the others in certain types of pectus and the controls. Level of Evidence II, Prognostic studies - investigating the effect of a patient characteristic on the outcome of disease.

RESUMO

Infectious diseases and the rapid development of pathogens resistant to conventional drugs are a serious global public health problem, which motivates the search for new pharmacological agents. In this context, cationic peptides without disulfide bridges from different species of scorpion venom have been the target of scientific studies due to their multifunctional activities. Stigmurin is a linear peptide composed of 17 amino acid residues (Phe-Phe-Ser-Leu-Ile-Pro-Ser-Leu-Val-Gly-Gly-Leu-Ile-Ser-Ala-Phe-Lys-NH2), which is present in the venom gland of the scorpion Tityus stigmurus. Here we present investigations of the in vitro antioxidant action of Stigmurin together with the in vivo antibacterial and healing activity of this peptide in a wound infection model induced by Staphylococcus aureus. In addition, we have reports for the first time of the three-dimensional structure determined by NMR spectroscopy of a peptide without disulfide bridges present in scorpion venom from the Tityus genus. Stigmurin showed hydroxyl radical scavenging above 70 % at 10 µM and antibiotic action in the skin wound, reducing the number of viable microorganisms by 67.2 % on the 7 day after infection. Stigmurin (1 µg / µL) increased the retraction rate of the lesion, with wound area reduction of 43 % on the second day after skin injury, which indicates its ability to induce tissue repair. Stigmurin in trifluoroethanol:water exhibited a random conformation at the N-terminus region (Phe1 to Pro6), with a helical structure from Ser7 to Phe16. This structural information, allied with the multifunctional activity of Stigmurin, makes it an attractive candidate for the design of novel therapeutic agents.

RESUMO

Anionic peptides of scorpions are molecules rich in aspartic and/or glutamic acid residues and correspond to a class of peptides without disulfide bonds that are still little explored. TanP is a linear anionic peptide (50 amino acid residues and net charge -20) present in the venom gland of the scorpion, Tityus stigmurus, with chelating properties for Cu2+ ion and immunomodulatory properties. The therapeutic application of chelating molecules is related to cases of acute or chronic intoxication by metals, neurodegenerative diseases, hematological diseases, healing of skin wounds, cardiovascular diseases, and cancer. In this approach, the chelating activity of TanP was evaluated in relation to new metal ions (Fe2+ and Zn2+) of biological importance, as well as its antioxidant, hemostatic, immunomodulatory, and healing potential, aiming to expand the biological and biotechnological potential of this peptide. TanP (25 µM) was able to form stable complexes with Fe2+ in a ratio of 1:5 (TanP: Fe2+). Theoretical results suggest that TanP can work as a sensor to identify and quantify Fe2+ ions. The fluorescence intensity of TanP (1.12 µM) decreased significantly after the addition of Fe2+, obtaining the highest ratio 1: 7.4 (TanP: Fe2+) that led to the lowest fluorescence intensity. For Zn2+, no relevant spectral change was noted. TanP (50 µM) showed a maximum of 3% of hemolytic activity, demonstrating biocompatibility, as well as exhibiting a 1,1-diphenyl-2-picrylhydrazyl radical-scavenging activity of above 70% at all the concentrations tested (1-25 µM), and 89.7% iron-chelating activity at 25 µM and 96% hydroxyl radical-scavenging activity at 73.6 µM. In addition, TanP (12.5 and 25 µM) revealed an anticoagulant effect, prolonging the clotting time in prothrombin time and activated partial thromboplastin time assays, with no fibrinogenolytic activity. TanP (12.5 and 25 µM) induced the release of TNF-α by murine macrophages, in the absence of lipopolysaccharides, with a concentration-dependent increase and also stimulated the migration of 3T3 cells in the in vitro healing assay. Thus, TanP revealed a multifunctional potential, being useful as a prototype for the development of new therapeutic and biotechnological agents.

RESUMO

Amasonia campestris (Aubl.) Moldenke, belongs to the Lamiaceae family, found in the North, Northeast, Midwestand Southeast regions of Brazil. A. campestris tea is popularly used for the fight against malaria, diabetes among otherdiseases. Its common names are mendoca, macaw tail, macaw bamboo. The present study evaluated the genotoxic andantigenotoxic potential of the methanol extract of the roots of Amasonia campestris in polychromatic erythrocytes of Swissmice. The animals were treated with different concentrations of Amasonia campestris (250, 500, 1000 and 2000 mg/kgb.w.) including positive control groups (doxorubicin, DXR 16 mg/kg body weight), negative (water) and solvent (dimethylsulfoxide, DMSO). Concentrations were administered to the animals via a gavage as well as the negative control andsolvent. The positive control was given intraperitoneally. The animals were treated daily with the respective doses for 15days for genotoxic evaluation. after that, samples of caudal peripheral blood were collected at the hours: 24 and 48hrs afterthe first gavage, and on days 7 and 15. For the antigenotoxic evaluation, mice were treated on day 14 with intraperitonealinjections of DXR and caudal peripheral blood samples were collected 24 and 48hrs after this intervention. After thetreatments, 2,000 polychromatic erythrocytes were counted per animal from each group to evaluate the frequency ofMicronuclei. The results showed that the methanol extracts of the roots of A. campestris were not genotoxic because nostatistically significant difference was observed when compared with the negative control. Animals treated with differentconcentrations of Amasonia campestris extracts associated with DXR had a significant reduction of micronucleatedpolychromatic erythrocytes when compared with the positive control. In conclusion, the methan...

RESUMO

A titanium surface nitrided by plasma contains nitrogen ions that guarantee resistance to corrosion and biocompatibility. Despite this, no descriptions concerning the influence of the expression of cell adhesion proteins and their influence on osteogenic cell differentiation are available. Thus, the present study aimed to assess the response of murine pre-osteoblastic cells (MC3T3-E1) cultured on nitrided titanium surfaces. Pre-osteoblastic cells were grown on polished titanium discs, used as controls, and on previously characterized plasma-nitrided titanium discs. Cells from both groups were submitted to the MTT cell viability test. The expressions of α5, α2, and ß1 integrin were assessed by flow cytometry and immunofluorescence, while osteocalcin expression was assessed by flow cytometry. The nitrided surface presented higher α2 and ß1 integrin expressions, as well as osteocalcin expression, when compared to the polished surface, with no alterations in cell viability. These findings seem to suggest that the plasma nitriding treatment produces a titanium surface with the potential for effective in vitro osseointegration.

Abstract Objective To obtain the oil of Astrocaryum aculeatum (A.a), and evaluate its genotoxicity/antigenotoxicity activities using the micronucleus test in peripheral blood cells. Methods The oil of Astrocaryum aculeatum was obtained by hydraulic pressing. The animals used were healthy Swiss male mice, at 6-7 weeks of age; there were six per group. The genotoxic and antigenotoxic activity of concentrations were 500, 1,000 and 2,000 mg/kg per 0.5 mL (oral), followed or not followed by intraperitoneal injection of doxorubicin (0.3 mL-15 mg/kg by body weight), in addition to a negative group (water) and dimethyl sulfoxide (600 μL). Peripheral blood samples were collected 24h and 48h after treatment. Results A statistically significant reduction was identified in the frequency of micronuclei in polychromatic cells ranging from 34.72% to 38.19% for 24-hour treatments, and from 63.70% to 66.12% for 48 hour. Conclusion The fixed oil of tucumã presented antigenotoxic potential for the concentrations used in acute treatments.

RESUMO

BACKGROUND: The cystic dilatation of the biliary tract is a rare disease and uncertain origin. It is recognized more frequently in children; however, its incidence comes increasing in adults, representing 20% of the cases. AIM: To evaluate morbimortality rates, evolution and handing of patients with cystic dilatation bile ducts in adults. METHODS: Were evaluated, retrospectively, five adults who had the diagnosis of choledochal cyst and that had been submitted to some surgical procedure. RESULTS: Abdominal pain was the commonest complain to all patients. Jaundice was present in 80%. Ultrasound scanning was done in all the cases as initial examination. CT scan, magnetic resonance imaging and endoscopic retrograde cholangiopancreatography were also done in some patients; however, the diagnosis was established intra-operatively in all cases. The cyst resection with reconstruction of the biliary tract was done in 60%; the cystojejunostomy in 20%; and in 20% biliary tract drainage. CONCLUSIONS: Biliary tract cystic dilatation is a rare disease. However, its incidence is increasing in the adult population, so, it must be thought as differential diagnosis when facing obstructive jaundice.

Samples of titanium were nitrided using plasma and a composition of two gases: 80% H2-20% N2 and 20% H2-80% N2. The influence of these treatments on their physical chemical properties and biological response when subjected to the primary cell culture of dental pulp were studied. Analyses by X-ray diffraction indicate the formation of TiN in both nitrided samples in contrast to the non-nitrided surface samples that contained only the Ti-α phase. Changes in hardness, topography and wettability of these samples compared to non-nitrided samples were found as well. Finally, the analysis of cell’s adhesion in these samples indicate that treatment increases its value being higher for samples treated at higher hydrogen concentration.

RESUMO

Artepillin C (3,5-diprenyl-p-coumaric acid) is one of the major phenolic compounds found in Brazilian green propolis, as well as in its botanical source, Baccharis dracunculifolia DC (Asteraceae). The present study evaluated the possible genotoxic and protective activities of artepillin C, in vitro, using methyl methanesulfonate (MMS) as a positive control, by comet and micronucleus assays. The cultures of Chinese hamster lung fibroblasts (V79 cells) were treated with different concentrations of artepillin C (2.5, 5.0, 10.0, and 20 µM). In antigenotoxicity assessment, the 3 concentrations of artepillin C (2.5, 5.0, and 10.0 µM) were associated with MMS (200 µM-comet assay and 400 µM-micronucleus assay). A statistically significant increase in the DNA damage and micronucleus frequencies was observed in the culture treated with the highest concentration of the artepillin C in comparison to the control group. All concentrations of artepillin C showed protective activity in relation to MMS-induced genotoxicity, which may be due to its antioxidant properties.

RESUMO

Copaifera lansdorffii Desf. is known as 'copaíba', 'copaiva' or 'paú-de-óleo', and is found in part of Brazil. The present study was undertaken to evaluate the genotoxic potential of C. langsdorffii leaf hydroalcoholic extract (CLE) and its influence on the genotoxicity induced by the chemotherapeutic agent doxorubicin (DXR) using the Swiss mouse peripheral blood micronucleus test. HPLC analysis of CLE using two monolithic columns linked in series allowed quantification of two major flavonoid heterosides, quercitrin and afzelin. Animals were treated with CLE by gavage at doses of 10, 20, 40 and 80 mg kg(-1) body weight per day, each for 20 days. Peripheral blood samples were collected at 24 and 48 h, and 7, 15 and 21 days after the beginning of the treatment. For the antigenotoxicity evaluation, the animals treated with different concentrations of CLE received DXR (15 mg kg(-1) body weight, intraperitoneal) at day 20. The peripheral blood samples were collected 24 and 48 h after the treatment with DXR. The results demonstrated that CLE itself was not genotoxic in the mouse micronucleus assay. In animals treated with CLE and DXR, the number of micronucleus was significantly decreased compared with animals receiving DXR alone. The putative antioxidant activity of one or more of the active compounds of CLE may explain the effect of this plant on DXR genotoxicity.
