Heavy metals such as cadmium (Cd), arsenic (As), and lead (Pb) pose significant health risks, particularly in Asia, where rice is a staple for nearly three billion people. Despite their known dangers and environmental prevalence, studies addressing their concentrations in rice across different regions and the associated health implications remain insufficient. This review systematically examines the occurrence and impact of these toxic elements, filling a critical gap in the literature. Data from seven countries indicate mean concentrations in the order of Pb > As>Cd, with values of 0.255 ± 0.013, 0.236 ± 0.317, and 0.136 ± 0.150 mg/kg, respectively. Uncertainty analysis shows extensive variability, especially for Cd, with a 95% confidence interval range from 0.220 to 0.992 mg/kg. The typical daily intake of heavy metals through rice consumption, in the order of As>Cd > Pb, frequently exceeds safe limits. Generally, data obtained from various studies showed that children were more prone to heavy metal contamination through rice consumption than adults. This review is fundamental for ongoing monitoring, future research, and management strategies to reduce heavy metal contamination in rice.

PURPOSE: Attenuated Salmonella typhimurium is a potential biotherapeutic antitumor agent because it can colonize tumors and inhibit their growth. The present study aimed to develop a doxycycline (Doxy)-inducible gene switch system in attenuated S. typhimurium and assess its therapeutic efficacy in various tumor-bearing mice models. PROCEDURES: A Doxy-inducible gene switch system comprising two plasmids was engineered to trigger the expression of cargo genes (Rluc8 and clyA). Attenuated S. typhimurium carrying Rluc8 were injected intravenously into BALB/c mice bearing CT26 tumors, and bioluminescence images were captured at specified intervals post-administration of doxycycline. The tumor-suppressive effects of bacteria carrying clyA were evaluated in BALB/c mice bearing CT26 tumors and in C57BL/6 mice bearing MC38 tumors. RESULTS: Expression of the fimE gene, induced only in the presence of Doxy, triggered a unidirectional switch of the POXB20 promoter to induce expression of the cargo genes. The switch event was maintained over a long period of bacterial culture. After intravenous injection of transformed Salmonella into mice bearing CT26 tumors, the bacteria transformed with the Doxy-inducible gene switch system for Rluc8 targeted only tumor tissues and expressed the payloads 2 days after Doxy treatment. Notably, bacteria carrying the Doxy-inducible gene switch system for clyA effectively suppressed tumor growth and prolonged survival, even after just one Doxy induction. CONCLUSIONS: These results suggest that attenuated S. typhimurium carrying this novel gene switch system elicited significant therapeutic effects through a single induction triggering and were a potential biotherapeutic agent for tumor therapy.

Type 2 diabetes mellitus (T2DM) has been increasing rapidly in Vietnam as well as world-wide. One of the major causes of the condition is low fiber intake. It is difficult to eat large amounts of vegetables every day to reach a sufficient amount of fiber but Textured Soybean Protein is rich in fiber. The study aimed to examine the effectiveness of Textured Soybean Protein consumption on T2DM patients. In this randomized controlled trial, 47 T2DM patients were divided into an intervention group (n=24) and a control group (n=23). The intervention group were asked to consume 40 g Textured Soybean Protein in 2 dishes for 4 wk. The control group continued their usual diet. Fasting blood samples were drawn before and after intervention to measure fasting plasma glucose (FPG), fructosamine, low density lipoprotein-cholesterol (LDL-C), high density lipoprotein-cholesterol (HDL-C), total cholesterol (T-C), and triglycerides (TG). A 3-day food record was conducted at 1 wk before (baseline) and at the last week (final) of the intervention period. In the Textured Soybean Protein consumption group, there was a significant decrease in fructosamine (363±86 µmol/L to 347±82 µmol/L, p=0.03), T-C (5.2±0.9 mmol/L to 4.8±0.8 mmol/L, p=0.02) and TG (3.5±2.2 mmol/L to 2.8±2.0 mmol/L, p=0.02). Total energy intake in the two groups did not change significantly. There was a shift in the dietary pattern of the Textured Soybean Protein consumption group; lipid intake showed a significant decrease (p=0.001) and fiber intake increased by 6 g (p<0.001). The consumption of Textured Soybean Protein in the diet could have favorable effects in improving glycemic and lipid concentrations in T2DM patients.

PURPOSE: In the programming of tumor-targeting bacteria, various therapeutic or reporter genes are expressed by different gene-triggering strategies. Previously, we engineered pJL87 plasmid with an inducible bacterial drug delivery system that simultaneously co-expressed two genes for therapy and imaging by a bidirectional tet promoter system only in response to the administration of exogenous doxycycline (Doxy). In this multi-cassette expression approach, tetA promoter (PtetA) was 100-fold higher in expression strength than tetR promoter (PtetR). In the present study, we developed pJH18 plasmid with novel Doxy-inducible gene expression system based on a tet promoter. PROCEDURES: In this system, Tet repressor (TetR) expressed by a weak constitutive promoter binds to tetO operator, resulting in the tight repression of gene expressions by PtetA and PtetR, and Doxy releases TetR from tetO to de-repress PtetA and PtetR. RESULTS: In Salmonella transformed with pJH18, the expression balance of bidirectional tet promoters in pJH18 was remarkably improved (PtetA:PtetR = 4~6:1) compared with that of pJL87 (PtetA:PtetR = 100:1) in the presence of Doxy. Also, the expression level by novel tet system was much higher in Salmonella transformed with pJH18 than in those with pJL87 (80-fold in rluc8 and 5-fold in clyA). Interestingly, pJH18 of the transformed Salmonella was much more stably maintained than pJL87 in antibiotic-free tumor-bearing mice (about 41-fold), because only pJH18 carries bom sequence with an essential role in preventing the plasmid-free population of programmed Salmonella from undergoing cell division. CONCLUSIONS: Overall, doxycycline-induced co-expression of two proteins at similar expression levels, we exploited bioluminescence reporter proteins with preclinical but no clinical utility. Future validation with clinically compatible reporter systems, for example, suitable for radionuclide imaging, is necessary to develop this system further towards potential clinical application.

The increasing accumulation of pharmaceuticals in aquatic ecosystems could impair freshwater quality and threaten human health. Despite the adsorption of pharmaceuticals on biochars is one of the most cost-effective and eco-friendly removal methods, the wide variation of experimental designs and research aims among previous studies pose significant challenge in selecting biochar for optimal removal. In this work, literature data of 1033 sets with 21 variables collected from 267 papers over ten years (2010-2020) covering 19 pharmaceuticals onto 88 biochars were assessed by different machine learning (ML) algorithms i.e., Linear regression model (LM), Feed-forward neural networks (NNET), Deep neutral networks (DNN), Cubist, K-nearest neighbor (KNN), and Random forest (RF), to predict equilibrium adsorption capacity (Qe) and explore adsorption mechanisms. LM showed the best performance on ranking importance of input variables. Except for initial concentration of pharmaceuticals, Qe was strongly governed by biochars' properties including specific surface area (BET), pore volume (PV), and pore structure (PS) rather than pharmaceuticals' properties and experimental conditions. The most accurate model for estimating Qe was achieved by Cubist, followed by KNN, RF, KNN, NNET and LM. The generalization ability was observed by the tuned Cubist with 26 rules for the prediction of the unseen data. This study not only provides an insightful evidence for data-based adsorption mechanisms of pharmaceuticals on biochars, but also offers a potential method to accurately predict the biochar adsorption performance without conducting any experiments, which will be of high interests in practice in terms of water/wastewater treatment using biochars.

This study was conducted to compare the exposure levels of five heavy metals via different pathways based on the field samplings and questionnaire surveys of children residing in an informal e-waste processing village and a reference village in Viet Nam. The findings revealed that levels of the total average daily intake (ADI) of the five heavy metals collected from a child at the exposed village were 3.90 times higher (p < 0.01) than that of a child at the reference village. Ingestion of cooked rice was the largest contributor to the total ADI of the children tested at both villages. However, the risks from water drinking and dermal contact of soil were negligible. The total non-carcinogenic risk and carcinogenic risk in an exposed child were significantly higher (p < 0.01) than their respective risks in a reference child. The non-carcinogenic risk to an exposed child was likely to occur, while the risk to a reference child was negligible. The carcinogenic risks found in children from both of the villages, however, were higher than the acceptable values, indicating the potential health risks to the children from both villages. The susceptibility of children to heavy metal contaminations shown in this study suggests that a mitigating measure need to be initiated jointly by a public agency and a private organization to prevent children from the risks of being exposed to the contaminants.
