Soil aggregates are the main sites for the decomposition of soil organic matter and the formation of humus. The composition characteristics of aggregates with different particle sizes are one of the indicators for soil fertility. We explored the effects of management intensity (frequency of fertilization and reclamation) on soil aggregates in moso bamboo forests, including mid-intensity management (T1, fertilization and reclamation every 4 years), high-intensity management (T2, fertilization and reclamation every 2 years), and extensive management (CK). The water-stable soil aggregates (0-10, 10-20, and 20-30 cm layers) from moso bamboo forest were separated by a combination of dry and wet sieving method and the distribution of soil organic carbon (SOC), total nitrogen (TN) and available phosphorus (AP) across different soil layers were determined. The results showed that management intensities had significant effects on soil aggregate composition and stability, and SOC, TN, AP distribution of moso bamboo forests. Compared with CK, T1 and T2 decreased the proportion and stability of macroaggregates in 0-10 cm soil layer, but increased that in 20-30 cm soil layer, while reduced the content of organic carbon in macroaggregates, the contents of organic carbon, TN and AP in microaggregates. Such results indicated that the intensified management was not conducive to formation of macroaggregates in 0-10 cm soil layer and carbon sequestration in macroaggregates. It was beneficial to the accumulation of organic carbon in soil aggregates and nitrogen and phosphorus in microaggregates with lower human disturbance. Mass fraction of macroaggregates and organic carbon content of macroaggregates was significantly positively correlated with aggregate stability, which best explained the variations of aggregate stability. Therefore, macroaggregates and organic carbon content of macroaggregates were the most important factors affecting the formation and stability of aggregates. Appropriate reduction of disturbance was beneficial to the accumulation of macroaggregates in the topsoil, the sequestration of organic carbon by macro-aggregates, and the sequestration of TN and AP by microaggregates, and improving soil quality and sustainable management in moso bamboo forest from the point of view of soil aggregate stability.

BACKGROUND: Acetaminophen (APAP) overdose represents one of the most common drug-induced liver injuries (DILI) worldwide. Oxidative damage to the hepatocytes and their resultant autophagy are the key components in the APAP-induced DILI. Echinacea purpurea polysaccharide (EPPS), the component extracted from the root of Echinacea purpurea (L.) Moench, shows various biological functions including immunoregulation and antioxidant activity. PURPOSE: This study aimed to elucidate the protective effect of EPPS against APAP-induced DILI and the underlying mechanisms. RESULTS: EPPS attenuates APAP overdose induced DILI in mice and ameliorates inflammation and oxidative stress in mice with APAP overdose-induced DILI. Furthermore, EPPS protected the hepatocytes against APAP-induced liver injury by suppressing apoptosis. EPPS ameliorates APAP-induced DILI via an autophagy-dependent mechanism in vivo and increases autophagy with a reduction in oxidative stress and inflammation in vitro. Parkin knockdown prevents the autophagic-dependent manner of EPPS effects in APAP-treated hepatocytes. CONCLUSIONS: EPPS exhibited a strong hepatoprotective effect against APAP-induced DILI and was correlated with reduction of autophagy-dependent oxidant response, inflammation, and apoptosis. Moreover, the findings indicated that EPPS exerts its hepatoprotective effect against APAP mainly via Parkin-dependent autophagy, and the use of EPPS can serve as a promising novel therapeutic strategy for APAP-induced DILI.

Moso bamboo (Phyllostachys edulis) is widely considered to be effective in capturing and sequestering atmospheric C, but the long-term effects of extensive management strategies on soil organic carbon (SOC), bacterial communities, fine root (FR, ø ≤ 2 mm) traits, and their inherent connection remain unclear. In this study, we simultaneously measured the SOC content of the bulk and rhizosphere soil fractions, the aggregate stability, the chemical composition of SOC (solid-state 13C nuclear magnetic resonance [NMR]), the bacterial community structure in the rhizosphere, and the FR morphological traits including biomass, specific root length (SRL), and root length density (RLD) along a chronosequence (stand age of 19, 37, and 64 years) of extensively managed Moso bamboo plantations and in an adjacent secondary forest as a control. The organic C content in both the rhizosphere and bulk soil increased rapidly with plantation age in the 0-20- and 20-40-cm soil layers, accompanied by an increase in the aggregate stability. FR traits including biomass, SRL, and RLD also increased continuously in response to soil C:N:P stoichiometry. All of these traits were significantly correlated with SOC, occluded particulate organic C (oPOC), and mineral-associated organic C (MOC), suggesting that FR traits could drive the soil C sequestration with the plantation age. Further analysis indicated that the microbial biomass C (MBC) content, MBC/total organic carbon (TOC) ratio, and bacterial abundance decreased with the plantation age, and the shift from soil oligotrophy to copiotrophy bacteria were mainly driven by changes in FR traits and SOC properties. Such a reassembly of bacterial communities combined with an increase in root biomass is favorable for the accumulation of stable C functional groups (alkyl C or aromatic C). Our findings indicate that extensive management regimes of Moso bamboo plantations could promote long-term soil C sequestration especially in the rhizosphere by promoting the formation of soil aggregates and organic-mineral complexes and by shifting bacterial community composition, and that these changes can be inferred through changes in the FR traits.

To examine the effects of management measures on carbon and nitrogen contents, as well as their distribution and structural characteristics of different soil fractions in Moso bamboo plantations, we compared three types of the bamboo forests (undisturbed, extensively managed, and intensively managed) and the control secondary broadleaved evergreen forest using the methods of physical fractionation, chemical and biological analysis and Fourier-transform infrared spectroscopy (FTIR). The results showed that soil total organic carbon (TOC) and total nitrogen (TN) content, as well as free particulate organic carbon and nitrogen, soluble organic carbon and nitrogen (DOC, DON), and mineral-associated organic carbon and nitrogen in the undisturbed and extensively managed Moso bamboo plantations were significantly increased compared with that in the control. The distribution ratio of free particulate organic carbon and nitrogen in the undisturbed Moso bamboo plantation significantly increased, with mineral-associated organic carbon being the largest reservoir of soil organic carbon (67.6%). Intensive management resulted in the decrease of soil organic carbon, total nitrogen storage, and the contents of each component, but significantly increased DOC/TOC, the ratio of microbial biomass nitrogen to TN as well as the ratio of microbial biomass carbon to TOC (microbial quotient). Management measures significantly affected the chemical structure of SOC. Compared with the control, the relative intensities of phenolic and alcoholic-OH, aliphatic methyl and methylene, aromatic C=C, and carbonyl C=O absorption were higher in the SOC of undisturbed and extensively managed Moso bamboo plantations, and soil hydrophobicity was significantly increased. Results from correlation analysis showed that soil hydrophobicity and the content of aliphatic and aromatic groups were negatively correlated with microbial quotient and positively correlated with TOC and TN content. In conclusion, the increased inputs of organic matter residues (such as litter and roots) could contribute to the relative accumulation of chemical resistance compounds with reduced human disturbance, which significantly enhanced chemical stability of soil organic carbon. Soil clay minerals played a key role in protecting soil organic carbon through the formation of mineral-organic compounds, which facilitate the stability of soil carbon storage and the long-term preservation of soil carbon.

MicroRNA deregulation is crucial for cancer development. Studies showed that polymorphisms in miRNA genes could affect miRNA expression, which might be associated with cancer development. In the current study, we investigated the association of seven single nucleotide polymorphisms (SNPs) in seven miRNA genes with the initiation and development of cervical cancer in a Chinese Han population. The SNPs of 358 cervical intraepithelial neoplasia (CIN) patients, 547 cervical cancer patients and 567 healthy individuals were genotyped using TaqMan assays. Moreover, we evaluated the association of the seven SNPs with the different stages of cervical cancer. Our results showed that rs4636297 in miR-126 was associated with susceptibility to CIN and cervical cancer (P=0.019 and 0.019, respectively) and that the T allele was associated with a higher risk of CIN (OR=1.334, 95% CI: 1.049-1.698) and cervical cancer (OR=1.296, 95% CI: 1.044-1.609). Similarly, rs11614913 in miR-125a was associated with CIN and cervical cancer (P=0.025 and 0.015, respectively), and the T allele might be the protective factor for CIN (OR=0.807, 95% CI: 0.669-0.974) and cervical cancer (OR=0.814, 95% CI: 0.689-0.961). Our results indicated that rs4636297 in miR-126 and rs11614913 in miR-196a2 play an important role only in the initiation of cervical cancer not in the development of CIN to cervical cancer.

The clinical value of the combined detection of miR-1202 and miR-195 in the early diagnosis of cervical cancer was studied. A retrospective analysis of 70 cervical cancer patients treated in the The Third Affiliated Hospital of Kunming Medical University and Yunnan Cancer Hospital from October 2015 to December 2017 was performed, and the lesion tissues were used as the experimental group. Normal cervical tissues from another 67 healthy females confirmed by physical examination at the same period were selected as the control group. The FIGO staging criteria were used for staging of the cervical cancer patients, reverse transcription-quantitative polymerase chain reaction (RT-qPCR) method was used for the detection of the expression of miR-1202 and miR-195 in different tissues, and the receiver operating curve (ROC) was used for the analysis of the application values of miR-1202 and miR-195 diagnosis alone and their combined diagnosis in early cervical cancer patients. The levels of miR-1202 and miR-195 in the experimental group were lower than those in the control group (P<0.05). The differences were significant in the different stages of cervical cancer tissues (P<0.05). The later the staging of cervical cancer tissues were, the lower the levels of miR-1202 and miR-195 were. The sensitivities and area under the curve (AUC) values of miR-1202 and miR-195 in the combined diagnosis of early cervical cancer were significantly higher than those of miR-1202 and miR-195 alone. The expression levels of miR-1202 and miR-195 in the cervical cancer patients are different in different stages. Guiding clinical treatment and prognosis according to the results of combined detection is beneficial for the development of treatment for cervical cancer patients and for prognostic judgement, worthy of popularization and application.
