RESUMO

An original set of 17 clones of hybridomas-producers of monoclonal antibodies (MCAB) against Fc-fragment of human IgG has been obtained. The possibility of using horseradish peroxidase conjugates of the obtained MCAB as a part of ELISA tests systems of the company "DiaProphMed" for diagnostics of syphilis, rubella, toxoplasmosis and cytomegalovirus infections has been comparatively studied. It has been shown, that the application of peroxidase conjugates of two monoclonal antibodies (156C10 and 153H11) as the part of five diagnostic ELISA test systems provides high specificity and sensitivity.

RESUMO

Hybridomes--producers of monoclonal antibodies (MAB) were obtained able to differentiate the variants Abent+ and Abent- poliovirus vaccine strain in the virus neutralizing reaction. Using the obtained panel the changes of the epitope structure of capsid proteins of poliovirus variants (dissociants) were found which appeared during reproduction in cell culture. It proves the fact that there exist essential antigenic differences of superficial virion's proteins, which appear during the process of dissociation.

A comparative evaluation was carried out of potentialities of the practical use of the virus neutralization reaction (VNR) and indirect immunoenzymatic analysis (IIEA) relying on the monoclonal antibodies (MKAB) for intratype differentiation of polioviruses. It has been proved that in each of the reactions MKAB can interact with different epitopes on the surface of virions. This brings about an unequal degree of activity and specificity of similar antibodies in the two serologic reactions. In VNR monoclonal antibodies "work" in a more specific manner than they do in IIEA. Therefore this test is still more suitable for clinical use in intratype differentiation of polioviruses.

RESUMO

A panel of murine hybridomas producing monoclonal antibodies is obtained by means of hybridization of splenocytes with Sp-2/0 myelomas. They are able to differentiate between vaccine and virulent poliovirus type II strains in the reaction of neutralization. The ability of monoclonal antibodies to differentiate polioviruses of different origins was investigated on reference strains as well as on the field poliovirus isolates compared to those obtained using the monoclonal antibodies from Pasteur Institute, polymerase chain reaction (PCR) test and ELISA tests with cross-adsorbent sera as recommended by World Health Organization. It is demonstrated that 4 of 6 obtained monoclonal antibodies allow precise differentiation of the field poliovirus isolates with the reliability comparable to PCR or ELISA test with cross-adsorbent sera. We also show that the obtained panel of monoclonal antibodies can be used for studying antigenic similarity in poliovirus vaccine type II strains.
